202. UV-Light-Activated (Gd,Y)VO4:Eu3+Nanoparticles for Radiotherapy Enhancement

Abstract

Cancerous cells could be resistant to radiotherapy (RT) and require enhanced doses of irradiation exceeding the tolerance of patients. Nanoparticles containing high-Z elements and possessing high X-ray absorption have been recently proposed as nano radio enhancers. Here, we investigate the mechanisms of generation of hydroxyl radicals (•OH) under X-ray irradiation of water solutions containing ultraviolet (UV)-light-pretreated (Gd,Y)VO4:Eu3+nanoparticles. The contributions of two mechanisms in the observed effect are analyzed. It has been concluded that UV-light pretreatment leads to the capture of photoinduced holes and the formation of hydrogen peroxide (H2O2) via photoinduced electron interaction with oxygen molecules, which provide a long-lasting •OH generation effect after the termination of UV irradiation. X-ray irradiation of UV-light-pretreated (Gd,Y)VO4:Eu3+nanoparticles stimulates both hole release and H2O2radiolysis, enhancing sufficiently the effect of •OH generation.

Chemical research shows that 54% of all oxygen atoms found in 4-(1-hydroxy-1-methylethyl)benzoic acid originate from water when 4-isopropylbenzoic acid undergoes monopersulfate oxidation in H2(super 18)O and metalloporphyrin catalysis. Radioactive labelling techniques were used to study the redox tautomerism reaction in the active hydroxo-metal-oxo porphyrin complex.

Abstract

Naphthoquinones such as 2-methyl-1,4-naphthoquinone and 6-methyl-1,4-naphthoquinone were synthesized by the metalloporphyrin-catalyzed oxidation of 2-methylnaphthalene. The reaction of 2-methylnaphthalene with potassium monopersulfate in aqueous solutions with water-soluble metalloporphyrins involved two oxygen transfer reactions. Furthermore, the reaction involves a redox tautomerism mechanism that leads to the formation of the naphthoquinones.

Abstract

Intramolecular kinetic isotope effects (KIEs) are detected in saturated alkane hydroxylations, which are catalyzed by iron and manganese tetraarylporphyrin complexes. Several ion porphyrins demonstrate a nature similar to that of the high-valent metal-oxo species when initiated by NaOCl and PhIO, as revealed by the KIE values of metalloporphyrin catalysts linked to various oxidants. A tunneling effect probably causes the high KIEs of Fe(TMP)Cl in association with PhIO or NaOCl.

Abstract

This study hypothesized that there is an association between chronic stress (as indexed by resting amygdalar activity [AmygA]), hematopoietic system activity (HMPA), and subclinical cardiovascular indexes (aortic vascular inflammation [VI] and noncalcified coronary plaque burden [NCB]) in psoriasis (PSO). The study also hypothesized that treatment of PSO would improve these parameters.PSO is a stress-related chronic inflammatory condition that is associated with increased prevalence of subclinical cardiovascular disease (CVD). In individuals without PSO, stress has been linked to CVD through a serial biological pathway that involves the amygdala, hematopoietic tissues, and atherosclerotic plaques.A total of 164 consecutive patients with PSO and 47 healthy volunteers underwent 18-fluorodeoxyglucose positron emission tomography/computed tomography scans for assessment of AmygA, HMPA, and VI, as well as coronary computed tomography angiography scans for quantifying NCB. Furthermore, a consecutive subset of 30 patients with severe PSO (Psoriasis Area Severity Index Score10) were followed at 1 year to assess the relationship between skin disease improvement and AmygA, HMPA, VI, and NCB.The PSO cohort was middle-aged (mean age: 50 years), had low cardiovascular risk (Framingham risk score: median: 3) and had mild to moderate PSO activity (median Psoriasis Area Severity Index Score: 5.6). AmygA was higher in patients with PSO compared to volunteer participants. AmygA was associated with HMPA (bone marrow activity: β = 0.20, p = 0.01) and subclinical CVD (VI: β = 0.31, p 0.001; NCB: β = 0.27, p 0.001) The AmygA-CVD association was in part mediated by HMPA (VI: 20.9%, NCB: 36.7%). Following 1 year of PSO treatment in those with severe disease, improvement in skin disease was accompanied by a reduction in AmygA, bone marrow activity, and VI, with no progression of NCB.In PSO, a chronic inflammatory disease state, AmygA, which is a manifestation of chronic stress, substantially contributes to the risk of subclinical CVD. Additional studies that use psychometric measures of stress are required to explore therapeutic impact.

