Abstract

The first phytochemical investigation of the flowers of Millettia dura resulted in the isolation of seven isoflavones, a flavonol and a chalcone. Eleven isoflavones and a flavonol isolated from various plant parts from this plant were tested for cytotoxicity against a panel of cell lines, and six of these showed good activity with IC50 values of 6-14 μM. Durmillone was the most active with IC50 values of 6.6 μM against A549 adenocarcinomic human alveolar basal epithelial cancer cell line with low cytotoxicity against the non-cancerous cell lines BEAS-2B (IC50 = 58.4 μM), LO2 hepatocytes (IC50 78.7 μM) and CCD19Lu fibroblasts (IC50 >100 μM). Abstract

Abstract Background The role of IL-7 and pre-TCR signaling during T cell development has been well characterized in murine but not in human system. We and others have reported that human BM hematopoietic progenitor cells (HPCs) display poor proliferation, inefficient double negative (DN) to double positive (DP) transition and no functional maturation in the in vitro OP9-Delta-like 1 (DL1) culture system. Results In this study, we investigated the importance of optimal IL-7 and pre-TCR signaling during adult human T cell development. Using a modified OP9-DL1 culture ectopically expressing IL-7 and Fms-like tyrosine kinase 3 ligand (Flt3L), we demonstrated enhanced T cell precursor expansion. IL-7 removal at various time points during T cell development promoted a slight increase of DP cells; however, these cells did not differentiate further and underwent cell death. As pre-TCR signaling rescues DN cells from programmed cell death, we treated the culture with anti-CD3 antibody. Upon pre-TCR stimulation, the IL-7 deprived T precursors differentiated into CD3+TCRαβ+DP cells and further matured into functional CD4 T cells, albeit displayed a skewed TCR Vβ repertoire. Conclusions Our study establishes for the first time a critical control for differentiation and maturation of adult human T cells from HPCs by concomitant regulation of IL-7 and pre-TCR signaling.

Abstract

Two new compounds Talaromycin A (1) and Talaromycin B (2) were isolated from a liquid culture of Talaromyces aurantiacus. The structures of 1 and 2 were elucidated by IR, MS, 1D and 2D NMR spectra and comparison of the experimental and calculated electronic circular dichroism spectra. Additional known compounds (3–6) were also isolated. These compounds were tested for monoamine oxidase, acetylcholinesterase and PI3K inhibitory activity, but showed only weak activity. Abstract

This paper theoretically examines the spatial linear instability of viscoelastic jets subjected to unrelaxed axial stress tension and moving within an inviscid stationary gas medium. Unlike the constant value assumption of previous studies, the effects of spatial decaying of the unrelaxed stress tension are included here. The Oldroyd-B constitutive equation has been adopted to model fluid viscoelasticity. Results indicated that the effects of unrelaxed stress tension were complicated and mainly dependent on stress relaxation time. When stress relaxation time was short, the maximum growth rates along the jet decreased to the constant value of the completely relaxed case; increasing unrelaxed tension slightly decreased the breakup length. When the stress relaxation time increased to exceed the critical value, the maximum growth rates continued to increase along the jet and larger unrelaxed tensions caused longer breakup lengths. This twofold effect can be explained by the competition between the stabilizing effects of the unrelaxed tension itself and the destabilizing effects of the spatial decay. Moreover, the fluid elasticity suppressed instability when the unrelaxed tension was great. Responses to the spatial decaying unrelaxed tension of the axisymmetric and nonaxisymmetric disturbances for high-speed viscoelastic jets were similar to those of the capillary case. Generally, the complex effects of the interplay between fluid elasticity and the spatially decaying unrelaxed tension may qualitatively explain the breakup behaviors of viscoelastic jets in experiments. Abstract

A ultra-small micro-ring phase shifter based on the silicon-on-insulator(SOI) material is designed.2π phase shift in the free spectral range and optimal linearization is achieved.According to the phase shift characteristics of the micro-ring resonator all-pass phase structure,phase shift of the micro-ring is π at the critical coupling area and 2π in the over coupling area.Two parameters(straight waveguide width and spacing) of the micro-ring are investigated which affect the phase shift.Finesse F also is used as a parameter for spacing,and micro-ring resonator' phase shift combined with the power of change is discussed.The experimental results show that the designed micro-ring phase shifter can achieve the RF phase shift of 0~4 rad,and RF power variation is less than 6 dB with a 40 GHz RF signal.

Abstract

This paper studies the narrow spectral feature appearing in a four-level system coupled by two strong coherent fields and probed by a weak laser field. The linewidth is examined as a function of the Rabi frequencies of coupling fields, and the result is explained by using the dressed-state formalism.
