This article describes a chemistry/art activity that originated in an National Science Foundation--funded two-week STEAM (Science, Technology, Engineering, Art, and Math) academy for grade 4-6 girls. The authors recommend using this investigation in conjunction with other activities focusing on chemical change as a step toward fulfilling the "Next Generation Science Standards" ("NGSS") performance expectation 5-PS1-4: "Conduct an investigation to determine whether the mixing of two or more substances results in new substances" (NGSS Lead States 2013). The investigation leverages the overlapping practices of art and science, including observation, experimentation, and the creation of solutions to design problems, as students use chemical reactions to develop a multicolored palette for painting. Using acids and bases to change the color of red cabbage pigments, students explore the disciplinary core idea that mixing substances can result in chemical reactions that form new substances with different properties.

Hydrogen peroxide has been used as a sanitation agent for many years. Recently, hydrogen peroxide products have been used to remove algae from irrigation lines and sanitize hydroponic systems between uses. However, hydrogen peroxide can have phytotoxic effects on plants at high concentrations. The goal of this research was to determine if hydrogen peroxide treatments affected plant and algae growth in the ebb and flow hydroponic systems. The research was conducted at the Department of Horticulture and Landscape Architecture greenhouses in Stillwater, OK. Two cultivars of lettuce, ‘Green Forest’ and ‘Tropicana’, and two cultivars of basil, ‘Aroma II’ and ‘Genovese’, were transplanted into the ebb and flow hydroponic systems, and three different hydrogen peroxide products, PERpose Plus, ZeroTol, and 3% hydrogen peroxide, were applied at different rates and combinations in two experiments. Shoot fresh weight in lettuce was found to be significantly greater in control and 3% hydrogen peroxide treatments for both cultivars; however, in ‘Tropicana’ those treatments were not different from any other treatment. Greater amounts of PERpose Plus and ZeroTol, such as 60 mL, restricted plant growth in lettuce, whereas only cultivar differences for SPAD and plant width were reported for basil. Algae growth was not significantly controlled by any treatment in this research based on algae counts, weights, or spectrometer readings. However, algae species quantification did show that Microspora tumidula was found in the greatest concentrations in control, with a 96.0%, 99.2%, 94.0%, and 97.9% reduction in the 15 mL ZeroTol, 60 mL ZeroTol, 15 mL PERpose Plus, and 3% hydrogen peroxide treatments, respectively. Other algae genera identified included Scenedesmus, Chlamydomonas, Gloeocystis, Tetraspora, Leptolyngbya, Pennate diatoms, and Centric diatoms.

Although reports of urban populations of amphibians and reptiles are increasingly common, the viability of such populations and information on threats to their continued existence remain less well known. From 2010–2018, we studied the nesting ecology of the imperiled Northern Map Turtle (Graptemys geographica) in an urbanized area in northeastern Maryland, USA. Mark-recapture data showed an estimated population size of 32 females with an annual survival of 89%. Females nested in highly altered areas such as flower gardens, along railroad rights-of-way, and along fence lines, but nest survival was much higher than in nearby natural areas, mainly due to very low predation rates from Raccoons. However, nesting females were subject to considerable disturbance from vehicle and foot traffic while nesting. To alleviate disturbance, we partnered with the town to construct a wildlife exclusion fence designed to isolate nesting Map Turtles from human disturbance. Due to the high annual survival of nests, this urban population may be important in the overall viability of Northern Map Turtles in this area.

RationaleIndirect airway hyperresponsiveness (AHR) is a fundamental feature of asthma that is manifest as exercise-induced bronchoconstriction (EIB). Secreted phospholipase A2 group X (sPLA2-X) plays a key role in regulating eicosanoid formation and the development of inflammation and AHR in murine models.ObjectivesWe sought to examine sPLA2-X in the airway epithelium and airway wall of patients with asthma, the relationship to AHR in humans, and the regulation and function of sPLA2-X within the epithelium.MethodsWe precisely phenotyped 34 patients with asthma (19 with and 15 without EIB) and 10 normal control subjects to examine in vivo differences in epithelial gene expression, quantitative morphometry of endobronchial biopsies, and levels of secreted protein. The regulation of sPLA2-X gene (PLA2G10) expression was examined in primary airway epithelial cell cultures. The function of epithelial sPLA2-X in eicosanoid formation was examined using PLA2 inhibitors and murine tracheal epithelial cells with Pla2g10 deletion.Measurements and main resultsWe found that sPLA2-X protein is increased in the airways of patients with asthma and that epithelial-derived sPLA2-X may be increased in association with indirect AHR. The expression of sPLA2-X increases during in vitro epithelial differentiation; is regulated by inflammatory signals including tumor necrosis factor, IL-13, and IL-17; and is both secreted from the epithelium and directly participates in the release of arachidonic acid by epithelial cells.ConclusionsThese data reveal a relationship between epithelial-derived sPLA2-X and indirect AHR in asthma and that sPLA2-X serves as an epithelial regulator of inflammatory eicosanoid formation. Therapies targeting epithelial sPLA2-X may be useful in asthma.

