Common thyme (Thymus vulgaris L., Lamiaceae) is regarded as one of the most important aromatic plant used both as a spice and medicine. The aim of this work was to investigate the yield and quality of common thyme grown for a fresh herb, cultivated in the organic farming system, within the temperate climate in Poland. Two methods of cultivation were applied: open field and foil tunnel. Three successive cuts of herb were possible to obtain during vegetation season, from the middle of July until the end of September. The raw material was analyzed for the content of essential oil (by hydrodestillation), phenolic acids and flavonoids according to Polish pharmacopeia (PP 6th). The composition of essential oil was determined by gas chromatography-mass spectrometry (GC-MS). Sensory evaluation was carried out by quantitative descriptive analysis (QDA) with a trained panelist. The yield of fresh herb was visibly higher when regards cultivation under foil tunnel in comparison to open field, reaching up to 6.74 kg FW × 10 m2. The content of essential oil as well as the percentage share of thymol (a dominant constituent, achieving up to 57.08%) decreased with consecutive herbal cuts. The opposite result was observed for phenolic compounds (phenolic acids and flavonoids). It was shown that the amount of flavonoids was similar in both variants of cultivation, while phenolic acids were accumulated at higher level in the open field conditions. Sensory analysis indicated on slight differences in odor and taste attributes, between samples of fresh herb collected from open field and foil tunnel.

The purpose of the mechanical root canal preparation is to clean it and give it the right shape. The preparation should be carried out in a way that maintains the original curvature and initial orientation of the apical end. Insufficient root canal preparation may prevent effective chemical decontamination and obturation to the full working length. The study aims to evaluate the shaping ability and effectiveness of the NiTi rotary and reciprocating endodontic instruments, compared to standard hand files using magnetic resonance imaging based on spin echo. Material and methods: A comparative study of severely curved root canals’ shaping abilities using three NiTi systems and K-type hand files was performed, with 40 training “endo-blocks” presenting with “L-shaped” canal. The root canal topography and geometry “before” and “after” mechanical preparation obtained by the magnetic resonance imaging based on the spin echo was used. The main measurement was made using the RARE sequence, with slice thickness reduced to 100 micrometers. In order to improve the signal-to-noise ratio, NA = 25 was used. To minimize the measurement time, the field of view was limited to a cuboid 17 × 16 × 3 mm, with a resolution of 33 × 31 × 100 micrometers. Each 3D image consisted of 512 × 512 × 30 voxels. The imaging plane has been selected in such a way as to fully illustrate the course of curvature of the model root canal. For TR = 5 s and effective TE = 36 ms, TA was 1.5 h. Measurements were performed twice, before and after the preparation of endo-blocs with a selected type of endodontic tools. Results: The use of rotary NiTi instruments caused a substantial alteration in the curvature topography and angle of the canals and change in the curvature length. The substantial discrepancy was observed during the preparation of simulated root canals with the reciprocating instruments and the use of WaveOne files led to the largest volume variation. No dependence between the amount of material removed and the measured intracanal side was observed when NiTi instruments were used. Preparation with hand K-type files revealed a relationship between the measuring side and the amount of material removed, with the lower values obtained for the canals’ internal curvatures. Conclusions: All the studied endodontic instruments allow a safe preparation of curved root canals in simulating in vivo conditions. The abbreviation of original root canals topography does not seem to be significantly altered following mechanical preparation of simulated, severely curved root canals. The spin echo-based magnetic resonance imaging technique can be utilized for visualization of the internal topography of the root’s canals in vitro before and after their mechanical preparation in in vitro conditions. In the future, magnetic resonance microscopy may become a diagnostic tool supporting the work of a clinician.

