202. Computationally designing a fluorescent probe to detect lipid membrane phase changes

- Abstract
Lipid membranes are responsible for the regulation of many key cellular processes. Some of these processes, such as signalling events, are thought to be induced by the presence of lipid rafts. Observation of rafts in vivo has therefore been a key area of research to gain a better insight to the existence and function of membrane rafts. One promising technique is the use of fluorescent molecular probes designed to report on phase changes within a lipid membrane. In this work, a computational protocol to investigate a novel probe of membrane phase has been developed. In this approach, spin-flip time-dependent density functional theory is carefully calibrated and employed to investigate conical intersections to understand the restriction of intramolecular movement mechanism, whereby the fluorescence of a molecule is dependent upon the freedom of rotation around a bond responsible for the access of non-radiative decay pathways. The probes tested in this study are methyl derivatives of 1,6-diphenylhexatriene and applied to quantum mechanics/molecular mechanics simulations of raft and non-raft environments, along with a preliminary study of molecules which could act as delivery vessels in vivo. It was found that the key dihedral angle of the 2-methyl derivative (2Me) could rotate sufficiently for non-radiative decay in the liquid-disordered non-raft environment whereas rotation was inhibited in the liquid-ordered raft environment promoting fluorescence making 2Me an ideal candidate for a simple phase sensitive fluorescent membrane probe.

203. Successional dynamics of carbon sequestration in forests of the eastern United States

Abstract Carbon sequestration in the forests of the eastern United States is an important offset to the country's CO2 emissions. Much of the eastern forestland is the product of reforestation of abandoned agricultural land or recovery following clear‐cutting over a century ago. This has led to concerns that eastern forests are even‐aged and that rates of carbon sequestration will decline as forests increase in carbon. Our objective was to examine the successional dynamics of forest carbon sequestration—using live tree carbon stocks as a proxy for successional status—for the six broadly defined forest types present in the region. We used datasets from the National Forest Inventory (NFI) for the 31 US states from Minnesota south to Louisiana and eastward and analyzed live tree net carbon increment for 2007–2021, the period for which NFI plot remeasurement data were available for all 31 states. Spruce–fir and southern pines were the only forest types for which carbon increment declined even modestly over a significant fraction of the range of live tree carbon observed in the region, and southern pine–hardwood forests were the only forests in which predicted sequestration in live tree carbon declined to zero within the range of carbon stocks observed in the region. Northern hardwood–conifer forests, oak–hickory forests, and lowland forests experienced either no decline or a slight increase in sequestration in live tree carbon across the range of successional status observed in the region. Thus, the average stocks of live tree carbon per unit area increased steadily over the study period. At some point in succession, rates of mortality are expected to increase and balance gross growth, leading to zero net sequestration in live tree carbon. Mature and old‐growth stands, however, are rare in all six forest types, and mortality as a fraction of live tree carbon for all six forest types declined across the range of successional status present in the region. Our results thus provide no support for the hypothesis that the successional dynamics of forests in this region can be expected to lead to near‐term declines in net carbon sequestration.

204. Do Vegetation Fuel Reduction Treatments Alter Forest Fire Severity and Carbon Stability in California Forests?

Abstract Forest fire frequency, extent, and severity have rapidly increased in recent decades across the western United States (US) due to climate change and suppression‐oriented wildfire management. Fuels reduction treatments are an increasingly popular management tool, as evidenced by California's plan to treat 1 million acres annually by 2050. However, the aggregate efficacy of fuels treatments in dry forests at regional and multi‐decadal scales is unknown. We develop a novel fuels treatment module within a coupled dynamic vegetation and fire model to study the effects of dead biomass removal from forests in the Sierra Nevada region of California. We ask how annual treatment extent, stand‐level treatment intensiveness, and spatial treatment placement alter fire severity and live carbon loss. We find that a ∼30% reduction in stand‐replacing fire was achieved under our baseline treatment scenario of 1,000 km2 year−1 after a 100‐year treatment period. Prioritizing the most fuel‐heavy stands based on precise fuel distributions yielded cumulative reductions in pyrogenic stand‐replacement of up to 50%. Both removing constraints on treatment location due to remoteness, topography, and management jurisdiction and prioritizing the most fuel‐heavy stands yielded the highest stand‐replacement rate reduction of ∼90%. Even treatments that succeeded in lowering aggregate fire severity often took multiple decades to yield measurable effects, and avoided live carbon loss remained negligible across scenarios. Our results suggest that strategically placed fuels treatments are a promising tool for controlling forest fire severity at regional, multi‐decadal scales, but may be less effective for mitigating live carbon losses.

