218. DBS-YOLO: A High-Precision Object Detection Algorithm for Hazardous Waste Images

Abstract

The adverse effects of hazardous waste on the natural environment and human health are evident. In real-life scenarios, the dense distribution, mutual occlusion, and complex image backgrounds of hazardous waste present significant challenges for existing object detection algorithms to achieve a high detection accuracy. In order to enhance the efficiency of hazardous waste sorting, this article proposes an improved detection model, DBS-YOLO, based on the YOLOv8n network. This model strikes a balance between accuracy and lightweight design. First, we replace part of the convolution modules in the C2f module with deformable convolutional networks version 3 (DCNv3) modules, proposing the DC2f module. Incorporating this module into YOLOv8n not only achieves a lightweight network design but also enhances the model’s adaptability to occluded hazardous waste. In addition, we introduce a bi-level routing attention (BRA) module to construct a global receptive field, capturing long-range dependencies and improving the model’s focus on targets. Finally, we employ a soft non-maximum suppression (Soft-NMS) algorithm to reduce instances of false positives (FPs) and false negatives (FNs) caused by mutual occlusion, further enhancing model accuracy. Experimental results demonstrate that compared to the original YOLOv8n network, DBS-YOLO achieves improvements of 2.0% in precision, 1.0% in recall, 2.1% in mean average precision (mAP)@0.5, and 4.9% in mAP@0.5:0.95, reaching a mAP@0.5 of 95.4%. The model also mitigates instances of FPs and FNs. Comparative analyses with SDD, Faster R-CNN, and YOLO series networks confirm the effectiveness of the proposed model.

Abstract

Cardiac conduction regulatory RNA (CCRR) has been documented as an antiarrhythmic lncRNA in our earlier investigation. This study aimed to evaluate the effects of CCRR on SERCA2a and the associated Ca2+homeostasis in myocardial infarction (MI). Overexpression of CCRR via AAV9-mediated delivery not only partially reversed ischemia-induced contractile dysfunction but also alleviated abnormal Ca2+homeostasis and reduced the heightened methylation level of SERCA2a following MI. These effects were also observed in CCRR over-expressing transgenic mice. A conserved sequence domain of CCRR mimicked the protective function observed with the full length. Furthermore, silencing CCRR in healthy mice led to intracellular Ca2+overloading of cardiomyocytes. CCRR increased SERCA2a protein stability by upregulating FTO expression. The direct interaction between CCRR and FTO protein was characterized by RNA-binding protein immunoprecipitation (RIP) analysis and RNA pulldown experiments. Activation of NFATc3 was identified as an upstream mechanism responsible for CCRR downregulation in MI. This study demonstrates that CCRR is a protective lncRNA that acts by maintaining the function of FTO, thereby reducing the m6A RNA methylation level of SERCA2a, ultimately preserving calcium homeostasis for myocardial contractile function in MI. Therefore, CCRR may be considered a promising therapeutic strategy with a beneficial role in cardiac pathology.

Abstract

The objective of this study is to analyse the quantitative relationship between the local Euler head distribution and the internal flow in a centrifugal impeller and provide guidance for the design specifications of inverse design methods. The local Euler head at the inlet is mainly affected by the clockwise vortex on the blade suction side. At the outlet, it is affected by the relative motion of clockwise and counterclockwise vortexes. The effect of the two types of vortexes on the local Euler head makes the pump head almost constant under deep part-load conditions, proving that the impeller has an effective diameter. The mean local Euler head distribution along the impeller passage is closely interrelated with the flow pattern. The position in which the mean local Euler head increases fastest coincides with the radial position of the clockwise vortex centre. The position of its maximum value is consistent with the radial position of the end of the clockwise vortex. The position of its maximum value moves towards the inlet as the flow rate decreases, confirming the impeller has an extreme impeller diameter. The inflection points of the mean local Euler head ratio curve correspond to the appearance of vortex structures.

Abstract

In March 2022, more than 600 million cases of Corona Virus Disease 2019 (COVID-19) and about 6 million deaths have been reported worldwide. Unfortunately, while effective antiviral therapy has not yet been available, chloroquine (CQ)/hydroxychloroquine (HCQ) has been considered an option for the treatment of COVID-19. While many studies have demonstrated the potential of HCQ to decrease viral load and rescue patients' lives, controversial results have also been reported. One concern associated with HCQ in its clinical application to COVID-19 patients is the potential of causing long QT interval (LQT), an electrophysiological substrate for the induction of lethal ventricular tachyarrhythmias. Yet, the mechanisms for this cardiotoxicity of HCQ remained incompletely understood. Adult New Zealand white rabbits were used for investigating the effects of HCQ on cardiac electrophysiology and expression of ion channel genes. HEK-293T cells with sustained overexpression of human-ether-a-go-go-related gene (hERG) K+ channels were used for whole-cell patch-clamp recordings of hERG K+ channel current (IhERG). Quantitative RT-PCR analysis and Western blot analysis were employed to determine the expression of various genes at mRNA and protein levels, respectively. electrocardiogram (ECG) recordings revealed that HCQ prolonged QT and RR intervals and slowed heart rate in rabbits. Whole-cell patch-clamp results showed that HCQ inhibited the tail current of hERG channels and slowed the reactivation process from inactivation state. HCQ suppressed the expression of hERG and hindered the formation of the heat shock protein 90 (Hsp90)/hERG complex. Moreover, the expression levels of connexin 43 (CX43) and Kir2.1, the critical molecular/ionic determinants of cardiac conduction thereby ventricular arrythmias, were decreased by HCQ, while those of Cav1.2, the main Ca2+ handling proteins, remained unchanged and SERCA2a was increased. HCQ could induce LQT but did not induce arrhythmias, and whether it is suitable for the treatment of COVID-19 requires more rigorous investigations and validations in the future. Abstract

Background: Angelman syndrome (AS) is a genetic disorder that affects neurodevelopment. The investigation of changes in the brain white matter network, which would contribute to a better understanding of the pathogenesis of AS brain, was lacking. Purpose: To investigate both local and global alterations of white matter in patients with AS. Study Type: Prospective. Subjects: A total of 29 AS patients (6.6 ± 1.4 years, 15 [52%] females) and 19 age‐matched healthy controls (HC) (7.0 ± 1.5 years, 10 [53%] females). Field Strength/Sequence: A 3‐T, three‐dimensional (3D) T1‐weighted imaging by using gradient‐echo‐based sequence, single shell diffusion tensor imaging by using spin‐echo‐based echo‐planar imaging. Assessment: Network metrics including global efficiency (Eg), local efficiency (Eloc), small world coefficient (Swc), rich‐club coefficient (Φ), and nodal degree (ND) were estimated from diffusion MR (dMR) data. Connections among highly connected (hub) regions and less connected (peripheral) regions were also assessed. Correlation between the topological parameters and age for each group was also calculated to assess the development of the brain. Statistical Tests: Linear regression model, permutation test. P values estimated from the regression model for each brain region were adjusted by false discovery rate (FDR) correction. Results: AS patients showed significantly lower Eg and higher swc compared to HC. Φn significantly increased at higher k‐levels in AS patients. In addition, the connections among hub regions and peripheral regions were significantly interrupted in AS patients.
