Abstract The research examined the ability of each treatment to recover a known quantity of particles and the impact on identifying polymer type using a microscope. By conducting extensive research on the changes induced in the chemical structures and morphologies of microplastics (MPs) there is potential for enhancing the detection and analysis of MPs by incorporating pre-treatment methods in the future. When treating environmental matrices with high organic content, chemical digestion treatment is required to get rid of microplastics and release particles. This type of treatment involves using a range of chemical agents, including acids, bases, and oxidizing agents. Unfortunately, there has been limited research into the chemical resistance of various types of microplastics to these substances. To address this issue, a study was conducted to examine the chemical resistance of four species of microplastics high-density polyethylene (HD-PE), low-density polyethylene (LD-PE), polypropylene (PP) and polyethylene terephthalate (PET) to hydrogen peroxide (H2O2), potassium hydroxide (KOH), nitric acid (HNO3) and acetic acid (CH3COOH). The results showed that acidic and alkaline substances were the most destructive to microplastics, while oxidative reagents resulted in fewer changes to plastic properties. These findings provide valuable insights into the properties of MPs and their response to strong acids, bases and oxidizing agents, which can serve as a reference for future studies on MP pre-treatment. In addition, used as a guideline to update current protocols and ensure that microplastics can be treated without causing damage.

Abstract Glyphosate (GLY), the most widely used herbicide active ingredient (AI) in the world, is frequently detected in aquatic environments where it can affect non-target organisms. Globally, more than 2000 commercial GLY-based herbicides (GBHs) are used to control weeds. Non-target organisms are exposed to complex pesticide formulations under real environmental conditions, but the co-formulants contained in GBHs are classified as so-called inert and inactive ingredients in terms of their biological effects. The main objective of this comprehensive review is to compile the results of aquatic ecotoxicological studies on the side-effects of GLY, GBHs, and their formulating agents. Based on the results demonstrated for a variety of plant and animal aquatic organisms, oxidative stress appears to be a major trigger for these adverse effects, affecting the integrity of DNA and other biochemical functions. Furthermore, there is evidence of impairment of various physiological and behavioral functions. Adverse effects of GLY and GBHs have been observed even at very low concentrations. There are also differences in the sensitivity of the aquatic organisms tested, even with similar lifestyles, habitats or identical taxa. The studies typically investigate the short-term effects of a single exposure to GLY/GBH on a single species, whilst in reality multiple applications of GBHs together with other pesticides are common during a cropping cycle. Moreover, the interactions between GLY/GBHs and other aquatic contaminants are rarely studied. Higher toxicity of GBHs compared to GLY alone has often been observed, demonstrating that co-formulants can be highly toxic on their own and markedly increase the toxicity of the GBH formulation. The possible impurities in GBHs, such as heavy metals, can cause additional problems for the environment and food safety. The widespread and massive use of GBHs leads to increased exposure and environmental hazards. In addition, the need for a revision of the risk assessment system is emphasized. According to the results of aquatic ecotoxicological studies, the current use and pollution of the aquatic environment by GLY/GBHs is highly problematic and cannot be considered environmentally sustainable. It is, therefore, necessary to at least tighten the permitted forms of use.

Abstract Background Parameters adversely affecting the contiguity and accuracy of the assemblies from Illumina next-generation sequencing (NGS) are well described. However, past studies generally focused on their additive effects, overlooking their potential interactions possibly exacerbating one another’s effects in a multiplicative manner. To investigate whether or not they act interactively on de novo genome assembly quality, we simulated sequencing data for 13 bacterial reference genomes, with varying levels of error rate, sequencing depth, PCR and optical duplicate ratios. Results We assessed the quality of assemblies from the simulated sequencing data with a number of contiguity and accuracy metrics, which we used to quantify both additive and multiplicative effects of the four parameters. We found that the tested parameters are engaged in complex interactions, exerting multiplicative, rather than additive, effects on assembly quality. Also, the ratio of non-repeated regions and GC% of the original genomes can shape how the four parameters affect assembly quality. Conclusions We provide a framework for consideration in future studies using de novo genome assembly of bacterial genomes, e.g. in choosing the optimal sequencing depth, balancing between its positive effect on contiguity and negative effect on accuracy due to its interaction with error rate. Furthermore, the properties of the genomes to be sequenced also should be taken into account, as they might influence the effects of error sources themselves.

