213. Unique Duplex Microstructure and Porosity Effect on Mechanical Properties of AlCoCrFeNi2.1Eutectic High-Entropy Alloys Processed by Selective Laser Melting

Abstract

Selective laser melting (SLM) was performed on AlCoCrFeNi2.1eutectic high-entropy alloys (EHEAs), and the unique duplex microstructure and porosity were investigated as well as the negative effect on mechanical properties. SLM printed AlCoCrFeNi2.1EHEAs is composed of face-centered cubic and BCC/B2 phases, and the surface texture of samples perpendicular to and parallel to the building direction is different. From the bottom of the molten pool to the inside, the structure changes from dendrites to columnar crystals and then to cellular crystals, the distribution of elements in and around the molten pool is uniform, and there is no element segregation. The surface hardness of as-printed AlCoCrFeNi2.1EHEAs is higher than that of as-cast ones due to the grain refinement strengthening, and there is anisotropy in different surface hardness, the highest hardness is 580 HV (X–Yplane) and 560 HV (X–Zplane). The reduction of bearing area and tip stress concentration caused by pores have adverse effects on the tensile strength and elongation of the alloy, the ultimate tensile strength increased from 1060 to 1289 MPa.

Abstract

ObjectiveTo provide a basis for human enteroviruses prevention and control by monitoring the enterovirus （EV） and its main virus types.MethodsSamples of hand-foot-and-mouth disease， herpetic angina and fever clinic patients in Dapeng New District of Shenzhen from 2016 to 2022 were tested for EV with real-time polymerase chain reaction （PCR）. To identify the isolates of EV， VP1 genes of EV were amplified with nested reverse transcription PCR， and then sequenced.A geneticphylogenetic tree was constructed based on the VP1 gene.ResultsAmong the 1 124 suspected hand-foot-and-mouth disease cases， 740 （65.84%） tested EV positive. Coxsackievirus A6 （CVA6） and Coxsackievirus A16 （CVA16） were the main two serotypes with regular cycle trends. Of the 137 suspected herpetic angina cases， 88 （64.23%） were EV positive， with Coxsackievirus A4 （CVA4） and CVA16 as the dominant serotypes. Among 428 respiratory infection specimens， 71 （16.59%） were EV positive. Coxsackievirus A4 （CVA4） was the predominant serotype which caused herpetic angina and respiratory infection. The epidemic EV isolates CVA6 from Shenzhen had a close genetic relationship with isolates in China’s mainland.ConclusionThe main serotypes EV CVA6 and CVA16 which caused hand-foot-and-mouth disease exhibit cyclical trends . The risk of EV transmitted from abroad is low， but their genetic variation and virulence change should be monitored continuously. In addition， the monitoring of dominant isolates CVA4 which cause herpetic angina and respiratory infection should be strengthened.

Abstract

Background: Immunotherapy has exhibited efficacy in thymic carcinoma patients; however, there are insufficient data to confirm this efficacy in thymoma. The toxicity of immunotherapy also remains to be determined. Methods: The efficacy and safety of immunotherapy were analyzed in 11 thymoma patients who received PD‐1 inhibitors according to a range of relevant indexes including the objective response rate (ORR), disease control rate (DCR), progression‐free survival (PFS), overall survival (OS), and immunotherapy‐related adverse events. Results: The PFS and OS rates for all patients were 12.8 and 56.5 months, respectively. No difference in efficacy was detected between monotherapy and combination therapy (PFS: 12.8 vs 2.2 months, P = 0.787; OS: 73.8 vs 56.5 months, P = 0.367). The ORRs and DCRs for all patients were 27.3% and 90.9%, respectively. The incidence of adverse events was 45.5% among the 11 thymoma patients, including immune‐related myocarditis (36.4%), immune‐related liver damage (18.2%), and myasthenia gravis (18.2%). In the whole cohort of patients, the rate of adverse events of grade 3 or higher was 36.4%. The rates of adverse events of grade 3 or 4 in B3‐type and non‐B3‐type thymoma patients were 0% and 62.5%, respectively. Conclusions: Immunotherapy elicited a response in thymoma patients; however, more attention should be paid to the immune‐related adverse events. [ABSTRACT FROM AUTHOR]

Abstract

Fibroblast growth factor receptor (FGFR) family includes four highly conserved receptor tyrosine kinases. Particularly, FGFR2 has been identified as a potential target for tyrosine kinase inhibitor (TKI) treatment. Except for immunohistochemistry and fluorescence in situ hybridization, next-generation sequencing (NGS) technology represents a novel tool for FGFR2 detection that covers a wide range of fusion genes. In the present work, we present a case of cholangiocarcinoma who had FGFR2-BICC1 rearrangement detected by NGS. A 76-year-old female diagnosed with cholangiocarcinoma underwent four cycles of chemotherapy. The NGS assay showed that the tumor had a FGFR2-BICC1 rearrangement. The patient had a favorable tumor response to sorafenib. Herein, we report the first case with cholangiocarcinoma harboring FGFR2-BICC1 who is sensitive to sorafenib therapy.