BackgroundDepressive symptoms are correlated with poor health outcomes in adults with chronic kidney disease (CKD). The prevalence, severity, and treatment of depressive symptoms and potential risk factors, including level of kidney function, in diverse populations with CKD have not been well studied.Study designCross-sectional analysis.Settings & participantsParticipants at enrollment into the Chronic Renal Insufficiency Cohort (CRIC) and Hispanic-CRIC (H-CRIC) Studies. Objectives Informing research participants of the results of studies in which they took part is viewed as an ethical imperative. However, there is little guidance in the literature about how to do this. The Fluoxetine Or Control Under Supervision trial randomised 3127 patients with a recent acute stroke to 6 months of fluoxetine or placebo and was published in the Lancet on 5 December 2018. The trial team decided to inform the participants of the results at exactly the same time as the Lancet publication, and also whether they had been allocated fluoxetine or placebo. In this report, we describe how we informed participants of the results.Design In the 6-month and 12-month follow-up questionnaires, we invited participants to provide an email address if they wished to be informed of the results of the trial. We re-opened our trial telephone helpline between 5 December 2018 and 31 March 2019.Setting UK stroke services.Participants 3127 participants were randomised. 2847 returned 6-month follow-up forms and 2703 returned 12-month follow-up forms; the remaining participants had died (380), withdrawn consent or did not respond.Results Of those returning follow-up questionnaires, a total of 1845 email addresses were provided and a further 50 people requested results to be sent by post. Results were sent to all email and postal addresses provided; 309 emails were returned unrecognised. Seventeen people replied, of whom three called the helpline and the rest responded by email.Conclusion It is feasible to disseminate results of large trials to research participants, though only around 60% of those randomised wanted to receive the results. The system we developed was efficient and required very little resource, and could be replicated by trialists in the future.Trial registration number ISRCTN83290762; Post-results.

Adoptive cell transfer of Chimeric Antigen Receptor (CAR)-T cells showed promising results in patients with B cell malignancies. However, the detailed mechanism of CAR-T cell interaction with the target tumor cells is still not well understood. This work provides a systematic method for analyzing the activation and degranulation of second-generation CAR-T cells utilizing antigen-presenting cell surfaces. Antigen-presenting cell surfaces composed of circular micropatterns of CAR-specific anti-idiotype antibodies have been developed to mimic the interaction of CAR-T cells with target tumor cells using micro-contact printing. The levels of activation and degranulation of fixed non-transduced T cells (NT), CD19.CAR-T cells, and GD2.CAR-T cells on the antigen-presenting cell surfaces were quantified and compared by measuring the intensity of the CD3ζ chain phosphorylation and the Lysosome-Associated Membrane Protein 1 (LAMP-1), respectively. The size and morphology of the cells were also measured. The intracellular Ca2+ flux of NT and CAR-T cells upon engagement with the antigen-presenting cell surface was reported. Results suggest that NT and CD19.CAR-T cells have comparable activation levels, while NT have higher degranulation levels than CD19.CAR-T cells and GD2.CAR-T cells. The findings show that antigen-presenting cell surfaces allow a quantitative analysis of the molecules involved in synapse formation in different CAR-T cells in a systematic, reproducible manner.

