Spring viraemia of carp (SVC) caused by spring viraemia of carp virus (SVCV) can infect almost all fish of cyprinids, which bring huge economic losses to aquaculture. Glycoprotein (G), as the most important antigenic determinant protein of SVCV, is widely considered as an effective method against SVCV. In our previous study, we found that G3 (131 aa) is the potential dominant antigen epitope that induces strong immune responses similar to G protein (510 aa). Here, in order to further improve the immune effect, we reported a subunit vaccine (PEG-G3) constructed by PEG-modified dominant epitope protein (G3). The results of serum antibody production, enzyme activities and immune-related genes expression showed that PEG-G3 induces significantly stronger immune protective responses against SVCV than G3. PEG modification significantly increased the serum antibody level of the vaccine, which increased significantly after immunization and reached the peak at 21 day post-vaccination. T-AOC and AKP activities in the lowest concentration group (5 μg) of PEG-G3 were significantly higher than those in the highest concentration group (20 μg) of G3. In PEG-G3 group, the expression of almost all genes increased at least 4 times compared with the control group. After 14-day challenge, the RPS (relative percentage survival) of the highest concentration of PEG-G3 group was 53.6%, while that of G3 group is 38.9%. Therefore, this work shows that PEG modification and dominant epitope screening may be effective methods to improve the immune protective effect of vaccines and to resist the infection of aquatic animal viral diseases.

Emerging studies have reported the mechanosensitive Piezo1 (piezo type mechanosensitive ion channel component 1) plays essential roles in regulating the vascular tone through mechanistic actions on intracellular calcium homeostasis. However, the specific roles of Piezo1 in pulmonary vessels remain incompletely understood. We aim to investigate whether and how Piezo1 regulates the intracellular calcium homeostasis in human pulmonary arterial smooth muscle cells (PASMCs) under normal and pulmonary arterial hypertension (PAH) conditions. Cultured human PASMCs isolated from both control donors and idiopathic PAH patients were used as cell models. Fura-2 based intracellular calcium imaging was performed to measure the intracellular free calcium concentration ([Ca 2+ ] i ). Results showed that activation of Piezo1 by Yoda1 increases [Ca 2+ ] i by inducing both intracellular calcium release from internal calcium stores through the intracellular (intra-) Piezo1 localized at the subcellular organelles, including endoplasmic reticulum/sarcoplasmic reticulum, mitochondria, and nucleus; as well as extracellular calcium influx through the plasma membrane-localized Piezo1 in a mechanism independent of the store-operated calcium entry. Moreover, the Piezo1-mediated increase of [Ca 2+ ] i is linked to increased contraction and proliferation of PASMCs. Yoda1 induces dose-dependent vasocontraction in endothelium-denuded rat intrapulmonary arteries. Significant upregulation and increased activity of Piezo1 were observed in idiopathic PAH-PASMCs versus donor-PASMCs, contributing to the increased [Ca 2+ ] i and excessive proliferation of idiopathic PAH-PASMCs. In summary, Piezo1 mediates the increase of [Ca 2+ ] i by triggering both intracellular calcium release and extracellular influx. The enhanced Piezo1 expression and activity accounts, at least partially, for the abnormally elevated [Ca 2+ ] i and proliferation in idiopathic PAH-PASMCs.

The adverse factors impacting the intestinal microbiota of newborns remain to be elucidated. We put forward a hypothesis that hyperoxia in combination with rituximab exhibits a synergistic effect that interferes with neonatal intestinal microbiota. Six C57BL/6J mice, aged 12 weeks and pregnant 18 days, were purchased. Their pups were breastfed and raised under a 75% oxygen or conventional environment. Low- (20 mg/kg) and high-dose (40 mg/kg) rituximab were intraperitoneally administered. Fecal genomic DNA was extracted and sequenced by a 16S rRNA platform. Severe intestinal dysbiosis in newborns were observed, whereas mild dysbiosis was caused by inducing hyperoxia alone, confirming the synergistic interference of the combination of hyperoxia and B-cell antagonist (rituximab) in neonatal intestinal microbiota disruption. Slight dysbiosis was observed in the intestinal microbiota of dams, indicating their much robust ability to confront hyperoxic conditions. The abundance of Akkermansia muciniphila was significantly and extensively altered in both pups and dams after being subjecting them to hyperoxic conditions with or without rituximab administration. In conclusion, this work demonstrated that the synergistic effect of hyperoxia and rituximab led to severe intestinal dysbiosis in newborns. More studies are recommended to explore the precise regulatory mode between hyperoxia and rituximab in intestinal microbiota.

