202. Maternal postpartum depression literacy subtypes: A latent profile analysis

Aim: To explore the potential categories and characteristic differences of maternal postpartum depression literacy. Design: Cross-sectional survey. Methods: From February 2023 to April 2023, convenience sampling was used to survey 278 women attending postnatal visits to three tertiary level A hospitals. The study included general demographic characteristics, postpartum depression literacy scale, and family caring index scale. Latent profile analysis was performed to identify the categories of maternal postpartum depression literacy, and multiple disordered logistic regression was used to analyze the influencing factors of different categories. Results: Maternal postpartum depression literacy was divided into three categories: low literacy (41.0 %), moderate literacy (32.4 %), and high literacy (26.6 %). The results showed that work status, education level, whether the pregnancy was planned, whether or not they had participated in mental health-related courses, and family functioning status were factors influencing the category of maternal postpartum depression literacy (P

203. CD22 blockade modulates microglia activity to suppress neuroinflammation following intracerebral hemorrhage

Microglia are first responders to acute brain insults and initiate neuroinflammation to drive secondary tissue injury. Yet the key molecular switches in control of the inflammatory activity of microglia remain poorly understood. Intracerebral hemorrhage (ICH) is a devastating stroke subtype whereby a hematoma is formed within the brain parenchyma and associated with high mortality. Using a mouse model of ICH, we found upregulation of CD22 that predominantly occurred in microglia. Antibody blockade of CD22 led to a reduction in neurological deficits, brain lesion and hematoma volume. This was accompanied by reduced inflammatory activity, increased expression of alternative activation markers (CD206 and IL-10) and enhanced phagocytosis activity in microglia after ICH. CD22 blockade also led to an increase of phosphorylated SYK and AKT after ICH. Notably, the benefits of CD22 blockade were ablated in ICH mice subjected to microglial depletion with a colony-stimulating factor 1 receptor inhibitor PLX5622. Additionally, the protective effects of CD22 blockade was diminished in ICH mice receiving a SYK inhibitor R406. Together, our findings highlight CD22 as a key molecular switch to control the detrimental effects of microglia after acute brain injury, and provide a novel strategy to improve the outcome of ICH injury.

The prediction of significant wave height (SWH) is crucial for managing wave energy. While many machine learning studies have focused on accurately predicting SWH values within hours in advance, the primary concern should be given to the level of the wave height for real-world applications. In this paper, a classification framework for the time-series of SWH based on Transformer encoder (TF) and empirical mode decomposition (EMD) is developed, which can provide a lead time of 6 to 48 h with the fixed thresholds of 2 m for high level waves and 1.5 m for low level waves. The performance of this approach is compared to that of three mainstream algorithms with and without EMD features. Results from the datasets collected from buoy measurements in the Atlantic Ocean indicate that the optimal mean accuracy at a lead time of 6 h was 99.1% and the average training time was 75 s, demonstrating the accuracy and efficiency of this proposed model. This study provides valuable tools and references for real-world SWH prediction applications.

205. SAP30 promotes breast tumor progression by bridging the transcriptional corepressor SIN3 complex and MLL1

SAP30 is a core subunit of the transcriptional corepressor SIN3 complex, but little is known about its role in gene regulation and human cancer. Here, we show that SAP30 was a nonmutational oncoprotein upregulated in more than 50% of human breast tumors and correlated with unfavorable outcomes in patients with breast cancer. In various breast cancer mouse models, we found that SAP30 promoted tumor growth and metastasis through its interaction with SIN3A/3B. Surprisingly, the canonical gene silencing role was not essential for SAP30’s tumor-promoting actions. SAP30 enhanced chromatin accessibility and RNA polymerase II occupancy at promoters in breast cancer cells, acting as a coactivator for genes involved in cell motility, angiogenesis, and lymphangiogenesis, thereby driving tumor progression. Notably, SAP30 formed a homodimer with 1 subunit binding to SIN3A and another subunit recruiting MLL1 through specific Phe186/200 residues within its transactivation domain. MLL1 was required for SAP30-mediated transcriptional coactivation and breast tumor progression. Collectively, our findings reveal that SAP30 represents a transcriptional dependency in breast cancer.

In East Asian archaeology, initial domestication and early dispersal of rice have continuously attracted scholarly interest in the recent decade, which has generated abundant new materials and revised opinions. This paper starts with a refreshed understanding of the domestication concept that emphasizes the dominant role of human behavior in the mutualistic relationship. A thorough review of the approaches to and data on reconstructing the rice story during 10,000–7,000 BP demonstrates the causally chained changes in phenotype, genotype, and human behavior in the establishment of domestication. Future studies will benefit from the revised paradigm, which has great potential to extract archaeological information to explain multiple mechanisms in rice domestication.

BackgroundUlcerative colitis (UC) is a chronic relapsing remitting disease of the colon. Appropriate monitoring of the disease status is necessary for patients to adopt optimal therapy and obtain a better prognosis. Finding an ideal non-invasive biomarker, which is suitable for long-term monitoring in clinical settings will bring a significant benefit to the individualized management of patients with UC. The aim of this study is to determine the clinical significance of a novel optimizing serological biomarker by integrating C-reactive protein (CRP) and bilirubin levels in monitoring disease activity.MethodsA total of 182 patients with UC were retrospectively enrolled. Clinical characteristics and laboratory parameters of the subjects were retrieved from the electronic medical record database of our hospital. The CRP-to-bilirubin ratio (CBR) was computed for clinical activity of UC defined by the partial Mayo score and endoscopic activity by the Mayo endoscopic score (MES).ResultsCBR was significantly elevated in patients with UC than that in healthy controls. Patients with clinically or endoscopically active UC showed evidently higher CBR levels compared to those with inactive disease, even in a subset of patients with normal CRP levels. Receiver operating characteristic (ROC) analysis showed that the area under the curve (AUC) of CBR was higher than that of CRP or bilirubin alone for determining clinical remission and endoscopic mucosal improvement. Furthermore, CBR levels were significantly decreased when patients achieved mucosal improvement compared with when they had active endoscopic inflammation.ConclusionCBR could be useful to reflect disease activity in patients with UC.

Abstract The accomplishment of condition monitoring and intelligent maintenance for cantilever structure‐based energy harvesting devices remains a challenge. Here, to tackle the problems, a novel cantilever‐structure freestanding triboelectric nanogenerator (CSF‐TENG) is proposed, which can capture ambient energy or transmit sensory information. First, with and without a crack in cantilevers, the simulations are carried out. According to simulation results, the maximum change ratios of natural frequency and amplitude are 1.1% and 2.2%, causing difficulties in identifying defects by these variations. Thus, based on Gramian angular field and convolutional neural network, a defect detection model is established to achieve the condition monitoring of the CSF‐TENG, and the experimental result manifests that the accuracy of the model is 99.2%. Besides, the relation between the deflection of cantilevers and the output voltages of the CSF‐TENG is first built, and then the defect identification digital twin system is successfully created. Consequently, the system is capable of duplicating the operation of the CSF‐TENG in a real environment, and displaying defect recognition results, so the intelligent maintenance of the CSF‐TENG can be realized.