Tumor necrosis factor (TNF)α is a major regulator of inflammation. However, the epigenetic regulation of TNFα in the context of an exercise intervention among older adults with cancer is understudied. In this exploratory analysis, we used data from a single-arm mobile health (mHealth) exercise intervention among older adults with myeloid malignancies to 1) assess changes in TNFα promoter methylation, TNFα mRNA expression, serum TNFα and other related-cytokine levels after intervention; and 2) assess correlations between blood markers and exercise levels. Twenty patients were included. From baseline to post-intervention, there was no statistical changes in TNFα promoter methylation status at seven CpG sites, TNFα mRNA expression, and serum TNFα levels. Effect sizes, however, were moderate to large for several CpG sites (−120, −147, −162, and −164; Cohen's d = 0.44–0.75). Median serum TNFα sR1 levels increased (83.63, IQR 130.58, p = 0.06; Cohen's d = 0.18) but not the other cytokines. Increases in average daily steps were correlated with increases in TNFα promoter methylation at CpG sites −147 (r = 0.48; p = 0.06) and −164 (r = 0.51; p = 0.04). Resistance training minutes were negatively correlated with TNFα promoter methylation at CpG site −120 (r = −0.62; p = 0.02). All effect sizes were moderate to large. In conclusion, after a mHealth exercise intervention, we demonstrated changes with moderate to large effect sizes in several CpG sites in the TNFα promoter region. Exercise levels were correlated with increases in TNFα promoter methylation. Larger exercise trials are needed to better evaluate TNFα regulation to inform interventions to augment TNFα regulation in order to improve outcomes in older adults with cancer.

Shortness of breath, chest pain, and palpitations occur as postacute sequelae of COVID-19, but whether symptoms are associated with echocardiographic abnormalities, cardiac biomarkers, or markers of systemic inflammation remains unknown. In a cross-sectional analysis, we assessed symptoms, performed echocardiograms, and measured biomarkers among adults more than 8 weeks after confirmed SARS-CoV-2 infection. We modeled associations between symptoms and baseline characteristics, echocardiographic findings, and biomarkers using logistic regression. We enrolled 102 participants at a median of 7.2 months following COVID-19 onset; 47 individuals reported dyspnea, chest pain, or palpitations. Median age was 52 years, and 41% of participants were women. Female sex, hospitalization, IgG antibody against SARS-CoV-2 receptor binding domain, and C-reactive protein were associated with symptoms. Regarding echocardiographic findings, 4 of 47 participants (9%) with symptoms had pericardial effusions compared with 0 of 55 participants without symptoms; those with effusions had a median of 4 symptoms compared with a median of 1 symptom in those without effusions. There was no strong evidence for a relationship between symptoms and echocardiographic functional parameters or other biomarkers. Among adults more than 8 weeks after SARS-CoV-2 infection, SARS-CoV-2 RBD antibodies, markers of inflammation, and, possibly, pericardial effusions are associated with cardiopulmonary symptoms. Investigation into inflammation as a mechanism underlying postacute sequelae of COVID-19 is warranted.

- Abstract
We present two results of a search for MeV-scale neutrino and anti-neutrino events correlated with gravitational wave events/candidates and large solar flares with KamLAND. The KamLAND detector is a large-volume neutrino detector using liquid scintillator, which is located at 1 km underground under the top of Mt. Ikenoyama in Kamioka, Japan. KamLAND has multiple reaction channels to detect neutrinos. Electron antineutrino can be detected via inverse-beta decay with 1.8 MeV neutrino energy threshold. All flavors of neutrinos can be detected via neutrino-electron scattering without neutrino energy threshold. KamLAND has continued the neutrino observation since 2002 March. We use the data set of 60 gravitational waves provided by the LIGO/Virgo collaboration during their second and third observing runs and search for coincident electron antineutrino events in KamLAND. We find no significant coincident signals within a ±500 s timing window from each gravitational wave and present 90% C.L. upper limits on the electron antineutrino fluence between 108–1013 cm−2 for neutrino energies of 1.8–111 MeV. For a solar-flare neutrino search at KamLAND, we determine the timing window using the solar X-ray data set provided by the GOES satellite series from 2002 to 2019 and search for the excess of coincident event rate on the all-flavor neutrinos. We find no significant event rate excess in the flare time windows and get 90% C.L. upper limits on the fluence of neutrinos of all flavors (electron anti-neutrinos) between 1010–1013 cm−2 (108–1013 cm−2) for neutrino energies in the energy range of 0.4–35 MeV.