Organophosphates (OPs) induce acute and chronic neurotoxicity, primarily by inhibiting acetylcholinesterase (AChE) activity as well as by necrosis, and apoptosis. Butyrylcholinesterase (BuChE), an exogenous bioscavenger of OPs, can be used as a treatment for OP exposure. It is prerequisite to develop in vitro brain models that can study BuChE post-treatment for acute OP exposure. In this study, we developed a three-dimensional (3D) brain-on-chip platform with human induced pluripotent stem cell (iPSC)-derived neurons and astrocytes to simulate human brain behavior. The platform consists of two compartments: 1) a hydrogel embedded with human iPSC-derived GABAergic neurons and astrocytes and 2) a perfusion channel with dynamic medium flow. The brain tissue constructs were exposed to Malathion (MT) at various concentrations and then treated with BuChE after 20 minutes of MT exposure. Results show that the iPSC-derived neurons and astrocytes directly interacted and formed synapses in the 3D matrix, and that treatment with BuChE improved viability after MT exposure up to a concentration of 10-3 M. We conclude that the 3D brain-on-chip platform with human iPSC-derived brain cells is a suitable model to study the neurotoxicity of OP exposure and evaluate therapeutic compounds for treatment.

A key challenge in the field of cognitive neuroscience is to identify discriminable cognitive functions, and then map these functions to brain activity. In the current study, we set out to explore the relationships between performance arising from different cognitive tasks thought to tap different domains of cognition, and then to test whether these distinct latent cognitive abilities also are subserved by corresponding "latent" brain substrates. To this end, we tested a large sample of adults under the age of 40 on twelve cognitive tasks as they underwent fMRI scanning. Exploratory factor analysis revealed 4-factor model, dissociating tasks into processes corresponding to episodic memory retrieval, reasoning, speed of processing and vocabulary. An analysis of the topographic covariance patterns of the BOLD-response acquired during each task similarity also converged on four neural networks that corresponded to the 4 latent factors. These results suggest that distinct ontologies of cognition are subserved by corresponding distinct neural networks.

Mechanical rigidity of a matrix, to which cells adhere, plays a significant role in regulating phenotypic cellular behaviors such as spreading and junction formation because vascular cells sense and respond to changes in their mechanical environment. Controlling mechanical properties of extracellular matrix by using a crosslinker is important for cell and tissue mechanobiology. In this paper, we explored genipin, a natural plant extract, to crosslink collagen-I in order to enhance mechanical properties with low cytotoxicity. We characterized the effects of genipin concentration on the mechanical properties, color change, degradation, structure, cell viability, and endothelial function such as transendothelial electrical resistance (TEER). Through the analysis of both material properties and endothelial response, it was found that genipin-based glycation caused an increase in viscoelastic moduli in collagen hydrogels, as well as increased fiber density in their structural morphology. Endothelial cells were found to form better barriers, express higher levels of tight junction proteins, and exhibit better adhesion on stiffer matrices.

Abstract Phthalates are known endocrine disruptors and associated with decreased fecundity, pregnancy loss, and adverse obstetrical outcomes, however the underlying mechanisms remain to be established. Environmental factors can influence gene expression and cell function by modifying epigenetic marks, impacting the developing embryo as well as future generations of offspring. The impact of phthalates on placental gene methylation and expression is largely unknown. We studied the effect of maternal phthalate exposure on the human placental DNA methylome and transcriptome. We determined epigenome-wide DNA methylation marks (Illumina Infinium Human Methylation 850k BeadChip) and gene expression (Agilent whole human genome array) associated with phthalate exposure in first trimester placenta. Integrative genomic analysis of candidate genes was performed to define gene methylation-expression relationships. We identified 39 genes with significantly altered methylation and gene expression in the high phthalate exposure group. Most of these relationships were inversely correlated. This analysis identified epidermal growth factor receptor (EGFR) as a critical candidate gene mediating the effects of phthalates on early placental function. Although additional studies are needed to determine the functional consequences of these changes, our findings are consistent with the model that phthalates impact placental function by modulating the expression of critical placental genes through epigenetic regulation.

The N ∼ 90 region of the nuclear chart has featured prominently as the spectroscopy of nuclei at extreme spin has progressed. This talk will present recent discoveries from investigations of high spin behavior in the N ∼ 90 Er, Tm and Yb nuclei utilizing the Gammasphere gamma-ray spectrometer. In particular it will include discussion of the beautiful shape evolution and coexistence observed in these nuclei along with the identification of a remarkable new family of band structures. The latter are very weakly populated rotational sequences with high moment of inertia that bypass the classic terminating configurations near spin 40-50h{stroke}, marking a return to collectivity that extends discrete γ-ray spectroscopy to well over 60h{stroke}. Establishing the nature of the yrast states in these nuclei beyond the oblate band-termination states has been a major goal for the past two decades. Cranking calculations suggest that these new structures most likely represent stable triaxial strongly deformed bands that lie in a valley of favored shell energy in deformation and particle-number space.

