Abstract

The Yuquan capsules is a commonly used traditional Chinese Patent Medicine used for the treatment of diabetes mellitus. In this study, a high‐throughput analytical method for identifying the chemical composition of Yuquan capsules was established for the first time by using ultra‐performance liquid chromatography–quadrupole time of flight mass spectrometry. The data obtained were subjected to fragment analysis and this was combined with UNIFI processing of natural products. One‐hundred sixteen compounds were characterized from Yuquan capsules. Twelve of the bioactive compounds were quantitatively analyzed by ultra‐performance liquid chromatography‐tandem triple quadrupole mass spectrometry. This study was undertaken to obtain a comprehensive chemical profile analysis as well as to evaluate the overall quality of Yuquan capsules. The results will provide a reference for the quality evaluation of different Yuquan preparations. Abstract

The lattice parameters, formation energy and electronic structure as well as elastic property and Debye temperature of Ca-doped Ti2Ni alloy have been calculated. The results show that the structural stability of Ca replacing the Ti atomic site is more stable than that of Ca replacing the Ni atom. This is consistent with the calculated energy band structure and electronic density of the state. The elastic properties approximated by Voigt–Reuss–Hill formulae show that the ratio of bulk modulus to shear modulus and Poisson's ratio of Ca replacing Ni system are greater than that of Ca replacing Ti system, the interstitial doping system and the pure Ti2Ni alloy, implying that Ca replacing Ni is more malleable and the ductility of Ti2Ni alloy can be tailored by Ca doping. The hardness and Debye temperature also indicate that both ratios of Ca replacing Ti system are higher than that of Ca replacing Ni system. [ABSTRACT FROM AUTHOR]

212. Regulating zinc deposition behaviors by using a functional PANI modification layer on a separator for high performance aqueous zinc-ion batteries.

Abstract

Aqueous zinc-ion batteries (AZIBs) are promising energy storage devices due to their high safety, abundant zinc reserves, high theoretical capacity and low redox potential. Glass fiber (GF) has been widely used as a separator in AZIBs; however, the strong affinity to zinc ions of GF easily induces the growth of zinc dendrites towards the separator and results in a short circuit of the battery. Herein, we propose a new strategy for regulating zinc deposition behaviors by in situ polyaniline (PANI) modification on a commercial GF separator (GF/PANI). The theoretical and experimental results indicate that a PANI modification layer weakens zincophilicity force, homogenizes the surface electric field distribution and induces uniform plating/stripping of zinc ions, and thus, the reversibility of the zinc anode is significantly improved. Typically, a Zn‖Zn symmetric battery with a GF/PANI-600 separator can sustain a longer cycle up to 3000 h at 1.0 mA cm−2 with 1.0 mA h cm−2, far exceeding than that with a GF separator (70 h). Additionally, a Zn‖MnO2 full battery with a GF/PANI-600 separator also delivers a higher capacity retention rate of 89.5% after 300 cycles at 0.5 A g−1 (50.7% that of the Zn‖GF‖MnO2 battery). Abstract

Neonatal jaundice refers to the clinical symptoms of yellowing of the skin, sclerosing and mucous membrane due to abnormal bilirubin metabolism, resulting in the blood bilirubin level to rise during the neonatal period. Physiological jaundice and pathological jaundice are two types of the disease. Western medicine treats pathological jaundice with drugs therapy, light therapy, and transfusion therapy, etc., all of which have a certain effect. But they often have rash, diarrhea and other adverse reactions. In recent years, more and more clinical studies have been made in the treatment of neonatal jaundice in Chinese medicine, and many achievements have been made, but there is a lack of evidence-based medical evidence on the effectiveness and safety of treatment methods. Therefore, this study uses meta-analysis method to compare the clinical effectiveness and safety of touching and acupoint massage as adjuvant therapy in combination with the traditional Chinese medicine Yinzhihuang preparation and traditional Chinese medicine Yinzhihuang preparation alone in the treatment of neonatal jaundice, which provides theoretical basis for finding safe and effective and inexpensive treatment of neonatal jaundice.

Abstract

Butyrylcholinesterase (BCHE) is a natural bioscavenger that protects humans against organophosphate toxicity. Due to the limited yield of human BCHE (hBCHE) when purifying from human plasma, it is necessary to find an alternative method to produce this protein. One potential method is to produce transgenic livestock that make modified milk containing high concentration of hBCHE. In this study, we cloned the hBCHEgene into a human lactoferrin (hLF) bacterial artificial chromosome (BAC) construct to make a hLF-hBCHE BAC construct. Subsequently, we injected the BAC construct into pronuclei of mouse fertilized embryos and generated transgenic mice. Expression analysis showed that recombinant hBCHE (rhBCHE) was expressed efficiently in the mammary gland of the transgenic mice and the concentration of rhBCHE in the milk of individual mice ranged from 76±12 to 159±28 mg·L-1. Protein function tests showed that rhBCHE has the same enzymatic activity as the native hBCHE. Our results pave the way for making transgenic livestock to produce large quantities of rhBCHE.

Abstract

Promoting blood–spinal cord barrier (BSCB) repair at the early stage plays a crucial role in treatment of spinal cord injury (SCI). Excessive activation of autophagy can prevent recovery of BSCB after SCI. Basic fibroblast growth factor (bFGF) has been shown to promote BSCB repair and locomotor function recovery in SCI. However, the therapeutic effect of bFGF via direct administration on SCI is limited because of its rapid degradation and dilution at injury site. Based on these considerations, controlled release of bFGF in the lesion area is becoming an attractive strategy for SCI repair. At present, we have designed a sustained-release system of bFGF (called ALG-bFGF) using sodium alginate hydrogel, which is able to load large amounts of bFGF and suitable for in situ administration of bFGF in vivo. Here, traumatic SCI mice models and oxygen glucose deprivation (OGD)–stimulated human brain microvascular endothelial cells were performed to explore the effects and the underlying mechanisms of ALG-bFGF in promoting SCI repair. After a single in situ injection of ALG-bFGF hydrogel into the injured spinal cord, sustained release of bFGF from ALG hydrogel distinctly prevented BSCB destruction and improved motor functional recovery in mice after SCI, which showed better therapeutic effect than those in mice treated with bFGF solution or ALG. Evidences have demonstrated that autophagy is involved in maintaining BSCB integrity and functional restoration in animals after SCI. In this study, SCI/OGD exposure–induced significant upregulations of autophagy activation-related proteins (Beclin1, ATG5, LC3II/I) were distinctly decreased by ALG-bFGF hydrogel near the baseline and not less than it both in vivo and in vitro, and this inhibitory effect contributed to prevent BSCB destruction. Finally, PI3K inhibitor LY294002 and KLF4 inhibitor NSC-664704 were applied to further explore the underlying mechanism by which ALG-bFGF attenuated autophagy activation to alleviate BSCB destruction after SCI. The results further indicated that ALG-bFGF hydrogel maintaining BSCB integrity by inhibiting autophagy activation was regulated by PI3K/Akt/FOXO1/KLF4 pathway. In summary, our current study revealed a novel mechanism by which ALG-bFGF hydrogel improves BSCB and motor function recovery after SCI, providing an effective therapeutic strategy for SCI repair.