208. Growing impact of wildfire on western US water supply

Streamflow often increases after fire, but the persistence of this effect and its importance to present and future regional water resources are unclear. This paper addresses these knowledge gaps for the western United States (WUS), where annual forest fire area increased by more than 1,100% during 1984 to 2020. Among 72 forested basins across the WUS that burned between 1984 and 2019, the multibasin mean streamflow was significantly elevated by 0.19 SDs (P < 0.01) for an average of 6 water years postfire, compared to the range of results expected from climate alone. Significance is assessed by comparing prefire and postfire streamflow responses to climate and also to streamflow among 107 control basins that experienced little to no wildfire during the study period. The streamflow response scales with fire extent: among the 29 basins where >20% of forest area burned in a year, streamflow over the first 6 water years postfire increased by a multibasin average of 0.38 SDs, or 30%. Postfire streamflow increases were significant in all four seasons. Historical fire-climate relationships combined with climate model projections suggest that 2021 to 2050 will see repeated years when climate is more fire-conducive than in 2020, the year currently holding the modern record for WUS forest area burned. These findings center on relatively small, minimally managed basins, but our results suggest that burned areas will grow enough over the next 3 decades to enhance streamflow at regional scales. Wildfire is an emerging driver of runoff change that will increasingly alter climate impacts on water supplies and runoff-related risks.

We present the results of a time-coincident event search for low-energy electron antineutrinos in the KamLAND detector with gamma-ray bursts (GRBs) from the Gamma-ray Coordinates Network and Fermi Gamma-ray Burst Monitor. Using a variable coincidence time window of ±500 s plus the duration of each GRB, no statistically significant excess above the background is observed. We place the world's most stringent 90% confidence level upper limit on the electron antineutrino fluence below 17.5 MeV. Assuming a Fermi-Dirac neutrino energy spectrum from the GRB source, we use the available redshift data to constrain the electron antineutrino luminosity and effective temperature.

We report on a search for electron antineutrinos ($\bar{\nu}_e$) from astrophysical sources in the neutrino energy range 8.3 to 30.8 MeV with the KamLAND detector. In an exposure of 6.72 kton-year of the liquid scintillator, we observe 18 candidate events via the inverse beta decay reaction. Although there is a large background uncertainty from neutral current atmospheric neutrino interactions, we find no significant excess over background model predictions. Assuming several supernova relic neutrino spectra, we give upper flux limits of 60--110 cm$^{-2}$ s$^{-1}$ (90% CL) in the analysis range and present a model-independent flux. We also set limits on the annihilation rates for light dark matter pairs to neutrino pairs. These data improves on the upper probability limit of $^{8}$B solar neutrinos converting into $\bar{\nu}_e$'s, $P_{\nu_e \rightarrow \bar{\nu}_e} < 3.5\times10^{-5}$ (90% CL) assuming an undistorted $\bar{\nu}_e$ shape. This corresponds to a solar $\bar{\nu}_e$ flux of 60 cm$^{-2}$ s$^{-1}$ (90% CL) in the analysis energy range., Comment: 21 pages, 9 figures, 4 tables, accepted for publication in Astrophysical Journal

The past few decades have seen major developments in the design and operation of cryogenic particle detectors. This technology offers an extremely good energy resolution - comparable to semiconductor detectors - and a wide choice of target materials, making low temperature calorimetric detectors ideal for a variety of particle physics applications. Rare event searches have continued to require ever greater exposures, which has driven them to ever larger cryogenic detectors, with the CUORE experiment being the first to reach a tonne-scale, mK-cooled, experimental mass. CUORE, designed to search for neutrinoless double beta decay, has been operational since 2017 at a temperature of about 10 mK. This result has been attained by the use of an unprecedentedly large cryogenic infrastructure called the CUORE cryostat: conceived, designed and commissioned for this purpose. In this article the main characteristics and features of the cryogenic facility developed for the CUORE experiment are highlighted. A brief introduction of the evolution of the field and of the past cryogenic facilities are given. The motivation behind the design and development of the CUORE cryogenic facility is detailed as are the steps taken toward realization, commissioning, and operation of the CUORE cryostat. The major challenges overcome by the collaboration and the solutions implemented throughout the building of the cryogenic facility will be discussed along with the potential improvements for future facilities. The success of CUORE has opened the door to a new generation of large-scale cryogenic facilities in numerous fields of science. Broader implications of the incredible feat achieved by the CUORE collaboration on the future cryogenic facilities in various fields ranging from neutrino and dark matter experiments to quantum computing will be examined., Comment: 45 pages, 14 figures