The preservation and promotion of rich urban environments demands more than logical and functional understandings alone. Although these types of understanding are important to the life of vital cities, what is often overlooked in these views is the role that the incomplete, the messy, and the complex play in constituting the wholeness and viscerality of real urbanity. Aided by perspectives from philosophy and film this article promotes the “residual” aspects of the urban experience and suggests why these aspects might be of even greater importance than more controlled elements of urban life to the continuation of thick, whole, urban settings.

• Little is known about nurse coaching's impact on nursing students' adaptation. • Nurse coaching shows promising benefits in well-being and empowerment. • Nurse coach interventions can assist students in the development of resilience. Nursing education is responsible for supporting student well-being and preparing resilient graduates to face challenges inherent to nursing. Nurse coaching may be a method to support student well-being and resilience. A program evaluation survey of recipients of interventions by the nurse coach was distributed. Fifty (50) students from a southwestern university responded to the survey. Of the 50 respondents, 18 received one-on-one nurse coaching, and 32 attended self-care presentations. Both groups found the interventions by the nurse coach beneficial in a variety of ways. The nurse coach may be a valuable student support resource in a variety of nursing programs. Objective: The purpose of this study was to investigate whether making fruit boxes available in the workplace is a successful health promotion strategy. Design: A quasi-experimental study involving three conditions--free fruit, 50c per piece of fruit and $1 per piece of fruit--to investigate the effect of a contribution scheme on employees' fruit purchase/consumption behaviours and willingness to contribute when in the paid conditions. Setting: Perth, Western Australia. Methods: In total, 36 workplaces participated and were randomly assigned to one of the three conditions. The results were analysed using generalised linear modelling. A qualitative follow-up was conducted with workplace representatives 6 weeks after the completion of the trial to investigate how many workplaces implemented the provision of fruit boxes after the trial and the factors influencing the decision to implement fruit boxes. Results: A significant difference in average fruit purchasing/consumption per person was found with respect to condition (p <0.001), with businesses in the free condition purchasing/consuming a significantly greater amount of fruit than businesses in the 50c contribution condition or $1 contribution condition. Following the trial, 13 workplaces continued providing their own fruit box, of which 7 were initially in the free condition. Qualitative findings revealed that management support, a receptive culture and sufficient resources were key to the implementation of fruit boxes. Conclusion: Having a fruit box may be a feasible health promotion strategy, and the financial burden of this strategy could be alleviated by asking employees to contribute to the cost of fruit.

Engaging students as partners in learning, teaching, and curriculum renewal is driven by different agendas, and takes multiple forms. Although the research is clear on the ethical necessity of student participation in these efforts, the institutional challenges remain ongoing and writ large. This article offers an account of an innovative students-as-partners initiative at the University of Sydney, in which six undergraduates were engaged as student ambassadors to participate in and research the institution's learning and teaching conference--the Sydney Teaching Colloquium. While students have regularly been sought out as contributors to, and representatives of, the student voice, the initiative marked a significant departure from the university's tendency to rely on student feedback surveys and committee representation as its main institutionalised forms of student voice. Over four months, these student ambassadors worked with the central academic development unit to plan, execute, research, and evaluate a programme of activity designed to enhance authentic student participation in curriculum renewal. This article--conceived and co-written with these students--is in part, their reflective account of what they learned about universities: how they are organised and enact assessment curriculum change designed to improve student learning. Their reflections remind us that in order for them to feel genuinely engaged as partners, more needs to be done to help students learn about how and why academics and universities operate as they do.

Objective: Colleges are at risk for communicable disease outbreaks because of the high degree of person-to-person interactions and relatively crowded dormitory settings. This report describes the US college student health screening requirements among US resident and international students for tuberculosis (TB) and vaccine-preventable diseases (VPDs) as they relate to the American College Health Association (ACHA) guidelines. Methods/Participants: In April 2012, US college health administrators (N = 2,858) were sent online surveys to assess their respective school's TB screening and immunization requirements. Results: Surveys were completed by 308 (11%) schools. Most schools were aware of the ACHA immunization (78%) and TB screening (76%) guidelines. Schools reported having policies related to immunization screening (80.4%), immunization compliance (93%), TB screening (55%), and TB compliance (87%). Conclusion: Most colleges were following ACHA guidelines. However, there are opportunities for improvement to fully utilize the recommendations and prevent outbreaks of communicable diseases among students in colleges.

