PurposeTo investigate the impact of N6-methyladenosine- (m6A) and neutrophil extracellular traps- (NETs) related lncRNAs (MNlncRNAs) on the prognosis of hepatocellular carcinoma (HCC).MethodsWe collected m6A and NETs-related genes from published studies. We identified the MNlncRNAs by correlation analysis. Cox regression and the least absolute selection operator (LASSO) method were used to select predictive MNlncRNAs. The expressions of predictive MNlncRNAs were detected by cell and tissue experiments. Survival, medication sensitivity, and immunological microenvironment evaluations were used to assess the model’s prognostic utility. Finally, we performed cellular experiments to further validate the model’s prognostic reliability.ResultsWe obtained a total of 209 MNlncRNAs. 7 MNlncRNAs comprised the prognostic model, which successfully stratifies HCC patients, with the area under the curve (AUC) ranging from 0.7 to 0.8. In vitro tests confirmed that higher risk patients had worse prognosis. Risk score, immunological microenvironment, and immune checkpoint gene expression were all significantly correlated with each other in HCC. In the group at high risk, immunotherapy could be more successful. Cellular assays confirmed that HCC cells with high risk scores have a higher proliferation and invasive capacity.ConclusionThe MNlncRNAs-related prognostic model aided in determining HCC prognosis, revealing novel therapeutic options, notably immunotherapy.

204. Sanhuang decoction inhibits autophagy of periodontal ligament fibroblasts during orthodontic tooth movement by activating PI3K-Akt-mTOR pathway

Background: Orthodontic tooth movement (OTM) is a typical treatment that corrects malaligned teeth by applying mechanical forces. However, mechanical overload often leads to damage of PDL fibroblasts. Sanhuang decoction (SHD) is commonly used to inhibit inflammation and oxidative stress. However, the mechanism of SHD for OTM treatment is still unclear. Therefore, this study attempts to explore the underlying mechanism through relevant experiments. Methods: In the present paper, we established a OTM rat model and further explored the effects of SHD on the PDL of OTM rats. The OTM model and effects of SHD were determined by micro-CT, and the PDL pathological changes, PDL width and capillaries in PDL were observed by H&E staining. Subsequently, the ROS levels in PDL was determined using flow cytometry analysis with DCFH-DA staining, MDA contents and antioxidative enzymes activities were also measured using commercial kits. Furthermore, the autophagy of PDL fibroblasts and proteins in the PI3K/Akt/mTOR pathway were detected using immunoluminescence, qPCR and western blotting assays. Results: The results showed SHD treatment can alleviate the decrease of PDL cells and capillaries induced by OTM, and improve the MDA and ROS levels in PDL, as well as enhance the activities of SOD and GSH-Px. Further experiments indicated SHD decreased the autophagy levels of PDL fibroblasts via promoting the phosphorylation levels of mTOR, PI3K and Akt proteins. Conclusion: SHD inhibited autophagy of periodontal ligament fibroblasts during orthodontic tooth movement by inhibiting oxidative stress via activating PI3K-Akt-mTOR pathway. Our present findings suggested SHD treatment would be useful for management of the possible disorders occurs in orthodontic tooth movement therapy.

IntroductionChanges in the gut microbiota of patients with partial small intestinal obstruction (PSIO) have not been widely clarified. We aimed to explore bacterial diversity in a PSIO mouse model.MethodsA PSIO mouse model was established using male C57BL/6 mice, and feces samples from the distal ileum and ileum epithelium tissues were collected. MiSeq sequencing of the 16S rRNA gene was conducted to characterize microbiota diversity and composition. RNA sequencing for differences in transcriptomic programming of the ileum tissue was performed between the PSIO and (Control) Ctrl groups.ResultsBacterial diversity in the PSIO group was significantly lower than that in the controls. Pseudomonadota was predominant in the feces of the PSIO group. Unclassified_Muribaculaceae (p = 0.008) and Akkermansia (p = 0.007) were more abundant in the Ctrl group than those in the PSIO group. Furthermore, Escherichia_Shigella (p = 0.008) was more predominant in the feces of the PSIO group. The Kyoto Encyclopedia of Genes and Genomes pathways related to metabolism were depleted in the PSIO group. Pathways associated with intestinal fibrosis, including extracellular matrix-receptor interaction, focal adhesion, phosphoinositide 3-kinase (PI3K)-Akt signaling pathway and transforming growth factor (TGF)-beta signaling pathway, which were enriched in ileum epithelial tissue in the PSIO group.ConclusionPSIO can lead to changes in the predominant intestinal bacterial groups. Depleted functional profiles of the gut microbiota were identified in the PSIO group. Functional pathways associated with intestinal fibrosis were activated by PSIO. The potential regulation by the microbiota needs to be explored in the future.

207. Empathy and bystander helping behavior in cyberbullying among adolescents: the mediating role of internet moral judgment and the moderating role of internet self-efficacy

IntroductionCyberbullying poses a significant challenge among adolescents. If bystanders stand up and help victims, their helping behavior may be able to reduce the frequency and negative impact of cyberbullying. This study investigates the association of empathy, internet moral judgment, and internet self-efficacy with bystander helping behavior among adolescents, building upon the empathy-altruism hypothesis, bystander intervention model, and dual-process model of morality.MethodsA sample of 919 Chinese adolescents from 3 schools in Hunan, Jiangxi and Guangdong provinces completed the Basic Empathy Scale, Internet Moral Judgment Questionnaire, Internet Self-Efficacy Questionnaire and Styles of Bystander Intervention Scale. And we constructed a moderated mediation model to examine the relationship between empathy and bystander helping behavior in cyberbullying and assessed the mediating role of internet moral judgment and the moderating role of internet self-efficacy.ResultsOur findings revealed a significant positive correlation between empathy and bystander helping behavior in cyberbullying. Internet moral judgment mediated the relationship between empathy and helping behavior, whereas internet self-efficacy moderated the latter half of the mediation pathway. Specifically, the association between internet moral judgment and helping behavior was stronger for bystanders with higher levels of internet self-efficacy compared with those that have lower levels.DiscussionThese results further deepen our understanding of the mechanisms involved in bystander helping behavior in cyberbullying, thus providing a basis for future interventions to encourage more helping actions from bystanders during cyberbullying incidents.

