201. The diagnostic value of metagenomic next-generation sequencing in critically ill patients with sepsis: A retrospective cohort study

Abstract

Metagenomic next-generation sequencing (mNGS) is a new high-throughput sequencing method that may have great importance in early diagnosis and clinical management of sepsis. This study aimed to detect the difference between mNGS and comprehensive routine microbiological test (CMT), and to explore the diagnostic efficacy of mNGS in septic patients. This study retrospectively analyzed 150 sepsis patients who were admitted to the intensive care units of 4 hospitals in Southwest China from October 1, 2018, to October 1, 2021, and underwent both blood mNGS and CMT. The demographic and clinical characteristics of the patients were recorded, and the distribution of pathogens was analyzed. Additionally, the diagnostic performance and concordance between mNGS and CMT were compared to evaluate the etiological diagnostic value of mNGS in sepsis patients. In this study of 150 sepsis patients, bacterial infections were identified in 126 (84.0%), viral in 15 (10.0%), and fungal in 9 (6.0%). Among the sample types, sputum was most common, representing 62% of the total cases. Bronchoalveolar lavage fluid constituted 58.7%, blood 56.0%, with other specimens including pleural fluid at 29.3%, pus at 19.3%, swabs at 9.3%, cerebrospinal fluid at 8.7%, tissue at 6.0%, and bone marrow at 5.3%. mNGS demonstrated a diagnostic accuracy of 56.0% for sepsis, with a sensitivity of 84.4%, specificity of 26.0%, a positive predictive value of 54.6%, a negative predictive value of 61.3%. Metagenomic testing enables the rapid and early identification of infectious pathogens in sepsis patients, especially fungi and viruses. The study found that mNGS has high sensitivity in diagnosing sepsis patients, particularly for fungal and viral infections. mNGS technology is beneficial for critically ill sepsis patients.

Abstract

Cubic Co9S8 fracture structure nanoshells and cubic Co3S4 nanocrystallines were prepared by an ultrasonic spray pyrolysis process using cobalt chloride (CoCl2·6H2O) and thiourea (CH4N2S) as the starting materials. The as-prepared Co9S8 nanoshells and Co3S4 nanocrystallines were characterized by X-ray diffraction (XRD), scanning electron microscope (SEM) with energy-disperse X-ray spectroscopy (EDX) attachment and Fourier-transform infrared (FTIR) spectrometer. It is found that the phase transition could be controlled by adjusting the reaction temperature, while morphologies of final products could be influenced by the molar ratio and reaction time of the precursor. And the possible mechanism for the formation of hollow and solid crystalline structures of the product was proposed.

Abstract

The responses of Vallisneria natans to different water depths were analyzed in potted plants cultivated in a pond. The aim of the present study was to outline the effects of turbid water (i.e. low light) on the morphological parameters and photosynthetic efficiency of V. natans. After 60 days, V. natans plants of morphological characteristics (blade length, width, area, and growth rate) at a depth of 140–170 cm showed a significant decrease with increasing water depth and each parameter was negatively correlated with water depth (p < 0.01); the growth rate was remarkably inhibited with increasing water depth. The portion of light energy absorbed for electron transfer through the antenna pigments gradually decreased at a water depth of 120–170 cm and the concentration of electrons involved in CO2 fixation and qP decreased. 205. ANALYSIS OF EF2~EF3 TORNADO ON JULY 22, 2020 IN NORTHERN JIANGSU PROVINCE.

Abstract

Based on the data from conventional observation, ground intensive observation, and the Doppler weather radars in Suqian, Huai'an and Yancheng, Jiangsu Province, the present study analyzed the tornado that happened on July 22, 2020 in northern Jiangsu Province. The research shows that: (1) The favorable environmental fields of this tornado included a strong 0~1 km vertical wind shear in the lower layer, sufficient water vapor in the middle and lower layers, low uplift condensation height, and strong convergence and upward movement. At the early stage, the continuous precipitation and cold air intrusion in the middle layer strengthened the stratification instability, providing a humid and unstable environment for the tornado. (2) The convective storm that caused the tornado originated near the surface convergence line. There were small-scale local vortices in the vicinity of the tornado and the ground convergence center moved eastward along the ground convergence line. Before and after the tornado, the surface wind speed near the surface vortex suddenly increased, the wind direction suddenly changed, the air pressure significantly decreased, and there was short-term heavy precipitation. (3) The supercell that caused the tornado had a life history of 3 hours. During its eastward movement, it was accompanied by deep mesocyclone and showed tornadic vortex signature (TVS). The first mesocyclone was detected in the middle of the storm, about 2.6 km in height, and then it gradually stretched vertically. The TVS bottom height was maintained at about 1km, and the intensity was maintained at about 60 ×10-3 s-1. When the base height of the mesocyclone is low and the shear value continues to be strong, combined with the evolution of the ground mesoscale system, it is possible to provide effective early warning of tornadoes. Abstract