OBJECTIVE This study evaluated the angiographic characteristics and clinical outcomes of chronic total occlusion (CTO) percutaneous coronary intervention (PCI) among patients with and without a history of myocardial infarction (MI). BACKGROUND The pathogenesis of CTO and myocardial viability are different in cases with or without previous MI. However, the lesion characteristics and clinical outcomes are unclear for these two groups. METHODS We reviewed consecutive patients who underwent single-vessel CTO PCI from 2010 to 2013. Patients were classified according to their history of MI. Acute procedural results were classified as optimal recanalization, suboptimal recanalization, or technical failure. The primary endpoint was the 5 year rate of cardiac death. RESULTS We identified 2,191 eligible patients, including 859 patients (39.2%) with previous MI. The overall technical success rate was 74.4%. Significance Axonal injury plays a major role in many neurodegenerative diseases. The dual leucine zipper kinase (DLK) signaling pathway is a key regulator of axonal injury-induced neuronal cell death; however, DLK also has an important role in promoting axonal outgrowth. Therefore, inhibiting DLK as a therapeutic approach for neurodegenerative diseases is limited to a neuroprotective outcome without axon regeneration, prohibiting restoration of function. In fact, there are currently no strategies that provide long-term neuroprotection and axonal regeneration after injury. Here, we identified the germinal cell kinase four (GCK-IV) family of kinases as targets to maximize neuroprotection while promoting axon regeneration, making it an attractive therapeutic approach for a subset of neurodegenerative diseases., Axon injury is a hallmark of many neurodegenerative diseases, often resulting in neuronal cell death and functional impairment. Dual leucine zipper kinase (DLK) has emerged as a key mediator of this process. However, while DLK inhibition is robustly protective in a wide range of neurodegenerative disease models, it also inhibits axonal regeneration. Indeed, there are no genetic perturbations that are known to both improve long-term survival and promote regeneration. To identify such a neuroprotective target, we conducted a set of complementary high-throughput screens using a protein kinase inhibitor library in human stem cell-derived retinal ganglion cells (hRGCs). Overlapping compounds that promoted both neuroprotection and neurite outgrowth were bioinformatically deconvoluted to identify specific kinases that regulated neuronal death and axon regeneration. This work identified the role of germinal cell kinase four (GCK-IV) kinases in cell death and additionally revealed their unexpected activity in suppressing axon regeneration. Using an adeno-associated virus (AAV) approach, coupled with genome editing, we validated that GCK-IV kinase knockout improves neuronal survival, comparable to that of DLK knockout, while simultaneously promoting axon regeneration. Finally, we also found that GCK-IV kinase inhibition also prevented the attrition of RGCs in developing retinal organoid cultures without compromising axon outgrowth, addressing a major issue in the field of stem cell-derived retinas. Together, these results demonstrate a role for the GCK-IV kinases in dissociating the cell death and axonal outgrowth in neurons and their druggability provides for therapeutic options for neurodegenerative diseases.

Microwave power transmission has traditionally been used for long-distance applications so far. For the first time, we experimentally demonstrate efficient short-distance wireless power transfer based on microwave radiation. To overcome the directivity deterioration of an antenna as the transfer distance decreases in the near field, an electromagnetic rectifying surface is proposed for reflection-less microwave reception and rectification. The rectifying surface is an array of metamaterial particles integrated with rectifiers, as a whole its impedance engineered to match with free space. Therefore, it keeps fully receiving the radiated power in the near field of a transmitting antenna without reflection; and maximally converts it into direct current. A rectifying surface of 14 × 14 unit cells is fabricated, and a short-distance wireless power transfer system is developed. Under an input power up to 45 dBm at 2.45 GHz, the measured transmission efficiency (from the microwave input to the direct current output) reaches 52.3%, 52.5%, 47.9%, and 41.7% over a distance of 5, 10, 15, and 20 cm, respectively. Even when the incident angle severely deviates to ±52.5° on E- and H-plane, the transmission efficiency over distance 15 cm merely declines 12% and 22.2% at most, respectively. The presented wireless power transfer based on microwave radiation via an electromagnetic rectifying surface provides a highly efficient and distance/angle-alignment insensitive alternative to the common coupling-based coil solution for short-distance applications.