The pyrrole moiety is an important structural motif in functional materials, natural products, and pharmaceuticals. More and more synthetic strategies toward pyrroles have emerged, where various efficient building blocks are developed and these synthons enable the syntheses of pyrroles with different numbers of components. However, no review specifically summarizes the syntheses of pyrroles according to the type and number of employed building blocks. To aid researchers to design appropriate substrates for pyrrole synthesis, herein we summarized the advances in pyrrole syntheses and classified these reactions into four categories according to the number of employed components, which may shed light on developing more efficient synthetic methods toward substituted pyrroles.

Abstract We study the existence of scalar fields outside neutral reflecting shells. We consider static massive scalar fields non-minimally coupled to the Gauss–Bonnet invariant. We analytically investigated properties of scalar fields through the scalar field equation. In the small scalar field mass regime, we derive a compact resonance formula for the allowed masses of scalar fields in the composed scalar field and shell configurations.

(RC) is a classic herbal medicine for the treatment of rheumatoid arthritis (RA) and has been used since ancient times. The ethanol extract of Rubia cordifolia L. (RCE) showed obvious anti-RA effects in our previous study. However, further potential mechanisms require more exploration. We aimed to investigate the mechanism of RCE for the treatment of RA by integrating metabolomics and network pharmacology in this study.Methods: An adjuvant-induced arthritis (AIA) rat model was established, and we evaluated the therapeutic effects of RCE. Metabolomics of serum and urine was used to identify the differential metabolites. Network pharmacology was applied to determine the key metabolites and potential targets. Finally, the potential targets and compounds of RCE were verified by molecular docking.Results: The results indicated that RCE suppressed foot swelling and alleviated joint damage and also had anti-inflammatory properties by inhibiting the expressions of tumor necrosis factor (TNF)-α, Interleukin (IL)-1β, prostaglandin E2 (PGE2), and P65. Ten and seven differential metabolites were found in the serum and urine, respectively, of rats. Six key targets, ie, phospholipase A2 group IIA (PLA2G2A), phospholipase A2 group X (PLA2G10), cytidine deaminase (CDA), uridine-cytidine kinase 2 (UCK2), charcot-leyden crystal galectin (CLC), and 5′,3′-nucleotidase, mitochondrial (NT5M), were discovered by network pharmacology and metabolite analysis and were found to be related to glycerophospholipid metabolism and pyrimidine metabolism. Molecular docking confirmed that the favorable compounds showed affinities with the key targets, including alizarin, 6-hydroxyrubiadin, ruberythric acid, and munjistin.Conclusion: This study revealed the underlying mechanisms of RCE and provided evidence that will allow researchers to further investigate the functions and components of RCE against RA.Keywords: inflammtion, AIA rats, molecular docking, chronic disease

212. COVID-19 vaccine uptake and hesitation among men and women preparing for pregnancy: a cross-section survey based on the theory of planned behavior

Abstract Background Given the accelerated speed of COVID-19 vaccine research and administration, the main barriers to herd immunity appear to be concerns about safety and efficacy. Men and women preparing for pregnancy may have the same concerns about COVID-19 vaccination, but few studies have focused on COVID-19 vaccine uptake and hesitation among them. Methods A cross-sectional study was conducted among men and women who were preparing for pregnancy in Southwest China. The questionnaire was designed based on the theory of planned behavior (TPB). Multiple logistic regression was used to explore the determinants of the behaviors of COVID-19 vaccination. Results A total of 2878 participants completed the survey. A total of 53.89% of participants received at least one dose of the COVID-19 vaccine. A total of 45.21% of participants would receive the COVID-19 vaccine in the future. A total of 0.90% of participants never thought about receiving the COVID-19 vaccine. Multiple logistic regression model 1 showed that female participants (OR:5.497, 95%CI: 4.292–7.041), participants who never received influenza vaccine (OR:2.664, 95%CI: 1.908–3.718), participants who had never been tested for COVID-19 (OR:2.244, 95%CI:1.504–3.349), participants who had higher score of negative attitude (OR:1.448, 95%CI: 1.219–1.719), participants who had lower scores of injunctive norms (OR:0.440, 95%CI: 0.360–0.537) and descriptive norms (OR:0.105, 95%CI: 0.088–0.126) were more likely to delay COVID-19 vaccination. Model 2 showed that participants who had lower scores for positive attitude (OR: 0.406, 95% CI: 0.230–0.716), injunctive norms (OR: 0.283, 95% CI: 0.130–0.614) and descriptive norms (OR: 0.060, 95% CI: 0.038–0.094) were more likely to refuse COVID-19 vaccination. Conclusions The COVID-19 vaccination rate of men and women preparing for pregnancy was significantly lower than the average vaccination rate of China. Gender, protective health behaviors, vaccination attitudes, and subjective norms had effects on the vaccination behaviors of couples preparing for pregnancy.

213. Reverse osmosis-based enrichment of virus in water samples: an experiment study

ObjectiveTo establish a reverse osmosis technology-based method for enrichment of virus in water samples. MethodsTwo inactivated virus vaccines and two viral nucleic acid plasmids were added into simulated water samples and detected with real-time quantitative reverse transcriptase PCR (qRT-PCR) after reverse osmosis-based enrichment. Reverse osmosis membrane material, membrane pore size, and filtration time were optimized in the enrichments. ResultsThe enrichment efficiency increased with the decrease of pore size of the reverse osmosis membrane. The average virus enrichment efficiency of reverse osmosis membrane with the pore size of 0.10 μm was 22.86 ± 0.84% for 2 simulated water samples. The virus enrichment efficiency of nylon, cellulose acetate and polyethersulfone membranes were similar between each other. The enrichment efficiency goes up with the increase of filtration times, but the difference in the efficiency was not significant between two and three times of filtration. ConclusionThe efficiency can be up to 32.64% for the enrichment of polio virus in water samples using cellulose acetate filter membrane with 0.10 μm aperture and two times of filtration.

In dynamic environments, achieving accurate and robust Visual SLAM (Simultaneous Localization and Mapping) remains a significant challenge, particularly for applications in robotic navigation and autonomous driving. This study introduces YG-SLAM, an innovative approach that integrates YOLOv8 and geometric constraints within the ORB-SLAM2 framework to adapt effectively to dynamic scenarios.YOLOv8 is employed for instance segmentation and dynamic object detection, enriching the semantic information while extracting image feature points. Geometric constraints, including epipolar geometry algorithms and Lucas-Kanade optical flow methods, are utilized to filter out dynamic objects effectively.The tracking thread exclusively relies on static feature points for camera pose estimation, substantially improving the system’s localization accuracy. Experimental results on the TUM dataset demonstrate that YG-SLAM significantly outperforms traditional ORB-SLAM2 in dynamic environments. Specifically, the Root Mean Square Error (RMSE) of the absolute trajectory errors reduced by 96.51% in comparison to ORB-SLAM2, and the RMSE of the relative pose errors decreased by 93.60% when compared to the performance of ORB-SLAM2.These notable reductions in errors demonstrate the promising performance enhancements of YG-SLAM over traditional ORB-SLAM2 in dynamic environments.

