Abstract

We performed small-angle and wide-angle X-ray scattering (SAXS/WAXS) measurements on poly(ethylene terephthalate) (PET) uniaxially drawn from amorphous state above the glass transition temperature. The measurements were done at a new beamline (BL) BL03XU (Frontier Softmaterial Bamline) in the synchrotron radiation X-ray facility, SPring-8, which is dedicated to development of advanced soft materials and belongs to the Advanced Softmaterial BL Consortium. We performed a calculation to reproduce the observed 2D SAXS patterns. Based on the results of the calculation we discussed the distribution of lamellar stacking direction and the relation between the lamellar length and the stacking direction.

Abstract

Recently,hemodialysis patients have been steadily increasing and for such patients,it is very important to control serumphosphate levels in order to decrease cardiovascular risks.Currently,a variety of therapeutic drugs,such as calcium carbonate,lanthanum carbonate and sevelamer hydrochloride,are administered to hemodialysis patients to enhance phosphateclearance from the body.Several anion-exchange resins,among them colestimide and colestyramine,have also been consideredfor the treatment of hyperphosphatemia.However,the relative effectiveness of these drugs in interacting with phosphatehas not been examined.In the present study,we compared the phosphate-binding capacities of therapeutic agents for hyperphosphatemia using aconventional centrifugation method.After mixing drug suspensions and a phosphate solution together,the pH of the mixturewas adjusted to between 1.2 and 8.0.It was then gently shaken for 30 min and subjected to high-speed centrifugation.By determining the phosphate concentration in the resultant supernatant fluid by the molybdenum blue method,the amountsof phosphate bound to each drug were estimated.The phosphate-binding capacity in descending order was as follows :sevelamer hydrochloride (614 mg/g)>lanthanum carbonate (557 mg/g)>calcium carbonate (535 mg/g)>colestimide (256mg/g)>cholestyramine (98 mg/g)=colestipol (93 mg/g),indicating that sevelamer hydrochloride was most potent in bindingphosphate.However,the interaction between sevelamer hydrochloride and phosphate was pH-dependent,as it was forcalcium carbonate and colestimide,and became weaker as the medium pH increased.In contrast,lanthanum carbonate hydrateexhibited potent binding that was independent of pH.The present results therefore suggest that lanthanum carbonatewould be most effective therapeutic agent for inhibiting phosphate absorption in hemodialysis patients.

Abstract

The mechanism and dynamics of the reaction of C-2(X-1 Sigma(+)(g)) with NH(X-3 Sigma(-)) have been investigated using electronic structure methods. The CASPT2 calculations show that C-2 can add to NH without a barrier. Several intermediates involved in the reaction on the lowest triplet potential energy surface were optimized at the B3LYP level and then the potential energy diagram was refined at more accurate levels of theory. In order to understand the reaction dynamics more quantitatively, direct dynamics calculations have been performed at the B3LYP level. It has been found that the HNC molecule is efficiently produced from the C-2 + NH reaction. (c) 2006 Elsevier B.V. All rights reserved.

Abstract

Background: Lenalidomide, an analogue of thalidomide, is frequently used to treat multiple myeloma (MM). Progressive multifocal leukoencephalopathy (PML) is an infectious disease of the central nervous system caused by the reactivation of the JC polyomavirus. Only a few reports have described PML in patients receiving lenalidomide therapy. Case presentation: A 76‐year‐old man with MM presented progressive visual and cognitive impairment during lenalidomide administration. Magnetic resonance imaging showed a lesion in the right parietal lobe. Brain biopsy confirmed the diagnosis of PML. Lenalidomide therapy was discontinued, and oral mefloquine was commenced. He was alive more than 1 year later with mild neurologic decline. Conclusions: Although rare, lenalidomide‐induced PML should be considered in myeloma patients treated with lenalidomide. Abstract

Autophagy is a lysosomal degradation pathway that is implicated in many viral infections. However, its role in West Nile virus (WNV) infection remains controversial. In the present study, we examined the relationship between WNV infection and autophagy in infected cells. We demonstrated that LC3-II expression, a molecular marker for autophagosomal membranes, was enhanced in WNV-infected cells 6 h post-infection. LC3-II expression was further enhanced in WNV-inoculated cells when treated with a lysosomal protease inhibitor. Meanwhile, WNV replication in cells lacking Atg5, an essential factor for autophagy, was increased compared with replication in wild-type cells. In addition, WNV replication was inhibited in cells lacking Atg5 when they were transfected with an ATG5 expression plasmid. These results suggest an antiviral role for autophagy in WNV-infected cells. We also examined which viral replication stages were affected by autophagy by using a Tat-beclin 1 peptide to induce autophagy and pseudo-infectious WNV reporter virus particles (WNV-RVPs) that monitor viral genome replication and gene expression stages via GFP expression. We found that autophagy induction in HeLa cells by Tat-beclin 1 peptide 3 h after WNV inoculation inhibited viral replication, and GFP expression was significantly inhibited in wild-type cells when compared with cells lacking Atg5. Taken together, these results suggest that autophagy is induced by WNV infection, and that this induction inhibits WNV replication at the viral genome replication and gene expression stages. Abstract

Katsushika-Harp Bridge, the curved cable-stayed bridge in Tokyo, Japan, has been monitored for its vibration during earthquakes since 1987 until present. Initially, the main purpose of the monitoring was to observe its dynamic behaviors for design verification. At that time the significant use of vibration data for health monitoring of the large scale structures, which requires a lot of sensors, was not recognized. For these reasons, only a limited number of accelerometers were installed and only limited amount of data are available. Recently, the health condition of the Katsushika-Harp Bridge has become the major concerns. By using the existing limited data, this paper proposes the simple methods to detect anomalies in the health condition. The health condition is described by the Frequency Response Functions (FRFs). The anomalies are described by: (1) the changes of the degree of non-linearity obtained from the Hilbert transform of the FRFs, and (2) the changes of the natural frequencies extracted from the FRFs. The results from the five recent earthquake records are discussed. According to the results, the proposed methods show possibilities to detect the anomalies in the health condition even from the limited data. Nevertheless, the validity of the proposed methods as well as the damage simulation based on the finite element method need to be further investigated.