Guideline-based treatment for sepsis and septic shock is difficult because sepsis is a disparate range of life-threatening organ dysfunctions whose pathophysiology is not fully understood. Early intervention in sepsis is crucial for patient outcome, yet those interventions have adverse effects and are frequently overadministered. Greater personalization is necessary, as no single action is suitable for all patients. We present a novel application of reinforcement learning in which we identify optimal recommendations for sepsis treatment from data, estimate their confidence level, and identify treatment options infrequently observed in training data. Rather than a single recommendation, our method can present several treatment options. We examine learned policies and discover that reinforcement learning is biased against aggressive intervention due to the confounding relationship between mortality and level of treatment received. We mitigate this bias using subspace learning, and develop methodology that can yield more accurate learning policies across healthcare applications., Comment: 25 pages, 8 figures

We present a comprehensive statistical analysis of 106 sheath regions driven by coronal mass ejections (CMEs) and measured near 1 AU. Using data from the STEREO probes, this extended analysis focuses on two discrete categorizations. In the first categorization, we investigate how the generic features of sheaths change with their potential formation mechanisms (propagation and expansion sheaths), namely, their associations with magnetic ejectas (MEs) which are primarily expanding or propagating in the solar wind. We find propagation sheaths to be denser and driven by stronger MEs, whereas expansion sheaths are faster. Exploring the temporal profiles of these sheaths with a superposed epoch technique, we observe that most of the magnetic field and plasma signatures are more elevated in propagation sheaths relative to expansion sheaths. The second categorization is based on speed variations across sheaths. Employing linear least squares regression, we categorize four distinct speed profiles of the sheath plasma. We find that the associated shock properties and solar cycle phase do not impact the occurrence of such variations. Our results also highlight that the properties of the driving MEs are a major source of variability in the sheath properties. Through logistic regression, we conclude that the magnetic field strength and the ME speed in the frame of the solar wind are likely drivers of these speed variations.

In-situ measurements carried out by spacecraft in radial alignment are critical to advance our knowledge on the evolutionary behavior of coronal mass ejections (CMEs) and their magnetic structures during propagation through interplanetary space. Yet, the scarcity of radially aligned CME crossings restricts investigations on the evolution of CME magnetic structures to a few case studies, preventing a comprehensive understanding of CME complexity changes during propagation. In this paper, we perform numerical simulations of CMEs interacting with different solar wind streams using the linear force-free spheromak CME model incorporated into the EUropean Heliospheric FORecasting Information Asset (EUHFORIA) model. The novelty of our approach lies in the investigation of the evolution of CME complexity using a swarm of radially aligned, simulated spacecraft. Our scope is to determine under which conditions, and to what extent, CMEs exhibit variations of their magnetic structure and complexity during propagation, as measured by spacecraft that are radially aligned. Results indicate that the interaction with large-scale solar wind structures, and particularly with stream interaction regions, doubles the probability to detect an increase of the CME magnetic complexity between two spacecraft in radial alignment, compared to cases without such interactions. This work represents the first attempt to quantify the probability of detecting complexity changes in CME magnetic structures by spacecraft in radial alignment using numerical simulations, and it provides support to the interpretation of multi-point CME observations involving past, current (such as Parker Solar Probe and Solar Orbiter), and future missions.

We present the first PSP-observed CME that hits a second spacecraft before the end of the PSP encounter, providing an excellent opportunity to study short-term CME evolution. The CME was launched from the Sun on 10 October 2019 and was measured in situ at PSP on 13 October 2019 and at STEREO-A on 14 October 2019. The small, but not insignificant, radial (~0.15 au) and longitudinal (~8 deg) separation between PSP and STEREO-A at this time allows for observations of both short-term radial evolution as well as investigation of the global CME structure in longitude. Although initially a slow CME, magnetic field and plasma observations indicate that the CME drove a shock at STEREO-A and also exhibited an increasing speed profile through the CME (i.e. evidence for compression). We find that the presence of the shock and other compression signatures at 1 au are due to the CME having been overtaken and accelerated by a high speed solar wind stream (HSS). We estimate the minimum interaction time between the CME and the HSS to be about 2.5 days, indicating the interaction started well before the CME arrival at PSP and STEREO-A. Despite alterations of the CME by the HSS, we find that the CME magnetic field structure is similar between the vantage points, with overall the same flux rope classification and the same field distortions present. These observations are consistent with the fact that coherence in the magnetic structure is needed for steady and continued acceleration of the CME.