Abstract Background The measurement of root dielectric response is a useful non-destructive method to evaluate root growth and function. Previous studies tracked root development throughout the plant growing cycle by single-time electrical measurements taken repeatedly. However, it is known that root conductivity and uptake activity can change rapidly, coupled with the day/night cycles of photosynthetic and transpiration rate. Therefore, the low-frequency dielectric monitoring of intact root–substrate systems at minute-scale temporal resolution was tested using a customized impedance measurement system in a laboratory environment. Electrical capacitance (CR) and conductance (GR) and the dissipation factor (DR) were detected for 144 h in potted maize, cucumber and pea grown under various light/dark and temperature conditions, or subjected to progressive leaf excision or decapitation. Photosynthetic parameters and stomatal conductance were also measured to evaluate the stress response. Results The CR and GR data series showed significant 24-h seasonality associated with the light/dark and temperature cycles applied. This was attributed to the diurnal patterns in whole-plant transpiration (detected via stomatal conductance), which is strongly linked to the root water uptake rate. CR and GR decreased during the 6-day dark treatment, and dropped proportionally with increasing defoliation levels, likely due to the loss of canopy transpiration caused by dark-induced senescence or removal of leaves. DR showed a decreasing trend for plants exposed to 6-day darkness, whereas it was increased markedly by decapitation, indicating altered root membrane structure and permeability, and a modified ratio of apoplastic to cell-to-cell water and current pathways. Conclusions Dynamic, in situ impedance measurement of the intact root system was an efficient way of following integrated root water uptake, including diurnal cycles, and stress-induced changes. It was also demonstrated that the dielectric response mainly originated from root tissue polarization and current conduction, and was influenced by the actual physiological activity of the root system. Dielectric measurement on fine timescale, as a diagnostic tool for monitoring root physiological status and environmental response, deserves future attention.

Sour cherry is a non-climacteric fruit that quickly loses its quality after harvest, so effective storage and packaging are essential to minimize postharvest decay. Our study aimed to (i) evaluate fruit decay incidence during shelf-life for both freshly harvested and six-week-cold-stored sour cherry fruits, comparing normal atmospheric conditions with modified atmosphere packaging across three sour cherry cultivars (‘Érdi bőtermő’, ‘Újfehértói fürtös’, and ‘Petri’); and (ii) assess postharvest fruit decay incidence across the following three production systems: conventional, integrated pest management (IPM), and reduced IPM, for the cultivar ‘Érdi bőtermő’. The results showed that modified atmosphere packaging effectively preserved or slightly increased fruit firmness (ranging from 27.1 to 46%) compared to control fruits across all cultivars. Fruit weight loss (ranging from 1.18 to 26.1%) was also significantly reduced under modified atmosphere packaging compared to normal atmospheric storage. Over a 14-day period, the decay incidence of harvested fruits consistently increased, with major losses from day 6 onward (ranging from 17.7 to 77.0% at shelf-life day 14) across all cultivars and years. After six weeks of normal atmospheric cold storage, decay incidence remained low (ranging from 0 to 9.4%) and was comparable across all cultivars. Both normal atmospheric and modified atmosphere packaging storage maintained sour cherry quality during cold storage, but the shelf-life of the fruits was significantly shorter at room temperature, regardless of the storage methods. Fruit decay incidence at harvest was highest in the reduced IPM system (ranging from 3.2 to 6.4%), significantly exceeding those observed in the conventional and IPM systems. In post-MAP cold storage, decay incidence followed the following order: conventional < IPM < reduced IPM, with significant differences observed only between the conventional and reduced IPM systems. In conclusion, our results suggest that sour cherries produced under conventional or IPM systems and stored in modified atmosphere packaging offer an effective postharvest strategy for preserving fruit quality.

