Network-based pharmacological analysis of the molecular mechanism of Gong Ying detoxification lotion with soaking agent in the treatment of diabetic foot ulcers. We used the traditional Chinese medicine system pharmacology analysis platform (Tcmsp) and the BATMAN-TCM database to retrieve the active ingredients of the drug in the Gong Ying detoxification lotion, and used the UniProt database to obtain the predicted target and to explore the potential targets of the Diabetic foot ulcers; the String platform was used to construct the protein interaction Action network; the data were analyzed by GO and KEGG pathway enrichment, and finally Cytoscape software used to construct the '' Gong Ying detoxification lotion component-Diabetic foot ulcers target-pathway'' network. The core active ingredients of Gong Ying detoxification lotion in the treatment of Diabetic foot ulcers were found to be quercetin, choline, wogonin. The biological pathways mainly involved involve classical pathways operating in cancer including the HIF-1 signaling pathway. This study in reveals the characteristics of multi-target and multi-path combination of Gong Ying to treat Diabetic foot ulcers, and provides research ideas and theoretical basis for further treatment of Diabetic foot ulcers.

Diabetes mellitus is a common disorder of endocrine and metabolic disorders. Long term hyperglycemia can lead to a variety of serious chronic complications, such as diabetic nephropathy, diabetic retinopathy, diabetes cardiovascular disease, and so on, threatening human life, health and safety. Scutellaria baicalensis Georgi is one of the most commonly used traditional Chinese medicine. Flavonoids are the main effective components of Scutellaria baicalensis Georgi. The flavonoids are mainly baicalin, baicalein, wogonoside and wogonin. The four have the effects of improving the renal function, insulin resistance and retinopathy of type 2 diabetic patients. The aim of this review is to summarize the pathogenesis and clinical research progress of baicalin, baicalein, wogonoside and wogonin in Scutellaria baicalensis Georgi in the treatment of type 2 diabetes mellitus and its complications.

Ecological restoration in dredging areas has attracted increasing attention. The reconstruction of a submerged plant ecosystem is an important method for aquatic ecosystem restoration. This study has systematically investigated the effect of biochar-improved sediment on the plant growth and decontamination efficiency of a constructed ecosystem. Microbial community composition and structure in the sediment were detected. The results showed that a supplement of 20 mg/g of biochar significantly increased the biomass of the submerged plants compared with other doses (0, 10, and 40 mg/g). The biomass and chlorophyll content were significantly inhibited by supplementing 40 mg/g of biochar. In the Ceratophyllum demersum L. system, TP and NH4+-N concentrations were significantly lower after treatment with 20 mg/g of biochar compared to other doses. In Vallisneria spiralis L. and Hydrilla verticillata (L. f.) Royle systems, NH4+-N, TP, and DO concentrations were significantly different among different biochar treatments. In general, 20 mg/g of biochar improved water quality in different submerged plant systems, while 40 mg/g of biochar had adverse effects on water quality, such as higher NH4+-N and TP concentrations. The dominant microbial community included Proteobacteria, Acidobacteria, Chloroflexi, Actinobacteriota, and Bacteroidota. The structure and function of microbial communities were different among submerged plants and biochar treatments. One of the pathological hallmarks of Alzheimer's disease (AD) is the presence of extracellular deposits of amyloid beta (Aβ) peptide. In addition to Aβ as the core component of the amyloid plaque, the amyloid precursor protein (APP) processing fragment Aβ was also found accumulated around the plaque. The APPη pathway, mainly mediated by membrane‐type 5 matrix metalloproteinase (MT5‐MMP), represents an important factor in AD pathogenesis. The proamyloidogenic features of MT5‐MMP could result from interactions with APP when trafficking between organelles, so determination of the location within the cell of APPη cleavage and interacting proteins of MT5‐MMP affecting this process will be of priority in understanding the role of MT5‐MMP in AD. In the present study, MT5‐MMP was found to be located in the nucleus, cytosol, and cytosolic subcellular granules of CHO cells that stably expressed wild‐type human APP751. MT5‐MMP fusion proteins were constructed that could localize enzyme production in the Golgi apparatus, endosome, ER, mitochondria, or plasma membrane. The fusion proteins significantly increased sAPPη when directed to the endosome, Golgi apparatus, plasma membrane, or mitochondria. Since the C‐terminal region of MT5‐MMP is responsible for its intracellular location and trafficking, this domain was used as the bait in a yeast two‐hybrid screen to identify MT5‐MMP protein partners in a human brain cDNA library. Identified binding partners included N4BP2L1, TMX3, EIG121, bridging Integrator 1 (BIN1), RUFY4, HTRA1, and TMEM199. The binding of N4BP2L1, EIG121, BIN1, or TMX3 to MT5‐MMP resulted in the most significant increase in sAPPη production. Thus, the action of MT5‐MMP on APP occurs in multiple locations within the cell and is facilitated by site‐specific binding partners. [ABSTRACT FROM AUTHOR]

BACKGROUND: Congenital heart disease (CHD) seriously affects children's health and quality of life, and early detection of CHD can reduce its impact on children's health. Tetralogy of Fallot (TOF) and ventricular septal defect (VSD) are two types of CHD that have similarities in echocardiography. However, TOF has worse diagnosis and higher morality than VSD. Accurate differentiation between VSD and TOF is highly important for administrative property treatment and improving affected factors' diagnoses. OBJECTIVE: TOF and VSD were differentiated using convolutional neural network (CNN) models that classified fetal echocardiography images. METHODS: We collected 105 fetal echocardiography images of TOF and 96 images of VSD. Four CNN models, namely, VGG19, ResNet50, NTS-Net, and the weakly supervised data augmentation network (WSDAN), were used to differentiate the two congenital heart diseases. The performance of these four models was compared based on sensitivity, accuracy, specificity, and AUC. RESULTS: VGG19 and ResNet50 performed similarly, with AUCs of 0.799 and 0.802, respectively. A superior performance was observed with NTS-Net and WSDAN specific for fine-grained image categorization tasks, with AUCs of 0.823 and 0.873, respectively. WSDAN had the best performance among all models tested. CONCLUSIONS: WSDAN exhibited the best performance in differentiating between TOF and VSD and is worthy of further clinical popularization. The purpose of this paper is to reveal the dynamic mass transfer effects between gas and liquid on pressure fluctuations in a space micropump using our proposed computational model of gas–liquid mass transfer. Complex dissolution and evolution processes were applied to achieve accurate dynamic gas–liquid mass transfer predictions in the micropump. The validation of experiments was conducted by measuring the performance characteristics of the micropump, and the mass transfer model was verified by a dissolved oxygen concentration experiment in plug discharge flow. Based on this, four conditions including unsteady single-phase, two-phase without mass transfer, dissolution, and coexisting dissolved–released flows calculations are discussed to clarify the frequency contents, generation reasons, and propagation law. Combined with entropy production analysis, the pressure fluctuation influenced by the dissolution and evolution is illustrated. The results exhibit that the evolution of the gas is located on the head of the long blade suction surface in the impeller. When a unidirectional gas-to-liquid dissolution process occurs, the fluctuating amplitude of the characteristic dominant 5 fn is significantly weakened; otherwise, when dissolution and evolution coexist, the amplitude is significantly promoted as the released gas increase the flow instability. In addition, the distributions of local high entropy production induced by mean and fluctuating velocity gradients overlap that of large mass transfer rates and high amplitude of the mixing frequency in the volute, exhibiting the relationship between entropy production, mass transfer, and flow instability. The current study provides a guidance that the dissolved gases' concentration must be controlled strictly to avoid the evolution of gas for the safety and stability of the space hydraulic system.