Abstract Physiological stress levels in response to sexual behavior stigma among men who have sex with men (MSM) in the United States (US) are understudied. The current study aims to explore the relationship between sexual behavior stigma and salivary cortisol both overall and stratified by race/ethnicity. If such an association exists, it may suggest that sexual behavior stigma can be physiologically measured or indicated by the presence of heightened salivary cortisol. A subsample of 667 MSM participants from the 2019 American Men’s Internet Survey (AMIS; N = 10,129) submitted morning (AM) and evening (PM) saliva cortisol samples using at-home mail-in collection kits. Average daily cortisol and daily cortisol change were calculated; simple linear regressions estimated associations between cortisol measures and sexual behavior stigma characterized in four different ways (ever and recent experience of individual stigma items; average ever and recent experience of three stigma scales: stigma from family and friends, anticipated healthcare stigma, general social stigma). Participants reported a mean age of 36.0 years (SD = 14.9), with most being non-Hispanic white (n = 480, 72.0%), Hispanic (n = 164, 12.3%), or Black/African American (n = 146, 10.9%), and identified as homosexual/gay (n = 562, 84.3%). Reporting ever experiencing healthcare providers gossiping was significantly associated with higher PM cortisol (β = 0.12, p = 0.001) and higher average daily cortisol (β = 0.11, p = 0.004), while reporting ever experiencing police refusing to protect was associated with higher AM cortisol (β = 0.08, p = 0.03) and higher average daily cortisol (β = 0.09, p = 0.02). Recent experiences of stigma were not significant predictors of any measure of cortisol. Measures of salivary cortisol may be used to characterize sexual behavior stigma among MSM populations, however more insight is needed to determine its exact relationship and strength.

Abstract Background CommunityRx is an evidence-based social care intervention delivered to family and friend caregivers (“caregivers”) at the point of healthcare to address health-related social risks (HRSRs). Two CommunityRx randomized controlled trials (RCTs) are being fielded concurrently on Chicago’s South Side, a predominantly African American/Black community. CommunityRx-Hunger is a double-blind RCT enrolling caregivers of hospitalized children. CommunityRx-Dementia is a single-blind RCT enrolling caregivers of community-residing people with dementia. RCTs with caregivers face recruitment barriers, including caregiver burden and lack of systematic strategies to identify caregivers in clinical settings. COVID-19 pandemic-related visitor restrictions exacerbated these barriers and prompted the need for iteration of the protocols from in-person to remote operations. This study describes these protocols and methods used for successful iteration to overcome barriers. Methods and findings CommunityRx uses individual-level data to generate personalized, local community resource referrals for basic, health and caregiving needs. In early 2020, two in-person RCT protocols were pre-tested. In March 2020, when pandemic conditions prohibited face-to-face clinical enrollment, both protocols were iterated to efficient, caregiver-centered remote operations. Iterations were enabled in part by the Automated Randomized Controlled Trial Information-Communication System (ARCTICS), a trial management system innovation engineered to integrate the data collection database (REDCap) with community resource referral (NowPow) and SMS texting (Mosio) platforms. Enabled by engaged Community Advisory Boards and ARCTICS, both RCTs quickly adapted to remote operations. To accommodate these adaptations, launch was delayed until November (CommunityRx-Hunger) and December (CommunityRx-Dementia) 2020. Despite the delay, 65% of all planned participants (CommunityRx-Hunger n = 417/640; CommunityRx-Dementia n = 222/344) were enrolled by December 2021, halfway through our projected enrollment timeline. Both trials enrolled 13% more participants in the first 12 months than originally projected for in-person enrollment. Discussion Our asset-based, community-engaged approach combined with widely accessible institutional and commercial information technologies facilitated rapid migration of in-person trials to remote operations. Remote or hybrid RCT designs for social care interventions may be a viable, scalable alternative to in-person recruitment and intervention delivery protocols, particularly for caregivers and other groups that are under-represented in traditional health services research. Abstract Tumors acquire alterations in oncogenes and tumor suppressor genes in an adaptive walk through the fitness landscape of tumorigenesis. However, the interactions between oncogenes and tumor suppressor genes that shape this landscape remain poorly resolved and cannot be revealed by human cancer genomics alone. Here, we use a multiplexed, autochthonous mouse platform to model and quantify the initiation and growth of more than one hundred genotypes of lung tumors across four oncogenic contexts: KRAS G12D, KRAS G12C, BRAF V600E, and EGFR L858R. We show that the fitness landscape is rugged—the effect of tumor suppressor inactivation often switches between beneficial and deleterious depending on the oncogenic context—and shows no evidence of diminishing-returns epistasis within variants of the same oncogene. These findings argue against a simple linear signaling relationship amongst these three oncogenes and imply a critical role for off-axis signaling in determining the fitness effects of inactivating tumor suppressors.

