Abstract

The prevalence of fatty liver disease (FLD), a chronic disorder that includes alcoholic liver disease (ALD) and non-alcoholic fatty liver disease (NAFLD) ranges from 1% to 50% and varies with age, gender and occupation in different districts of China. Some studies have shown that FLD especially NAFLD, has a relationship with cardiac disease, metabolic syndrome, dyslipidemia and obesity. To determine the prevalence of FLD and related diseases in the Dehui City of Jilin Province in the Northeastern region of China, we surveyed the population of Dehui City of Jilin Province in China. The objective of this study was to evaluate the prevalence of FLD in relation to different factors, such as age, location, occupations and educational levels, and analyze the risk factors of FLD. The study sample was separated into three groups according to alcohol consumption: non-alcoholic fatty liver disease (NAFLD), alcoholic liver disease (ALD) and suspected alcoholic liver disease (SALD) groups. All subjects (18- 78 years) completed a standard questionnaire, physical examination and hepatic ultrasound examination. Multivariate logistic regression was used to screen risk factors of FLD. The study indicated that the prevalence of FLD was 23.1%, strati ed by NAFLD 14.5%, SALD 5.5% and ALD 3.0%. Abstract

Trehalose‐6‐phosphate synthase (TPS), an enzyme that hydrolyzes two glucose molecules to yield trehalose, plays a pivotal role in various physiological processes. In this study, we cloned the trehalose‐6‐phosphate synthase gene (HvTPS) and investigated its expression patterns in various tissues and developmental stages in Heortia vitessoides Moore (Lepidoptera: Crambidae). HvTPS was highly expressed in the fat body and after pupation or before molting. We knocked down TPS in H. vitessoides by RNA interference and found that 3.0 μg of dsHvTPS resulted in optimal interference at 24 h and 36 h post‐injection and caused a sharp decline in the survival rate during the 5th instar larval–pupal stage and obviously abnormal or lethal phenotypes. Additionally, compared to the controls, TPS activity and trehalose contents were significantly lower and the glucose content was significantly higher 24 h or 36 h after injection with 3.0 μg of dsHvTPS. Furthermore, the silencing of HvTPS suppressed the expression of six key genes in the chitin biosynthesis pathway and one key gene related to lipid catabolism. The expression levels of two genes associated with lipid biosynthesis were upregulated. These results strongly suggest that HvTPS is essential for the normal growth and development of H. vitessoides and provide a reference for further studies of the utility of key genes involved in chitin and lipid biosynthesis for controlling insect development. Abstract

Hydrogen sulfide (H2S) plays antidepressant‐like roles in diabetic rats. However, the underlying mechanisms remain unclear. Brain‐derived neurotropic factor (BDNF), a neurotrophic factor, plays important regulatory roles in depression by its high‐affinity tropomysin‐related kinase B (TrkB) receptor. Autophagy also is implicated in modulation of depression. Previous work confirmed the modulatory roles of H2S in BDNF protein expression and autophagy. Thus, in this study, we explored whether the BDNF‐TrkB pathway mediates the antidepressant‐like effects of H2S in diabetic rats and whether this process is achieved via promoting hippocampal autophagy. We demonstrated that H2S upregulated the expressions of BDNF and p‐TrkB proteins in the hippocampus of streptozotocin (STZ)‐induced diabetic rats. K252a (an inhibitor of BDNF‐TrkB pathway) reversed the antidepressant‐like roles of H2S, as evidenced by the tail suspension, forced swimming, novelty suppressed feeding, and elevated plus‐maze tests. Furthermore, K252a abolished H2S‐promoted hippocampal autophagy in diabetic rats, as evidenced by a decrease in the number of autolysosome, downregulation of Beclin‐1 (a regulator of autophagy in the early stage of the formation of autophagosomal membranes and its level is positively correlated with autophagic activity) expression, and upregulation of P62 (a substrate of autophagic degradation and its level is inversely correlated with autophagic activity) expression, in the hippocampus of rats co‐treated with NaHS and STZ. Taken together, these data indicated that the BDNF‐TrkB pathway mediates the antidepressant‐like roles of H2S in diabetic rats by enhancing hippocampal autophagy. [ABSTRACT FROM AUTHOR]

Abstract

Dual-modal imaging by combining magnetic resonance (MR) and near-infrared (NIR) fluorescence can integrate the advantages of high-resolution anatomical imaging with high sensitivity in vivo fluorescent imaging, which is expected to play a significant role in biomedical researches. Here we report a dual-modality imaging probe (NIR/MR-MSNs) fabricated by conjugating NIR fluorescent heptamethine dyes (IR-808) and MR contrast agents (Gd-DTPA) within highly aminated mesoporous silica nanoparticles (MSNs-NH2). The dual-modality imaging probes NIR/MR-MSNs possess a size of ca. 120 nm. The NIR/MR-MSNs show not only near-infrared fluorescence imaging property with an emission peak at 794 nm, but also highly MRT1relaxivity of 14.54 mM−1 s−1, which is three times more than Gd-DTPA. In vitro experiment reveals high uptake and retention abilities of the nanoprobes, while cell viability assay demonstrates excellent cytocompatibility of the dual-modality imaging probe. After intratumor injection with the NIR/MR-MSNs, MR imaging shows clear anatomical border of the enhanced tumor region while NIR fluorescence exhibits high sensitive tumor detection ability. These intriguing features suggest that this newly developed dual-modality imaging probes have great potential in biomedical imaging.

