201. 53 Mn and 60 Fe in iron meteorites—New data, model calculations

Abstract: We measured specific activities of the long‐lived cosmogenic radionuclides ⁶⁰Fe in 28 iron meteorites and ⁵³Mn in 41 iron meteorites. Accelerator mass spectrometry was applied at the 14 MV Heavy Ion Accelerator Facility at ANU Canberra for all samples except for two which were measured at the Maier‐Leibnitz Laboratory, Munich. For the large iron meteorite Twannberg (IIG), we measured six samples for ⁵³Mn. This work doubles the number of existing individual ⁶⁰Fe data and quadruples the number of iron meteorites studied for ⁶⁰Fe. We also significantly extended the entire ⁵³Mn database for iron meteorites. The ⁵³Mn data for the iron meteorite Twannberg vary by more than a factor of 30, indicating a significant shielding dependency. In addition, we performed new model calculations for the production of ⁶⁰Fe and ⁵³Mn in iron meteorites. While the new model is based on the same particle spectra as the earlier model, we no longer use experimental cross sections but instead use cross sections that were calculated using the latest version of the nuclear model code INCL. The new model predictions differ substantially from results obtained with the previous model. Predictions for the ⁶⁰Fe activity concentrations are about a factor of 2 higher, for ⁵³Mn, they are ~30% lower, compared to the earlier model, which gives now a better agreement with the experimental data.

Abstract: Based on the collected data on the diversity of microalgae and environmental indicators in dry and wet seasons during 2011–2018, from 45 samples, 59 species of microalgae were identified in the ornithological object—Lake Agmon in the Hula Valley. In the samples of periphyton and microphytobenthos, diatoms predominated. Bioindication analysis and statistical mapping revealed the most pronounced zones of influence on the lake ecosystem, as well as indicators of the environment and diversity that clearly demonstrate them. The correlation between the distribution of TDS of water over the lake surface and the distribution of green, diatom microalgae and cyanobacteria detected two areas of impact from the old channel of the Jordan River in the northwestern part and from the drainage channel in the northeastern parts of the lake. The area on the east coast, in contact with the resting fields of migratory birds, has provided nutrients that stimulate the development of green algae and cyanobacteria. This showed implicit links in the lake ecosystem using bioindicators make it possible to recommend them for monitoring in combination with statistical mapping, which visualizes the distribution of data and is easily accessible for the decision-making system for the management of a protected ornithological lake.

203. Croup during the COVID-19 Omicron Variant Surge

Abstract: Croup is a common upper respiratory disease usually associated with parainfluenza virus, resulting in stridor, hoarse voice, barky cough, and variable respiratory distress. Here we examine the data at our center confirming a sharp increase in cases of croup associated with the Omicron variant. Data was retrospectively extracted from patient charts among those seen in the Emergency Department at Seattle Children’s Hospital. Inclusion criteria were patients who were assigned a diagnosis containing “croup” during either 5/30/2021-11/30/2021, a time period correlating with predominance of the COVID-19 Delta variant (B.1.617.2), or the initial phase of the Omicron variant surge (12/1/2021-1/15/2022). Contemporaneous publicly available local data on the proportion of SARS-CoV-2 samples in surrounding King County, Washington, with spike gene target failure on TaqPath PCR assays was used as a proxy for the proportion of infections caused by the Omicron variant. A total of 401 patients were diagnosed with croup during the Delta surge and 107 patients were diagnosed with croup during the Omicron surge. Patients who presented during the Omicron surge were more likely to test positive for COVID-19 (48.2% vs 2.8%, p < 0.0001). Children with a clinical diagnosis of croup during the Omicron surge were more likely to be prescribed racemic epinephrine as part of their care (21.5% vs 13.0%, p = 0.032). There were no differences in presenting age, rate of admission, rate of return to the ED within 72 hours, or admission among those who returned within 72 hours. During the Omicron surge, the incidence of croup nearly doubled compared to the rate in prior months, while at the same time the number of cases of parainfluenza virus identified decreased. Consistent with prior case reports, we have identified a sharp rise in cases of croup seen in our pediatric ED in parallel with the replacement of the SARS-CoV-2 Delta variant by Omicron as the dominant variant in our community.