Nowadays, online education has been a more general demand in context of COVID-19 epidemic. The intelligent educational evaluation systems assisted by intelligent techniques are in urgent demand. To deal with this issue, this paper introduces the strong information processing ability of deep learning, and proposes the design of an intelligent educational evaluation system using deep learning. Inside the algorithm part, the low-complexity offset minimal sum (OMS) is selected as the front-end processor of deep neural network, so as to reduce following computational complexity in deep neural network. And the deep neural network is adopted as the major calculation backbone. In this paper, our OMS deep neural network parameters are 23 and 57 compared with other parameters, which can save about 59.64% of the network parameters, and the training time is 11270 s and 25000 s respectively, which saves the training time 54.92%. It can be also reflected from experiments that the proposal further improves the performance of unbalanced data classification in this problem scenario.

216. The assessment value of pathological condition of serum adiponectin and amylin in primary osteoporosis and its correlation analysis with bone metabolism indexes

Background: This paper explores the assessment value of pathological condition of serum adiponectin (APN) and amylin in primary osteoporosis (POP) and their correlation with bone metabolism indexes. Methods: From January 2019 to June 2021, 79 cases of POP patients were selected as the research objects. A test of the patients' bone density was conducted, and clinical grading of POP was via T value (normal, mild, moderate, severe). The analysis of the assessment value of pathological condition of serum APN and amylin for POP and their association with bone metabolism indexes in patients was performed. Results: APN and amylin in patients were declined with POP's aggravation. APN of 5.15 mg/mL or less and amylin of 15.38 pmol/L or less were risk factors influencing the aggravation of pathological condition of POP (P< 0 .0 5). The area under the curve (AUC) of combined detection of APN and amylin to assess the severity of POP was elevated vs. alone test of amylin (P< 0.05). 25-hydroxyvitamin D (25-(OH) D) and total type 1 procollagen amino-terminal propeptide (t-PINP) in patients were descended with the aggravation of pathological condition of osteoporosis (P < 0.05). At the same time, no distinct differences were presented in the three groups of type I collagen hydroxyl terminal peptide b degradation product (b-CTX) and N-terminal osteocalcin (N-MID) (P> 0.05). APN, amylin, 25(OH)D, b-CTX, and t-PINP were negatively linked with POP clinical grade (P< 0.05). APN and amylin were associated with 25-(OH) D, b-CTX, t-PINP (P< 0.05), and APN and amylin were not linked with N-MID (P> 0.05). Conclusions: Serum APN and amylin are provided with evaluation values for the severity of POP and are associated with bone metabolism in patients.

Abstract Background Secretory breast carcinoma (SBC) is a rare malignant breast neoplasm with distinct histological features, including solid, microcystic, tubular, and rarely papillary structures, traditionally characterized by a t (12;15) (p13:q25) translocation, which usually leads to ETV6-NTRK3 fusion, suggesting an early event in tumorigenesis. Due to the rarity of this disease, very few genome sequencing studies have been performed on a series of cases, especially progressive cases. Methods Seven lesions from 5 patients diagnosed at the Third Affiliated Hospital of Soochow University from 2007 to 2021 were included. Clinicopathological features and prognosis/survival data were collected. Next-generation DNA sequencing was performed on six of the seven lesions. Results In total, 3/7 (42.9%) lesions demonstrated estrogen receptor (ER) expression, including weak, moderate to strong staining, and no lesion demonstrated progesterone receptor (PR) expression. There were no cases of human epidermal growth factor (HER2) overexpression, and the Ki-67 index was low. S-100 and pan-TRK protein were diffusely positively expressed in all cases. All lesions were characterized by a t(12;15) (p13:q25) translocation, leading to ETV6-NTRK3 fusion confirmed by fluorescence in situ hybridization (FISH). The sequencing results showed that ETV6-NTRK3 fusion was the main driver of early tumorigenesis, while SBC with invasive biological behavior had more complex genomic variation in which TERT promoter mutation was detected. Conclusions Immunohistochemical staining of a biomarker panel, including ER, PR, HER2, Ki-67, S-100 and pan-TRK, can be used as an auxiliary diagnostic tool, and FISH detection can be used as a diagnostic tool. ETV6-NTRK3 gene fusion involving multiple sites may drive tumorigenesis, while mutations in the TERT promoter region may be a factor driving tumor progression.

Abstract Background Cattle industry is critical for China’s livestock industry, whereas E. coli infection and relevant diseases could lead huge economic loss. Traditional mammalian models would be costly, time consuming and complicated to study pathological changes of bovine E. coli. There is an urgent need for a simple but efficient animal model to quantitatively evaluate the pathological changes of bovine-derived E. coli in vivo. Caenorhabditis elegans (C. elegans) has a broad host range of diverse E. coli strains with advantages, including a short life cycle, a simple structure, a transparent body which is easily visualized, a well-studied genetic map, an intrinsic immune system which is conservable with more complicated mammalians. Results Here, we considered that O126 was the dominant serotype, and a total of 19 virulence factors were identified from 41 common E. coli virulence factors. Different E. coli strains with diverse pathogenicity strengths were tested in C. elegans in E. coli with higher pathogenicity (EC3/10), Nsy-1, Sek-1 and Pmk-1 of the p38 MAPK signaling pathway cascade and the expression of the antimicrobial peptides Abf-3 and Clec-60 were significantly up-regulated comparing with other groups. E. coli with lower pathogenicity (EC5/13) only activated the expression of Nsy-1 and Sek-1 genes in the p38 MAPK signaling pathway, Additionally, both groups of E. coli strains caused significant upregulation of the antimicrobial peptide Spp-1. Conclusion Thirteen E. coli strains showed diverse pathogenicity in nematodes and the detection rate of virulence factors did not corresponding to the virulence in nematodes, indicating complex pathogenicity mechanisms. We approved that C. elegans is a fast and convenient detection model for pathogenic bacteria virulence examinations.