The CUPID-Mo experiment to search for 0$u\beta\beta$ decay in $^{100}$Mohas been recently completed after about 1.5 years of operation at LaboratoireSouterrain de Modane (France). It served as a demonstrator for CUPID, a nextgeneration 0$u\beta\beta$ decay experiment. CUPID-Mo was comprised of 20enriched Li$_2$$^{100}$MoO$_4$ scintillating calorimeters, each with a mass of$\sim$ 0.2 kg, operated at $\sim$20 mK. We present here the final analysis withthe full exposure of CUPID-Mo ($^{100}$Mo exposure of 1.47 kg$\times$yr) usedto search for lepton number violation via 0$u\beta\beta$ decay. We report onvarious analysis improvements since the previous result on a subset of data,reprocessing all data with these new techniques. We observe zero events in theregion of interest and set a new limit on the $^{100}$Mo 0$u\beta\beta$ decayhalf-life of $T^{0u}_{1/2} > 1.8 \times 10^{24}$ year (stat.+syst.) at 90%CI. Under the light Majorana neutrino exchange mechanism this corresponds to aneffective Majorana neutrino mass of $\left <(0.28$--$0.49)$ eV, dependent upon the nuclear matrix element utilized.

We report on a search for electron antineutrinos ( ) from astrophysical sources in the neutrino energy range 8.3-30.8 MeV with the KamLAND detector. In an exposure of 6.72 kton-year of the liquid scintillator, we observe 18 candidate events via the inverse beta decay reaction. Although there is a large background uncertainty from neutral current atmospheric neutrino interactions, we find no significant excess over background model predictions. Assuming several supernova relic neutrino spectra, we give upper flux limits of 60-110 cm-2 s-1 (90% confidence level, CL) in the analysis range and present a model-independent flux. We also set limits on the annihilation rates for light dark matter pairs to neutrino pairs. These data improve on the upper probability limit of 8B solar neutrinos converting into , (90% CL) assuming an undistorted shape. This corresponds to a solar flux of 60 cm-2 s-1 (90% CL) in the analysis energy range.

We report the result of a search for neutrinos in coincidence with solar flares from the GOES flare database. The search was performed on a 10.8 kton-year exposure of KamLAND collected from 2002 to 2019. This large exposure allows us to explore previously unconstrained parameter space for solar flare neutrinos. We found no statistical excess of neutrinos and established 90% confidence level upper limits of 8.4 × 107 cm-2 (3.0 × 109 cm-2) on the electron antineutrino (electron neutrino) fluence at 20 MeV normalized to the X12 flare, assuming that the neutrino fluence is proportional to the X-ray intensity.

LKB1 is among the most frequently altered tumor suppressors in lung adenocarcinoma. Inactivation of Lkb1 accelerates the growth and progression of oncogenic KRAS-driven lung tumors in mouse models. However, the molecular mechanisms by which LKB1 constrains lung tumorigenesis and whether the cancer state that stems from Lkb1 deficiency can be reverted remains unknown. To identify the processes governed by LKB1 in vivo, we generated an allele which enables Lkb1 inactivation at tumor initiation and subsequent Lkb1 restoration in established tumors. Restoration of Lkb1 in oncogenic KRAS-driven lung tumors suppressed proliferation and led to tumor stasis. Lkb1 restoration activated targets of C/EBP transcription factors and drove neoplastic cells from a progenitor-like state to a less proliferative alveolar type II cell-like state. We show that C/EBP transcription factors govern a subset of genes that are induced by LKB1 and depend upon NKX2-1. We also demonstrate that a defining factor of the alveolar type II lineage, C/EBPα, constrains oncogenic KRAS-driven lung tumor growth in vivo. Thus, this key tumor suppressor regulates lineage-specific transcription factors, thereby constraining lung tumor development through enforced differentiation.

