Abstract

Air pollution is the leading environmental risk factor for ill health, estimated to account for 4.9 million deaths and 147 million years of healthy life lost in 2017. As people spend the majority of their time indoors, there has been great concerns about the ubiquitous exposure to indoor air pollution, particularly fine particulate matter (PM2.5). Accumulating evidence suggests that both short- and long-term exposure to PM2.5 are associated excess risks of cardiovascular disease (CVD) and mortality. The use of air purifiers to reduce indoor air pollution exposure is increasingly recommended in air quality control strategies, but the cardiovascular impact of such intervention remain to be investigated in randomized controlled trials (RCTs). Therefore, we aimed to conduct a systematic review and meta-analyses to synthesize the results of all randomized controlled trials assessing the cardiovascular effects of indoor air purification interventions.

Abstract

Selective laser melting (SLM) is an emerging layer-wise additive manufacturing technique that can generate complex components with high performance. Particulate-reinforced aluminum matrix composites (PAMCs) are important materials for various applications due to the combined properties of Al matrix and reinforcements. Considering the advantages of SLM technology and PAMCs, the novel SLM PAMCs have been developed and researched in recent years. Therefore, the current research progress about the SLM PAMCs is reviewed. Firstly, special attention is paid to the solidification behavior of SLM PAMCs. Secondly, the important issues about the design and fabrication of high-performance SLM PAMCs, including the selection of reinforcement, the influence of parameters on the processing and microstructure, the defect evolution and phase control, are highlighted and discussed comprehensively. Thirdly, the performance and strengthening mechanism of SLM PAMCs are systematically figured out. Finally, future directions are pointed out on the advancement of high-performance SLM PAMCs.

226. The mutation features and geographical distributions of the surface glycoprotein (S gene) in SARS-CoV-2 strains: A comparative analysis of the early and current strains

Abstract

The surface glycoprotein (S protein) of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was used to develop coronavirus disease 2019 (COVID-19) vaccines. However, SARS-CoV-2, especially the S protein, has undergone rapid evolution and mutation, which has remained to be determined. Here, we analyzed and compared the early (12 237) and the current (more than 10 million) SARS-CoV-2 strains to identify the mutation features and geographical distribution of the S gene and S protein. Results showed that in the early strains, most of the loci were with relative low mutation frequency except S: 23403 (4486 strains), while in the current strains, there was a surge in the mutation strains and frequency, with S: 23403 constantly being the highest one, but tremendously increased to approximately 1050 times. Furthermore, D614 (S: 23403) was one of the most highly frequent mutations in the S protein of Omicron as of March 2022, and most of the mutant strains were still from the United States, and the United Kingdom. Further analysis demonstrated that in the receptor-binding domain, most of the loci with low mutation frequency in the early strains, while S: 22995 was nowadays the most prevalent loci with 3 122 491 strains in the current strains. Overall, we compare the mutation features of the S region in SARS-CoV-2 strains between the early and the current stains, providing insight into further studies in concert with emerging SARS-CoV-2 variants for COVID-19 vaccines. © 2022 Wiley Periodicals LLC.

Abstract

Persistent HPV infection is recognized as the main etiologic factor for cervical cancer. HPV expresses the oncoproteins E6 and E7, both of which play key roles in maintaining viral infection and promoting carcinogenesis. While siRNA- mediated targeting of E6 and E7 transcripts temporarily induces apoptosis in HPV-positive cells, it does not eliminate viral DNA within the host genome, which can harbor escape mutants. Here, we demonstrated that specifically targeting E6 and E7 within host DNA with transcription activator-like effector nucleases (TALENs) induces apoptosis, inhibits growth, and reduces tumorigenicity in HPV-positive cell lines. TALEN treatment efficiently disrupted E6 and E7 oncogenes, leading to the restoration of host tumor suppressors p53 and retinoblastoma 1 (RB1), which are targeted by E6 and E7, respectively. In the K14-HPV16 transgenic mouse model of HPV-driven neoplasms, direct cervical application of HPV16-E7-targeted TALENs effectively mutated the E7 oncogene, reduced viral DNA load, and restored RB1 function and downstream targets transcription factor E2F1 and cycling-dependent kinase 2 (CDK2), thereby reversing the malignant phenotype. Abstract

We have developed a simple, convenient and inexpensive voltammetric method for determining trace Sudan I contamination in chili powder, based on the catalyzed electrochemical reduction of Sudan I at the carbon nanotube modified electrode. Under optimized conditions, the method exhibited acceptable analytical performance in terms of linearity (over the concentration range 6.0×10–7 to 7.5×10–5 M, r = 0.9967), detection limit (2.0×10–7 M) and reproducibility (RSD = 4.6%, n=10, for 2.0×10–5 M Sudan I).