Fraternity members are exposed to developmental opportunities through service, leadership, and involvement, guided by organizational values. The purpose of this study was to learn more about how members interpret their experiences in relation to civic values. Data sources were responses from essays (n = 196) written by graduate and undergraduate members of an international fraternity. Results from thematic analysis evidenced a range of applications, from a literal interpretation of rules to a broader sense of agency in the world. Discussion focuses on addressing gaps in what members are learning based on the core tenets of NASPA's Civic Learning and Democratic Engagement (CLDE).

Abstract Background Few controlled clinical trials exist to support oral combination therapy in pulmonary arterial hypertension (PAH). Methods Patients with PAH (idiopathic [IPAH] or associated with connective tissue disease [APAH-CTD]) taking bosentan (62.5 or 125 mg twice daily at a stable dose for ≥3 months) were randomized (1:1) to sildenafil (20 mg, 3 times daily; n = 50) or placebo (n = 53). The primary endpoint was change from baseline in 6-min walk distance (6MWD) at week 12, assessed using analysis of covariance. Patients could continue in a 52-week extension study. Abstract Frankia sp. NRRL B-16219 was directly isolated from a soil sample obtained from the rhizosphere of Ceanothus jepsonii growing in the USA. Its host plant range includes members of Elaeagnaceae species. Phylogenetically, strain NRRL B-16219 is closely related to “Frankia discariae” with a 16S rRNA gene similarity of 99.78%. Because of the lack of genetic tools for Frankia, our understanding of the bacterial signals involved during the plant infection process and the development of actinorhizal root nodules is very limited. Since the first three Frankia genomes were sequenced, additional genome sequences covering more diverse strains have helped provide insight into the depth of the pangenome and attempts to identify bacterial signaling molecules like the rhizobial canonical nod genes. The genome sequence of Frankia sp. strain NRRL B-16219 was generated and assembled into 289 contigs containing 8,032,739 bp with 71.7% GC content. Annotation of the genome identified 6211 protein-coding genes, 561 pseudogenes, 1758 hypothetical proteins and 53 RNA genes including 4 rRNA genes. The NRRL B-16219 draft genome contained genes homologous to the rhizobial common nodulation genes clustered in two areas. The first cluster contains nodACIJH genes whereas the second has nodAB and nodH genes in the upstream region. Phylogenetic analysis shows that Frankia nod genes are more deeply rooted than their sister groups from rhizobia. PCR-sequencing suggested the widespread occurrence of highly homologous nodA and nodB genes in microsymbionts of field collected Ceanothus americanus.

Abstract Plant volatile signatures are often used as cues by herbivores to locate their preferred hosts. Here, we report on the volatile organic compounds used by the subterranean root-knot nematode (RKN) Meloidogyne incognita for host location. We compared responses of infective second stage juveniles (J2s) to root volatiles of three cultivars and one accession of the solanaceous plant, Capsicum annum against moist sand in dual choice assays. J2s were more attracted to the three cultivars than to the accession, relative to controls. GC/MS analysis of the volatiles identified common constituents in each plant, five of which were identified as α-pinene, limonene, 2-methoxy-3-(1-methylpropyl)-pyrazine, methyl salicylate and tridecane. We additionally identified thymol as being specific to the accession. In dose-response assays, a blend of the five components elicited positive chemotaxis (71–88%), whereas individual components elicited varying responses; Methyl salicylate (MeSA) elicited the highest positive chemotaxis (70–80%), α-pinene, limonene and tridecane were intermediate (54–60%), and 2-methoxy-3-(1-methylpropyl)-pyrazine the lowest (49–55%). In contrast, thymol alone or thymol combined with either the preferred natural plant root volatiles or the five-component synthetic blend induced negative chemotaxis. Our results provide insights into RKN-host plant interactions, creating new opportunities for plant breeding programmes towards management of RKNs.