BackgroundThe presence of parental arterial disease (PAD) is correlated with the outcomes of patients with a single subcortical infarction (SSI). Due to the relatively low incidence of PAD, the predictors of outcomes seem to be limited for SSI patients without PAD. This study aims to investigate the association between asymptomatic intracranial atherosclerotic stenosis (aIAS) and outcomes in patients with SSI and in the subgroup without PAD.MethodsPatients with SSI were consecutively enrolled. aIAS referred to a stenosis of ≥50% in intracranial arteries irrelevant to SSI by using magnetic resonance angiography. A poor outcome refers to a modified Ranking Scale >2 points at discharge.ResultsIn total, 298 participants were enrolled. The presence of aIAS could predict a poor outcome for all SSI patients [adjusted relative risk (aRR) = 2.14, 95% confidence interval (CI) = 1.17–3.93, p = 0.014] and in the subgroup without PAD (aRR = 3.12, 95% CI = 1.47–6.62, p = 0.003), but not in the subgroup with PAD. Compared with participants with neither aIAS nor PAD, the risk of a poor outcome increased approximately 2-fold in those with aIAS only (aRR = 2.95, 95% CI = 1.55–5.60, p = 0.001) and in those with concomitant aIAS and PAD (aRR = 3.10, 95% CI = 1.62–5.95, p = 0.001).ConclusionThe presence of aIAS is a predictor of a poor outcome in SSI patients, especially in those without PAD.

Summary: Drug-target interactions (DTIs) prediction is an important step in drug discovery. As traditional biological experiments or high-throughput screening are high cost and time-consuming, many deep learning models have been developed. Overfitting must be avoided when training deep learning models. We propose a simple framework, called OverfitDTI, for DTI prediction. In OverfitDTI, a deep neural network (DNN) model is overfit to sufficiently learn the features of the chemical space of drugs and the biological space of targets. The weights of trained DNN model form an implicit representation of the nonlinear relationship between drugs and targets. Performance of OverfitDTI on three public datasets showed that the overfit DNN models fit the nonlinear relationship with high accuracy. We identified fifteen compounds that interacted with TEK, a receptor tyrosine kinase contributing to vascular homeostasis, and the predicted AT9283 and dorsomorphin were experimentally demonstrated as inhibitors of TEK in human umbilical vein endothelial cells (HUVECs).

Cadmium (Cd) exposure is a risk factor for endothelial dysfunction and cardiovascular disease. Ferroptosis is a type of cell death that relies on lipid peroxidation. Whether ferroptosis acts in Cd-induced vascular endothelial damage and the underlying mechanisms remain unclear. Herein, we found that Cd resulted in ferroptosis of vascular endothelial cells (ECs) in vivo and in vitro. In the visualized zebrafish embryos, Cd accumulated in vascular ECs, ROS and lipid peroxidation levels were increased, and the oxidoreductase system was disturbed after exposure. Moreover, Cd decreased Gpx4 in ECs and caused smaller mitochondria with increased membrane density. Accompanied by ferroptosis, the number of ECs and the area of the caudal venous plexus in zebrafish embryos were reduced, and the survival rate of HUVECs decreased. These effects were partially reversed by ferrostatin-1 and aggravated by erastin. Mechanistically, an excessive increase in Heat Shock Protein 70 (Hsp70) was identified by transcriptomics after Cd exposure. Inhibition of Hsp70 by VER-155008 or siRNA ameliorated Cd-induced ferroptosis, thereby alleviating endothelial injury. Furthermore, Hsp70 regulated Cd-induced ferroptosis by targeting multiple targets, including Gpx4, Fth1, Nrf2 and Acsl4. Our findings provide a new approach to investigating the endothelial damage of Cd and indicate that regulation of Hsp70 is an important target for alleviating this process.

As the additive manufacturing provides an advanced method for fabricating polymeric composite components in thermal engineering applications, the thermal contact conductance (TCC) between the contact composites can become an important problem. In order to understand the interfacial heat transfer mechanism between thermoplastic composites, TCC between additively manufactured carbon fiber reinforced polyamide 12 (CF/PA12) composites was experimentally studied with steady-state measurements in this work. By considering the material surface microstructure, thermal and mechanical properties, the effects of interface temperature, contact pressure, surface morphology, carbon fiber (CF) content and orientation in composites on TCC were analyzed. The results show that the TCC is dependent on the actual contact area and the thermal conduction path inside the material. In general, high surface roughness decreases the TCC because of the reduction of contact area; while it is the opposite of temperature and pressure. The CF distribution affects the internal thermal conduction path and surface roughness, leading to the variations in TCC.

ObjectiveTo explore the difference in CT values between pulmonary thromboembolism and postmortem clot in postmortem CT pulmonary angiography (CTPA) to further improve the application value of virtual autopsy.MethodsPostmortem CTPA data with the definite cause of death from 2016 to 2019 were collected and divided into pulmonary thromboembolism group (n=4), postmortem clot group (n=5), and control group (n=5). In order to improve the anti-corrosion performance of epoxy coatings, modified nano-ZrO2/Zn-Al-C epoxy resin composite coatings were prepared by using γ-aminopropyltriethoxysilane (KH-550) to improve the agglomeration phenomenon of nano-ZrO2 particles in combination with Zn-Al-C alloy powder. X-ray diffraction (XRD), Fourier infrared spectroscopy (FT-IR) and scanning electron microscopy (SEM) were used to characterize the structural changes of nano-ZrO2 before and after the modification. With electrochemical AC impedance spectroscopy (EIS) and polarization curves (Tafel), the corrosion behavior of the composite coatings in simulated geothermal water environment was analyzed. The results show that the nano-ZrO2 surface is successfully grafted with KH-550; the number of corrosion scaling products on the surface of the composite coating is significantly reduced compared with the original sheet after full immersion in simulated geothermal water for 720 h; the self-corrosion current density is 6.113×10-10 (A·cm-2), and the electrochemical performance is good. The modified epoxy composite coating effectively utilizes the superior properties of nanomaterials and metallic materials, and its hardness and corrosion resistance are improved significantly, which provides a reference for the long-term use of epoxy coating in geothermal water environment.