Abstract: IntroductionChildhood cancer is a leading cause of death. It is unclear whether the COVID-19 pandemic has impacted childhood cancer mortality. In this study, we aimed to establish all-cause mortality rates for childhood cancers during the COVID-19 pandemic and determine the factors associated with mortality.MethodsProspective cohort study in 109 institutions in 41 countries. Inclusion criteria: children ResultsAll-cause mortality was 3.4% (n=71/2084) at 30-day follow-up, 5.7% (n=113/1969) at 90-day follow-up and 13.0% (n=206/1581) at 12-month follow-up. The median time from diagnosis to multidisciplinary team (MDT) plan was longest in low-income countries (7 days, IQR 3–11). Multivariable analysis revealed several factors associated with 12-month mortality, including low-income (OR 6.99 (95% CI 2.49 to 19.68); pConclusionsChildren with cancer are more likely to die within 30 days if infected with SARS-CoV-2. However, timely treatment reduced odds of death. This report provides crucial information to balance the benefits of providing anticancer therapy against the risks of SARS-CoV-2 infection in children with cancer.

207. Mouse Norovirus Uses Host Metabolites to Enhance Receptor Binding and Evade Immune Recognition

Abstract: Noroviruses are the major cause of epidemic gastroenteritis in humans, causing ~20 million cases annually, resulting in more than 70,000 hospitalizations and 570-800 deaths in the United States alone. The T=3 icosahedral calicivirus capsid is composed of viral protein 1 (VP1) with three major domains: the N-terminus (N), shell (S), and C-terminal protruding (P) domains. The S domain forms a shell around the viral RNA genome, while the P domains dimerize to form protrusions on the capsid surface. The P domain is subdivided into P1 and P2 subdomains, with the latter containing the binding sites for cellular receptors and neutralizing antibodies. Mouse norovirus (MNV) is a widely used system for study of norovirus biology since we have a cell culture system, reverse genetic tools, and small animal model to eventually correlate structural information to whole animal pathology Mouse norovirus is a surprisingly dynamic virus that switches between receptor and antibody binding structures depending upon the in-vivo environment. In the circulation, the P domain floats above the shell by more than 15Å and the P domain loops (A’B’/E’F’) at the very tip are splayed apart in an ‘open’ conformation that antibodies learn to recognize. Upon ingestion, the low pH environment with high metal and bile salt concentrations in the alimentary canal each independently trigger the P domains to rotate 90° and contract by 15 Å onto the capsid surface. This hides any epitopes at base of the P domain. During this reversible collapse, the two P domains within the dimer rotate about each other and the A’B’/E’F’ loops adopt the ‘closed’ conformation. This opens the receptor binding site while burying the epitopes at the tip of the P domain. Therefore, rather than only depending on escape mutations to block antibody binding, MNV aggressively uses host conditions to remodel itself to enhance receptor binding while blocking antibody recognition. This review will describe the structural processes and biological consequences of the virus responding to activating host cues in the gut while these same triggers bury the epitopes presented in the circulation. This is an aggressive and unique mode of immune escape that has been subsequently shown in other viruses such as COVID-19. Therefore, a deeper understanding of the dynamic processes of virus capsids will improve vaccine design by understanding how to present the epitope conformations at the site of infection rather than what is presented to the immune system.