The purpose of the study was to determine the differences between two subspecies: O. vulgare L. subsp. hirtum (Link) Ietswaart (Greek oregano) and O. vulgare L. subsp. vulgare (common oregano) growing in cultivation conditions within temperate climate of Central Europe. The characteristic of the subspecies was undertaken in terms of selected morphological parameters and the quality of the raw material. The herb of both subspecies was evaluated on the content and composition of essential oil by hydrodistillation followed by GC-MS and GC-FID (gas chromatography coupled with mass spectrometry and flame ionization detector), the total content of phenolic acids (according to PP 6th ed.) and the content of rosmarinic acid (by HPLC). The sensory evaluation (QDA) was performed, as well. Greek oregano was distinguished by visibly higher number of glandular trichomes on the leaves (up to 4.85 per 1 mm2) followed by higher content of essential oil in the herb (up to 3.36 g × 100 g−1 DW) in comparison to common oregano. Based on the essential oil composition, Greek oregano was classified as mixed carvacrol/γ-terpinene chemotype, while common oregano as mixed sabinyl/cymyl type rich in sesquiterpenes. Greek oregano was also characterized by higher total content of phenolic acids (up to 6.16 g × 100 g−1 DW) and rosmarinic acid (up to 6787.2 mg × 100 g−1 DW) than common oregano. Essential oil content reached the maximum at the beginning of blooming (common oregano) and at the full blooming stage (Greek oregano). In turn, the amount of phenolic acids followed by rosmarinic acid was the highest at the beginning of seed-setting stage, in the case of both subspecies. The differences between subspecies concerning chemical composition (especially essential oil) were reflected in the sensory attributes, where both odor and taste notes were found at higher level for Greek oregano. Results of our work indicate that Greek oregano is well adapted to grow in the temperate zone conditions. Such adaptation was reflected mainly in the satisfied yield and maintaining characters typical for the Mediterranean plant, e.g., a high essential oil content followed by high carvacrol share, traits the most important from practice viewpoint.

The application of the Three-Dimensional Ultra-Short Echo Time (3D UTE)pulse sequence at a high magnetic field for visualization of the distribution of 19F loaded theranostic core-shell nanocapsules with Nafion® (1,1,2,2-tetrafluoroethene; 1,1,2,2-tetrafluoro-2- [1,1,1,2,3,3-hexafluoro-3-(1,2,2-trifluoroethenoxy)propan-2-yl] oxyethanesulfonic acid) incorporated into the shell is presented. The nanocarriers were formed via the layer-by-layer technique with biodegradable polyelectrolytes: PLL (Poly-L-lysine), and with Nafion®: polymer with high 19F content. Before imaging, an MR (magnetic resonance) spectroscopy and T1 and T2 measurements were performed, resulting in values of T2 between 1.3 ms and 3.0 ms, depending on the spectral line. To overcome limitations due to such short T2, the 3D UTE pulse sequence was applied for 19F MR imaging. First Nafion® solutions of various concentrations were measured to check the detection limit of our system for the investigated molecule. Next, the imaging of a phantom containing core-shell nanocapsules was performed to assess the possibility of visualizing their distribution in the samples. Images of Nafion® containing samples with SNR ≥ 5 with acquisition time below 30 min for 19F concentration as low as 1.53 × 10−2 mmol 19F/g of sample, were obtained. This is comparable with the results obtained for molecules, which exhibit more preferable MR characteristics.

Common valerian (Valeriana officinalis L.) is one of the most important medicinal plants, with a mild sedative, nervine, antispasmodic and relaxant effect. Despite a substantial number of studies on this species, the genetic diversity and population structure have not yet been analyzed. Here, we use a next-generation sequencing-based Diversity Array Technology sequencing (DArT-seq) technique to analyze Polish gene bank accessions that originated from wild populations and cultivars. The major and, also, the most astounding result of our work is the low level of observed heterozygosity of individual plants from natural populations, despite the fact that the species is widespread in the studied area. Inbreeding in naturally outcrossing species such as valerian decreases reproductive success. The analysis of the population structure showed the potential presence of a metapopulation in the central part of Poland and the formation of a distinct gene pool in the Bieszczady Mountains. The results also indicate the presence of the cultivated gene pool within wild populations in the region where the species is cultivated for the needs of the pharmaceutical industry, and this could lead to structural and genetic imbalances in wild populations.

The article presents calculations and power flow of a real virtual power plant (VPP), containing a fragment of low and medium voltage distribution network. The VPP contains a hydropower plant (HPP), a photovoltaic system (PV) and energy storage system (ESS). The purpose of this article is to summarize the requirements for connection of generating units to the grid. Paper discusses the impact of the requirements on the maximum installed capacity of distributed energy resource (DER) systems and on the parameters of the energy storage unit. Firstly, a comprehensive review of VPP definitions, aims, as well as the characteristics of the investigated case study of the VPP project is presented. Then, requirements related to the regulation, protection and integration of DER and ESS with power systems are discussed. Finally, investigations related to influence of DER and ESS on power network condition are presented. One of the outcomes of the paper is the method of identifying the maximum power capacity of DER and ESS in accordance with technical network requirements. The applied method uses analytic calculations, as well as simulations using Matlab environment, combined with real measurement data. The obtained results allow the influence of the operating conditions of particular DER and ESS on power flow and voltage condition to be identified, the maximum power capacity of ESS intended for the planed VPP to be determined, as well as the influence of power control strategies implemented in a PV power plant on resources available for the planning and control of a VPP to be specified. Technical limitations of the DER and ESS are used as input conditions for the economic simulations presented in the accompanying paper, which is focused on investigations of economic efficiency.