Abstract Impaired protein N-glycosylation leads to the endoplasmic reticulum (ER) stress, which triggers adaptive survival or maladaptive apoptosis in renal tubules in diabetic kidney disease (DKD). Therapeutic strategies targeting ER stress are promising for the treatment of DKD. Here, we report a previously unappreciated role played by ENTPD5 in alleviating renal injury by mediating ER stress. We found that ENTPD5 was highly expressed in normal renal tubules; however, ENTPD5 was dynamically expressed in the kidney and closely related to pathological DKD progression in both human patients and mouse models. Overexpression of ENTPD5 relieved ER stress in renal tubular cells, leading to compensatory cell proliferation that resulted in hypertrophy, while ENTPD5 knockdown aggravated ER stress to induce cell apoptosis, leading to renal tubular atrophy and interstitial fibrosis. Mechanistically, ENTPD5-regulated N-glycosylation of proteins in the ER to promote cell proliferation in the early stage of DKD, and continuous hyperglycemia activated the hexosamine biosynthesis pathway (HBP) to increase the level of UDP-GlcNAc, which driving a feedback mechanism that inhibited transcription factor SP1 activity to downregulate ENTPD5 expression in the late stage of DKD. This study was the first to demonstrate that ENTPD5 regulated renal tubule cell numbers through adaptive proliferation or apoptosis in the kidney by modulating the protein N-glycosylation rate in the ER, suggesting that ENTPD5 drives cell fate in response to metabolic stress and is a potential therapeutic target for renal diseases.

Abstract Background The triglyceride-glucose (TyG) index and triglyceride to high-density lipoprotein cholesterol (TG/HDL-C) ratio, two simple surrogate indicators of insulin resistance, have been demonstrated to predict cardiovascular disease (CVD). However, very few studies have investigated their associations with CVD in European populations. Methods A total of 403,335 participants from the UK Biobank with data for TyG index and TG/HDL-C ratio and free from CVD at baseline were included. Cox models were applied to evaluate the association between TyG index and TG/HDL-C ratio and incident CVD. Mediation analyses were performed to evaluate the contribution of prevalent diabetes, hypertension, and dyslipidemia to observed associations. Results During a median follow-up of 8.1 years, 19,754 (4.9%) individuals developed CVD, including 16,404 (4.1%) cases of CHD and 3976 (1.0%) cases of stroke. The multivariable-adjusted hazard ratios of total CVD in higher quartiles versus the lowest quartiles were 1.05, 1.05, and 1.19, respectively, for TyG index, and 1.07, 1.13, and 1.29, respectively, for TG/HDL-C ratio. There were significant trends toward an increasing risk of CVD across the quartiles of TyG index and TG/HDL-C ratio. In mediation analyses, dyslipidemia, type 2 diabetes, and hypertension explained 45.8%, 27.0%, and 15.0% of TyG index’s association with CVD, respectively, and 40.0%, 11.8%, and 13.3% of TG/HDL-C ratio’s association with CVD, respectively. Conclusions Elevated baseline TyG index and TG/HDL-C ratio were associated with a higher risk of CVD after adjustment for the well-established CVD risk factors. These associations were largely mediated by greater prevalence of dyslipidemia, type 2 diabetes, and hypertension.

Abstract Background Functional diversity is important to maintain ecosystem functioning. Species with different ecomorphological traits may display distinct functional roles in ecosystems. Accordingly, functionally extreme species are more important as they can exhibit specific strategies. However, little is known about the distribution patterns of functionally extreme species at a local scale and whether the prior extinction of extreme species can cause significant effects on functional diversity. In addition, no empirical studies have been conducted on the microhabitat determinants of extreme species to maintain the functional diversity. Results This study collected 1470 tadpoles belonging to 6 families and 20 anuran species. These species were subsequently divided into 65 functional entities based on their developmental stages to incorporate intraspecific traits variability. As a result, we detected seven extreme functional entities, accounting for 10.7% of the total number of entities. Moreover, the prior extinction of extreme entities can lead to a significant decrease in functional diversity compared with the random extinction of entities. Microhabitat variables such as conductivity, water depth, and current velocity determined the distribution of extreme entities. Conclusion Although the functionally extreme entities only represented a small proportion of the total number of tadpoles, they played irreplaceable roles in maintaining functional diversity. Their extinction may induce high functional vulnerability in tadpole communities. Therefore, anuran species with extreme tadpole traits need to be projected for amphibian conservation.

218. Warming changes the composition and diversity of fungal communities in permafrost

Abstract Purpose It is the data support and theoretical basis for the response mechanism of soil fungi to climate warming in permafrost areas in the Greater Xing’an Mountains. Methods We collected permafrost from the Greater Xing’an Mountains for indoor simulation experiments and took the natural permafrost as the control (CK) and the test groups of 0 °C (T1), 2 °C (T2), and 4 °C (T3) were set. Illumina MiSeq high-throughput sequencing technology was used to understand the changes in characteristics of fungal communities, and the correlations were analyzed combined with the soil physicochemical properties. Results Compared with CK, the value of pH and the content of available potassium (AK) in the three warming treatment groups were significantly lower (P < 0.05), and the microbial biomass carbon (MBC) content was significantly higher (P < 0.05). The content of total nitrogen (TN) and available nitrogen (AN) in the T1 and T3 groups was significantly lower than that in the CK group (P < 0.05). A total of 11 phyla, 39 classes, 89 orders, 187 families, 361 genera, and 522 species were obtained through fungal sequencing and divided into 1463 amplicon sequence variants (ASVs). Ascomycota and Dimorphospora were the dominant phylum and genus, respectively, and there were differences in the response of relative abundance of various groups at the phylum and genus levels to warming. Warming significantly decreased the Sobs and ACE indexes of the treatment groups (P < 0.05), and the Shannon and Shannoneven indexes also showed a downward trend. Moreover, warming significantly changed the fungal beta diversity (P < 0.01), while the value of pH and the content of TN, MBC, and AK could significantly affect the community structure (P < 0.05), and the correlation between fungi at different phyla levels and soil physicochemical properties was different. Conclusions These results can provide a reference for further study on the changes in composition and structure of fungal communities and the influence factor in permafrost in the Greater Xing’an Mountains under the background of warming.