208. P-BN07 Identifying gallstones in acute pancreatitis. Are we undermanaging our patients?

Abstract: Background Acute pancreatitis (AP) is a common surgical presentation with a wide spectrum of severity and outcome. The most common cause of AP is gallstones, accounting for approximately 50% of cases, followed by alcohol excess. Reliable identification of gallstones is crucial as patients can be offered cholecystectomy to prevent recurrence. Current UK guidelines recommend a minimum of two negative ultrasounds to rule out gallstones. The aim of this study was to assess the pickup rate of gallstones on ultrasound for patients admitted with AP and audit our compliance with UK guidelines. Methods All patients admitted with acute pancreatitis between the start of January 2019 to the end of December 2020 were retrospectively analysed. All patients with a known pre-existing cause for pancreatitis such as alcohol excess, chronic pancreatitis, CBD stricture and pancreatic mass were excluded. Electronic records were examined to identify subsequent imaging investigations and final diagnosis. Particular interest was given to whether gallstones were identified, and adherence to UK guidelines. Results 206 patients were identified following the exclusion criteria. 189 underwent an ultrasound on admission, 111 were positive for gallstones. Of the negative ultrasounds (78), 15 underwent a further USS (4 positive), 29 underwent an MRCP (12 positive), 15 had a CT (3 positive) and 3 had an ERCP (3 positive). This left 16 with an unknown aetiology after 1 ultrasound and did not undergo further imaging and therefore did not comply with the current guidelines. Of the 11 patients who had 2 negative ultrasounds 5 had further imaging and all were negative for gallstones. Conclusions In conclusion Gallstone pancreatitis is a common acute surgical presentation of which morbidity and mortality can be significant. Following our retrospective assessment, we deem secondary imaging in the form of USS or MRCP to be necessary in the investigation of acute pancreatitis due to the high pickup rate. Compliance with current guidelines aids diagnostics and ensures appropriate and timely management of this condition leading to improved patient care.

Abstract: An 88-year-old male with a history of hypertension, ischaemic heart disease and Bell’s palsy presented with symptoms and signs of an acute ischaemic stroke. National Institutes of Health Stroke Scale (NIHSS) was 19 at presentation, indicative of potential large vessel occlusion. The initial CT scan revealed evidence of small vessel disease and arterial calcification. As there were no contraindications, he received thrombolytic treatment. CT angiography and CT perfusion imaging were performed in preparation for possible thrombectomy. There was no evidence of a large vessel thrombus, and changes on CT perfusion were suggestive of seizure activity, with relative hyperperfusion on the cerebral hemisphere of interest. Post thrombolysis, his NIHSS was 5. An MR scan revealed evidence of bilateral thalamic infarcts. After a period of rehabilitation, he was discharged home and independently mobile but with cognitive impairment.Acute stroke care increasingly uses multimodal imaging to confirm the clinical diagnosis and help optimise initial emergency management. Such imaging is useful in determining whether the presentation is a vascular event or stroke mimic. Moreover, seizures complicate and mimic acute strokes, which can lead to therapeutic uncertainty. This case highlights the increasingly sophisticated investigation of patients presenting with suspected acute stroke, with the attendant need for accurate interpretation by experienced clinicians.

Abstract: Bacteriophages screened and isolated from sewage water samples exhibited antibacterial activities against multi-drug-resistant Escherichia coli strains. Five different water samples from Canadian habitats such as Kamloops Wastewater Treatment Center, Domtar, the Pacific Ocean, Bisaro Anima Cave, and alkali ponds, were used in this study. Four Enterobacteriaceae strains including one non-resistant and three clinical multi-drug Escherichia coli strains (E. coli 15-102, E. coli 15-124, and E. coli 15-318) were selected as target bacteria to screen for the bacteriophages from these collected water samples. Seeded agar assay technique was implemented for the screening. It was found that only sewage water sample exhibited a significant number of plaques count with the E. coli 15-318 (1.82 × 102 plaques/plate) cells in comparison to E. coli non-resistant strain K12 (8 plaques/plate). The phage did not produce plaques in the E. coli 15-124 and E. coli 15-102 strains. The bacteriophage, designated EMCL318, was isolated, purified, characterized, and identified to belong to the G4 species of the Family Microviridae, GenBank accession number MG563770. This is an explorative study conducted in order to reveal the viruses as alternative potentials to fight against emerging and existing multi-drug-resistant infectious diseases.

