Simple Summary: Papillary thyroid carcinoma is still the most common endocrine tumor, most of which can be diagnosed by pathological results. However, there is still a small number of cases that are difficult to judge malignancy. In this study, we performed an integrative analysis of DNA methylation and RNA array from our cohort for potential papillary thyroid cancer-specific indicators. SERINC2, one of the differentially methylated and expressed genes, was first identified as a potential tumor-driven indicator in papillary thyroid carcinoma. We confirmed the influence of SERINC2 on proliferation and apoptosis in vitro after intervention or overexpression. Furthermore, we found tryptophan metabolism as a potential pathway targeted by SERINC2 through the investigation of data from the Cancer Dependency Map. In conclusion, our results demonstrate the whole-genome DNA methylation and gene expression profiles of papillary thyroid carcinoma, identify a new tumor-driven indicator, and provide a novel insight into the etiology of papillary thyroid cancer. Most papillary thyroid carcinomas (PTCs) can be diagnosed preoperatively by routine evaluation, such as thyroid ultrasonography and fine-needle aspiration biopsy. Nevertheless, understanding how to differentiate indolent thyroid tumors from aggressive thyroid cancers remains a challenge, which may cause overtreatment. This study aimed to identify papillary thyroid cancer-specific indicators with whole-genome DNA methylation and gene expression profiles utilizing Infinium Methylation EPIC BeadChip (850k) and RNA arrays. In this paper, we report SERINC2 as a potential tumor-driven indicator in PTC. The up-regulated expression levels of SERINC2 were verified in PTC cell lines via qPCR. Then, cell counting kit 8 (CCK-8), wound healing, and flow cytometric assays were performed to confirm the influence of SERINC2 on proliferation and apoptosis in PTC cell lines after intervention or overexpression. Moreover, the investigation of data from the Cancer Dependency Map (DepMap) provided a potential pathway targeted by SERINC2. The activation of the tryptophan metabolic pathway may reduce the dependency of SERINC2 in thyroid cancers. In conclusion, our results demonstrate the whole-genome DNA methylation and gene expression profiles of papillary thyroid carcinoma, identify SERINC2 as a potential tumor-driven biomarker, and preliminarily verify its function in PTC. Background: The outcome of hepatitis B virus (HBV) infection is significantly affected by host immune response; herein, we aim to dissect the effect of L-carnitine (L-Cn) on germinal center (GC)-related immune cells and the influence on the prognosis of HBV infection.Methods: In vitro and in vivo experiments were performed in patients with chronic HBV infection and a hydrodynamic injection mouse model.Results: In vitro assays revealed that L-Cn significantly reduced GC-related immune responses and enhanced immunosuppressive profiles. Intriguingly, L-Cn released from lysed hepatocytes was associated with the degree of liver damage. Besides, the administration of L-Cn in an HBV mouse model resulted in delayed clearance of hepatitis B surface antigen (HBsAg) in serum and decreased GC formation in the spleen. Notably, patients with HBsAg loss showed decreased plasma L-Cn levels, and longitudinal observations found that low baseline levels of L-Cn were associated with a favorable treatment response in patients with chronic hepatitis B.Conclusions: The suppressive effect of hepatocyte-derived L-Cn on GC-related immune cells may contribute to the inability of HBsAg clearance in chronic HBV infection, indicating that L-Cn might serve as a potential therapeutic target for the treatment of HBV infection. Jerusalem artichoke (JA) has increasingly established feral populations with its introduction worldwide. In mid-June, 2019, white colonies of powdery mildew (PM) appeared on wild JA in Shenyang, Liaoning, China. ITS sequences retrieved from the pathogen clustered within the Golovinomyces lineage, and holomorph characteristics agreed with those of G. latisporus rather than G. ambrosiae. Pathogenicity tests confirmed that this pathogen caused the PM symptoms observed in the field. The PM pathogen was eventually identified as G. latisporus, the sole species in a clade including our combined PMITS (powdery mildew specific ITS), IGS (intergenic spacer), TUB2 (beta-tubulin) and CHS1 (chitin synthase I) sequences. In three JA invaded plant communities, G. latisporus showed high specificity for this introduced host among plants of 21 genera from 15 families. This is a first report of G. latisporus infecting feral JA in Liaoning, Northeast China. Cross-infective risks of G. latisporus between wild and cultivar JA, as well as its related sunflowers, deserve more attention as worldwide feralization of JA is increasing. [ABSTRACT FROM AUTHOR]

Purpose: To study the vibration characteristics of porous functionally graded material (FGM) circular plate in temperature field by using Voigt modified porous material mixed power-law model and the relationship between physical parameters and temperature. Methods: First, based on the first-order shear deformation theory, physical neutral plane and Hamilton variational principle, the dimensionless differential equation of motion control of porous FGM circular plate in temperature field is derived. Then, the differential equations and boundary conditions are analyzed by using the differential transformation method (DTM), and the algebraic characteristic equation for calculating the dimensionless natural frequency is obtained. The calculation results are compared with the existing literature to verify its effectiveness. Results: The effects of gradient index, thickness diameter ratio, temperature rise and porosity on dimensionless natural frequency under different boundary conditions were calculated and analyzed. Conclusion: Gradient index affects the distribution of material components, porosity weakens the overall stiffness of the structure, and then leads to the change of dimensionless natural frequency of free vibration of porous FGM circular plate in temperature field.