Abstract

Background: MicroRNAs (miRNAs), a large family of short endogenous RNAs known to post-transcriptionally repress gene expression, participate in the regulation of almost every cellular process. Changes in miRNA expression are associated with many pathologies. Ovarian folliculogenesis and testicular spermatogenesis are complex and coordinated biological processes, in which tightly regulated expression and interaction of a multitude of genes could be regulated by these miRNAs. Identification and preliminary characterization of gonad-specific miRNAs would be a prerequisite for a thorough understanding of the role that miRNA-mediated posttranscriptional gene regulation plays in mammalian reproduction.Method: Here, we present the identification of a repertoire of porcine miRNAs in adult ovary and testis using deep sequencing technology. A bioinformatics pipeline was developed to distinguish authentic mature miRNA sequences from other classes of small RNAs represented in the sequencing data.Results: Using this approach, we detected 582 precursor hairpins (pre-miRNAs) encoding for 732 mature miRNAs, of which 673 are unique. Statistically, 224 unique miRNAs (out of 673, 33.28%) were identified which had significant differential expression (DE) between ovary and testis libraries (P < 0.001). Most of DE miRNAs located on the X chromosome (X-linked miRNAs) (24 out of 34, 70.59%) significantly up-regulated in ovary versus testis (P < 0.001). Predictably, X-linked miRNAs are expressed in a testis-preferential or testis-specific pattern. To explore the potential for co-expression among genomic location clusters of X-linked miRNAs, we surveyed the relationship between the distance separating miRNA loci and the coordinate expression patterns of 32 high confidence X-linked miRNAs in seven normal pig tissues using the real-time quantitative PCR (q-PCR) approach. Our results show that proximal pairs of miRNAs are generally co-expressed implying that miRNAs within 50 kb of genomic bases are typically derived from a common transcript.Conclusions: The present study characterizes the miRNA transcriptome of adult porcine gonads, with an emphasis on the co-expression patterns of X-linked miRNAs. Our report should facilitate studies of the organ-specific reproductive roles of miRNAs.

237. Comprehensive study of hot deformation behavior and fracture mechanism of Al/Cu laminated composite

Abstract

The hot deformation behavior of Al/Cu laminated composite was studied by isothermal tensile experiments in the temperature range of 200–500 °C and strain rate range of 0.001–1/s. The Arrhenius constitutive model was established to predict the flow stress by determining the constants of composites. In addition, based on the dynamic material model (DMM), the hot working map is established. The results show that the power dissipation efficiency is high under the conditions of high temperature, medium and low strain rate. When the strain is 0.25, there are few areas of stability due to the increasing frictional resistance at the interface. It is found that the dynamic recrystallization (DRX) is promoted by hot tensile and laminate structure. The softening mechanism of Al side is mainly dynamic recovery (DRV) and discontinuous dynamic recrystallization (DDRX), while that of Cu side is DRX. During the tensile process, the Al side grains always maintain stable Brass, Copper and S textures; the Cu side grains always maintain the Cube texture with stable-end orientation and when the deformation exceeds 60 %, the Goss unstable texture appears. It is conducive to dislocation movement and increases the recrystallization ratio. Then, the fracture mechanism of Al/Cu laminated composite is revealed. Due to the presence of the interface, the Cu side shows brittle fracture at room temperature. The snake-like sliding occurs on both sides of Al and Cu under hot deformation, showing ductile fracture, and the defects on Cu side are more serious than those on Al side. These results lay a foundation for optimizing the hot working parameters, improving the performance of the forming parts and clarifying the fracture damage mechanism of Al/Cu laminated composite.

238. Significant transitions of microstructure and mechanical properties in laser additive manufacturing AlCoCrFeNi2.1eutectic high-entropy alloy under heat treatment

Abstract

In this work, the AlCoCrFeNi2.1eutectic high-entropy alloys (EHEAs) were fabricated by laser additive manufacturing and subsequently heat-treated at various temperatures between 800 and 1200 °C to achieve excellent strength-plasticity synergy. The high solidification rate of selective laser melting (SLM) leads to instability of the eutectic growth front, resulting in a typical melt pool morphology of equiaxed and columnar crystals for AlCoCrFeNi2.1. The body-centered cubic (BCC) content of ∼82% in the SLM specimens leads to a higher tensile strength of ∼1175 MPa and poorer plasticity of ∼6.42%. After heat treatment, the BCC transforms to face-centered cubic (FCC) and the microstructure changes to a dendrite containing eutectic lamellar structure. As the temperature increases, Ni–Al aggregates, B2 phase particles precipitate and grow in the BCC matrix. Meanwhile, the uniform heating mode eliminates the orientation of the SLM melt pool structure, reduces the internal stress and dislocation density of the SLM alloy. 239. Microstructures and wear behaviors of WC particle reinforced nickel-based composites fabricated by selective laser melting

Abstract

To improve the mechanical performance of popular Nickel alloys, in this study, selective laser melting method was introduced to print IN718 alloy composites (WC/IN718) by using Tungsten carbide (WC) as reinforcement particles. The effects of WC content (10-20 wt%) on microstructure evolution and mechanical performance of the printed composites were analyzed comprehensively. It was found, with proper addition of WC reinforcement particles, desired microstructures and mechanical properties can be achieved in the printed composites. WC particles could distribute uniformly in the matrix and excellent metallurgical bonding could be obtained at the heterogeneous interface between the WCPand the IN718 matrix. Under strong laser irradiation, the surface of WC particles could be melted and the decomposed W and C atoms could diffuse into the matrix under strong laser irradiation. WC-W2C/MxC/IN718 (M representing Ni, Cr, Fe, Ti, Nb) sandwich structure was formed due to the substitution of matrix element for W and C element in WC particles. When the added WC particle content increases from 0 to 20 wt%, the hardness of the printed WC/IN718 composite can increase from 304.8 HV to 421.3 HV. Its wear resistance was found to first increase and then decrease due to the transition of the wear mechanism. Among all the printed composites, 15 %WC/IN718 has the best wear resistance and smallest coefficient of friction. The ranking of the wear resistance of the printed composites was found to be 15 %WC/IN718 > 10 %WC/IN718 > 20 %WC/IN718 > IN718.