Abstract: BACKGROUND: One key element of lung-protective ventilation is the use of a low tidal volume (V(T)). A sex difference in use of low tidal volume ventilation (LTVV) has been described in critically ill ICU patients. OBJECTIVES: The aim of this study was to determine whether a sex difference in use of LTVV also exists in operating room patients, and if present what factors drive this difference. DESIGN, PATIENTS AND SETTING: This is a posthoc analysis of LAS VEGAS, a 1-week worldwide observational study in adults requiring intra-operative ventilation during general anaesthesia for surgery in 146 hospitals in 29 countries. MAIN OUTCOME MEASURES: Women and men were compared with respect to use of LTVV, defined as V(T) of 8 ml kg(−1) or less predicted bodyweight (PBW). A V(T) was deemed ‘default’ if the set V(T) was a round number. A mediation analysis assessed which factors may explain the sex difference in use of LTVV during intra-operative ventilation. RESULTS: This analysis includes 9864 patients, of whom 5425 (55%) were women. A default V(T) was often set, both in women and men; mode V(T) was 500 ml. Median [IQR] V(T) was higher in women than in men (8.6 [7.7 to 9.6] vs. 7.6 [6.8 to 8.4] ml kg(−1) PBW, P*** < 0.001). Compared with men, women were twice as likely not to receive LTVV [68.8 vs. 36.0%; relative risk ratio 2.1 (95% CI 1.9 to 2.1), P*** < 0.001]. In the mediation analysis, patients’ height and actual body weight (ABW) explained 81 and 18% of the sex difference in use of LTVV, respectively; it was not explained by the use of a default V(T). CONCLUSION: In this worldwide cohort of patients receiving intra-operative ventilation during general anaesthesia for surgery, women received a higher V(T) than men during intra-operative ventilation. The risk for a female not to receive LTVV during surgery was double that of males. Height and ABW were the two mediators of the sex difference in use of LTVV. Abstract: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is an emerging RNA virus causing COVID-19 disease across the globe. SARS-CoV-2 infected patients exhibit acute respiratory distress syndrome which can be compounded by endemic respiratory viruses and thus highlighting the need to understand the genetic bases of clinical outcome under multiple respiratory infections. In this study, 42 individual datasets and a multi-parametric based selected list of over 12,000 genes against five medically important respiratory viruses (SARS-CoV-2, SARS-CoV-1, influenza A, respiratory syncytial virus (RSV) and rhinovirus were collected and analysed in an attempt to understand differentially regulated gene patterns and to cast genetic markers of individual and multiple co-infections. While a certain cohort of virus-specific genes were regulated (negatively and positively), notably results revealed a greatest correlation among gene regulation by SARS-CoV-2 and RSV. Furthermore, out of analysed genes, the MAP2K5 and NFKBIL1 were specifically and highly upregulated in SARS-CoV-2 infection in vivo or in vitro. In contrast, several genes including GPBAR1 and SC5DL were specifically downregulated in SARS-CoV-2 datasets. Additionally, we catalogued a set of genes that were conserved or differentially regulated across all the respiratory viruses. These finding provide foundational and genome-wide data to gauge the markers of respiratory viral infections individually and under co-infection.

Abstract: Advances in neonatal care have resulted in improved outcomes for high-risk newborns with technologies playing a significant part although many were developed for the neonatal intensive care unit. The care provided in the delivery room (DR) during the first few minutes of life can impact short- and long-term neonatal outcomes. Increasingly, technologies have a critical role to play in the DR particularly with monitoring and information provision. However, the DR is a unique environment and has major challenges around the period of foetal to neonatal transition that need to be overcome when developing new technologies. This review focuses on current DR technologies as well as those just emerging and further over the horizon. We identify what key opinion leaders in DR care think of current technologies, what the important DR measures are to them, and which technologies might be useful in the future. We link these with key technologies including respiratory function monitors, electoral impedance tomography, videolaryngoscopy, augmented reality, video recording, eye tracking, artificial intelligence, and contactless monitoring. Encouraging funders and industry to address the unique technological challenges of newborn care in the DR will allow the continued improvement of outcomes of high-risk infants from the moment of birth. IMPACT: Technological advances for newborn delivery room care require consideration of the unique environment, the variable patient characteristics, and disease states, as well as human factor challenges. Neonatology as a speciality has embraced technology, allowing its rapid progression and improved outcomes for infants, although innovation in the delivery room often lags behind that in the intensive care unit. Investing in new and emerging technologies can support healthcare providers when optimising care and could improve training, safety, and neonatal outcomes.