In this communication, a superstrate for the high radiation performance of a Fabry–Perot antenna (FPA) is proposed that consists of a nonuniform partially reflective surface (NPRS) and a phase correcting structure (PCS). The NPRS and the PCS function to uniformize the amplitude and phase distributions, successively, of the FPA’s aperture field, thereby achieving a high directivity and a high aperture efficiency. Due to the independent amplitude and phase manipulations, the antenna design procedure is relatively rapid and effective. As a proof of concept, an FPA sample at an operating frequency of 5.8 GHz with a diameter of 200 mm is designed. The NPRS, comprising metallic square rings with dissimilar dimensions printed on a printed circuit board (PCB), is designed according to the leaky-wave method. A two-layer PCB structure is adopted as the PCS, which is designed by the transmission line model integrated with full-wave simulations. The FPA with the NPRS and the PCS is fabricated and measured for verification. The simulated and measured results are in good agreement. The directivity of the FPA with the proposed superstrate is 21.49 dB and the corresponding aperture efficiency is as high as 95.49% at 5.8 GHz.

Cultivated caladiums (Caladium × hortulanum Birdsey) are popular ornamental plants. Although somaclonal variation occurs frequently in caladium during tissue culture, little research has been conducted on obtaining and detecting variants from long-term callus cultures. Herein, plants were regenerated from ‘Red Flash’ caladium calluses subcultured for approximately 40 mo, and 116 out of 520 established plants were grouped into 14 somaclonal variation types based on their morphological differences. Nuclear DNA content of six types (SVT1, SVT2, SVT4, SVT5, SVT8, and SVT10) varied from − 1.08% to 0.33% compared with the wild type, and these variants shared a similar chromosome number to the wild caladium (2n = 2x = 30). Three types (SVT3, SVT7, and SVT9) containing 2.82 to 5.42% less nuclear DNA content was the result of losing one or two chromosomes, and one type (SVT6) with significantly lower cellular DNA content was due to losing four chromosomes. Four types (SVT11–SVT14) contained 85.16 to 101.52% more DNA content and the SVT12 and the SVT13 had a double number of chromosomes (2n = 4x = 60), while the SVT11 and SVT14 had four more chromosomes and six less chromosomes as compared with a typical tetraploid, respectively. Correlation analysis suggested that leaf thickness, leaf index, and stomatal characteristics could be used as indicators of plant ploidy in caladium. A wide variation of pigment content was found among the variation types, and the content of chlorophyll, flavonoid, and anthocyanin had a significant positive correlation with the color parameters a* and b*. Leaf color variants created by prolonged in vitro callus cultures might hold great promise for caladium breeding.

A simple chiral primary-tertiary diamine derived from C2-symmetric 1,2-diphenylethane-1,2-diamine as the organocatalyst in combination with the trifluoroacetic acid additive for the asymmetric Mannich reaction of cyclic N-sulfonyl trifluoromethylated ketimines and methyl ketones afforded the desired product with high enantioselectivity (73-96% ee). The reactions proceeded well for a variety of different substituted cyclic N-sulfonyl trifluoromethyl ketimines and various alkyl methyl ketones, providing access to diverse enantioenriched benzo-fused cyclic sulfamidate N-heterocycles bearing a trifluoromethylated α-tetrasubstituted carbon stereocenter. This study also investigated the diastereoselective reduction of the carbonyl group and ring cleavage reduction of the sulfamidate group of the corresponding Mannich product.

The development of biomaterials, stem cells and bioactive factors has led to cartilage tissue engineering becoming a promising tactic to repair cartilage defects. Various polymer three-dimensional scaffolds that provide an extracellular matrix (ECM) mimicking environment play an important role in promoting cartilage regeneration. In addition, numerous growth factors have been found in the regenerative process. However, it has been elucidated that the uncontrolled delivery of these factors cannot fully exert regenerative potential and can also elicit undesired side effects. Considering the complexity of the ECM, neither scaffolds nor growth factors can independently obtain successful outcomes in cartilage tissue engineering. Therefore, collectively, an appropriate combination of growth factors and scaffolds have great potential to promote cartilage repair effectively; this approach has become an area of considerable interest in recent investigations. Of late, an increasing trend was observed in cartilage tissue engineering towards this combination to develop a controlled delivery system that provides adequate physical support for neo-cartilage formation and also enables spatiotemporally delivery of growth factors to precisely and fully exert their chondrogenic potential. This review will discuss the role of polymer scaffolds and various growth factors involved in cartilage tissue engineering. Several growth factor delivery strategies based on the polymer scaffolds will also be discussed, with examples from recent studies highlighting the importance of spatiotemporal strategies for the controlled delivery of single or multiple growth factors in cartilage tissue engineering applications.

