207. Structural insights into variation of intramolecular acid site using phase transition experiments and computation.

Abstract

Mass transfer from liquid to gas phase is a crucial process in chemical engineering. Due to the heterogeneity and dynamic nature of the phase transition process, monitoring the structural changes of compounds during this process is challenging. In this work, a combining of the mass spectrometry‐based method and quantum chemistry calculations was used to study the structural changes during liquid‐to‐gas phase transitions. Electrospray is used to induce the transition of phenolic acid molecules from the liquid phase to the gas phase. The deprotonated structures were captured and separated using ion mobility mass spectrometry. Deprotonated structures helped to identify the most acidic functional group within the molecule. These deprotonated structures were confirmed by collision‐induced dissociation and molecular collision cross‐section measurements. Quantum chemistry calculations revealed the mechanisms about the variation of intramolecular acid site and "kinetic trapping." Three liquid‐to‐gas phase transformation models were proposed, demonstrating that the variation in free energy among different deprotonated structures governs the thermodynamic/kinetic process of the liquid‐to‐gas phase conversion. Abstract

Background: Parkinson's disease (PD) is a common neurodegenerative disorder. Microglia‐mediated neuroinflammation has emerged as an involving mechanism at the initiation and development of PD. Activation of adenosine triphosphate (ATP)‐sensitive potassium (KATP) channels can protect dopaminergic neurons from damage. Sodium butyrate (NaB) shows anti‐inflammatory and neuroprotective effects in some animal models of brain injury and regulates the KATP channels in islet β cells. In this study, we aimed to verify the anti‐inflammatory effect of NaB on PD and further explored potential molecular mechanisms. Methods: We established an in vitro PD model in BV2 cells using 1‐methyl‐4‐phenylpyridinium (MPP+). The effects of MPP+ and NaB on BV2 cell viability were detected by cell counting kit‐8 assays. The morphology of BV2 cells with or without MPP+ treatment was imaged via an optical microscope. The expression of Iba‐1 was examined by the immunofluorescence staining. The intracellular ATP content was estimated through the colorimetric method, and Griess assay was conducted to measure the nitric oxide production. The expression levels of pro‐inflammatory cytokines and KATP channel subunits were evaluated by reverse transcription–quantitative polymerase chain reaction and western blot analysis. Results: NaB (5 mM) activated the KATP channels through elevating Kir6.1 and Kir6.1 expression in MPP+‐challenged BV2 cells. Both NaB and pinacidil (a KATP opener) suppressed the MPP+‐induced activation of BV2 cells and reduced the production of nitrite and pro‐inflammatory cytokines in MPP+‐challenged BV2 cells. Conclusion: NaB treatment alleviates the MPP+‐induced inflammatory responses in microglia via activation of KATP channels. [ABSTRACT FROM AUTHOR]

Abstract

Continuous cropping is the main form of cultivation in Chinese agriculture. The bacterial community plays an important role in maintaining the healthy growth of plants. However, there are few reports on the composition and dynamics of the bacterial community structure under continuous cropping of Lonicera japonica Thunb. High-throughput sequencing was used to monitor the variation in the soil bacterial community structure of different monocropping years of Lonicera japonica Thunb., as well as the correlation between soil characteristics and bacterial community. Meanwhile, antagonistic bacteria for Fusarium oxysporum pathogens were isolated and functionally verified by culture-dependent techniques and pot experiments. Bacterial community diversity and structure changed significantly with the increase in the years of Lonicera japonica Thunb. succession. However, some beneficial bacteria, such as Bacillus and Nitrosospira, were gradually depleted. The complexity of the bacteria co-occurrence networks decreased with increasing years of cropping. FAPROTAX-based functional prediction showed that the abundance of genes related to carbon, nitrogen, sulfur metabolism and chitinlysis were reduced with the extended crop succession. Furthermore, the three Bacillus strains that were strongly antagonistic toward Fusarium oxysporum and the pot experiment demonstrated they significantly promoted Lonicera japonica Thunb. growth. Abstract

In harsh operating environments such as mines and gas stations, intrinsically safe power converters are required for electronics devices. Reliable communication is the key ensuring good performance of monitoring and coordinated control for intrinsically safe converters. This paper proposes a power and signal dual modulation (PSDM)‐based communication method for intrinsically safe parallel‐connected Buck converters. The bidirectional communication between two parallel Buck converters is realized by combining the frequency shift keying (FSK) and phase shift keying (PSK) methods. In addition, switching frequencies are employed to identify the signals received from the power converters interfacing different sources. According to the intrinsic safety requirement, inductors and capacitors are redesigned based on the modulated voltage ripples during communication. Simulations and prototype experiments are performed to verify the feasibility of the proposed bidirectional communication for intrinsically safe converters. The amplitude of voltage ripples modulated for carrying data is restricted to less than 1.5% of the bus voltage. The transmission speed can be achieved from 0.5 to 2kbps depending on the adopted switching frequencies. The demodulation delay is less than 5% duration of one bit of transmitted data. Abstract

Long‐term patient and graft survival has been achieved in organ transplantation but at the expense of toxic side effects that are associated with long‐term use of nonspecific immunosuppressive drugs. Discovering new regulators of dendritic cells is the key for development of an ideal treatment to prevent immune rejection. We hypothesized that knockdown of circMAP2K2 induces immunosuppressive DCs and that treatment with circMAP2K2 silenced‐DCs can prevent alloimmune rejection. DCs were cultured and transfected with siRNA for circMAP2K2. circMAP2K2 levels were measured by qRT‐PCR. DC's maturation and immune function were assessed by flow cytometry and mixed lymphocyte reactions. The function of circMAP2K2 was illustrated by a series of RIP and IP. The therapeutics of engineered DCs was tested in a mouse heart transplantation model. We found that circMAP2K2 was highly expressed in mature DCs. Knockdown of circMAP2K2 reduced expression of MHCII, CD40 and CD80, attenuated the ability of DCs to activate allogeneic naïve T cells, and enhanced CD4+CD25+FOXP3+ regulatory T cells (Treg). circMAP2K2‐induced immunosuppressive DCs by interacting with SENP3. Treatment with circMAP2K2‐knockdown DCs attenuated alloimmune rejection and prolonged allograft survival in a murine heart transplantation model. The immune suppression induced in vivo was donor‐antigen specific. In conclusion, knockdown of circMAP2K2 can induce immunosuppressive DCs which are able to inhibit overactive immune response, highlighting a new promising therapeutic approach for immune disorder diseases.
