The article presents a study which aims to examine the accuracy of ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) technology in validating non-targeted and target assays for postmortem drug screening. Targeted detection of over 200 analytes by UPLC-MS-MS was executed with double transition ion monitoring. The results of the confirmation testing reveal a high accuracy rate across all screening assays and detection sensitivity.

Background: Several measures have been developed to examine acute psychedelic effects (e.g. mystical-type and challenging experiences), but no measure assesses acute psychologically insightful experiences that may occur during psychedelic experiences. Aim: The purpose of this study was to develop and examine the psychometric properties of the Psychological Insight Questionnaire. Method: A cross-sectional survey study among psilocybin and LSD users. Respondents (n =1661; M age=22.9, standard deviation=8.5; Caucasian/White=83%; non-Hispanic=91%; men=72%; United States resident=66%) completed an Internet-based survey. Results: The Psychological Insight Questionnaire consists of 23 items with two subscales: (a) Avoidance and Maladaptive Patterns Insights and (b) Goals and Adaptive Patterns Insights. Construct validity of the Psychological Insight Questionnaire was supported by strong correlations of the Psychological Insight Questionnaire (and Avoidance and Maladaptive Patterns Insights and Goals and Adaptive Patterns Insights subscales) scores with the insight subscale of the Session Impacts Scale, and weak-to-moderate correlations with the Mystical Experiences and Challenging Experiences Questionnaires. Furthermore, Psychological Insight Questionnaire (and Avoidance and Maladaptive Patterns Insights and Goals and Adaptive Patterns Insights subscales) scores were moderately-to-strongly correlated with retrospectively reported increases in psychological flexibility, and well-being/life satisfaction that were attributed to a memorable psychedelic experience. Lastly, incremental validity was established showing that the Psychological Insight Questionnaire (and Avoidance and Maladaptive Patterns Insights subscale) scores predict unique variance in changes in psychological flexibility, and Psychological Insight Questionnaire (and Avoidance and Maladaptive Patterns Insights and Goals and Adaptive Patterns Insights subscales) scores predict changes in well-being and life satisfaction, beyond measures of acute mystical-type and challenging effects. Conclusions: The Psychological Insight Questionnaire has the potential to extend the understanding of the acute and enduring effects of psychedelics. Further longitudinal research is necessary to determine the long-term predictive validity of the Psychological Insight Questionnaire and to examine the role of psychological insight in predicting therapeutic outcomes. The clinical application of composites seeks to exploit the mechanical and chemical properties of materials which make up the composite, and in researching polymer composites for biomedical applications the aim is usually to enhance the bioactivity of the polymer, while maintaining the mechanical properties. To that end, in this study medical grade Poly(L-lactic) acid (PLLA) has been reinforced with short phosphate-based glass fibers (PGF). The materials were initially mixed by melting PLLA granules with the short fibers, before being extruded to form a homogenous filament, which was pelletized and used as feedstock for compression moulding. As made the composite materials had a bending strength of 51 MPa ± 5, and over the course of eight weeks in PBS the average strength of the composite material was in the range 20–50 MPa. Human mesenchymal stromal cells were cultured on the surfaces of scaffolds, and the metabolic activity, alkaline phosphatase production and mineralization monitored over a three week period. The short fiber filler made no significant difference to cell proliferation or differentiation, but had a clear and immediate osteoinductive effect, promoting mineralization by cells at the material surface. It is concluded that the PLLA/PGF composite material offers a material with both the mechanical and biological properties for potential application to bone implants and fixation, particularly where an osteoinductive effect would be valuable. • Fabrication via compression moulding of PLLA filled with phosphate glass short fibers, as a new biocomposite material • The use of 5% short fiber filling allows the phosphate glass to present at the surface of the composite. • Minerals released by the phosphate glass provide a clear and immediate osteoinductive effect in vitro. [Display omitted] Vat photopolymerization has garnered interest from pharmaceutical researchers for the fabrication of personalised medicines, especially for drugs that require high precision dosing or are heat labile. However, the 3D printed structures created thus far have been insoluble, limiting printable dosage forms to sustained-release systems or drug-eluting medical devices which do not require dissolution of the printed matrix. Resins that produce water-soluble structures will enable more versatile drug release profiles and expand potential applications. To achieve this, instead of employing cross-linking chemistry to fabricate matrices, supramolecular chemistry may be used to impart dynamic interaction between polymer chains. In this study, water-soluble drug-loaded printlets (3D printed tablets) are fabricated via digital light processing (DLP) 3DP for the first time. Six formulations with varying ratios of an electrolyte acrylate monomer, [2-(acryloyloxy)ethyl]trimethylammonium chloride (TMAEA), and a co-monomer, 1-vinyl-2-pyrrolidone (NVP), were prepared to produce paracetamol-loaded printlets. 1H NMR spectroscopy analysis confirmed the integration of TMAEA and NVP in the polymer, and residual TMAEA monomers were found to be present only in trace amounts (0.71 – 1.37 %w/w). The apparent molecular mass of the photopolymerised polymer was found to exceed 300,000 Da with hydrodynamic radii of 15 – 20 nm, estimated based on 1H DOSY NMR measurements The loaded paracetamol was completely released from the printlets between 45 minutes to 5 hours. In vivo single-dose acute toxicity studies in rats suggest that the printlets did not cause any tissue damage. The findings reported in this study represent a significant step towards the adoption of vat photopolymerization-based 3DP to produce personalised medicines. [ABSTRACT FROM AUTHOR]