International audience; Understanding the transcriptional function of thyroid hormone receptors implies a precise analysis of their interactions with chromatin and other protein components of the cells. We present here two protocols that are routinely used in our laboratory. The first co-immunoprecipitation procedure allows addressing the capacity of proteins to form stable multiprotein complexes with TRs in cells. The chromatin affinity purification enables us to define the sites occupied by TRs on chromatin. In this case the lack of high quality antibodies is circumvented by introducing an N-terminal tag in TR, with unspecific affinity for immunoglobulins.

Abstract

Aim: Previous studies have indicated that early metastasis is a major cause of mortality in patients with choriocarcinoma. However, what determines whether early metastasis of choriocarcinoma has occurred is unknown. The emerging role of miRNA in regulating cancer development and progression has been recognized. miR‐373 has been shown to play pivotal roles in tumorigenesis and metastasis. However, whether miR‐373 functions to promote choriocarcinoma metastasis is not clear. The purpose of this study is to determine the function of miR‐373‐3p in the progression of this cancer. Methods: In this study, we first compared epithelial–mesenchymal transition (EMT)‐related markers, which were inversely correlated with miR‐373‐3p expression in trophoblast and choriocarcinoma cell lines. Using PCR and Western blot, upregulation of miR‐373‐3p was observed to inhibit EMT progression. Similarly, gain‐ and loss‐of‐function studies revealed that ectopic miR‐373‐3p overexpression inhibited the migration by transwell methods of choriocarcinoma cells. Results: Our results revealed that miR‐373‐3p acted as an EMT inhibitor in JEG‐3 and JAR cells; this was due to its mediation of the transforming growth factor‐β (TGFβ) signaling pathway, which was responsible for EMT. miRNA microarray analysis demonstrated that miR‐373‐3p interacted with the 3′ untranslated region of TGFβR2 mRNA, and then Western blot and dual‐luciferase reporter gene assays verified this interaction. Conclusion: Our findings suggest that miR‐373‐3p upregulation partly accounts for TGFβR2 downregulation and leads to a restraint of EMT and migration. miR‐373‐3p may therefore serve as a valuable potential target in the treatment of choriocarcinoma. Abstract

BACKGROUND: Penicillic acid (PA) is produced by Aspergillus spp. and Penicillium spp., which are common postharvest and storage fungi of fruits. PA can be of concern for human health because of its toxicity and high fruit consumption by the population. However, no data on PA occurrence in various fruits have yet been reported. A quick, easy, cheap, effective, rugged and safe (QuEChERS) approach for PA determination in various fruits was developed and applied to explore PA incidence in fruits. RESULTS: The modified QuEChERS procedure with extraction by ethyl acetate and purification by multi‐walled carbon nanotubes (MWCNTs), primary secondary amine (PSA) and octadecyl silane (C18) was established to determine PA in various fruits by high‐performance liquid chromatography–tandem mass spectrometry (HPLC‐MS/MS). The average recoveries were 72.9–102.2% and relative standard deviations (RSDs) were 1.3–7.9%. A total of 161 fruits samples, including kiwi, apple, peach, grape and mandarin/orange, were collected in southern China. The incidence of PA in fruits was 14.9% and the levels of PA contamination were 0.200–0.596 μg kg−1. Our results suggested that orange/mandarin, grape and kiwi were favorable matrices for Aspergillus spp. and Penicillium spp. to produce PA, rather than peach and apple. CONCLUSION: To the best of our knowledge, this is the first report about PA contamination in various fruits in China. Abstract

Osteogenesis imperfecta (OI) is a rare heritable disease with systemic connective tissue disorder. Most of the patients represent autosomal dominant form of OI, and are usually resulting from the mutations in type I collagen genes. However, the gene mutations reported previously only account for ∼70% of the OI cases. Here, in a Chinese OI family, we examined seven patients and nine normal individuals using the whole genome sequencing and molecular genetic analysis. The mutation of rs66612022 (COL1A2:p.Gly328Ser) related to glycine substitution was found in the seven patients. Moreover, we identified a novel missense mutation (HMMR:p.Glu2Gln). Interestingly, the individuals of this family with both the mutations were suffering from OI, while the others carried one or none of them are normal. The mutations of COL1A2 and HMMR and their combined effect on OI would further expand the genetic spectrum of OI. [ABSTRACT FROM AUTHOR]
