Forage legumes are contributing in sustainable crop production apart from nutritional security to the livestock. Forage legumes are crucial for the nutritional security for mankind as they are integral component for increased availability of animal protein and product which has higher biological value compared to plant proteins. The inclusion of forage legumes in crop production systems is more useful as these can not only provide food and feed to animals but also improves soil productivity and act as soil-conserving components of agricultural and agroforestry systems. Production system as a whole and tremendous deficit of forage nutritious resources demand in particular to give some importance to the forage crops especially leguminous forages. Therefore, critical assessment is necessary for determining the direction and magnitude of change in agricultural management practice with inclusions of forage legume. Forage legumes have good capacity as a feed to promote sufficient quantities and qualities required for different productive animals. Therefore, these crops can contribute to achieving the objectives of sustainable food/ fodder and environmental security. Hence, inclusion of legumes forages in cropping system is inevitable in advancing soil sustainability and food and nutritional security without compromising the long-term soil fertility base of the soil resources. Rational soil management practices must involve forage legume-based rotations and intercropping considerations for restoring soil health, and soil sustainability should be given due emphasis. Besides, forage legumes can also provide a wide range of benefits such as restoration of soil fertility, nitrogen fixation and fertilizer saving, enhancement of soil biology and biodiversity, improving soil carbon sequestration and by neutralizing negative impact of climate change. This review summarizes the potential role of forage legume in animal nutrition, soil fertility building, nitrogen fixation, soil biology and biodiversity, carbon sequestration, climate change and other ecological services provided.

The cotton in India provides direct livelihood to 6 million farmers and about 40-50 million people are employed in the cotton trade and its processing in India. Cotton is a tropical and subtropical crop. For successful germination of its seeds, a minimum temperature of 15ºC is required. It is grown mainly as a dry crop in black cotton and medium black soil. The physical properties of the cotton plant are very important to optimize the design parameters of cottonuprooter cum shredder. Physical properties such asstalk stem diameter, plant height, and taproot depth were determined for the development of cotton uprooter cum shredder. The obtained results for stalk stem diameter, plant height, and taproot depth were2.2 cm,117.4 cm, and 32.7 cm, respectively. The moisture content of the cotton stalks is 27.3% and the soil moisture content was found to be 11.1%. The maximum deviation of the cotton plant was found to be 7.5 cm.

Complete, reproducible extraction of protein material is essential for comprehensive and unbiased proteome analyses. A current gold standard is single-pot, solid-phase-enhanced sample preparation (SP3), in which organic solvent and magnetic beads are used to denature and capture proteins, with subsequently washes allowing contaminant removal. However, SP3 is dependent on effective protein immobilisation onto beads, risks losses during wash steps, and experiences a drop-off in protein recovery at higher protein inputs. Magnetic beads may also contaminate samples and instruments, and become costly for larger scale protein preparations. Here, we propose solvent precipitation SP3 (SP4) as an alternative to SP3, omitting magnetic beads and employing brief centrifugation—either with or without low-cost inert glass beads—as the means of aggregated protein capture. SP4 recovered equivalent or greater protein yields for 1–5000 µg preparations and improved reproducibility (median protein R2 0.99 (SP4) vs. 0.97 (SP3)). Deep proteome profiling (n = 9,076) also demonstrated improved recovery by SP4 and a significant enrichment of membrane and low-solubility proteins vs. SP3. The effectiveness of SP4 was verified in three other labs, each confirming equivalent or improved proteome characterisation over SP3. This work suggests that protein precipitation is the primary mechanism of SP3, and reliance on magnetic beads presents protein losses, especially at higher concentrations and amongst hydrophobic proteins. SP4 represents an efficient and effective alternative to SP3, provides the option to omit beads entirely, and offers virtually unlimited scalability of input and volume—all whilst retaining the speed and universality of SP3.BRIEFSolvent precipitation SP3 (SP4) captures aggregated protein for proteomics sample clean-up by omitting magnetic beads, instead employing brief centrifugation—with or without low-cost inert glass beads. Sandercyanin, a blue homo-tetrameric lipocalin protein purified from Canadian walleye (Stizostedion vitreus), is the first far-red fluorescent protein reported in vertebrates(1–3). Sandercyanin binds non-covalently to biliverdin IXα (BLA) and fluoresces at 675nm on excitation at 375nm and 635nm(1). Sandercyanin fluorescence can be harnessed for many in vivo applications when engineered into a stable monomeric form. Here, we report the spectral properties and crystal structures of engineered monomeric Sandercyanin-BLA complexes. Compared to wild-type protein, monomeric Sandercyanin (∼18kDa) binds BLA with similar affinities and show a broad red-shifted absorbance spectra but possess reduced quantum efficiency. Crystal structures reveal D-ring pyrrole of BLA rotated around the C14-C15 bond, which is stabilized by neighboring aromatic residues and increased water-mediated polar contacts in the BLA-binding pocket. A tetrameric Sandercyanin variant (Tyr-142-Ala) co-displaying red- and far-red absorbing states, and reduced fluorescence shows similar conformational changes in BLA binding pocket. Our results suggest that D-ring flexibility of BLA and its rearrangement reduces the fluorescence quantum-yield of monomeric Sandercyanin. Structures of monomeric Sandercyanin could be utilized as prototypes to generate bright BLA-inducible fluorescent proteins. Further, our study postulates a mechanism for modulating photo-states in BLA-bound lipocalins, known only in phytochromes till date.Significance StatementSandercyanin is a tetrameric red fluorescent protein from a blue variant of walleye (Stizostedion vitreum) that binds to biliverdin IXα (BLA). Its biophysical properties and structures have been published earlier(1). A bright and stable monomeric Sandercyanin could be utilized as a fusion protein for fluorescence-based applications. Here we report the first structures and spectral properties of fluorescent monomeric Sandercyanin-BLA complexes and describe the molecular basis of modulated spectral properties due to rotated D-ring pyrrole around C14-C15 bond and re-shuffling of BLA-binding pocket. BLA-bound monomeric Sandercyanin could be engineered into brighter variants for in-vivo applications. Our study also reveals an unfamiliar mechanism in BLA-binding lipocalins that regulates red- and far-red absorbance states.

The objective of the present article is to discuss a numerical method based on wavelets for finding the solution of pantograph differential equations with proportional delays. First, the pantograph differential equation is converted into system of linear algebraic equations and then unknown coefficients are induced by solving the linear system. The convergence of the approximate solution is also derived along with its error estimate. Some numerical examples are considered to demonstrate the superiority of Bernoulli wavelet over Haar, Chebyshev and Legendre wavelets etc. Publisher's Version