Objective Myocardial fibrosis is a potential mechanism of light-chain myocardial amyloidosis(AL-CA). This research aimed at exploring the correlation between multiparameter cardiac magnetic resonance (CMR) and myocardial fibrosis by relating the CMR myocardial tissue characteristics, the morphological and the functional parameters with gallium-68-labeledfibroblast activation protein inhibitor 04 positron emission tomography (68Ga-FAPI PET). Methods We gave the patients diagnosed with AL-CA in Peking Union Medical College Hospital from August to December 2021 the examinations of CMR and 68Ga-FAPI PET/CT. We recorded and analyzed the information on clinical manifestations and examinations of the patients. Results A total of 23 patients with AL-CA were included, 15 (65.2%)of which were male and the mean age was 58.3±6.5 years. Patients with high 68Ga-FAPI-04 uptake had shown growth in myocardial extracellular volume (ECV), significantly higher than those in the negative group (P=0.047). In addition, patients' myocardial ECV was positively correlated with myocardial FAPI uptake (r=0.628, P=0.001;r=0.727, P < 0.001;r=0.661, P=0.001). Patients in the positive group showd reduced left ventricular (LV) ejection fraction (EF)(P < 0.001).LVEF (r=-0.798, P < 0.001;r=-0.794, P < 0.001; r=-0.795, P < 0.001) and right ventricular (RV)EF (r=-0.735, P < 0.001;r=-0.739, P < 0.001;r=- 0.684, P < 0.001) showd negatively correlated with myocardial FAPI uptake, LV circumferential strain (r=0.668, P < 0.001;r=0.708, P < 0.001;r=0.705, P < 0.001), LV longitudinal strain (r=0.629, P=0.001;r=0.635, P=0.001; r=0.597, P=0.003), and RV longitudinal strain (r=0.575, P=0.004; r=0.792, P < 0.001;r=0.673, P < 0.001) were negatively correlated with myocardial FAPI uptake. Conclusions FAPI-related fibroblast activation is concurrent with CMR-related abnormal myocardial interstitial characteristics that leads to the decreased function of the myocardial movement. Patients with increased FAPI uptake present with increased ECV, decreased EF, and decreased strain with morphological abnormalities.

Hypertrophic cardiomyopathy (HCM) is cardiomyopathy with a clinical phenotype of cardiac hypertrophy. The etiology includes genetically defective encoding sarcomeres, congenital metabolic diseases such as lysosomal storage diseases, systemic amyloidosis such as transthyretin amyloidosis(ATTR), and Fabry disease. Previous therapies did not target the etiology and pathogenesis and therefore were less effective. In recent years, treatments targeting different mechanisms of myocardial hypertrophy have achieved good results. Mavacamten can reduce myocardial contractility by inhibiting ATP activity, thereby significantly improving left ventricular outflow tract(LVOT) obstruction, cardiac contractility, ventricular tension, and limitting myocardial damage. By inhibiting the dissociation of transthyretin(TTR) and subsequent formation and deposition of the amyloid fibril, tafamidis can reduce the mortality and morbidity of patients with transthyretin cardiac amyloidosis(ATTR-CA). Gene silencing and gene editing technology can reduce abnormal TTR levels. Synthesis of α-galactosidase A by gene recombination technology in vitro can effectively reduce left ventricular mass index(LVMi), improve cardiac function, reduce angina attacks and decrease mortality of Fabry disease.

Homozygous familial hypercholesterolemia (HoFH) is a rare and serious autosomal genetic metabolic disease. Patients without intervention often die younger than 30 years old from early atherosclerotic cardiovascular disease (ASCVD)incurred by extremely high levels of low-density lipoprotein cholesterol (LDL-C). We present a case of HoFH, a child with compound heterozygous mutation in this study. The effect of conventional lipid-lowering therapy through diet control and lipid-lowering drugs was unsatisfactory. The blood-lipid purification proves effective but has poor compliance and difficult to maintain for a longer time. The patient received orthotopic liver transplantation and had been followed for 2 years, with the patient shows normal LDL-C, well growth and development. We hope the case will provide the clinician with better understanding of the diagnosis and treatment of the rare disease of HoFH.

Transthyretin cardiac amyloidosis (ATTR-CA) is caused by the deposition of transthyretin(TTR) in the myocardial interstitium. Its clinical manifestations are mainly heart failure and arrhythmia, leading to poor life quality and low survival rate. Diagnosis is often delayed or missed due to the lack of disease awareness, the non-specific clinical symptom presentation of the disease, and inadequacy of non-invasive diagnostic methods and medications in the past. The recent availability of effective treatments makes the early recognition and diagnosis especially critical, because treatment is likely more effective earlier in the disease course. Therefore, it is crucial to establish a diagnosis and treatment strategy to facilitate the rapid and accurate identification of the disease. Based on the advances in research and experiences gained ATTR-CA, our team has developed a consensus on diagnosis and treatment for the disease. In this article, we interpret the key points and present the update of diagnostic process, providing clinicians with an overview of key aspects of ATTR-CA in China.

Ambient light interference stands as the primary source of noise encountered by the light detection and ranging (LiDAR) systems based on single-photon avalanche diode (SPAD), which seriously restricts the detection range and accuracy of sensors. Multi-event mode and time-gating method are extensively employed due to their effective suppression ability of the ambient light interference with easily implemented circuit structure. However, the current research on these two operational modes is limited to qualitative description, lacking a comprehensive theoretical analysis. This paper investigates the impact of the multi-event mode and time-gating method on enhancing the imaging capabilities of the sensor, employing a probability distribution perspective. A behavioral model is established, enabling the simulation of the comprehensive functionality of a SPAD-based LiDAR system in either first-hit mode or multi-event mode, including the histogramming, coincident detection, time- gating functions. The model can serve as a substitute for the actual circuit in system function validation and parameter optimization, thereby significantly reducing the design and simulation cycle.

Standard Multi-Object Tracking (MOT) frameworks are currently categorised into three categories: tracking-by-detection, joint detection, and tracking and attention mechanisms. Infrequently, the latter two frameworks require substantial computing resources. The difficulty of implementing real-time tracking does not apply to vehicle detection at traffic crossings. Not only is it essential to meet real-time requirements for vehicle tracking and detection at traffic intersections, but it is also necessary to address common MOT issues such as target occlusion, repetition technology, error detection, etc. Detection-based tracking has a great deal of potential. This study proposed a shallow feature fusion algorithm based on SORT, called SFFSORT and developed an innovative architecture for vehicle monitoring and counting based on detection tracking. This tracking method is more efficient than both SORT and DeepSORT. It achieved 60.9% MOTA and 65.5% IDF1 in MOT16 while MOTA achieved 60.1% and 64.7% IDF1 in MOT17. Utilizing this tracking method as a foundation, we have developed a vehicle counting framework and successfully implemented it on road traffic videos sourced from the Malaysian transportation department. The tracking algorithm presented here effectively addresses the tracking challenges arising from both detection errors and inaccuracies, providing a robust solution. The experimental findings demonstrate that the deep learning framework is capable of achieving lane-level vehicle counting even in scenarios with limited labelled data.