We study two interplanetary coronal mass ejections (ICMEs) observed at Mercury and 1 AU by spacecraft in longitudinal conjunction, investigating the question: what causes the drastic alterations observed in some ICMEs during propagation, while other ICMEs remain relatively unchanged? Of the two ICMEs, the first one propagated relatively self-similarly, while the second one underwent significant changes in its properties. We focus on the presence or absence of large-scale corotating structures in the ICME propagation space between Mercury and 1 AU, that have been shown to influence the orientation of ICME magnetic structures and the properties of ICME sheaths. We determine the flux rope orientation at the two locations using force-free flux rope fits as well as the classification by Nieves-Chinchilla et al. (2019). We also use measurements of plasma properties at 1 AU, the size evolution of the sheaths and ME with heliocentric distance, and identification of structures in the propagation space based on in situ data, remote-sensing observations, and simulations of the steady-state solar wind, to complement our analysis. Results indicate that the changes observed in one ICME were likely caused by a stream interaction region, while the ICME exhibiting little change did not interact with any transients between Mercury and 1 AU. This work provides an example of how interactions with corotating structures in the solar wind can induce fundamental changes in ICMEs. Our findings can help lay the foundation for improved predictions of ICME properties at 1 AU.

The KamLAND-Zen 800 experiment is searching for the neutrinoless double-beta decay of $^{136}$Xe by using $^{136}$Xe-loaded liquid scintillator. The liquid scintillator is enclosed inside a balloon made of thin, transparent, low-radioactivity film that we call Inner Balloon (IB). The IB, apart from guaranteeing the liquid containment, also allows to minimize the background from cosmogenic muon-spallation products and $^{8}$B solar neutrinos. Indeed these events could contribute to the total counts in the region of interest around the Q-value of the double-beta decay of $^{136}$Xe. In this paper, we present an overview of the IB and describe the various steps of its commissioning minimizing the radioactive contaminations, from the material selection, to the fabrication of the balloon and its installation inside the KamLAND detector. Nuclear Experiment - Abstract
The possibility that neutrinos may be their own antiparticles, unique among the known fundamental particles, arises from the symmetric theory of fermions proposed by Ettore Majorana in 1937. Given the profound consequences of such Majorana neutrinos, among which is a potential explanation for the matter-antimatter asymmetry of the universe via leptogenesis, the Majorana nature of neutrinos commands intense experimental scrutiny globally; one of the primary experimental probes is neutrinoless double beta ($0 \nu \beta \beta$) decay. Here we show results from the search for $0 \nu \beta \beta$ decay of $^{130}$Te, using the latest advanced cryogenic calorimeters with the CUORE experiment. CUORE, operating just 10 millikelvin above absolute zero, has pushed the state of the art on three frontiers: the sheer mass held at such ultra-low temperatures, operational longevity, and the low levels of ionising radiation emanating from the cryogenic infrastructure. We find no evidence for $0 \nu \beta \beta$ decay and set a lower bound of $T_{1/2}^{0 \nu} > 2.2 \times 10^{25}$ years at a 90% credibility interval. We discuss potential applications of the advances made with CUORE to other fields such as direct dark matter, neutrino and nuclear physics searches and large-scale quantum computing, which can benefit from sustained operation of large payloads in a low-radioactivity, ultra-low temperature cryogenic environment.

We defined PASC as the presence of 1 or more coronavirus disease 2019 (COVID-19)-attributed symptoms beyond 90 days. We compared fold-changes in marker values between those with and without PASC using mixed-effects models with terms for PASC and early and late recovery time periods.ResultsDuring early recovery, those who went on to develop PASC generally had higher levels of cytokine biomarkers including tumor necrosis factor-α (1.14-fold higher mean ratio [95% confidence interval {CI}, 1.01-1.28]; P = .028) and interferon-γ-induced protein 10 (1.28-fold higher mean ratio [95% CI, 1.01-1.62]; P = .038). Among those with PASC, there was a trend toward higher interleukin 6 levels during early recovery (1.29-fold higher mean ratio [95% CI, .98-1.70]; P = .07), which became more pronounced in late recovery (1.44-fold higher mean ratio [95% CI, 1.11-1.86]; P

Two of the most pressing questions in physics are the microscopic nature of the dark matter that comprises 84% of the mass in the universe and the absence of a neutron electric dipole moment. These questions would be resolved by the existence of a hypothetical particle known as the quantum chromodynamics (QCD) axion. In this work, we probe the hypothesis that axions constitute dark matter, using the ABRACADABRA-10cm experiment in a broadband configuration, with world-leading sensitivity. We find no significant evidence for axions, and we present 95% upper limits on the axion-photon coupling down to the world-leading level $g_{a\gamma\gamma}<3.2 \times10^{-11}$ GeV$^{-1}$, representing one of the most sensitive searches for axions in the 0.41 - 8.27 neV mass range. Our work paves a direct path for future experiments capable of confirming or excluding the hypothesis that dark matter is a QCD axion in the mass range motivated by String Theory and Grand Unified Theories., Comment: 17 pages, 12 figures