Retinal ischemia–reperfusion (I/R) injury is a critical pathogenic mechanism in various eye diseases, and an effective therapeutic strategy remains unresolved. Natural derivatives have recently reemerged; therefore, in our present study, we examined the potential therapeutic effects of a stilbenoid that is chemically related to resveratrol. Pterostilbene, recognized for its anti-inflammatory, anti-carcinogenic, anti-diabetic, and neuroprotective properties, counteracts oxidative stress during I/R injury through various mechanisms. This study explored pterostilbene as a retinoprotective agent. Male Sprague Dawley rats underwent retinal I/R injury and one-week reperfusion and were treated with either vehicle or pterostilbene. After this functional electroretinographical (ERG) measurement, Western blot and histological analyses were performed. Pterostilbene treatment significantly improved retinal function, as evidenced by increased b-wave amplitude on ERG. Histological studies showed reduced retinal thinning and preserved the retinal structure in the pterostilbene-treated groups. Moreover, Western blot analysis revealed a decreased expression of glial fibrillary acidic protein (GFAP) and heat shock protein 70 (HSP70), indicating reduced glial activation and cellular stress. Additionally, the expression of pro-apoptotic and inflammatory markers, poly(ADP-ribose) polymerase 1 (PARP1) and nuclear factor kappa B (NFκB) was significantly reduced in the pterostilbene-treated group. These findings suggest that pterostilbene offers protective effects on the retina by diminishing oxidative stress, inflammation, and apoptosis, thus preserving retinal function and structure following I/R injury. This study underscores pterostilbene’s potential as a neuroprotective therapeutic agent for treating retinal ischemic injury and related disorders.

In 2019, random samples of Panicum miliaceum growing as a weed were surveyed to uncover their virus infections at two locations in Hungary. This pilot study revealed infection with three viruses, two appearing for the first time in the country. As follow-up research, in the summer of 2021, we collected symptomatic leaves of several monocotyledonous plants in the same locations and determined their viromes using small RNA high-throughput sequencing (HTS). As a result, we have identified the presence of wheat streak mosaic virus (WSMV), barley yellow striate mosaic virus (BYSMV), barley virus G (BVG), and two additional viruses, namely Aphis glycines virus 1 (ApGlV1) and Ljubljana dicistrovirus 1 (LDV1), which are described for the first time in Hungary. New hosts of the viruses were identified: Cynodon dactylon is a new host of BYSMV and LDV1, Echinocloa crus-galli is a new host of BVG, ApGlV1 and LDV1, Sorghum halepense is a new host of ApGlV1, and Panicum miliaceum is a new host of LDV1. At the same time, Zea mays is a new host of ApGlV1 and LDV1. Small RNA HTS diagnosed acute infections but failed to detect persistent ones, which could be revealed using RT-PCR. The infection rates at the different locations and plant species were different. The phylogenetic analyses of the sequenced virus variants suggest that the tested monocotyledonous weeds can host different viruses and play a virus reservoir role. Viral spread from the reservoir species relies on the activity of insect vectors, which is why their management requires an active role in plant protection strategies, which need careful planning in the changing environment.

This study explores the efficacy of bio-efficient solutions, specifically plant growth-promoting microorganisms (PGPMs), in sustainable soil management. This research was conducted in 2020. It evaluates the impact of various single microbial inoculants, including Enterobacter ludwigii, Bacillus subtilis, Pseudomonas fluorescens, Kosakonia cowanii, and Trichoderma harzianum, on plant growth soil enzyme activity and organism abundance. Perennial ryegrass and mustard were used as test plants, in controlled environmental conditions. The results show generally positive effects of microbial inoculants on plant biomass (E. ludwigii increased ryegrass biomass by 9.75%, and P. fluorescens increased mustard biomass by up to 38.81% compared to the control) and on soil microbial activities. Our study further investigated the combined application of all these strains in five different soil types and textures. The results highlight the significance of soil physicochemical properties in determining inoculant efficacy; we found that clayey soils with higher colloid content support more robust microbial activity. Additionally, using natural clay minerals like alginite for enhancing soil conditions showed promising interactions with microbial inoculants, although application requires further optimization. These findings suggest that integrating microbial inoculants in sustainable agricultural practices could enhance plant growth, improve soil health, and reduce the need of chemical fertilizers. Future research should aim to refine the combinations and application methods of these bio-efficient solutions for broader agricultural applicability.

Intestinal digestion and absorption are complex processes; thus, it is a challenge to imitate them realistically. There are numerous approaches available, with different disadvantages and advantages. The simplest methods to mimic absorption are the non-cell-based transport models but these lack important characteristics of enterocytes of the intestine. Therefore, the most often used method is to measure absorption through viable mammalian cells (most commonly Caco-2 cells, cultured on membrane insert plates), which not only assures the incorporation of brush border enzymes (responsible for the final digestion of peptides and disaccharides), it also simulates the absorption process. This means that influx/efflux transporter-facilitated transport, carrier-mediated transport, endocytosis, and transcytosis is also imitated besides passive diffusion. Still, these also lack the complexity of intestinal epithelium. Organoids or ex vivo models are a better approach if we want to attain precision but the highest accuracy can be achieved with microfluidic systems (gut-on-a-chip models). We propose that more research is necessary, and food absorption should also be studied on gut-on-a-chips, especially with fragmented organoids. Our review supports the choices of a proper intestinal epithelium model, which may have a key role in functional food development, nutrition studies, and toxicity assessment.