Abstract: A taxonomic survey of the lamp flora from electrically lit passages in Mammoth Cave, Mammoth Cave National Park, identified 28 species. Overall, cyanobacteria were dominant represented by 14 species (50% of the total), green algae had eight species (29%), and six diatoms species (21%) were present. There was not a correlation between species diversity and temperature, but there is a general trend of increasing diversity with warmer temperatures. There were two algal or cyanobacterial species identified in this study that overlapped with previous studies. There is a lack of continuity between previous studies only having one species identified in more than one study. This suggests a high algal turnover and possible colonization rates.

Abstract: The eye movement system is sensitive to reward. However, whilst the eye movement system is extremely flexible, the extent to which changes to oculomotor behaviour induced by reward paradigms persist beyond the training period or transfer to other oculomotor tasks is unclear. To address these issues we examined the effects of presenting feedback that represented small monetary rewards to spatial locations on the latency of saccadic eye movements, the time-course of learning and extinction of the effects of rewarding saccades on exogenous spatial attention and oculomotor IOR. Reward feedback produced a relative facilitation of saccadic latency in a stimulus driven saccade task which persisted for 3 blocks of extinction trials. However this hemifield-specific effect failed to transfer to peripheral cueing tasks. We conclude that rewarding specific spatial locations is unlikely to induce long-term, systemic changes to the human oculomotor or attention systems.

232. Effective coverage and systems effectiveness for malaria case management in sub-Saharan African countries.

Abstract: Scale-up of malaria preventive and control interventions over the last decade resulted in substantial declines in mortality and morbidity from the disease in sub-Saharan Africa and many other parts of the world. Sustaining these gains will depend on the health system performance. Treatment provides individual benefits by curing infection and preventing progression to severe disease as well as community-level benefits by reducing the infectious reservoir and averting emergence and spread of drug resistance. However many patients with malaria do not access care, providers do not comply with treatment guidelines, and hence, patients do not necessarily receive the correct regimen. Even when the correct regimen is administered some patients will not adhere and others will be treated with counterfeit or substandard medication leading to treatment failures and spread of drug resistance. We apply systems effectiveness concepts that explicitly consider implications of health system factors such as treatment seeking, provider compliance, adherence, and quality of medication to estimate treatment outcomes for malaria case management. We compile data for these indicators to derive estimates of effective coverage for 43 high-burden Sub-Saharan African countries. Parameters are populated from the Demographic and Health Surveys and other published sources. We assess the relative importance of these factors on the level of effective coverage and consider variation in these health systems indicators across countries. Our findings suggest that effective coverage for malaria case management ranges from 8% to 72% in the region. Different factors account for health system inefficiencies in different countries. Significant losses in effectiveness of treatment are estimated in all countries. The patterns of inter-country variation suggest that these are system failures that are amenable to change. Identifying the reasons for the poor health system performance and intervening to tackle them become key priority areas for malaria control and elimination policies in the region.

Abstract: The BLSA is an invaluable resource for the study of human aging with uniquely rich measures taken longitudinally on a continuous replenishment cohort since 1958. Tremendous interest has been expressed by the United States and international research communities in expanded access to and use of BLSA data to address emerging scientific questions. Here, we will describe recent study leadership initiatives into: (1) making the BLSA data more FAIR (Findable, Accessible, Interoperable, and Reusable), (2) growing a BLSA data ecosystem that enables scalable sharing, and (3) leveraging and developing platforms for accessing the ecosystem. Lastly, we will demonstrate current platform functions that empower researchers to find and access BLSA data, metadata, protocols, code and supporting materials to make new discoveries.