However, data remain fraught with heterogeneity and bias from comorbid conditions. Additionally, data on the impact of COPD-specific factors, such as pre-hospital medications and pulmonologist involvement, remain sparse.Objective: We report a single-center analysis of COPD patients hospitalized with COVID-19 compared to those without COPD. Primary outcomes include ICU admission, mechanical ventilation, and in-hospital mortality.Methods: We evaluated all patients ≥ 40 years admitted with PCR-confirmed COVID-19 between February 2020 and February 2021. COPD was defined by documented ICD-10 diagnosis of COPD, confirmed smoking history, and active bronchodilator use. We compared outcomes between COPD patients and the remainder of the COVID-19 cohort. Multivariable analyses were adjusted for age, sex, smoking status, and comorbid conditions.Results: Of 1537 hospitalized COVID-19 patients, 122 (7.9%) carried a diagnosis of COPD. The COPD cohort was older (74 ± 13 vs 66 ± 15 years, P < 0.001) and more often former smokers (P < 0.001). Comorbid conditions including diabetes, cardiovascular disease, and kidney disease were more prevalent in the COPD group (P < 0.001). After adjusting for comorbid conditions, the COPD cohort had higher severity scores and trended towards fewer hospital-free days. Among patients with COPD, pre-hospital use of aspirin was associated with decreased ICU admissions (aHR 0.56, P = 0.049) and mechanical ventilation (aHR 0.25, P = 0.008), while LAMAs (long-acting muscarinic antagonists) were associated with decreased in-hospital mortality (aHR 0.34, P = 0.047). Involvement of pulmonology in pre-hospital management of COPD was not found to significantly affect outcomes.Conclusion: When corrected for comorbid illnesses, COPD was associated with more severe disease but not with increased ICU admission, mechanical ventilation, or in-hospital mortality rates. Objective To investigate the clinical efficacy and safety of high intensity focused ultrasound (HIFU) combined with ultrasound-guided suction curettage in patients with type I/II/III cesarean scar pregnancy (CSP).Methods A total of 153 patients with CSP were enrolled and classified according to the type of CSP. All of them were treated by HIFU combined with ultrasound-guided suction curettage. When active uterine bleeding was observed after curettage, a Foley balloon was used for hemostasis by compression. Baseline characteristics, technical parameters of HIFU, intraoperative blood loss in suction curettage, the time for serum β-HCG to return to normal levels, reproductive outcomes, and adverse effects were recorded and analyzed.Results 152 patients completed one session of HIFU combined with suction curettage except one patient transferred to surgery. Total energy used for ablation and the time for serum β-HCG return to normal level in type II and III were significantly higher than type I (p < .05). The treatment time and sonication time of HIFU in type III were significantly longer than type I (p < .05). Vaginal bleeding after curettage and the rate of using Foley catheter balloon in type III was larger than type I and II.Conclusions HIFU combined with ultrasound-guided suction curettage is a safe and effective treatment option for patients with type I/II/III CSP and desire for fertility. Patients with type III CSP were more dependent on Foley catheter balloon compression therapy than the other two types after HIFU combined with curettage.

Background Abdominal wall endometriosis(AWE)is an unusual extra-pelvic endometriosis. Currently, multiple treatment modalities are available, but no clear guidelines exist for the management of large AWE.Materials and methods We present a 36-year-old female patient with a large AWE lesion who underwent cesarean section due to abnormal fetal position 8 years ago. The mass lesion of AWE located in rectus muscle fascia and rectus muscle with a size of 61 × 25 × 49mm.Results HIFU treatment was completed in one session. One day post-HIFU MRI showed the mass was completely ablated. After HIFU treatment, the cyclical abdominal pain disappeared. The mass lesion shrank during follow-up period and disappeared in 1 year after HIFU. No complication was observed after HIFU.Conclusion Surgical resection of AWE remains the standard of care. In patients with large AWE lesion located in rectus muscle fascia and rectus muscle where the muscle and fascia must be excised, HIFU treatment should be considered to avoid mesh implantation.

Background Acute upper respiratory infections are the most common reason for primary physician visits in the community. This study investigated whether the type of antibiotic used to treat streptococcal tonsillitis can reduce the burden by affecting the number of additional visits.Objectives To assess the effect of different antibiotic treatments for tonsillitis on the number of additional primary physician visits and the development of infectious or inflammatory sequels.Methods This retrospective study included first cases of culture-confirmed streptococcal tonsillitis (n = 242,366, 55.3% females, 57.6% aged 3–15 years) treated in primary clinics throughout Israel between the years 2010 and 2019. The cotton bollworm, Helicoverpa armigera, is set to become the most economically devastating crop pest in the world, threatening food security and biosafety as its range expands across the globe. Key to understanding the eco-evolutionary dynamics of H. armigera, and thus its management, is an understanding of population connectivity and the adaptations that allow the pest to establish in unique environments. We assembled a chromosome-scale reference genome and re-sequenced 503 individuals spanning the species range to delineate global patterns of connectivity, uncovering a previously cryptic population structure. Using a genome-wide association study (GWAS) and cell line expression of major effect loci, we show that adaptive changes in a temperature- and light-sensitive developmental pathway enable facultative diapause and that adaptation of trehalose synthesis and transport underlies cold tolerance in extreme environments. Incorporating extensive pesticide resistance monitoring, we also characterize a suite of novel pesticide and Bt resistance alleles under selection in East China. These findings offer avenues for more effective management strategies and provide insight into how insects adapt to variable climatic conditions and newly colonized environments.

Abstract The existence of null circular geodesics in the background of non-extremal spherically symmetric asymptotically flat black holes has been proved in previous works. An interesting question that remains, however, is whether extremal black holes possess null circular geodesics outside horizons. In the present paper, we focus on the extremal spherically symmetric asymptotically flat hairy black holes. We show the existence of the fastest circular trajectory around an extremal black hole. As the fastest trajectory corresponds to the position of null circular geodesics, we prove that null circular geodesics exist outside extremal spherically symmetric asymptotically flat hairy black holes.

Objective: The research is to explore the enzyme inhibition of Perilla crude extract, and to preliminarily elucidate the pharmacological substance basis of them on hyperglycemia, hyperuric acid and hyperlipidemia. Methods: The content of flavonoids and phenolic acid in UV extract was determined and showed the active components associated with enzyme inhibition. In vitro experiments, the inhibition of Perilla extracts on different enzyme activity was determined and the inhibitory activity was evaluated by IC50. Molecular docking was used to predict the targets of Perilla crude extract on pancreatic lipase and amylase inhibition. Results: Flavone content in the crude UV extract was 0.309 8 mg/mg, and the total phenolic acid content was 58.59 mg/g. The IC50 values of Perilla crude extractd for α-amylase, xanthine oxidase, pancreatic lipase and acetylcholinesterase were 0.273, 0.244, 0.347 and 0.018 mg/mL, respectively.Apigenin-5-O-β-D-glucosine, xyrhinin-7-diglucosotide, apigenin-7-glucosine, cannol-3-O-ruassin, rosemary acid, rutin and others were the effective active components of perilla crude extracts. Conclusion: The Perilla crude extracts all have certain inhibitory activities on four enzymes, and presents a dose-effect relationship.

229. Changes in soil faunal density and microbial community under altered litter input in forests and grasslands

Root and foliar litter inputs are the primary sources of carbon and nutrients for soil fauna and microorganisms, yet we still lack a quantitative assessment to evaluate the effects of root and foliar litter on various groups of soil organisms across terrestrial ecosystems. Here, we compiled 978 paired observations from 68 experimental sites to assess the directions and magnitudes of adding and removing foliar and root litter on the soil faunal density and microbial biomass that was evaluated by phospholipid fatty acids (PLFAs) across forests and grasslands worldwide. We found that litter addition had only a marginal effect on soil faunal density but significantly increased the soil total microbial-, fungal- and bacterial-PLFAs by 13%, 14%, and 10%, respectively, across ecosystems, suggesting that the soil microbial community is more sensitive to carbon source addition than soil fauna, particularly in soils with low carbon to nitrogen ratios. In contrast, removing litter significantly decreased the soil faunal density by 17% but had few effects on soil microorganisms. Compared with foliar litter, root litter input had a more positive effect on the development of soil fungal taxa. The effect of both litter addition and removal on soil faunal density and microbial biomass did not differ between humid and arid regions, but a greater influence was observed in grasslands than in forests for soil microbial community. Our results highlight that the increasing litter production under a global greening scenario would stimulate microbial activity in grasslands more than in forests, and this stimulation would be greater for soil microbes than soil fauna.