Alterations to intestinal microbiota are assumed to occur in the pathogenesis of inflammatory bowel disease (IBD). This study aims to analyze the association of fecal microbiota composition, body composition, and lipid characteristics in patients with Crohn’s disease (CD). In our cross-sectional study, patients with CD were enrolled and blood and fecal samples were collected. Clinical and endoscopic disease activity and body composition were assessed and laboratory tests were made. Fecal bacterial composition was analyzed using the shotgun method. Microbiota alterations based on obesity, lipid parameters, and disease characteristics were analyzed. In this study, 27 patients with CD were analyzed, of which 37.0% were obese based on visceral fat area (VFA). Beta diversities were higher in non-obese patients (p < 0.001), but relative abundances did not differ. C. innocuum had a higher abundance at a high cholesterol level than Bacillota (p = 0.001, p = 0.0034). Adlercreutzia, B. longum, and Blautia alterations were correlated with triglyceride levels. Higher Clostridia (p = 0.009) and B. schinkii (p = 0.032) and lower Lactobacillus (p = 0.035) were connected to high VFA. Disease activity was coupled with dysbiotic elements. Microbiota alterations in obesity highlight the importance of gut microbiota in diseases with a similar inflammatory background and project therapeutic options.

With the global population projected to reach 8.6 billion by 2050 and urbanization on the rise, sustainable food production in cities becomes imperative. Vertical farming presents a promising solution to meet this challenge by utilizing space-efficient, controlled-environment agriculture techniques. In a vertical farming system, high quality, high nutritional value products can be produced with minimum water consumption, using LEDs as energy-efficient light sources. Microgreens are a new market category of vegetables among sprouts and baby leaf greens. The most critical challenge in their cultivation is the choice of growing medium, lighting, and light spectrum, which affect photosynthesis, plant growth, and yield. This review explores various cultivation methods, including hydroponics, within the context of vertical farming. Using current research, it investigates the effect of LED lighting on the physiological properties and growth of microgreens and baby leaf lettuce, but further research is needed to determine the response of the varieties and the optimal light spectrum ratios to meet their needs.

Purpose: Based on the general formula, which depends on the length and difficulty of the test, the number of respondents and the number of ability levels, this study aims to provide a closed formula for the adaptive tests with medium difficulty (probability of solution is p = 1/2) to determine the accuracy of the parameters for each item and in the case of calibrated items, determine the required test length given number of respondents. Design/methodology/approach: Empirical results have been obtained on computerized or multistage adaptive implementation. Simulation studies and classroom/experimental results show that adaptive tests can measure test subjects' ability to the same quality over half the test length compared to linear versions. Due to the complexity of the problem, the authors discuss a closed mathematical formula: the relationship between the length of the tests, the difficulty of solving the items, the number of respondents and the levels of ability. Findings: The authors present a closed formula that provides a lower bound for the minimum test length in the case of adaptive tests. The authors also present example calculations using the formula, based on the assessment framework of some student assessments to show the similarity between the theoretical calculations and the empirical results. Originality/value: With this formula, we can form a connection between theoretical and simulation results.

Grasping and object manipulation have been considered key domains of Cyber-Physical Systems (CPS) since the beginning of automation, as they are the most common interactions between systems, or a system and its environment. As the demand for automation is spreading to increasingly complex fields of industry, smart tools with sensors and internal decision-making become necessities. CPS, such as robots and smart autonomous machinery, have been introduced in the meat industry in recent decades; however, the natural diversity of animals, potential anatomical disorders and soft, slippery animal tissues require the use of a wide range of sensors, software and intelligent tools. This paper presents the development of a smart robotic gripper for deployment in the meat industry. A comprehensive review of the available robotic grippers employed in the sector is presented along with the relevant recent research projects. Based on the identified needs, a new mechatronic design and early development process of the smart gripper is described. The integrated force sensing method based on strain measurement and magnetic encoders is described, including the adjacent laboratory and on-site tests. Furthermore, a combined slip detection system is presented, which relies on an optical flow-based image processing algorithm using the video feed of a built-in endoscopic camera. Basic user tests and application assessments are presented.