To effectively solve the problem of high dust concentration during coal cutting and frame shifting in fully mechanized mining faces, based on the theory of gas–solid two-phase flow, a geometric model of a fully mechanized mining face was established by using COMSOL numerical simulation software. Simulations were performed for the movement characteristics of wind flow and the law of dust diffusion. Results show that the air flow at the junction of the working face, the air inlet, the hydraulic support moving area, and the vicinity of the shearer has accelerated movement, and the maximum wind speed zone of about 3 m/s can be formed. Under the influence of wind flow, dust particles above 35 μm settle faster, while dust particles below 35 μm are very vulnerable to the influence of wind flow, and the settling speed is slower. Using a custom experimental platform, the atomization characteristics and wind resistance of a pressure fan nozzle, a supersonic nozzle, and an ultrasonic nozzle were tested, and the nozzle that was suitable for the scheme was selected and applied in the field. Comparing the dust concentration before and after the application of the dust removal scheme at the sampling point, results show that the dust removal efficiency of the proposed scheme exceeds 85%, and the treatment effect is good.

Stockpile monitoring has been recently conducted with the help of modern remote sensing techniques—e.g., terrestrial/aerial photogrammetry/LiDAR—that can efficiently produce accurate 3-D models for the area of interest. However, monitoring of indoor stockpiles still requires more investigation due to unfavorable conditions in these environments such as a lack of global navigation satellite system signals and/or homogenous texture. This article develops a fully automated image/LiDAR integration framework that is capable of generating accurate 3-D models with color information for stockpiles under challenging environmental conditions. The derived colorized 3-D point cloud can be subsequently used for volume estimation and visual inspection of stockpiles. The main contribution of the developed strategy is using automatically derived conjugate image/LiDAR linear features for simultaneous registration and camera/LiDAR system calibration. Data for this article are acquired using a camera-assisted LiDAR mapping platform—denoted as stockpile monitoring and reporting technology—which was recently designed as a time-efficient and cost-effective bulk material tracking. Experimental results on three datasets show that the developed framework outperforms a classical planar feature-based registration technique in terms of the alignment of acquired point cloud. Results also indicate that the proposed approach can lead to a high relative accuracy between image lines and their corresponding back-projected LiDAR features in the range of 4–7 pixels.

Anomaly detection has become one of the crucial tasks in hyperspectral images processing. However, most deep learning-based anomaly detection methods often suffer from the incapability of utilizing spatial–spectral information, which decreases the detection accuracy. To address this problem, we propose a novel hyperspectral anomaly detection method with a spatial–spectral dual-window mask transformer, termed as S2DWMTrans, which can fully extract features from global and local perspectives, and suppress the reconstruction of anomaly targets adaptively. Specifically, the dual-window mask transformer aggregates background information of the entire image from a global perspective to neutralize anomalies, and uses neighboring pixels in a dual-window to suppress anomaly reconstruction. An adaptive-weighted loss function is designed to further suppress anomaly reconstruction adaptively during network training process. According to our investigation, this is the first work to apply transformer to hyperspectral anomaly detection. Comparative experiments and ablation studies demonstrate that the proposed S2DWMTrans achieves competitive performance.

Abstract The mechanical properties of bulk-heterojunction (BHJ) films play critical roles in the operational stability of flexible polymer solar cells (PSCs). In this study, the multi-scale mechanical properties of the BHJ films containing three representative electron acceptors are systematically evaluated. Interestingly, the tensile moduli of the films based on PC71BM, Y6, and PRi-C39 decrease sequentially, and their fracture strain and toughness show an increasing trend. Moreover, the nanoindentation moduli of the films based on these acceptors vary in accordance with the results of tensile characterizations. Creep analyses illustrate that the occurrence of creep deformation in the PC71BM, Y6, and PRi-C39-based films decrease sequentially. Contact angle tests demonstrate that the donor-acceptor interactions greatly affect the mechanical properties of these films. All results above demonstrate the effects of acceptor type on the multi-scale mechanical properties of the BHJ films, which are important for the development of reliable flexible PSCs.

Much research has been conducted to improve the defect-detection rate and detection accuracy of the imaging technology used in terahertz nondestructive testing. Due to the power limit of light sources and noise interference in terahertz equipment, images have low resolution and fuzzy defect edges. Hence, improving the resolution is crucial for detecting defects. We designed an edge detection network structure based on a traditional deep neural network. Besides, we devised a node-fusing strategy to train the network. It demonstrates significant improvement of the resolution of the terahertz defect contour. A quartz fiber composites with embedded defects was tested with our network. The results showed that the proposed super-resolution reconstruction algorithm improves resolution, particularly on the edges of defect contours.

Background and purpose: To evaluate the efficiency and safety of immunotherapy combined with or without radiotherapy (RT) for metastatic or recurrent esophageal squamous cell carcinoma (ESCC). Methods: We retrospectively reviewed data of 127 patients with metastatic or recurrent ESCC, who received immunotherapy with or without RT at Tianjin Medical University Cancer Institute between 2017 and 2021. Results: The median follow-up time was 15.7 months (95 % confidence interval (CI): 12.42–18.99). The median PFS of the RT and NRT groups was 5.45 months (95 % CI: 2.89–8.28) and 4.60 months (95 % CI: 3.75–7.06), respectively (P = 0.660). The median OS was 11.9 (95 % CI: 8.61–19.2) and 10.3 (95 % CI: 7.56–15.8) months, respectively (P = 0.890). The median PFS of locoregional recurrence patients in the RT and NRT groups was 11.27 months (95 % CI: 2.45–20.09) and 4.17 months (95 % CI: 2.64–5.71), respectively (P = 0.081). The median OS of locoregional recurrent patients in the RT and NRT groups was 19.48 months (95 % CI: 8.37–30.60) and 7.69 months (95 % CI: 3.45–11.93), respectively (P = 0.026). 64 % of patients in the RT group and 30 % of patients in the NRT group experienced an improvement in dysphagia (P = 0.033). No significant increase in treatment-related toxicity was observed in the RT group compared with the NRT group, except for some hematological complications. Conclusions: Locoregional recurrent patients gained survival benefits from immunotherapy combined with RT. The combination of immunotherapy and RT was safe in metastatic/recurrent ESCC patients. RT for the esophagus leads to the improvement of dysphagia compared to immunotherapy alone.