The purpose of this study was to explore the possible correlation of VFA with DPN in such a population.Methods: A total of 2498 hospitalized patients with type 2 diabetes mellitus (T2DM) undergone VFA measurement, and were divided into DPN group (n=900) and non-DPN group (n=1594). The association of VFA with the presence of DPN was evaluated by correlation and multiple logistic regression analyses, generalized additive model with a smooth curve fitting, and receiver operating characteristic (ROC) curve analysis.Results: The VFA was significantly lower in the DPN group than in the non-DPN group (P < 0.001). VFA was significantly and positively associated with sural nerve conduction velocity (SNCV) and amplitude potential (SNAP) and negatively associated with the presence of DPN (all P< 0.001); there was no significant difference in the curve fitting (P = 0.344). Multivariate logistic regression analysis showed that the risk of presence of DPN decreased progressively across the VFA quartiles (P for trend < 0.001) and was significantly lower in patients in the highest VFA quartile than in those in the lowest quartile (OR: 0.382, 95% CI 0.151– 0.968, P< 0.001) after multivariate adjustment. 231. Identifying the key genes regulating mesenchymal stem cells chondrogenic differentiation: an in vitro study

Abstract Background Mesenchymal stem cells (MSCs) possess the potential to differentiate into chondrocytes, which makes them an ideal source for healing cartilage defects. Here, we seek to identify the essential genes participating in MSCs chondrogenesis. Methods Human MSCs were induced for chondrogenesis for 7, 14, and 21 days using a high-density micromass culture system, and RNA was extracted for RNA-seq. Results A total of 6247 differentially expressed genes (DEGs) were identified on day 7, and 85 DEGs were identified on day 14. However, no significant DEGs was identified on day 21. The top 30 DEGs at day 7, including COL9A3, COL10A1, and CILP2, are closely related to extracellular matrix organization. While the top 30 DEGs at day 14 revealed that inflammation-related genes were enriched, including CXCL8, TLR2, and CCL20. We also conducted protein–protein interaction (PPI) networks analysis using the search tool for the retrieval of interacting genes (STRING) database and identified key hub genes, including CXCL8, TLR2, CCL20, and MMP3. The transcriptional factors were also analyzed, identifying the top 5 TFs: LEF1, FOXO1, RORA, BHLHE41, and SOX5. We demonstrated one particular TF, RORA, in promoting early MSCs chondrogenesis. Conclusions Taken together, our results suggested that these DEGs may have a complex effect on MSCs chondrogenesis both synergistically and solitarily.

Abstract Background Regulation of gene expression plays an essential role in controlling the phenotypes of plants. Brassica napus (B. napus) is an important source for the vegetable oil in the world, and the seed oil content is an important trait of B. napus. Results We perform a comprehensive analysis of the transcriptional variability in the seeds of B. napus at two developmental stages, 20 and 40 days after flowering (DAF). We detect 53,759 and 53,550 independent expression quantitative trait loci (eQTLs) for 79,605 and 76,713 expressed genes at 20 and 40 DAF, respectively. Among them, the local eQTLs are mapped to the adjacent genes more frequently. The adjacent gene pairs are regulated by local eQTLs with the same open chromatin state and show a stronger mode of expression piggybacking. Inter-subgenomic analysis indicates that there is a feedback regulation for the homoeologous gene pairs to maintain partial expression dosage. We also identify 141 eQTL hotspots and find that hotspot87-88 co-localizes with a QTL for the seed oil content. To further resolve the regulatory network of this eQTL hotspot, we construct the XGBoost model using 856 RNA-seq datasets and the Basenji model using 59 ATAC-seq datasets. Using these two models, we predict the mechanisms affecting the seed oil content regulated by hotspot87-88 and experimentally validate that the transcription factors, NAC13 and SCL31, positively regulate the seed oil content. Conclusions We comprehensively characterize the gene regulatory features in the seeds of B. napus and reveal the gene networks regulating the seed oil content of B. napus.

The uncertain power flow analysis is essential to assessing the operating states of distribution networks under various uncertainties. The power flow calculations considering the single uncertainty have been well developed. However, different types of uncertainties may coexist in power systems. In order to comprehensively assess the impacts of mixed uncertainties, this paper proposes a surrogate-assisted point estimate method to solve the hybrid probabilistic and interval power flow. By taking advantage of the point estimate method and high-fidelity surrogate model, the proposed method can accurately obtain the relevant results of power flow calculation with high efficiency. The numerical studies in IEEE 33-bus and 141-bus systems verify the accuracy and efficiency of the proposed method by comparing it with other well-known methods.

Energy harvesters based on a piecewise linear system make it possible to extract more energy from environment vibrations. The amplitude truncation effect is a phenomenon of an energy harvester with a piecewise linear system. It shows a wider energy harvesting bandwidth and a lower voltage response than the purely linear counterpart. This work investigates the influence of spring length on the amplitude truncation effect and power density of electromagnetic energy harvesters. We first establish an analytical model of the piecewise linear system. Then, we apply the averaging method to explore the voltage responses in the frequency domain and compare the influence of various parameters. Later, we conduct a finite element simulation to analyze the voltage response at a constant frequency. The results obtained by the finite element simulation and averaging method match well with the experimental measurements. The experimental results demonstrate that the amplitude truncation effect is more visible at the shorter spring. Spring length plays a key role in the power density of an electromagnetic energy harvester. The case of 40 mm yields a maximum average power of 45.13 mW. The case of 30 mm displays a maximum power density of 2.35 mW/cm 3.

235. Missing wind speed data reconstruction with improved context encoder network

Missing wind speed data are mainly caused by harsh weather, wind turbine failures, and data transmission errors, which have adverse effects on the performance of wind power forecasting, power curve modeling, and energy assessment. Inspired by context encoders (CE), this paper proposes an improved context encoder network (ICE) for missing wind speed data reconstruction. An auto-encoder architecture with multiple one-dimensional convolutional layers is established for data generation. During network training, a joint loss function that includes reconstruction loss and adversarial loss is presented to obtain the stable and near-real reconstructed wind speed data. We add an Inception layer to the generator network to automatically select the appropriate convolutional filters and then recalibrate the channel relationship between feature maps via the squeeze-and-excitation network. At last, this paper uses wind speed data collected from an on-shore wind farm to verify the effectiveness of the proposed network. The results show that the mean absolute error (MAE) and root mean square error (RMSE) of the ICE network in different data missing rates are 0.019–0.021 and 0.021–0.025, respectively. It has the lowest reconstruction errors compared with six typical data reconstruction methods.

