202. Interferon Regulatory Factor 4: An Alternative Marker for Plasma Cells in Daratumumab‐Treated Patients With Multiple Myeloma.

Abstract

ABSTRACT Introduction Methods Results Conclusions Anti‐CD38 therapeutic modalities (e.g., daratumumab) can impede classical CD38 and CD138 gating use for plasma cell (PC) detection in multiple myeloma (MM) patients with minimal residual disease (MRD). We assessed the applicability of CD229, CD269, and interferon regulatory factor (IRF‐4) for PC detection in MM MRD patients.Bone marrow samples were collected from patients with MM. Through multiparameter flow cytometry, we evaluated the suitability of CD229, CD269, and IRF‐4 for distinguishing PCs from other hematopoietic cells and compared their expression pattern on normal PCs (nPCs) and aberrant PCs (aPCs). We also assessed IRF‐4 expression stability after sample storage under different conditions. A 10‐color MRD antibody panel was used to determine whether IRF‐4 is an alternative primary PC‐gating marker for MM MRD assessment.IRF‐4 was expressed specifically on all PCs; its mean fluorescence intensity (MFI) was highest on PCs among all hematopoietic cells. This MFI did not decrease even after sample storage at 4°C or 25°C for 72 h. In all 42 MRD assessment samples, except for samples (n = 10) with no PCs, the use of IRF‐4 enabled accurate nPC (n = 12), aPC (n = 13), and nPC + aPC (n = 7) identification. Even samples from daratumumab‐treated patients had high IRF‐4 MFI, with no difference between pre‐treatment and post‐treatment (n = 7; p = 0.610).IRF‐4 demonstrates high MFI on PCs, and it is not expressed on other leukocytes. In MM patients with MRD, daratumumab treatment does not affect IRF‐4 expression. IRF‐4 is a promising marker for PC identification in MRD assessment of MM patients undergoing anti‐CD38 therapy. Abstract

To improve the nutritional value and the palatability of air-dried rice straw, culture broth of the lactic acid bacteria community SFC-2 was used to examine the effects of two different treatments, fermentation and adsorption. Air-dried and chopped rice straw was treated with either fermentation for 30 d after adding 1.5 L nutrient solution (50 mL inocula L−1, 1.2×1012 CFU mL−1 inocula) kg−1 straw dry matter, or spraying a large amount of culture broth (1.5 L kg−1 straw dry matter, 1.5×1011 CFU mL−1 culture broth) on the straw and allowing it to adsorb for 30 min. The feed quality and aerobic stability of the resulting forage were examined. Both treatments improved the feed quality of rice straw, and adsorption was better than fermentation for preserving nutrients and improving digestibility, as evidenced by higher dry matter (DM) and crude protein (CP) concentrations, lower neutral detergent fiber (NDF), acid detergent fiber (ADF) and NH3-N concentrations, as well as higher lactic acid production and in vitro digestibility of DM (IVDMD). The aerobic stability of the adsorbed straw and the fermented straw was 392 and 480 h, respectively. After being exposed to air, chemical components and microbial community of the fermented straw were more stable than the adsorbed straw.

Abstract

Artesunate-transferrin(ATS-Tf) adduct was prepared through drug-ligand self-assembly.UV-vis spectrum analysis showed that ATS-Tf adduct can be easily formed with relatively high binding constant at neutral pH(3.4×105 L/mol at pH=7.4).However,the adduct became less stable with low binding constant at acidic condition(1.7×104 L/mol at pH=5.5).Proliferation inhibition studies of ATS-Tf adduct on cancer cells and normal cells showed that the ATS-Tf adduct had better antitumor activity on human to hepatocellular carcinoma cell (HepG2),lung adenocarcinoma cell (A549),and gastric carcinoma cell(MGC-803),while it had low toxicity on normal human liver cell(L-02).HPLC analysis confirmed that the incorporation of Tf increased the uptake of ATS by cancer cells.The interactive model and mechanism of ATS with Tf were further studied by molecular docking and fluorescence spectroscopy analysis.The possible inhibition mechanism of ATS-Tf adduct on cancer cells was also proposed.

Abstract

The present study investigated the degradation of un-pretreated wheat straw, corn stalk, and rice straw by a lignocellulose-degrading microbial consortium XDC-2. Following six days of cultivation, exocellular xylanase activities were 414.9, 491.9, and 335 U/mL, respectively. After 12 days, the rice straw had lost 39.71% of its weight, hemicellulose and cellulose losses of 78.27% and 14.08%, respectively. The total amount of volatile products reached a maximum on day six for rice straw degradation. The four major types of volatile products were acetic acid, propionic acid, butanoic acid, and glycerin, all of which would be suitable substrates for conversion to methanol by anaerobic digestion. According to PCR-DGGE analysis, XDC-2 remained stable during the degradation process of untreated lignocellulosic biomass. These results demonstrate the potential for further development and application of XDC-2; it is capable of degrading un-pretreated lignocellulosic materials, and has a low cost of operation.

Abstract

Returning crop straw to field is one of the common methods to improve soil organic matter and achieve sustainable development of agriculture.However,corn straw is composed mainly of highly polymerized lignocellulose with a high carbon to nitrogen(C:N) ratios,and it degrades slowly in the field.The difficulty of straw degradation in a short time hampers the seedling growth and farming operation.In some place farmers burn crop straw after harvest in order to immediately sow the next crop,which contributes to environmental pollution and greenhouse gas emissions.Acceleration of straw degradation in the field is urgently needed,and utilization of microbial inoculants is a safe and effective method.To reveal the effects of microbial inoculants on degrading process of corn straw and soil microbial diversity after straw was returned to the field,an incubation experiment was carried out in the field.The experiment was conducted in the winter Wheat-maize rotation of the high yield area located around Hebei Academy of Agriculture and forestry Sciences,North China Plain from Oct 2009 to Apr 2010,and three different microbial inoculants(ND,NK and NS) were used in this study.The residual weights and C/N ratios of corn straw in different inoculated treatments were measured to estimate the degrading enhancement of the three inoculants at four different sampling times of 15d,25d,145d and 160d.The results showed that the degradation of corn straw could be enhanced by inoculation in early period(first 25d),and residual percentages of corn straw in ND,NK and NS were 14.3%,7.7% and 1.6% lower than that in CK,respectively.C/N ratios of corn straw in inoculant treatments decreased more rapidly than that in CK,and the decreases were also showed up in early stage.In order to find the effects of microbial inoculants on soil microbial diversity,V3 region of bacterial 16S rDNA and D1 region of fungal 26S rDNA gene sequences were detected by denaturing gradient gel electrophoresis(DGGE),and Cluster analyses based on the band similarity in migration and intensity between lanes were used to understand the distance of each lanes.Furthermore bands of the three inoculants were used to study the relationships between inoculation and soil microbial diversity,and dominant bands of bacteria and fungi were excised from DGGE gels and sequenced to get further microbial information.The results showed that most bands from inoculants,which matched to Bacillus fusiformis(97%),Chloroflexus bacterium(87%),Uncultured Clostridiales bacterium(99%) and Uncultured compost bacterium(92%) were detected in early stage and disappear in later stage,and some band from inoculants were not detected.Soil bacterial and fungal composition responded to each added microbial inoculants in early stage.Compared with CK,adding microbial inoculants after straw returning to the field could effectively accelerate corn straw degradation in early period,and soil microbial diversity was accordingly different in early period and gradually went to the same level in late stage of the inoculation.