Abstract Genome editing is an advanced genetic modification tool that facilitates the removal, insertion, or substitution of nucleotides in the genome of an organism for mutation induction. CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR‐associated protein 9) is the most widely used genome editing tool that can precisely modify the specific sequence of a genome. Significant progress has been made in the use of CRISPR/Cas9 for the genetic improvement of food crops. However, in forage crops, the use of CRISPR/Cas9 is still in the infant stage with limited understanding and application. This article reviews current advances in the establishment and application of the CRISPR/Cas9 system in alfalfa, summarizes the application of CRISPR/Cas9 in several other forage legume and grass species, and discusses the prospects of CRISPR/Cas9 in the genetic improvement of forage crops.

Abstract Retinal degeneration is a kind of neurodegeneration characterized by progressive neuronal death and dysfunction of retinal pigment epithelium (RPE) cells, leading to permanent visual impairment. It still lacks effective therapeutic options and new drugs are highly warranted. In this study, we found the expression of IL-4, a critical regulator of immunity, was reduced in both patients and mouse models. Importantly, exogenous intravitreal IL-4 application could exert a novel neuroprotective effect, characterized by well-preserved RPE layer and neuroretinal structure, as well as amplified wave-amplitudes in ERG. The RNA-seq analysis revealed that IL-4 treatment suppressed the essential oxidative and pro-inflammatory pathways in the degenerative retina. Particularly, IL-4 upregulated the IL-4Rα on RPE cells and induced a reparative phenotype via the activation of Nrf2 both in vitro and in vivo. Furthermore, the Nrf2-/- mice displayed no recovery in response to IL-4 application, highlighting a significant role of Nrf2 in IL-4-mediated protection. Our data provides evidence that IL-4 protects against retinal neurodegeneration by its antioxidant and anti-inflammatory property through IL-4Rα upregulation and Nrf2 activation in RPE cells. The IL-4/IL-4Rα-Nrf2 axis maybe the potential targets for the development of novel therapies for neurodegenerative diseases.

Abstract The human estrogen receptor has been used for about thirty years, in the yeast S. cerevisiae, as a component of chimeric transcription factors. Its ligand, β-estradiol, permits to control the protein translocation into the nucleus and, as a consequence, the expression of the gene(s) targeted by the synthetic transcription factor. Activators that are orthogonal to the yeast genome have been realized by fusing the human estrogen receptor to an activation and a DNA-binding domain from bacteria, viruses, or higher eukaryotes. In this work, we optimized the working of a β-estradiol-sensing device—in terms of detection range and maximal output signal—where the human estrogen receptor is flanked by the bacterial protein LexA and either the strong VP64 (from herpes simplex virus) or the weaker B42 (from E. coli) activation domain. We enhanced the biosensor performance by thoroughly engineering both the chimeric activator and the reporter protein expression cassette. In particular, we constructed a synthetic promoter—where transcription is induced by the chimeric activators—based on the core sequence of the yeast CYC1 promoter, by tuning parameters such as the length of the 5′ UTR, the distance between adjacent LexA binding sites (operators), and the spacing between the whole operator region and the main promoter TATA box. We found a configuration that works both as a highly sensitive biosensor and a sharp switch depending on the concentration of the chimeric activator and the strength of its activation domain.

Abstract Clustering and layering are widely employed in large‐scale wireless sensor networks (WSNs) to alleviate data implosion, reduce transmission delay and improve energy efficiency. It is vital to conduct performance analysis in clustered WSNs for better defining and designing networks. Here, a general analytical framework based on a randomly spacial model is proposed to conduct inter‐cluster interference analysis and physical‐layer security analysis for clustered WSNs. It is assumed that two adjacent cluster heads exchange confidential massage with a passive eavesdropper and the non‐head nodes in clusters could be selected as cooperative jamming (CJ) nodes to disturb the eavesdropper. All mentioned nodes are randomly located in their specific areas. Under the above settings, three scenarios, namely interference, eavesdropping and CJ are considered, and a stochastic geometry tool based on kinematic measure is employed for analysis. Comparing to existing stochastic geometry methods, the proposed analytical framework can handle arbitrarily shaped, disjoint and tiered networks. The results obtained from extensive simulations have verified the rationality of the framework and demonstrated the impact of different parameters on the performance metrics of interest. The comparison with PPP also shows the advantages of this proposed model.

Compelling data have indicated menopause-associated increase in cardiovascular disease in women, while the underlying mechanisms remain largely unknown. It is established that changes of intestinal microbiota affect cardiovascular function in the context of metabolic syndrome. We here aimed to explore the possible link between host intestinal function, microbiota, and cardiac function in the ovariectomy (OVX) mouse model. Mice were ovariectomized to induce estrogen-related metabolic syndrome and cardiovascular defect. Microbiota was analyzed by 16s rRNA sequencing. miRNA and mRNA candidates expression were tested by qPCR. Cardiac function was examined by echocardiography. Colon specific delivery of miRNA candidates was achieved by oral gavage of Eudragit S100 functionalized microspheres. In comparison with the sham-operated group, OVX mice showed compromised cardiac function, together with activated inflammation in the visceral adipose tissue and heart. Lactobacillus abundance was significantly decreased in the gut of OVX mice. Meanwhile, miR-155 was mostly upregulated in the intestinal epithelium and thus the feces over other candidates, which in turn decreased Lactobacillus abundance in the intestine when endocytosed. Oral delivery of miR-155 antagonist restored the protective microbiota and thus protected the cardiac function in the OVX mice. This study has established a possible regulatory axis of intestinal miRNAs-microbiota-estrogen deficiency related phenotype in the OVX model. Colon specific delivery of therapeutic miRNAs would possibly restore the microbiota toward protective phenotype in the context of metabolic syndrome.