Abstract

IMPORTANCE: Lymphopenia is common and correlates with poor clinical outcomes in patients with coronavirus disease 2019 (COVID-19). OBJECTIVE: To determine whether a therapy that increases peripheral blood leukocyte and lymphocyte cell counts leads to clinical improvement in patients with COVID-19. DESIGN, SETTING AND PARTICIPANTS: Between February 18 and April 10, 2020, we conducted an open-label, multicenter, randomized clinical trial at 3 participating centers in China. The main eligibility criteria were pneumonia, a blood lymphocyte cell count of 800 per μL (to convert to ×109/L, multiply by 0.001) or lower, and no comorbidities. Severe acute respiratory syndrome coronavirus 2 infection was confirmed with reverse-transcription polymerase chain reaction testing. EXPOSURES: Usual care alone, or usual care plus 3 doses of recombinant human granulocyte colony-stimulating factor (rhG-CSF, 5 μg/kg, subcutaneously at days 0-2). MAIN OUTCOMES AND MEASURES: The primary end point was the time from randomization to improvement of at least 1 point on a 7-category disease severity score. RESULTS: Of 200 participants, 112 (56%) were men and the median (interquartile range [IQR]) age was 45 (40-55) years. There was random assignment of 100 patients (50%) to the rhG-CSF group and 100 (50%) to the usual care group. Time to clinical improvement was similar between groups (rhG-CSF group median of 12 days (IQR, 10-16 days) vs usual care group median of 13 days (IQR, 11-17 days); hazard ratio, 1.28; 95% CI, 0.95-1.71; P = .06). For secondary end points, the proportion of patients progressing to acute respiratory distress syndrome, sepsis, or septic shock was lower in the rhG-CSF group (rhG-CSF group, 2% vs usual care group, 15%; difference, −13%; 95%CI, −21.4% to −5.4%). At 21 days, 2 patients (2%) had died in the rhG-CSF group compared with 10 patients (10%) in the usual care group (hazard ratio, 0.19; 95%CI, 0.04-0.88). At day 5, the lymphocyte cell count was higher in the rhG-CSF group (rhG-CSF group median of 1050/μL vs usual care group median of 620/μL; Hodges-Lehmann estimate of the difference in medians, 440; 95% CI, 380-490). Serious adverse events, such as sepsis or septic shock, respiratory failure, and acute respiratory distress syndrome, occurred in 29 patients (14.5%) in the rhG-CSF group and 42 patients (21%) in the usual care group. CONCLUSION AND RELEVANCE: In preliminary findings from a randomized clinical trial, rhG-CSF treatment for patients with COVID-19 with lymphopenia but no comorbidities did not accelerate clinical improvement, but the number of patients developing critical illness or dying may have been reduced. 采用“交替循环浸泡法(ALIM)”在TiO 2 纳米管管内填充类骨磷灰石, 使磷灰石涂层由层状结构变为嵌入式结构, 并考察填充量与TiO 2 纳米管结构间的关系, 通过模拟体液浸泡实验评价其生物活性. 结果表明, 甘油体系(60V电压)中制备的纳米管填充效果最好; 无定型的TiO 2 纳米管相比锐钛矿相的TiO 2 纳米管更能诱导类骨磷灰石的填充; 采用ALIM于TiO 2 纳米管上填充类骨磷灰石可大大提高其生物活性.

Abstract

Using the third-order WKB approximation, we evaluate the quasinormal frequencies of massless Dirac field perturbation around a Reissner-Nordstrom black hole surrounded by a static and spherically symmetric quintessence. We study the variation of quasinormal frequencies with the quintessential state parameter , the total angular momentum number |k|, the charge Q, and the overtone number n changes, respectively. Moreover, from the results we obtained, we find that the massless Dirac field damps more slowly due to the presence of quintessence.

Abstract

We discuss the entropy of the Garfinkle-Horowitz-Strominger dilaton black hole by using the thin film brickwall model, and the entropy obtained is proportional to the horizon area of the black hole confirming the Bekenstein-Hawking's area-entropy formula. Then, by comparing with the original brick-wall method, we find that the result obtained by the thin film method is more reasonable avoiding some drawbacks, such as little mass approximation, neglecting logarithm term, and taking the term L3 as a contribution of the vacuum surrounding the black hole, and the physical meaning of the entropy is more clearer.