Introduction. The performance and durability of asphalt pavements are majorly conditioned by the quality of the bituminous binders. One of the main disadvantages of asphalt pavements is the permanent change in their properties in time due to the aging of the binder. Various laboratory methods of aging are used to predict the intensity of changes in the properties of bituminous binders that occur during the asphalt mixing at plants and the lifetime of asphalt concrete in the pavement. Problem Statement. In Ukraine, with the implementation of European principles of standardization and the transition to harmonized European standards, there is a gradual replacement of the aging method ГОСТ 18180, which has long been used in the domestic road industry, with the world-accepted RTFOT aging method. Due to the different conditions set in these methods of aging, the question of the impact of differences in aging regimes on the properties of bituminous binders, the possibility of comparing the results obtained by these two methods and the interchangeability of methods is topical. Purpose. The aim of the work is to obtain the influence of the methods of aging adopted in the road industry of Ukraine on the change of properties of road viscous bitumens and bituminous binders, modified with various additives. Materials and methods. Viscous bitumens and bituminous binders modified with various additives (adhesive additives, polymers, structuring additives) used in the domestic road industry are used as objects of research. For methods of aging the domestic method ГОСТ 18180 (for a long time this method was the only one in Ukraine used for estimation of change of properties of bituminous binders under the influence of technological temperatures and which, under test conditions, is close to the standardized method TFOT), as well as the world’s widespread RTFOT method of aging are used. Results. Based on the obtained experimental data, it is found that aging by the ГОСТ 18180 method has a slightly greater effect on the change of properties than after aging by the RTFOT method. The least sensitive indicator of aging conditions is the breaking point temperature — the difference between the values of this indicator after aging by different methods is within the convergence of the method. The conditions of the aging method have the greatest effect on the quality of bitumens modified with adhesives. The relatively short heating time of binders in accordance with the conditions of the RTFOT aging method leads to less change in the adhesion of binders to the glass surface, which may contribute to misleading in thermal stability of adhesives. Conclusions. When revising the domestic standards for technical conditions for bituminous binders, which provide for the replacement of the aging method according to GOST 18180 by the RTFOT method and setting of limits for changing the standard quality of binders after aging, should take into account the slightly lower aging intensity of RTFOT binders. This is especially true for the adhesion requirements of bitumens modified with adhesives.

Abstract Chronic morphine administration alters gene expression in different brain regions, an effect which may contribute to plastic changes associated with addictive behavior. This change in gene expression is most possibly mediated by addictive drug-induced epigenetic remodeling of gene expression programs. Our previous studies showed that chronic morphine-induced decrease of miR-105 in the medial prefrontal cortex (mPFC) contributed to context-induced retrieval of morphine withdrawal memory. However, how chronic morphine treatment decreases miR-105 in the mPFC still remains unknown. The present study shows that chronic morphine induces addiction-related change in miR-105 in the mPFC via two kinds of transcription factors: the first transcription factor is CREB activated by mu receptors-ERK-p90RSK signaling pathway and the second transcription factor is glucocorticoid receptor (GR), which as a negative transcription factor, mediates chronic morphine-induced decrease in miR-105 in the mPFC of rats.

Our previous study revealed that the combination of Rapamycin liposomes (Rapa/Lps) and 5-Fluorouracil (5-FU) has anti-colorectal cancer effects. However, the therapeutic efficacy of Rapa/Lps and 5-FU in other colorectal cancer mice models is yet to be thoroughly explored. The purpose of this study was to investigate the anti-tumor effect of Rapa/Lps combined with 5-FU in vivo and in vitro.Methods: In this study, we evaluated the effect of Rapa/Lps and 5-FU on APC (Min/+) mice and AOM/DSS-induced colorectal cancer mice. The small intestine, colorectum, serum, and plasma of mice in each group were collected following sacrifice to record the number of tumors. HE staining was utilized for observing pathological damage to intestine tissues. Tube formation assay, Transwell assay, wound healing assay, Western Blot were used to explore the anti-angiogenesis effect of drugs in HUVECs.Results: As expected, Rapa/Lps and 5-FU significantly suppressed tumor formation, decreased the number of tumors, and tumor load both in two mouse models, and had no influence on mouse weight. Mechanically, the anti-tumor effect of the drug also was associated in inhibiting angiogenesis and proliferation. Furthermore, we found that Rapa/Lps obviously inhibited HUVECs tube formation and migration.Conclusion: Altogether, we revealed the Rapa/Lps synergism with 5-FU decreased colon and small intestinal tumorigenesis in AOM/DSS-treated and APC (Min/+) mice, respectively, and correlated with anti-angiogenesis.Keywords: rapamycin liposomes, 5-fluorouracil, angiogenesis, APC (Min/+) mice, AOM/DSS, colorectal cancer

239. Genomic characteristics of two breast malignant phyllodes tumors during pregnancy and lactation identified through whole-exome sequencing

Abstract Background The genomic landscape of breast malignant phyllodes tumors (PTs) is not well defined, especially pregnancy-related malignant PTs. To clarify this topic, whole-exome next-generation sequencing (NGS) was performed on tumor samples and paired normal breast tissues from two pregnancy-related malignant PTs, followed by a functional analysis of the genetic alterations. Methods DNA from malignant PT samples and matched normal breast tissues of both patients were subjected to molecular profiling. NGS of the whole-exome was performed in a commercial molecular pathology laboratory. Predictive tools were used to estimate genetic variation in somatic and germline genes. Results In total, 29 somatic genomic alterations and 18 germline alterations were found in both patients. In Patient 1, 12 aberrations were identified in the tumor tissue, and 9 alterations were identified in matched normal breast tissue. One pathogenic variant in tumor suppressor genes (TP53) was detected in patient 1. In Patient 2, 18 and 10 variants were found in the tumor and matched normal breast tissue, respectively. In Patient 2, pathogenic alterations were identified in two tumor suppressor genes (PTEN and TP53). PTEN and TP53 may be potential drug targets. The functional predictive tools showed that genes of unknown significance for PTs, including FCHO1 in Patient 1, and LRP12 and PKM in Patient 2, were pathogenic. Several genes, including FCHO1, LRP12 and PKM, were shown for the first time to be altered in malignant PTs. A potentially pathogenic germline variant in PRF1, was detected in Patient 1. Conclusion Our study first demonstrated somatic and germline gene alterations in two malignant PTs during pregnancy and lactation. These two PTs shared major genetic events, including TP53 mutation, which commonly occurs in malignant PTs; additionally, we identified two potential genes for targeted therapy, TP53 and PTEN. One germline mutation in PRF1 was also detected. These results provide clues regarding tumor pathogenesis and precision therapy development.