As the therapeutic landscape for metastatic clear cell renal cell carcinoma (mccRCC) expands to include vascular endothelial growth factor receptor tyrosine kinase inhibitors (VEGFR TKIs) and immunotherapies, new challenges are in place for evaluating and treating refractory disease. Assessing and managing refractory disease has several elements: (1) the mechanism(s) of front-line treatment, (2) timing of progressive disease, (3) rapidity and sites of progressing disease, (4) use of adjuvant therapy, and (5) incorporation of surgical and radiation techniques. These variables all have distinct impact on the biology of refractory or resistant mccRCC. A better understanding of the essential mechanisms of both primary and secondary immunotherapy resistance will inform biomarker development and therapeutic strategies in the refractory setting. This paper addresses the current understanding of treatment sequencing in refractory mccRCC, focusing on treatment options with prospective clinical trial data, considers refractory mccRCC after adjuvant immunotherapy, and incorporates radiation or surgical resection for oligoprogressive disease.

Terahertz detectors based on two-dimensional Dirac materials offer a new approach for room-temperature terahertz detection with high response and low noise. However, these devices can hardly show high response over a broad frequency range, mainly due to the poor absorption caused by their ultrathin nature. Here we apply metallic gratings to enhance the excitation efficiency of graphene plasmons. When nonzero source-drain bias is applied, graphene plasmons can generate terahertz response orthogonal to the polarization of the incidence. The response is attributed to the orthogonal overdamped plasmon rectification effect, and graphene plasmons-enhanced photo-thermoelectric effect. By comparing the normalized on/off ratio, the metallic gratings are found to effectively enhance the coupling efficiency between graphene plasmons and THz incidence, and thus the absorption and responsivity. The results are beneficial for improving the response of room temperature THz detectors.

Abstract Background Calcineurin B-like proteins (CBLs) are ubiquitous Ca2+ sensors that mediate plant responses to various stress and developmental processes by interacting with CBL-interacting protein kinases (CIPKs). CBLs and CIPKs play essential roles in acclimatization of crop plants. However, evolution of these two gene families in the genus Medicago is poorly understood. Results A total of 68 CBL and 135 CIPK genes have been identified in five genomes from Medicago. Among these genomes, the gene number of CBLs and CIPKs shows no significant difference at the haploid genome level. Phylogenetic and comprehensive characteristic analyses reveal that CBLs and CIPKs are classified into four clades respectively, which is validated by distribution of conserved motifs. The synteny analysis indicates that the whole genome duplication events (WGDs) have contributed to the expansion of both families. Expression analysis demonstrates that two MsCBLs and three MsCIPKs are specifically expressed in roots, mature leaves, developing flowers and nitrogen fixing nodules of Medicago sativa spp. sativa, the widely grown tetraploid species. In particular, the expression of these five genes was highly up-regulated in roots when exposed to salt and drought stress, indicating crucial roles in stress responses. Conclusions Our study leads to a comprehensive understanding of evolution of CBL and CIPK gene families in Medicago, but also provides a rich resource to further address the functions of CBL-CIPK complexes in cultivated species and their closely related wild relatives.

With the decrease in energy storage costs and the popularization of the shared economy principle, shared energy storage (SES) has been gradually deployed in the residential community to reduce energy costs. However, centralized trading platforms for SES trading are rare in the residential community, so it is difficult for users with the trading demands to search for appropriate trading partners and make a deal. To address the above problem, a decentralized trading mechanism of SES considering the preference of trading subjects is proposed to enable the users in the residential community to trade in the absence of centralized trading platforms. The cost-based price models are built to determine the acceptable price ranges for SES users, DG users and power consumers, which will be used as the basis for the quotation in the process of decentralized trading. Moreover, based on a multinomial logit model and an improved Rubinstein model, the trading partner selection method and the negotiation strategy are proposed to be adopted in the decentralized trading mechanism. Through the autonomous selection of trading partners and multiple rounds of negotiation and quotation, the trading probability of the users can be improved. Numerical simulation studies show that the proposed decentralized trading mechanism can achieve a higher sale rate of SES trading. All types of users can obtain revenue or reduce energy costs through SES trading.

Building-level load forecasting has become essential with the support of fine-grained data collected by widely deployed smart meters. It acts as a basis for arranging distributed energy resources, implementing demand response, etc. Compared to aggregated-level load, the electric load of an individual building is more stochastic and thus spawns many probabilistic forecasting methods. Many of them resort to artificial neural networks (ANN) to build forecasting models. However, a well-designed forecasting model for one building may not be suitable for others, and manually designing and tuning optimal forecasting models for various buildings are tedious and time-consuming. This paper proposes an adaptive probabilistic load forecasting model to automatically generate high-performance NN structures for different buildings and produce quantile forecasts for future loads. Specifically, we cascade the long short term memory (LSTM) layer with the adjusted Differential ArchiTecture Search (DARTS) cell and use the pinball loss function to guide the model during the improved model fitting process. A case study on an open dataset shows that our proposed model has superior performance and adaptivity over the state-of-the-art static neural network model. Besides, the improved fitting process of DARTS is proved to be more time-efficient than the original one.

246. Effect of PEG-rhG-CSF in Preventing Chemotherapy-induced Neutropenia in Locally Advanced Non-small Cell Lung Cancer Patients at Nutritional Risk

Objective To explore the efficacy and safety of polyethylene glycol recombinant human granulocyte colony-stimulating factor (PEG-rhG-CSF) in preventing chemotherapy-induced neutropenia in locally advanced non small cell lung cancer (NSCLC) patients at nutritional risk. Methods A total of 337 locally advanced NSCLC patients at nutritional risk were selected. They were randomly divided into three groups: 112 cases in the non-prophylactic drug group (control group), 112 cases in the prophylactic use of rhG-CSF treatment group (rhG-CSF group), and 113 cases in the prophylactic use of PEG-rhG-CSF treatment group (PEG-rhG-CSF group). The incidence and duration of neutropenia after chemotherapy and the ratio of CD4+/CD8+T cells in peripheral blood were observed. Results The incidences of neutropenia in the control group, rhG-CSF group, and PEG-rhG-CSF group were 67.97%, 41.57%, and 38.98% (P < 0.05), respectively. The incidences of grade Ⅲ-Ⅳ neutropenia in the three groups were 22.39%, 14.25%, and 11.14% (P < 0.05); moreover, the incidence of febrile neutropenia in the three groups was 3.55%, 1.84%, and 1.21% (P < 0.05); in addition, the ratios of CD4+/CD8+T cells in peripheral blood were 1.27±0.44, 1.41±0.52, and 1.49±0.42 (P < 0.05). The duration of grade Ⅲ-Ⅳ neutropenia and the time required for the neutrophil value to reach 2.0×109/L from the lowest value in the PEG-rhG-CSF group were lower than those in the control and rhG-CSF groups (P < 0.05). Conclusion The PEG-rhG-CSF preventive treatment used in the course of chemoradiotherapy in locally advanced NSCLC patients at nutritional risk can reduce the incidence of neutropenia and improve immunologic function. PEG-rhG-CSF preventive treatment is worthy of clinical recommendation.