The patch currently on the US market contains 150 µg norelgestromin and 35 µg ethinylestradiol (EE). The 20 cm2 patch is applied once weekly for 3 weeks, followed by a patch-free week, for a 21–7 cycle. Typical failure rates are similar to that of combined oral contraceptives (COCs). Transdermal delivery results in less peaks and troughs of estrogen, but a higher total estrogen exposure compared with COCs. Though studies show mixed results, the risk of developing venous thromboembolism (VTE) is about twice as high with the patch as with COCs; however, the absolute risk of VTE remains low. The side effect profile is similar to that of COCs, with slightly higher rates of breast tenderness plus a unique adverse effect of application site reactions. Two new patches have been developed, one containing gestodene and EE in Europe and another containing levonorgestrel and EE. Overall, the patch provides an alternative to COCs for women who want autonomy and the benefit of not needing to take a pill daily, with similar efficacy and tolerability. Abstract Background Group 3 pulmonary hypertension (PH) encompasses PH owing to lung diseases and/or hypoxia. Treatment patterns, healthcare resource use, and economic burden to US payers of Group 3 PH patients were assessed. Methods This retrospective observational study extracted data from July 1, 2010 to June 30, 2013 from two Truven Health Analytics MarketScan databases. Adult Group 3 PH patients were identified based on claims for PH (ICD-9-CM 416.0/416.8), a related lung disease, and an echocardiogram or right heart catheterization (RHC). The index date was the date of the first PH claim; data were collected for 12 months pre- and post-index. A difference-in-difference approach using generalized estimating equations was done to account for baseline differences. Results Group 3 PH patients (n = 2,236) were matched 1:1 to controls on lung disease. PH patients had higher all-cause resource utilization and annual healthcare costs ($44,732 vs. $7,051) than controls. Costs were driven by inpatient admissions (35.4% of total costs), prescriptions (33.0%), and outpatient care (26.5%). Abstract: T-helper 17 (Th17) cells have been widely implicated as drivers of autoimmune disease. In particular, Th17 cytokine plasticity and acquisition of an interleukin-17A+(IL-17A+)interferon γ(IFNγ)+ cytokine profile is associated with increased pathogenic capacity. Donor Th17 polarization is known to exacerbate graft-versus-host disease (GVHD) after allogeneic stem cell transplantation (allo-SCT); however, donor Th17 cytokine coexpression and plasticity have not been fully characterized. Using IL-17 “fate-mapping” mice, we identified IL-6-dependent Th17 cells early after allo-SCT, characterized by elevated expression of proinflammatory cytokines, IL-17A, IL-22, granulocyte-macrophage colony-stimulating factor, and tumor necrosis factor. This population did not maintain lineage fidelity, with a marked loss of IL-17A and IL-22 expression late posttransplant. Th17 cells were further segregated based on IFNγ coexpression, and IL-17A+IFNγ+ Th17 displayed an enhanced proinflammatory phenotype. Th17 cytokine plasticity and IFNγ production were critically dependent upon donor-derived IL-12p40, and cyclosporine (CsA) treatment regulated this differentiation pathway. This observation was highly concordant with clinical samples from allo-SCT recipients receiving CsA-based immune suppression where although the IFNγ-negative–Th17 subset predominated, IFNγ+-Th17 cells were also present. In sum, Th17 polarization and ensuing differentiation are mediated by sequential inflammatory signals, which are modulated by immunosuppressive therapy, leading to distinct phenotypes within this lineage.

In the past, DOT stormwater drainage facilities have been designed solely on the conveyance of runoff from DOT facilities in terms of immediate discharge and runoff volume. Since the emergence of the National Pollutant Discharge Elimination System (NPDES) Phase I requirements, DOTs have relied on Best Management Practices (BMPs) for controlling stormwater. The idea has been that as long as one of the BMPs was being utilized, then the quality of the treatment and discharge was sufficient. Now, with the onset of the Phase II mandates, municipalities are scrambling to meet numeric concentration and mass limit restrictions on stormwater discharge. The implications in terms of implementation cost, monitoring, land-use planning, and acquisition could be staggering. These issues are weighing heavily on the minds of DOT personnel who are searching for solutions that are achievable within the logistic realm of current practices. This paper discusses the feasibility, pros, and cons of using the expansive areas of land available at large highway interchanges, as a resource for treating stormwater runoff. Current practices for these areas do not typically utilize the available space for water quality. These large grassed areas require frequent mowing and weed control. In some instances, reforestation and landscape projects are being implemented, but with a little more attention to detail and very little added cost, they could perform a functional need. These large areas at interchanges could be valuable resources in terms of meeting Phase II requirements, and doing their part to service the entire watershed.