235. NUMERACY SKILL USE AMONG MIDDLE-AGED AND OLDER WORKERS IN THE US

Abstract: In numeric information-rich societies, numeracy is essential both at work and in everyday life (e.g., calculating a budget). Numeracy skills generally decline with aging. Thus, among middle-aged and older workers, maintaining and improving numeracy skills is crucial to securing employment as well as managing everyday life. One of the counteracting strategies is to practice numeracy. However, little is known about how and what kinds of numeracy skills are used among older workers. We analyzed a nationally representative sample of U.S. workers aged 45 to 74 (n = 3,850) from the 2012/2014/2017 Program for International Assessment of Adult Competencies (PIAAC) restricted-use-file. Six dichotomous numeracy use indicators (e.g., calculating a budget, using advanced statistics) at work and in everyday life were considered. Survey weighted latent class analysis (LCA) identified three subgroups with distinctive numeracy skill use patterns, including ubiquitous (both at work and in everyday life) users, occupational users, and non-users. For example, calculating a budget was common both in non-users (40%) and ubiquitous users (84%), whereas uncommon among occupational users (25%). Also, only ubiquitous users practiced advanced math and statistics (14%), while others did not (nearly 0%). The subsequent regression analysis revealed that higher educational attainment, higher income, certain racial group (i.e., Whites), and better self-rated health were associated with greater numeracy skill use. The lack of numeracy skill use leads to lower skill levels and, in turn, social and economic disadvantages in later life. In addition to more detailed LCA results interpretations, possible policy and educational interventions are evaluated.

Abstract: Establishing accurate electrical load prediction is vital for pricing and power system management. However, the unpredictable behavior of private and industrial users results in uncertainty in these power systems. Furthermore, the utilization of renewable energy sources, which are often variable in their production rates, also increases the complexity making predictions even more difficult. In this paper an interval type-2 intuitionist fuzzy logic system whose parameters are trained in a hybrid fashion using gravitational search algorithms with the ridge least square algorithm is presented for short-term prediction of electrical loading. Simulation results are provided to compare the performance of the proposed approach with that of state-of-the-art electrical load prediction algorithms for Poland, and five regions of Australia. The simulation results demonstrate the superior performance of the proposed approach over seven different current state-of-the-art prediction algorithms in the literature, namely: SVR, ANN, ELM, EEMD-ELM-GOA, EEMD-ELM-DA, EEMD-ELM-PSO and EEMD-ELM-GWO.

Abstract: A search is performed for rare and forbidden charm decays of the form $D_{(s)}^+ \to h^\pm \ell^+ \ell^{(\prime)\mp}$, where $h^\pm$ is a pion or kaon and $\ell^{(')\pm}$ is an electron or muon. The measurements are performed using proton-proton collision data, corresponding to an integrated luminosity of $1.6\text{fb}^{-1}$, collected by the LHCb experiment in 2016. No evidence is observed for the 25 decay modes that are investigated and $90\%$ confidence level limits on the branching fractions are set between $1.4\times10^{-8}$ and $6.4\times10^{-6}$. Abstract: Management scholarship is increasingly paying attention to “grand challenges” such as climate and demographic changes, sociopolitical uncertainty, poverty alleviation, and global health improvement. Drawing upon a review of grand challenges research, we highlight the lack of a coherent conceptualization of a “grand challenge” and examine the diversity of the conceptual, policy, and empirical research. We problematize the failure in current research to distinguish between qualitatively different types of grand challenges—in our view, the resolution of different challenges calls for distinct research, policy, and practice approaches. We propose a novel framework that encompasses four distinct forms of grand challenge, situate the existing empirical literature within our framework, describe how the contributions to our special issue advance grand challenges research, and explore implications for future management research that responds to the imperatives of grand challenges.JEL Classification: D81, G01, M14, I18, Q54, Q55, Q56, Q57

Abstract: Akathisia is a relatively common adverse effect that may emerge during treatment with antipsychotics and other medication classes. We present a case of akathisia that may have been induced by the abrupt discontinuation of varenicline and review existing literature related to this phenomenon. A 46-year-old female with a past psychiatric history of bipolar disorder and borderline personality disorder was admitted to the acute psychiatric services department for suicidal ideation after 3 weeks of a new course of varenicline. This was prescribed for smoking cessation and titrated to 1 mg twice daily. Upon admission, the varenicline was discontinued. Roughly 3 days later, the patient began to complain of akathisia. The patient had experienced akathisia previously while taking antipsychotics for her bipolar disorder and was able to recognize its emergence. As the akathisia worsened, propranolol 10 mg 3 times daily was ordered and was effective in relieving her symptoms. A PubMed search using the terms varenicline, akathisia, withdrawal, and discontinuation was conducted. No literature of this phenomenon was found; however, reports of other extrapyramidal symptoms were noted. Considering the timing of varenicline's discontinuation and its mechanism, a pharmacological link between its use and akathisia is possible. Akathisia is a severely uncomfortable sequela of medications that may produce severe outcomes, such as suicidal ideation. In this case, it is possible that the discontinuation of varenicline after 3 weeks of therapy led to akathisia, which was successfully treated with propranolol.