Abstract Background Leukocyte phytohemagglutinin (PHA-L), derived from the L4 tetramer of PHA, has been frequently employed as a mitogen to induce T lymphocyte proliferation in vitro. The biological application of PHA-L in cancer diagnosis and treatment has gained traction in recent years. However, it has been noted that PHA-L obtained using traditional procedures has a massive amount of impurities or toxic components, which interfere with the activity of PHA-L. Preparation of a monoclonal antibody against active PHA-L is a significant tool for studying PHA-L's function and therapeutic potential. Results We successfully prepared monoclonal antibodies against the active components of PHA-L based on the whole PHA-L protein as an antigen, and found that monoclonal antibody 3C1C6G11 can be employed in western blot, immunofluorescence, and immunohistochemistry detection. Importantly, preliminary result shows that the mAb 3C1C6G11 may prevent PHA-L-induced cell aggregation and AICD (activation-induced cell death). Conclusions The monoclonal antibody mAb 3C1C6G11 prepared in this study can be used as an effective tool for detecting PHA-L active components, investigating PHA-L's function and antineoplastic application.

Penelitian ini bertujuan untuk mengetahui apakah model pembelajaran artikulasi berpengaruh penggunaanya terhadap hasil belajar siswa pada mata pelajaran IPA terpadu. Metode penelitian ini menggunakan metode penelitian kuantitatif dan jenis penelitian adalah quasi eksperimen. Populasi dalam penelitian adalah seluruh siswa kelas IX SMP Negeri 1 Toma yang berjumlah 116 orang. Sampel penelitian adalah kelas IX-B sebagai kelas eksperimen dengan jumlah 29 orang dan kelas IX-C sebagai kelas kontrol berjumlah 27 orang. Instrumen yang digunakan adalah tes hasil belajar siswa yaitu tes awal dan tes akhir. Berdasarkan pengolahan data yang telah dilaksanakan, maka ada beberapa yang menjadi kesimpulan yaitu (1) berdasarkan perhitungan nilai rata-rata tes awal pada kelas eksperimen diperoleh sebesar 42.93 sedangkan kelas kontrol sebesar 54.26. (2) berdasarkan perhitungan nilai rata-rata tes akhir pada kelas eksperimen diperoleh sebesar 66.04 sedangkan pada kelas kontrol sebesar 45.66. (3) berdasarkan perhitungan uji hipotesis thitung = 8,228 dan ttabel = 1.671, sehingga thitung > ttabel yaitu 8,228 > 1.671. Maka Ha diterima dan Ho ditolak. Dapat disimpulkan bahwa ada pengaruh model pembelajaran artikulasi terhadap hasil belajar siswa pada mata pelajaran IPA Terpadu di SMP Negeri 1 Toma.

242. Dynamic recrystallization behavior of low-carbon steel during hot rolling process: modeling and simulation

DRX (dynamic recrystallization) behavior, which can improve the product's microstructure and enhance its performance, is vital in the high-temperature plastic deformation of metal materials. In this paper, single-pass hot compression tests were carried out on a Gleeble-3800 thermo-mechanical simulator to investigate the DRX behavior of a low carbon steel at a deformation temperature of 900°C–1100 °C and a strain rate of 0.01s−1-0.1s−1. The effect of temperature and strain rate on DRX was investigated based on the direct connection between DRX and dynamic softening behavior. The Zener–Hollomon model revealed the relationship between temperature, strain rate, and stress was established. A DRX kinetic model was established for the hot-rolled condition based on the relationship between the characteristic parameters of DRX and temperature and strain rate. The results showed that the predicted and tested values were in good agreement. The established DRX kinetic model was embedded into DEFORM-3D software to simulate the DRX behavior of the hot rolling process of a round bar. The results showed that the fraction of DRX of the whole section of the bar after hot rolling showed noticeable non-uniformity. The DRX ability of the surface area in direct contact with the roller is significantly lower than in other areas. This phenomenon is related to the high-temperature deformation behavior during the rolling process. Reducing the roller speed has almost no effect on this phenomenon; increasing the rolling temperature can significantly improve this phenomenon.

Abstract Liquid metals (LMs) have compelling applications in stretchable electronics, wearable devices, and soft robotics ascribing to the unique combination of room temperature fluidity and metallic electrical/thermal conductivity. Adding metallic elements in gallium‐based LMs can produce heterophasic (i.e., solid and liquid) LMs with altered properties including morphology, surface energy, rheology, electrical/thermal conductivity, and chemical reactivity. Importantly, heterophasic LMs can respond to external stimuli such as magnetic fields, temperature, and force. Thus, heterophasic LMs can broaden the potential applications of LMs. This report reviews the recent progress about heterophasic LMs through metallic elements in the periodic table and discusses their functionalities. The heterophasic LMs are systematically organized into four categories based on their features and applications including electrical/thermal conductivity, magnetic property, catalysis/energy management, and biomedical applications. This comprehensive review is aimed to help summarize the field and identify new opportunities for future studies.

244. Rapid test to detect insecticide resistance in field populations of Spodoptera frugiperda (Lepidoptera: Noctuidae)

Introduction: The development of insecticide resistance in Spodoptera frugiperda populations is a serious threat to the crop industry. Given the spread of invasive resistant populations, prospective monitoring should be accelerated, and the development of diagnostic tools for rapid and accurate assessments of insecticide resistance is essential.Methods: First, the discriminating dose and diagnostic time of the kit were determined by the glass vial method based on a susceptible strain. Then, pests that were collected from field populations were used to determine their susceptibility to seven insecticides by using the diagnostic kit. Finally, the accuracy of the kit was verified based on correlation analyses and the likelihood of insecticide control failure was assessed.Results: Here, we describe a diagnostic kit that enables the rapid detection of resistance to chlorpyrifos, bifenthrin, deltamethrin, lambda-cyhalothrin, phoxim, chlorantraniliprole and chlorfenapyr within 1-2 h in S. frugiperda at diagnostic doses of 0.98, 0.84, 0.38, 1.64, 0.0082, 1.75 and 0.65 μg/cm2, respectively. The linear equation between mortalities under diagnostic doses and actual resistance ratios measured by the diet-overlay bioassay was determined. The high correlation indicates that the insecticide resistance levels diagnosed by the kit were consistent with the results of the diet-overlay bioassay. Moreover, we found a significant negative correlation between diagnostic mortality and the likelihood of control failure for bifenthrin (r = −0.899, p = 0.001), deltamethrin (r = −0.737, p = 0.024) and lambda-cyhalothrin (r = −0.871, p = 0.002).Discussion: The insecticide resistance diagnostic kit for S. frugiperda is a user-friendly tool (portable, short detection time). Its excellent performance qualifies the kit as a reliable screening tool for identifying effective insecticides in sustainable resistance management.

The development of urbanization and the transformation of green lands into impermeable land increase temperature and create urban heat islands (UHIs). Our observations with remote sensing instruments of Landsat platforms show considerable changes in land use types in Beijing city with the shrinking of green lands, expansion of built environments, and a slight increase in the temperature during the recent four decades. Using remote sensing instruments of Landsat platforms and registered data from two meteorological stations in Beijing, this study finds the relationship between land surface temperature (LST) and the increasing conversion of cultivated lands into built-up areas. This article presents innovative research that shows the mutual correlation well and recommends revisions in the land use policies for better weather. The geographically weighted regression model (GWR) with a Gaussian weighting kernel function analyzes the impact of various urban land use types on the LST and the increase UHIs. In Beijing city, green lands show fewer standard deviations (SD) in the average temperatures equal to 0.109, while the industrial spaces exhibit a high SD equal to 0.212. The outcomes of this paper contribute to finding optimal land use policies everywhere in the world with the increasing urbanization through simulating its model for a more comfortable life.