Occupational activity represents a large percentage of people’s daily activity and thus likely is as impactful for people’s general and cognitive health as other lifestyle components such as leisure activity, sleep, diet, and exercise. Different occupations, however, require different skills, abilities, activities, credentials, work styles, etc., constituting a rich multidimensional formative exposure with likely consequences for brain development over the lifespan. In the current study, we were interested in how different occupations with their different attributes relate to five variables: structural brain health, duration of early-life education, gender, IQ, and age, although the main focus was the relationship to brain health. To this end, we used the Occupation Information Network (O∗NET), which provides quantification of occupations along 246 items. Occupational patterns with different loadings for these 246 items were derived from 277 community-dwelling adults, ranging in age from 40 to 80, based upon the five subject measures. We found significant patterns underlying four of our variables of interest, with gender and education predictably showing the most numerous and strongest associations, while brain health and intelligence showed weaker associations, and age did not manifest any associations. For the occupational pattern associated with brain health, we found mainly positive associations on items pertaining to rigorous problem-solving, leadership, responsibility, and information processing. We emphasize that the findings are correlational and cannot establish causation. Future extensions of this work will assess the influence of occupation on future cognitive brain status and cognitive performance.

ABSTRACT Here, we report our educational approach and learner evaluations of the first 5 years of the Explorations in Data Analysis for Metagenomic Advances in Microbial Ecology (EDAMAME) workshop, held annually at Michigan State University’s Kellogg Biological Station from 2014 to 2018. We hope this information will be useful for others who want to organize computing-intensive workshops and will encourage quantitative skill development among microbiologists. IMPORTANCE High-throughput sequencing and related statistical and bioinformatic analyses have become routine in microbiology in the past decade, but there are few formal training opportunities to develop these skills. A weeklong workshop can offer sufficient time for novices to become introduced to best computing practices and common workflows in sequence analysis. We report our experiences in executing such a workshop targeted to professional learners (graduate students, postdoctoral scientists, faculty, and research staff).

Abstract A common response of plant communities to increased nitrogen (N) deposition is a shift in species’ abundances. Multiple factors have been proposed to explain the changes in abundance, notably competition and soil acidification. We hypothesized that a plant species that decreased in abundance with elevated N would have lower ectomycorrhizal fungi, altered root‐associated bacteria communities, and/or greater susceptibility to Al toxicity than a species that increased in abundance with increasing N deposition. We examined changes in plant–microbe associations and Al toxicity in two dominant species from an alpine dry meadow community subjected to long‐term low‐level N addition. Carex rupestris has increased in cover over time with N addition and Kobresia myosuroides has decreased. We conducted field sampling of soil microbes from treatment plots and tested whether field levels of Al have toxic effects on sedge species in a greenhouse study. Declines in ectomycorrhizal infection of Cenococcum geophilum occurred on Kobresia with increasing N treatment. In contrast, neither Al level nor changes in bacteria community composition corresponded with the change in cover of sedge species. Decreased ectomycorrhizal infection may have contributed to the decrease in abundance of Kobresia. This study contributes to an understanding of the types of plant–soil interactions that may influence how plant species respond to N deposition and rejects Al toxicity and changes in bacteria composition as factors that likely play a role in changes in sedge abundance.

Chronic thromboembolic pulmonary hypertension (CTEPH), a rare pulmonary vascular disease, is often misdiagnosed due to nonspecific symptoms. The objective of the study was to develop, refine and validate a case ascertainment algorithm to identify CTEPH patients within the French exhaustive hospital discharge database (PMSI), and to use it to estimate the annual number of hospitalized patients with CTEPH in France in 2015, as a proxy for disease prevalence. As ICD-10 coding specifically for CTEPH was not available at the time of the study, a case ascertainment algorithm was developed in close collaboration with an expert committee, using a two-step process (refinement and validation), based on matched data from PMSI and hospital medical records from 2 centres. The best-performing algorithm (specificity 95%, sensitivity 70%) consisted of ≥1 pulmonary hypertension (PH) diagnosis during 2015 and any of the following criteria over 2009-2015: (i) CTEPH interventional procedure, (ii) admission for PH and pulmonary embolism (PE), (iii) PE followed by hospitalization in competence centre then in reference centre, (iv) history of PE and right heart catheterization. Patients with conditions suggestive of pulmonary arterial hypertension were excluded. A total of 3,138 patients hospitalized for CTEPH was estimated for 2015 (47 cases/million, range 43 to 50 cases/million). Assuming that patients are hospitalized at least once a year, the present study provides an estimate of the minimal prevalence of CTEPH and confirms the heavy burden of this disease.

