Abstract

Polycomb group (PcG) proteins are critical chromatin regulators for cell fate control. The mono-ubiquitylation on histone H2AK119 (H2AK119ub1) is one of the well-recognized mechanisms for Polycomb repressive complex 1 (PRC1)-mediated transcription repression. Unexpectedly, the specific H2AK119 deubiquitylation complex composed by additional sex comb-like proteins and BAP1 has also been genetically characterized as Polycomb repressive deubiquitnase (PR-DUB) for unclear reasons. However, it remains a mystery whether and how PR-DUB deficiency affects chromatin states and cell fates through impaired PcG silencing. Here through a careful epigenomic analysis, we demonstrate that a bulk of H2AK119ub1 is diffusely distributed away from promoter regions and their enrichment is positively correlated with PRC1 occupancy. Upon deletion of Asxl2 in mouse embryonic stem cells (ESCs), a pervasive gain of H2AK119ub1 is coincident with increased PRC1 sampling at chromatin. Accordingly, PRC1 is significantly lost from a subset of highly occupied promoters, leading to impaired silencing of associated genes before and after lineage differentiation of Asxl2-null ESCs. Abstract

MYC stimulates both metabolism and protein synthesis, but how cells coordinate these complementary programs is unknown. Previous work reported that, in a subset of small-cell lung cancer (SCLC) cell lines, MYC activates guanosine triphosphate (GTP) synthesis and results in sensitivity to inhibitors of the GTP synthesis enzyme inosine monophosphate dehydrogenase (IMPDH). Here, we demonstrated that primary [MYC.sup.hi] human SCLC tumors also contained abundant guanosine nucleotides. We also found that elevated MYC in SCLCs with acquired chemoresistance rendered these otherwise recalcitrant tumors dependent on IMPDH. Unexpectedly, our data indicated that IMPDH linked the metabolic and protein synthesis outputs of oncogenic MYC. Coexpression analysis placed IMPDH within the MYC-driven ribosome program, and GTP depletion prevented RNA polymerase I (Pol I) from localizing to ribosomal DNA. Furthermore, the GTPases GPN1 and GPN3 were upregulated by MYC and directed Pol I to ribosomal DNA. Constitutively GTP-bound GPN1/3 mutants mitigated the effect of GTP depletion on Pol I, protecting chemoresistant SCLC cells from IMPDH inhibition. GTP therefore functioned as a metabolic gate tethering MYC-dependent ribosome biogenesis to nucleotide sufficiency through GPN1 and GPN3. Abstract

Conventional evaluation of fish freshness based on physiological and biochemical methods was destructive, complicated and costly. In this study, the new model was trained on the eye images of 100 large yellow croakers along with their total volatile basic nitrogen (TVB-N) value as freshness indicators in the storage of nine consecutive days at 4 °C. The experiment was divided into three stages (0–2 days, 3–6 days, and 7–8 days) based on TVB-N value, about 1000 images in each stage were used for freshness classification. A non-destructive and rapid fish freshness detection method based on the eye region images of large yellow croaker was proposed by mathematical modeling. The features of large yellow croaker images were extracted automatically by ResNet-34 structure, and then the key extracted feature was focused on the pupil of the fish eye by mixed attention mechanism. Finally, the features of pupil were used to classify the freshness of large yellow croaker. The results showed the accuracy of the model to classify the fish freshness was reached to 99.4%. The model constructed based on the eye images was non-destructive, and could well monitor and distinguish the freshness of large yellow croakers at different storage stages.

Abstract

Invasive fungal infections, with high morbidity and mortality, have become one of the most serious threats to human health. There are a few kinds of clinical antifungal drugs but large amounts of them are used, so there is an urgent need for a new structural type of antifungal drug. In this study, we carried out three rounds of structural optimisation and modification of the compound YW-01, which was obtained from the preliminary screening of the group, by using the strategy of scaffold hopping. A series of novel phenylpyrimidine CYP51 inhibitors were designed and synthesised. In vitroantifungal testing showed that target compound C6exhibited good efficacy against seven common clinically susceptible strains, which was significantly superior to the clinical first-line drug fluconazole. Subsequently in vitrotests on metabolic stability and cytotoxicity revealed that C6was safe and stable for hepatic microsomal function. Finally, C6warranted further exploration as a possible novel structural type of CYP51 inhibitor.