Abstract High‐efficiency and low‐loss processing is the mainstay to reduce the cost and deepen the application of 4H silicon carbide (4H‐SiC) wafers in high‐power and high‐frequency electronics. In this study, the high‐yield slicing of 4H‐SiC wafers is realized by combining femtosecond laser irradiation and bandgap‐selective photo‐electrochemical (PEC) exfoliation. By combining light‐absorption measurements, micro‐Raman, and micro‐photoluminescence characterizations, it is found that the damage layer formed inside 4H‐SiC after femtosecond‐laser irradiation consists of amorphous silicon and amorphous carbon. This indicates that the femtosecond‐laser irradiation leads to phase separation in 4H‐SiC. The bandgap of the damage layer is 0.4 eV. Taking advantage of the different bandgap energies of the damage layer and the perfect 4H‐SiC region, the damage layer is removed from the perfect region of 4H‐SiC by using bandgap‐selective PEC etching. During the PEC etching, light‐generated holes can selectively oxidize and corrode the damaged layer with the assistance of the HF solution, and leave the upper and lower perfect 4H‐SiC layers being intact. The current work contributes to the development of the high‐yield and high‐throughput femtosecond laser slicing of 4H‐SiC wafers.

Acute myeloid leukemia (AML) is a complex mixed entity composed of malignant tumor cells, immune cells and stromal cells, with intra-tumor and inter-tumor heterogeneity. Single-cell RNA sequencing enables a comprehensive study of the highly complex tumor microenvironment, which is conducive to exploring the evolutionary trajectory of tumor cells. Herein, we carried out comprehensive analyses of aggrephagy-related cell clusters based on single-cell sequencing for patients with acute myeloid leukemia. A total of 11 specific cell types (T, NK, CMP, Myeloid, GMP, MEP, Promono, Plasma, HSC, B, and Erythroid cells) using t-SNE dimension reduction analysis. Several aggrephagy-related genes were highly expressed in the 11 specific cell types. Using Monocle analysis and NMF clustering analysis, six aggrephagy-related CD8+ T clusters, six aggrephagy-related NK clusters, and six aggrephagy-related Mac clusters were identified. We also evaluated the ligand-receptor links and Cell–cell communication using CellChat package and CellChatDB database. Furthermore, the transcription factors (TFs) of aggrephagy-mediated cell clusters for AML were assessed through pySCENIC package. Prognostic analysis of the aggrephagy-related cell clusters based on R package revealed the differences in prognosis of aggrephagy-mediated cell clusters. Immunotherapy of the aggrephagy-related cell clusters was investigated using TIDE algorithm and public immunotherapy cohorts. Our study revealed the significance of aggrephagy-related patterns in tumor microenvironment, prognosis, and immunotherapy for AML.

248. Treatment of immunoglobulin-resistant kawasaki disease: a Bayesian network meta-analysis of different regimens

BackgroundThis study aimed to gather evidence from clinical trials on the efficacy and safety of the available treatments for intravenous immunoglobulin (IVIG)-resistant Kawasaki disease (KD) in children.MethodsThis work adopted the Newcastle–Ottawa scale to analyse the quality of the enrolled articles. A network meta-analysis was performed using clinical trials that compared drugs used to treat IVIG-resistant KD. Aggregate Data Drug Information System software v.1.16.5 was employed to analyse whether infliximab, second IVIG infusions, and intravenous pulse methylprednisolone (IVMP) were safe and effective.ResultsTen studies, involving 704 patients with IVIG-resistant KD, were identified and analysed. Overall, infliximab exhibited remarkable antipyretic activity compared with the second IVIG infusions (2.46, 1.00–6.94). According to the drug rank, infliximab was the best option against IVIG-resistant KD. Regarding adverse effects, the infliximab group was more prone to hepatomegaly. A second IVIG infusion was more likely to result in haemolytic anaemia. IVMP treatment was more susceptible to bradycardia, hyperglycaemia, hypertension, and hypothermia. In addition, infliximab, IVMP, and the second IVIG infusions showed no significant differences in the risk of developing a coronary artery aneurysm (CAA).ConclusionInfliximab was the best option against IVIG-resistant KD, and IVMP, infliximab, and second IVIG infusions have not significant differences of prevent CAA in patients with IVIG-resistant KD.Systematic Review RegistrationIdentifier: https://osf.io/3894y.

249. Development of o-aminobenzamide salt derivatives for improving water solubility and anti-undifferentiated gastric cancer

Background: Gastric cancer is one of the cancers with wide incidence, difficult treatment and high mortality in the world, especially in Asia and Africa. In our previous work, a novel o-aminobenzamide analogue F8 was identified as an early preclinical candidate for treatment of undifferentiated gastric cancer (IC50 of 0.26 μM for HGC-27). However, the poor water solubility of compound F8 prevents its further progress in preclinical studies.Aim: To improve the water solubility and drug-likeness of F8 via salt formation.Method: Different acids and F8 were reacted to obtain different salt forms. Physicochemical property screening, pharmacokinetic property research, and antitumor biological activity evaluation in vitro and in vivo were used to obtain the optimal salt form with the best druggability.Results: our continuous efforts have finally confirmed F8·2HCl as the optimal salt form with maintained in vitro antitumor activity, improved water solubility and pharmacokinetic properties. Importantly, the F8·2HCl displayed superior in vivo antitumor efficacy (TGI of 70.1% in 75 mg/kg) in HGC-27 xenograft model. The further immunohistochemical analysis revealed that F8·2HCl exerts an antitumor effect through the regulation of cell cycle-related protein (CDK2 and p21), apoptosis-related protein Cleaved Caspase-3, proliferation marker Ki67, and cell adhesion molecule E-cadherin. In addition, F8·2HCl showed acceptable safety in the in vivo acute toxicity assay.Conclusion: Salting is an effective means to improve the drug-like properties of compound F8, and F8·2HCl can serve as a promising therapeutic agent against undifferentiated gastric cancer.

Mechanisms for cellular detoxification of drug compounds are of significant interest in human health. Cyclosporine A (CsA) and tacrolimus (FK506) are widely known antifungal and immunosuppressive microbial natural products. However, both compounds can result in significant side effects when used as immunosuppressants. The insect pathogenic fungus Beauveria bassiana shows resistance to CsA and FK506. However, the mechanisms underlying the resistance have remained unknown. Here, we identify a P4-ATPase gene, BbCRPA, from the fungus, which confers resistance via a unique vesicle mediated transport pathway that targets the compounds into detoxifying vacuoles. Interestingly, the expression of BbCRPA in plants promotes resistance to the phytopathogenic fungus Verticillium dahliae via detoxification of the mycotoxin cinnamyl acetate using a similar pathway. Our data reveal a new function for a subclass of P4-ATPases in cell detoxification. The P4-ATPases conferred cross-species resistance can be exploited for plant disease control and human health protection.