246. Assessing worker performance using dynamic cost functions in human robot collaborative tasks

Abstract: The aim of this research is to develop a framework to allow efficient human robot collaboration on manufacturing assembly tasks based on cost functions that quantify capabilities and performance of each element in a system and enable their efficient evaluation. A proposed cost function format is developed along with initial development of two example cost function variables, completion time and fatigue, obtained as each worker is completing assembly tasks. The cost function format and example variables were tested with two example tasks utilizing an ABB YuMi Robot in addition to a simulated human worker under various levels of fatigue. The total costs produced clearly identified the best worker to complete each task with these costs also clearly indicating when a human worker is fatigued to a greater or lesser degree than expected.

Abstract: En esta contribución, los autores traducen y analizan una página de la llamada Probanza de Santo Domingo Petapa, que es un título primordial, escrito en zapoteco del Istmo a finales del siglo XVII. La importancia histórica del texto se encuentra en su fuerte relación con el famoso Lienzo de Guevea, uno de los documentos pictográficos zapotecos más estudiados. Esta relación hace posible un análisis comparativo entre el texto alfabético y el pictórico, que tienen fuertes semejanzas, pero también diferencias notables. Los autores, además, muestran la belleza retórica y literaria de la Probanza. Un aspecto notable es el uso de elaborados difrasismos que hasta ahora no se han encontrado en otros textos zapotecos publicados.

Abstract: Clonal hematopoiesis (CH aka CHIP) is the process whereby otherwise healthy individuals accumulate clones bearing low-frequency somatic mutations in hematologic malignancy (HM) driver genes. Historically thought to be a phenomenon of the elderly that is associated with increased risk of HM and cardiovascular disease, CH has been observed in increasingly younger cohorts as sequencing technology has advanced. CH in hematopoietic stem cell transplant (HSCT) donors has also been associated with adverse recipient outcomes. In this retrospective analysis, we asked whether CH mutations preferentially expanded in recipients vs. donors in paired samples collected a median of 34 years post-HSCT (range 6-45). 16 donor-recipient pairs were analyzed, 11 of which had a donor sample collected pre-HSCT. All individuals were healthy at the time of contemporary sampling. Median donor age was 26 years (range 12-84) at time of HSCT. We took advantage of the ultra-low error rate ( The mean DS error-corrected molecular depth across all samples was 26,731x for AML-associated genes and 32,812x in the neutral regions. CH was detected in 100% of donors and recipients, with a trend toward more CH clones in older vs. younger donors. We defined CH as mutations observed in at least 2 unique DNA molecules, rather than by a threshold variant allele frequency (VAF). Among the 11 donors with matched historical and contemporary DNA samples, CH mutations in the latter had a higher average VAF and comprised more unique mutations than in the former, reflecting clonal expansions over time. The magnitude of CH expansions was greater for the AML-associated genes than for neutral loci, consistent with positive selection. Post-HSCT the overall number of unique mutations was similar between donors and recipients. Most donor CH variants did not preferentially expand in the recipient. A minority of recipients had a modestly increased overall average VAF relative to paired donors in AML genes. While the VAF of some likely pathogenic variants increased in the recipient vs the donor, this was not the case for most CH mutations in most recipients and the reciprocal was sometimes observed. DS represents a powerful tool for the high-resolution study of CH evolution. In a unique historical cohort comprising longitudinal samples from some of the earliest patients and donors to undergo HSCT, we observed limited instances of accelerated CH in recipients relative to donors. While this cohort is biased due to enriched inclusion of long-term survivors, our data exemplifies how healthy hematopoiesis, indistinguishable between donors and recipients, can be maintained almost half a century following transplant.