Nuclear Experiment - Abstract
The CUORE experiment is a large bolometric array searching for the lepton number violating neutrino-less double beta decay ($0\nu\beta\beta$) in the isotope $\mathrm{^{130}Te}$. In this work we present the latest results on two searches for the double beta decay (DBD) of $\mathrm{^{130}Te}$ to the first $0^{+}_2$ excited state of $\mathrm{^{130}Xe}$: the $0\nu\beta\beta$ decay and the Standard Model-allowed two-neutrinos double beta decay ($2\nu\beta\beta$). Both searches are based on a 372.5 kg$\times$yr TeO$_2$ exposure. The de-excitation gamma rays emitted by the excited Xe nucleus in the final state yield a unique signature, which can be searched for with low background by studying coincident events in two or more bolometers. The closely packed arrangement of the CUORE crystals constitutes a significant advantage in this regard. The median limit setting sensitivities at 90\% Credible Interval (C.I.) of the given searches were estimated as $\mathrm{S^{0\nu}_{1/2} = 5.6 \times 10^{24} \: \mathrm{yr}}$ for the ${0\nu\beta\beta}$ decay and $\mathrm{S^{2\nu}_{1/2} = 2.1 \times 10^{24} \: \mathrm{yr}}$ for the ${2\nu\beta\beta}$ decay. No significant evidence for either of the decay modes was observed and a Bayesian lower bound at $90\%$ C.I. on the decay half lives is obtained as: $\mathrm{(T_{1/2})^{0\nu}_{0^+_2} > 5.9 \times 10^{24} \: \mathrm{yr}}$ for the $0\nu\beta\beta$ mode and $\mathrm{(T_{1/2})^{2\nu}_{0^+_2} > 1.3 \times 10^{24} \: \mathrm{yr}}$ for the $2\nu\beta\beta$ mode. These represent the most stringent limits on the DBD of $^{130}$Te to excited states and improve by a factor $\sim5$ the previous results on this process., Comment: 13 pages, 6 figures

Developing sustainable software for the scientific community requires expertise in software engineering and domain science. This can be challenging due to the unique needs of scientific software, the insufficient resources for software engineering practices in the scientific community, and the complexity of developing for evolving scientific contexts. While open-source software can partially address these concerns, it can introduce complicating dependencies and delay development. These issues can be reduced if scientists and software developers collaborate. We present a case study wherein scientists from the SuperNova Early Warning System collaborated with software developers from the Scalable Cyberinfrastructure for Multi-Messenger Astrophysics project. The collaboration addressed the difficulties of open-source software development, but presented additional risks to each team. For the scientists, there was a concern of relying on external systems and lacking control in the development process. For the developers, there was a risk in supporting a user-group while maintaining core development. These issues were mitigated by creating a second Agile Scrum framework in parallel with the developers' ongoing Agile Scrum process. This Agile collaboration promoted communication, ensured that the scientists had an active role in development, and allowed the developers to evaluate and implement the scientists' software requirements. The collaboration provided benefits for each group: the scientists actuated their development by using an existing platform, and the developers utilized the scientists' use-case to improve their systems. This case study suggests that scientists and software developers can avoid scientific computing issues by collaborating and that Agile Scrum methods can address emergent concerns., Comment: Revisions: in response to peer-review recommendations, most sections have been substantially expanded and reworked, five new figures have been added, and the title has been changed. There are many theories where a dark matter particle is part of a multiplet with an electrically charged state. If WIMP dark matter ($\chi^{0}$) is accompanied by a charged excited state ($\chi^{-}$) separated by a small mass difference, it can form a stable bound state with a nucleus. In supersymmetric models, the $\chi^{0}$ and the $\chi^{-}$ could be the neutralino and a charged slepton, such as the neutralino-stau degenerate model. The formation binding process is expected to result in an energy deposition of {\it O}(1--10 MeV), making it suitable for detection in large liquid scintillator detectors. We describe new constraints on the bound state formation with a xenon nucleus using the KamLAND-Zen 400 Phase-II dataset. In order to enlarge the searchable parameter space, all xenon isotopes in the detector were used. For a benchmark parameter set of $m_{\chi^{0}} = 100$ GeV and $\Delta m = 10$ MeV, this study sets the most stringent upper limits on the recombination cross section $\langle\sigma v\rangle$ and the decay-width of $\chi^{-}$ of $2.0 \times 10^{-31}$ ${\rm cm^3/s}$ and $1.1 \times 10^{-18}$ GeV, respectively (90\% confidence level).