The subtractive manufacturing of machining and the additive manufacturing of 3D printing rely on special processing equipment, and the produced parts have inherent flaws such as single function and non-reusability. Combining with the development of self-assembly in swarm robotics, a programmable forming method with flexible task is previously proposed. However, self-assembly of hollow shapes which have a hole inside has proved to be difficult to achieve, because robots in the hollow shape have less communication and reference to make decisions, while individuals make decisions purely through local interactions. In this paper, we propose a novel distributed self-assembly approach for hollow shape, which employs the stratified mechanism and uses a chain forming approach. The novel approach mainly includes the methods of state update, follow-up motion, rule extension, and trapped planning, respectively, solving the message blocking, motion separation, and robot trapped problems when the motion-chains fill up the hollow shape. We evaluate the feasibility and flexibility of this approach in simulation, and demonstrate the self-assembly algorithm on the hardware robotic platform designed in our lab. The formation of the hollow shape expands the range in which self-assembly can be formed, making it suitable for various types of parts.

The micro-arc oxidization (MAO) ceramic layer on an LA103Z Mg-Li alloy substrate was treated with a hydrothermal treatment at 90 °C for various times. The effect of the hydrothermal treatment time on the microstructure and corrosion behavior of MAO/LDH composite coatings in 3.5 wt.% NaCl solution was investigated, and the mechanism of hydrothermal film formation and corrosion was discussed. The results show that MgO on the surface of the MAO ceramic coating was partially dissolved during the hydrothermal treatment, and the released Mg2+ ions combined with OH− ions in the hydrothermal solution to form Mg(OH)2 nanosheets, which were deposited on the surface of the ceramic coating and its pores. Pulmonary hypertension (PH) is responsible for premature death caused by progressive and severe heart failure. A simple, feasible, and reproducible animal model of PH is essential for the investigation of the pathogenesis and treatment of this condition. Previous studies have demonstrated that the vascular endothelial growth factor receptor 2 (VEGFR-2) inhibitor SU5416 combined with hypoxia could establish an animal model of PH. Here, we investigated whether SU5416 itself could induce PH in rats. The effects of SU5416 treatment followed by 5 weeks of normoxia were examined. Hemodynamic measurements and histological assessments of the pulmonary vasculature and the heart were conducted to evaluate the physiological and pathophysiological characteristics of PH. Compared with the control rats, the SU5416-treated rats showed significantly increased right ventricle systolic pressure, right ventricle mass, total pulmonary vascular resistance, and total pulmonary vascular resistance index, while the cardiac output and cardiac index were substantially decreased. Moreover, the degree of occlusion and the muscularization levels of the distal small pulmonary vessels and the medial wall thickness of larger vessels (OD > 50 μm) simultaneously increased. SU5416 inhibited pulmonary vascular endothelial cell apoptosis in rats, as shown by immunostaining of cleaved caspase-3. Furthermore, changes in the right ventricle, myocardial hypertrophy, myocardial edema, myocardial necrosis, striated muscle cell atrophy, vessel muscularization, neointimal occlusion, and increased collagen deposition were observed in the SU5416 group compared with the control group. Thus, treatment with SU5416 alone plus 5 weeks of normoxia could be sufficient to induce PH in rats, which may provide a good and convenient model for future investigation of PH.

Acclimatization of tissue cultured plants to greenhouse or field conditions is the final and most crucial step of micropropagation. Hydroponic system has the potential to harden in vitro plants in an efficient and convenient way. Herein, in vitro regenerated plants of Trichosanthes kirilowii were cultured in 1/4, 1/2 and full strength Hoagland nutrient solution under a simple hydroponic system during acclimatization, respectively, and soil culture was prepared as a control. Morphological, physiological, anatomical and histochemical alterations of the plants treated by different concentrations of nutrient solution and soil conditions were analyzed after 30 days of growth. Results showed that all the three concentrations of nutrient solution significantly improved the survival rate, plant growth, pigment content, photosynthesis capacity, and stomatal density compared with those grown in soil, and there were no significant differences of the plant morphological parameters and the stomatal characteristics among the three hydroponic treatments. The plants grown in the 1/4 Hoagland solution yielded the highest net photosynthesis rate, transpiration rate and stomatal conductance, as might be attributed to their highest photosynthetic pigment content among the treatments. Anatomical and histochemical studies revealed that enlarged air spaces, multiple lignified Φ thickenings, and heavier suberin and lignin were formed in the cortex of the adventitious roots grown in 1/4 Hoagland nutrient solution as compared with those cultured in soil conditions, indicating that T. kirilowii had enough phenotypic plasticity in the root structure to grow in hydroponic conditions by formation of larger aerenchyma spaces and apoplastic barriers. This study clearly demonstrates that hydroponics is an alternative for the tissue cultured plants of T. kirilowii to adapt to great environmental changes during acclimatization. Micropropagated plants of T. kirilowii can adapt to hydroponic conditions through changes in plant morphological, physiological and stomatal characteristics, and anatomical and histochemical features of roots during acclimatization.