Common thyme is regarded as one of the most important culinary plants. The purpose of the work was to determine the intraspecific variability of common thyme with respect to morphological and chemical characters including the content and composition of essential oil and phenolic compounds in the herb. The objects of the study were 12 clones, vegetatively multiplied from randomly selected individual plants of cultivar ‘Standard Winter’. The morphological observations and harvest of raw materials were carried out in the first year plants’ vegetation. The highest differences between clones were on fresh and dry weight of herb (CV = 0.38 and 0.36, respectively), width of leaves (CV = 0.21), and density of glandular trichomes on the abaxial surface of leaves (CV = 0.29). Examined clones were also differentiated as to the chemical features. Essential oil content (performed by hydrodestillation) and composition (by GC-MS and GC-FID) were determined and they ranged from 2.10 to 4.38 g × 100 g−1 DW. Here, thymol, γ-terpinen, and p-cymen were the dominant compounds. Clone no. 4 was distinctive as to the highest content of essential oil followed by the highest share of thymol (54.59%). The total content of phenolic acids and flavonoids (determined according to PPh 6th) also differed among clones (CV = 0.38 and 0.36, respectively). Using a validated HPLC-DAD method, the following compounds were identified: caffeic, rosmarinic, p-coumaric acids, luteolin 7-O-glucoside, naryngenin, and (−)-epicatechin. Here, rosmarinic acid followed by luteolin 7-O-glucoside were present in the highest amounts (611.47–2675.59 and 46.77–325.11 mg × 100 g−1 DW, respectively). The highest differences between clones were the contents of p-coumaric acid (CV = 0.59), luteolin 7-O-glucoside (CV = 0.50) and rosmarinic acid (CV = 0.40). Such a high range of variability can provide problems with raw material standardization. Nevertheless, it opens possibilities for breeders, whereas individual plants/clones may become valuable components for breeding.

The aim of the study was to assess the effects of Melittis melissophyllum shading on its development and accumulation of phenolics. Their content (verbascoside, apiin, luteolin-7-O-glucoside, coumarin, 3,4-dihydroxycoumarin, o-coumaric acid 2-O-glucoside as well as o-coumaric, p-coumaric, chlorogenic, caffeic, ferulic and cichoric acid) was determined in the herb using HPLC-DAD. The results showed that the content of abovementioned flavonoids and phenolic acids was highest in plants grown under full sunlight. On the other hand, a higher content of coumarin was observed in shaded plants, especially after the seed-setting stage. A similar tendency was noted for the amount of chlorophyll a and b. The content of hydrogen peroxide and malondialdehyde, the activity of polyphenol oxidase and catalase and the antioxidant capacity of plant extracts (measured using DPPH, ABTS and FRAP assays) were found to be the highest in the plants grown in full sunlight. However, the plants grown in moderate (30%) shade were found to thrive best.

The phytochemical diversity of Melittis melissophyllum was investigated in terms of seasonal changes and age of plants including plant organs diversity. The content of phenolics, namely: coumarin; 3,4-dihydroxycoumarin; o-coumaric acid 2-O-glucoside; verbascoside; apiin; luteolin-7-O-glucoside; and o-coumaric; p-coumaric; chlorogenic; caffeic; ferulic; cichoric acids, was determined using HPLC-DAD. Among these, luteolin-7-O-glucoside, verbascoside, chlorogenic acid, and coumarin were the dominants. The highest content of flavonoids and phenolic acids was observed in 2-year-old plants, while coumarin in 4-year-old plants (272.06 mg 100 g–1 DW). When considering seasonal changes, the highest content of luteolin-7-O-glucoside was observed at the full flowering, whereas verbascoside and chlorogenic acid were observed at the seed-setting stage. Among plant organs, the content of coumarin and phenolic acids was the highest in leaves, whereas verbascoside and luteolin-7-O-glucoside were observed in flowers. The composition of essential oil was determined using GC-MS/GC-FID. In the essential oil from leaves, the dominant was 1-octen-3-ol, whilst from flowers, the dominant was α-pinene.