Composites have clinical application for their ability to mimic the hierarchical structure of human tissues. In tissue engineering applications the use of degradable biopolymer matrices reinforced by bioactive ceramics is seen as a viable process to increase osteoconductivity and accelerate tissue regeneration, and technologies such as additive manufacturing provide the design freedom needed to create patient-specific implants with complex shapes and controlled porous structures. In this study a medical grade poly(l -lactide) (PLLA) was used as matrix while apatite-wollastonite (AW) was used as reinforcement (5 wt% loading). Premade rods of composite were pelletized and processed to create a filament with an average diameter of 1.6 mm, using a twin-screw extruder. The resultant filament was 3D printed into three types of porous woodpile samples: PLLA, PLLA reinforced with AW particles, and PLLA with short AW fibres. None of the samples degraded in phosphate buffered solution over a period of 8 weeks, and an average effective modulus of 0.8 GPa, 1 GPa and 1.5 GPa was obtained for the polymer, particle and fibre composites, respectively. Composite samples immersed in simulated body fluid exhibited bioactivity, producing a surface apatite layer. Furthermore, cell viability and differentiation were demonstrated for human mesenchymal stromal cells for all sample types, with mineralisation detected solely for biocomposites. It is concluded that both composites have potential for use in critical size bone defects, with the AW fibre composite showing greater levels of ion release, stimulating more rapid cell proliferation and greater levels of mineralisation. Highly branched poly( N -isopropyl acrylamide) additives chain end functionalised with vancomycin have been designed to agglutinate and report on targetted Gram-positive strains of bacteria ( S. aureus ). These branched systems selectively desolvate with temperature or binding interactions depending on their chain architecture. We have prepared samples with three different degrees of branching which have incorporated Nile red acrylate as a low concentration of co-monomer to report upon their solution properties. A linear analogue polymer functionalised with vancomycin along the chain instead of the termini is presented as a control which does not bind to targeted bacteria. These samples were analysed by diffusion NMR spectrometry (DOSY), calorimetry, fluorescence lifetime measurements, optical microscopy and scanning electron microscopy to gain a full understanding of their solution properties. The branched polymers are shown conclusively to have a core-shell structure, where the chain ends are expressed from the desolvated globule even above the lower critical solution temperature - as demonstrated by NMR measurements. The level of desolvation is critically dependent on the degree of branching, and as a result we have found intermediate structures provide optimal body temperature bacterial sensing as a consequence of the Nile red reporting dye.

Whilst the development of advanced organic dots with aggregation-induced emission characteristics (AIE-dots) is being intensively studied, their clinical translation in efficient biotherapeutic devices has yet to be tackled. This study explores the synergistic interplay of oligo(styryl)benzenes (OSBs), potent fluorogens with an increased emission in the aggregate state, and Indocyanine green (ICG) as dual Near Infrared (NIR)-visible fluorescent nanovesicles with efficient reactive oxygen species (ROS) generation capacity for cancer treatment using photodynamic therapy (PDT). The co-loading of OSBs and ICG in different nanovesicles has been thoroughly investigated. The nanovesicles' physicochemical properties were manipulated via molecular engineering by modifying the structural properties of the lipid bilayer and the number of oligo(ethyleneoxide) chains in the OSB structure. Diffusion Ordered Spectroscopy (DOSY) NMR and spectrofluorometric studies revealed key differences in the structure of the vesicles and the arrangement of the OSB and ICG in the bilayer. The in vitro assessment of these OSB-ICG nanovesicles revealed that the formulations can increase the temperature and generate ROS after photoirradiation, showing for the first time their potential as dual photothermal/photodynamic (PTT/PDT) agents in the treatment of prostate cancer.