Abstract

In this study, an accurate tool is provided for the evaluation of the effect of joint motion effect on gait stability. This quantitative gait evaluation method relies exclusively on the analysis of data acquired using acceleration sensors. First, the acceleration signal of lower limb motion is collected dynamically in real-time through the acceleration sensor. Second, an algorithm based on improved dynamic time warping (DTW) is proposed and used to calculate the gait stability index of the lower limbs. Finally, the effects of different joint braces on gait stability are analyzed. The experimental results show that the joint brace at the ankle and the knee reduces the range of motions of both ankle and knee joints, and a certain impact is exerted on the gait stability. In comparison to the ankle joint brace, the knee joint brace inflicts increased disturbance on the gait stability. Compared to the joint motion of the braced side, which showed a large deviation, the joint motion of the unbraced side was more similar to that of the normal walking process. In this paper, the quantitative evaluation algorithm based on DTW makes the results more intuitive and has potential application value in the evaluation of lower limb dysfunction, clinical training and rehabilitation. 249. English language development in P.R. China : a study of the impact of some learner-internal and learner-external factors

Abstract

English learning in China belongs in what is called \foreign language\" learning which has so far been little studied in the field of second language acquisition research. This study investigated within a single theoretical framework the English language development of Chinese university students as revealed from their oral production and how some learner-internal and learner-external factors contributed to this development. The oral English development was investigated by examining how the subjects used the English they learnt in the classroom to express the discourse functions of \"foreground\" and \"background\" and the concept of \"past time\" in oral narrative. The learner-internal factors considered were: the subjects' attitudes both in relation to English learning and to the classroom learning environment their motivations for learning English and their learning strategies. The learner-external factors under investigation were: the subjects' classroom learning experiences and their out-of-class contact with English. Based on some representative theories of second language acquisition and relevant empirical studies a conceptual framework was first established which delineated the possible relationships between the chosen factors and their relevant concepts. Data for the study were collected in an 11-month period from 20 students majoring in English in Foreign Languages Department of Fujian Teachers University in China. The subjects were chosen by random sampling from 98 of the 109 students enrolled in 1989. The data comprised (i) three administrations of four types of questionnaires which provided information concerning the subjects' attitudes motivations learning strategies and out-of-class contacts with English; (ii) the subjects' performance of a metalinguistic judgment test; and (iii) orthographic transcriptions of the subjects' speech elicited at an interval of three to four weeks two narratives for each subject on each of the 14 occasions. Data analyses reveal the following main findings. First discourse functions had selective impact on the subjects' choice of linguistic features to express them on their expression of the notion \"past time\" and on their adoption of self-corrections and communication strategies. Second the subjects' oral English development can be described linguistically and non-linguistically. Linguistically the development was reflected in the subjects' growing ability to use more types of linguistic features. Non-linguistically the development manifested itself in (i) an ability to break chronological order of the events described in oral narratives (ii) a growing desire to be both conceptually and linguistically accurate in oral production as indicated by the use of irregular verbs in past tense after a growing number of auxiliaries and inflectional forms of the link verb \"to be\" and by the self-correction of more types of linguistic features and (iii) a growing ability to cope with communication problems. Third the subjects' intrinsic interests in English-speaking people and in learning English appeared to be channeled by the knowledge-oriented language instruction into the adoption of the type of learning strategies which enabled them to extend only the knowledge about the target language. The lack of oral practice both in and out of the classroom resulted in a great gap between a highly analytical knowledge about the target language and an under-developed procedural knowledge. The under-developed procedural knowledge prevented the subjects from engaging in fluent oral communication. The possible dissatisfaction with their own oral learning outcomes appeared to prevent the subjects from further participating in oral practice."