We investigated a potential use of a 3D tetraculture brain microphysiological system (BMPS) for neurotoxic chemical agent screening. This platform consists of neuronal tissue with extracellular matrix (ECM)-embedded neuroblastoma cells, microglia, and astrocytes, and vascular tissue with dynamic flow and membrane-free culture of the endothelial layer. We tested the broader applicability of this model, focusing on organophosphates (OPs) Malathion (MT), Parathion (PT), and Chlorpyrifos (CPF), and chemicals that interact with GABA and/or opioid receptor systems, including Muscimol (MUS), Dextromethorphan (DXM), and Ethanol (EtOH). We validated the BMPS platform by measuring the neurotoxic effects on barrier integrity, acetylcholinesterase (AChE) inhibition, viability, and residual OP concentration. The results show that OPs penetrated the model blood brain barrier (BBB) and inhibited AChE activity. DXM, MUS, and EtOH also penetrated the BBB and induced moderate toxicity. The results correlate well with available in vivo data. In addition, simulation results from an in silico physiologically-based pharmacokinetic/pharmacodynamic (PBPK/PD) model that we generated show good agreement with in vivo and in vitro data. In conclusion, this paper demonstrates the potential utility of a membrane-free tetraculture BMPS that can recapitulate brain complexity as a cost-effective alternative to animal models.

Introduction The science of patient safety demonstrates that good communication is essential for effective interprofessional collaboration. Methods We created a low-stakes, formative assessment with which medical students, pharmacy students, and nursing students could practice several of the Interprofessional Education Collaborative competencies. We aimed to enable students to practice collaborative care, respect for other disciplines, and shared accountability. Senior students from medicine, nursing, and pharmacy worked in teams to disclose a medical error to a standardized patient. The activity began with an icebreaker exercise wherein students learned about each other. Next, each team planned a strategy for error disclosure and collaboratively disclosed the error. Standardized patients evaluated the team's performance. Subsequently, students regrouped for a debriefing. The participating institutions administered a survey to their students. Results In total, 1,151 students participated: 464 fourth-year students from the University of Houston College of Pharmacy, 450 third- and fourth-year students from Baylor College of Medicine, and 237 fourth-year students from Texas Woman's University Nelda C. Stark College of Nursing, all in Houston, Texas. Postsession survey data showed that students thought they achieved the relevant competencies. Students’ understanding of the perspectives of the other two disciplines improved. Students found the simulation encounter and debriefing effective in helping them consider the contributions of other disciplines to patient care. Discussion This interprofessional standardized patient activity enabled collaborative problem solving. The debriefing discussion broadened students’ understanding of the expertise of the other disciplines and promoted shared accountability. Students found this activity engaging and effective.

Background: Professional identity formation (PIF), a foundational process in becoming a physician, includes establishment of values, moral principles, and self-awareness. The purpose of this report is to examine challenges in establishing the validity of measures of identity fusion as one facet of PIF. Method: Utilizing the modern approach of validity as a unitary concept, the authors generated six hypotheses to examine the evidence for the construct validity of the scores of Physician Professional Identity (PPI) and Identity Integration (IdIn), considering relationships of these measures with each other, year of training and data from a larger survey. Results: Responses from 3473 students at 8 medical schools revealed a weak association between the measures with distributions varying by cohort. PPI had a stronger relationship to cohort and IdIn was moderately associated with students’ attitudes relevant to social media use. Responses were independent of response format and evidence supported the interpretation of scores for IdIn as indications of integration of identity. Discussion : Sufficient evidence was found to suggest that these measures assess aspects of PIF. Use of these measures as part of a multidimensional, longitudinal approach to refining understanding of the construct of PIF and developing effective assessment strategies.