Osteosarcoma (OS) is an uncommon tumour that mainly affects bone in children and adolescents. The current treatment options of OS are of limited significance due to their immense side effects. In the present manuscript, we have developed a novel series of 1,2,3-triazole chalcone derivatives as potential agents against OS. The compounds were synthesized and evaluated for their PI3K and mTOR inhibitory activity using luminescent kinase assay, and Lance ultra assay, resp. The entire set of compounds showed significant to moderate inhibition of both kinases in the nanomolar range. The three most active compounds: 4e (N-(4-(3-(1-(4-bromophenyl)-1H-1,2,3-triazol-4-yl)acryloyl)phenyl)-4-nitrobenzamide), 4f (N-(4-(3-(1-(4-bromophenyl)-1H-1,2,3-triazol-4-yl)acryloyl)phenyl)-4-chlorobenzamide) and 4g (4-bromo-N-(4-(3-(1-(4-bromophenyl)-1H-1,2,3-triazol-4-yl)acryloyl)phenyl)benzamide), were evaluated for anticancer activity against human OS cancer cell line (MG-63), liver cancer cell line (HepG2), lung cancer cell line (A549) and cervical cancer (HeLa), using MTT assay. Among the tested series, compound 4e showed a better inhibitory profile than gedatolisib against PI3K and was approximately comparable to that of gedatolisib against mTOR. The most significant inhibitory activity was observed for compound 4e against all cell lines (MG-63, HepG2, A549 and HeLa), still somewhat lower to comparable to that of gedatolisib, but with the highest potency against MG-63 cells. Compound 4e was further tested for anti-cancer activity against other OS cells and showed to be equipo-tent to gedatolisib against U2OS and Saos-2 cells. Moreover, it was also found non-toxic to normal cells (BEAS-2B and MCF 10A). The effect of compound 4e was further determined on apoptosis of Saos-2 cells by Annexin-PI assay, where it significantly amplified the percentage of apoptotic cells. Novel 1,2,3-triazole chalcone derivatives are potential agents against OS.

The overwhelming impact of complex-phase microstructures to mechanical response in multiphase steels requires accurate constitutive properties of the individual phases. However, precise prediction of individual phase properties to their mechanical response is critical and sophisticated, and commonly requires multiscale characterizations and numerous approximations. In this work, by employing full phase information and semi-empirical analytical models, we accurately predict the yield strength of deformed variants of Ce-modified SAF2507 super duplex stainless steel (SDSS). High energy synchrotron X-ray diffraction (HE-SXRD) reveals the phase fractions of major phases along with secondary phases of Cr2N and eutectic CexFey. Average lattice strain/crystallite size of the austenite and ferrite/martensite phases from the measured volume is estimated through the Williamson-Hall method. A unique composite strengthening type analytical model is used to estimate yield strength by taking individual strengthening contributions from all phases, their grain sizes, stored dislocation densities, solid solution, and precipitates. Close agreement between reconstructed and experimental yield strength is observed for several cold and cryogenic rolled SDSS. A combination of HE-SXRD and analytical model offers a time-effective virtual design pathway to engineer high-strength steel.

Abstract Multifocal microscopy enables high-speed three-dimensional (3D) volume imaging by using a multifocal grating in the emission path. This grating is typically designed to afford a uniform illumination of multifocal subimages for a single emission wavelength. Using the same grating for multicolor imaging results in non-uniform subimage intensities in emission wavelengths for which the grating is not designed. This has restricted multifocal microscopy applications for samples having multicolored fluorophores. In this paper, we present a multicolor multifocal microscope implementation which uses a Spatial Light Modulator (SLM) as a single multifocal grating to realize near-uniform multifocal subimage intensities across multiple wavelength emission bands. Using real-time control of an in-situ-optimized SLM implemented as a multifocal grating, we demonstrate multicolor multifocal 3D imaging over three emission bands by imaging multicolored particles as well as Escherichia coli (E. coli) interacting with human liver cancer cells, at $$\sim 2.5$$ ∼ 2.5 multicolor 3D volumes per second acquisition speed. Our multicolor multifocal method is adaptable across SLM hardware, emission wavelength band locations and number of emission bands, making it particularly suited for researchers investigating fast processes occurring across a volume where multiple species are involved.

Abstract Background Growing oocytes acquire the ability to mature through two-way communication between gametes and surrounding somatic cumulus cells (CCs). Granulosa cells (GCs) support oocyte growth, regulate meiosis progression, and modulate global oocyte transcription activity. However, the proliferation and differentiation of the yak ovary in GCs and CCs remain unclear. To characterize the important roles of long non-coding RNA, (lncRNA), microRNA (miRNA), and messenger RNA (mRNA), whole-transcriptome analysis was performed. Real-time quantitative fluorescence PCR was performed to verify the selected RNA sequences. Results Important gene ontology terms and Kyoto Encyclopedia of Genes and Genomes pathways related to differentiation and oocyte development were identified for the target genes of differentially expressed lncRNAs, miRNAs, and mRNAs. In total,6223 mRNAs (2197 upregulated, 4026 downregulated), 643 lncRNAs (204 upregulated, 479 downregulated), and 559 miRNAs (311 upregulated, 248 downregulated) were significantly altered between the two groups. Target genes involved in cell adhesion, cell differentiation, regulation of developmental processes, cell proliferation, embryo development, signal transduction, apoptosis, and aromatic compound biosynthetic processes were significantly enriched. These RNAs were involved in ECM-receptor interaction, MAPK signaling, Hippo signaling, PI3K-Akt signaling, cell cycle, cell adhesion, leukocyte trans-endothelial migration, and actin cytoskeleton regulation. Conclusions A comprehensive analysis of the co-expression network of competing endogenous RNAs (ceRNAs) will facilitate the understanding of the process of granulosa cell proliferation and differentiation and offer a theoretical basis for the development of oocytes.