Abstract

A combination of MS and computation on μ-nitrido bridged diiron complexes reveals H2O2 binding to the complex and generates an oxidant capable of oxidizing methane., Iron(iv)–oxo species have been identified as the active intermediates in key enzymatic processes, and their catalytic properties are strongly affected by the equatorial and axial ligands bound to the metal, but details of these effects are still unresolved. In our aim to create better and more efficient oxidants of H-atom abstraction reactions, we have investigated a unique heteroleptic diiron phthalocyanine complex. We propose a novel intramolecular approach to determine the structural features that govern the catalytic activity of iron(iv)–oxo sites. Heteroleptic μ-nitrido diiron phthalocyanine complexes having an unsubstituted phthalocyanine (Pc1) and a phthalocyanine ligand substituted with electron-withdrawing alkylsulfonyl groups (PcSO2R) were prepared and characterized. A reaction with terminal oxidants gives two isomeric iron(iv)–oxo and iron(iii)–hydroperoxo species with abundances dependent on the equatorial ligand. Cryospray ionization mass spectrometry (CSI-MS) characterized both hydroperoxo and diiron oxo species in the presence of H2O2. Sufficient amounts of these transient species were trapped in the quadrupole region of the mass-spectrometer and underwent a CID-MS/MS fragmentation. Analyses of fragmentation patterns indicated a preferential formation of hydroperoxo and oxo moieties at more electron-rich iron sites of both heteroleptic μ-nitrido complexes. DFT calculations show that both isomers are close in energy. However, the analysis of the iron(iii)–hydroperoxo bond strength reveals major differences for the (Pc1)FeN(PcSO2R)FeIIIOOH system as compared to (PcSO2R)FeN(Pc1)FeIIIOOH system, and, hence binding of a terminal oxidant will be preferentially on more electron-rich sides. Subsequent kinetics studies showed that these oxidants are able to even oxidize methane to formic acid efficiently.

Abstract

Abstract Background An evolutionary classification of genes from sequenced genomes that distinguishes between orthologs and paralogs is indispensable for genome annotation and evolutionary reconstruction. Shortly after multiple genome sequences of bacteria, archaea, and unicellular eukaryotes became available, an attempt on such a classification was implemented in Clusters of Orthologous Groups of proteins (COGs). Rapid accumulation of genome sequences creates opportunities for refining COGs but also represents a challenge because of error amplification. One of the practical strategies involves construction of refined COGs for phylogenetically compact subsets of genomes. Results New Archaeal Clusters of Orthologous Genes (arCOGs) were constructed for 41 archaeal genomes (13 Crenarchaeota, 27 Euryarchaeota and one Nanoarchaeon) using an improved procedure that employs a similarity tree between smaller, group-specific clusters, semi-automatically partitions orthology domains in multidomain proteins, and uses profile searches for identification of remote orthologs. The annotation of arCOGs is a consensus between three assignments based on the COGs, the CDD database, and the annotations of homologs in the NR database. The 7538 arCOGs, on average, cover ~88% of the genes in a genome compared to a ~76% coverage in COGs. The finer granularity of ortholog identification in the arCOGs is apparent from the fact that 4538 arCOGs correspond to 2362 COGs; ~40% of the arCOGs are new. The archaeal gene core (protein-coding genes found in all 41 genome) consists of 166 arCOGs. The arCOGs were used to reconstruct gene loss and gene gain events during archaeal evolution and gene sets of ancestral forms. The Last Archaeal Common Ancestor (LACA) is conservatively estimated to possess 996 genes compared to 1245 and 1335 genes for the last common ancestors of Crenarchaeota and Euryarchaeota, respectively. It is inferred that LACA was a chemoautotrophic hyperthermophile that, in addition to the core archaeal functions, encoded more idiosyncratic systems, e.g., the CASS systems of antivirus defense and some toxin-antitoxin systems. Conclusion The arCOGs provide a convenient, flexible framework for functional annotation of archaeal genomes, comparative genomics and evolutionary reconstructions. Genomic reconstructions suggest that the last common ancestor of archaea might have been (nearly) as advanced as the modern archaeal hyperthermophiles. Abstract

This article is dedicated to the problem of interaction between regional geological practices and geological education in the classical university exemplified by Siberian educational entities. Turning to real geological works in classical universities can reveal new aspects of geological knowledge for historians and methodologists of geology. It is important to emphasize that regional works are fundamental for all types of geological practices. It was proved that decline in funding for regional geological works that resulted from Russia’s transition to a market economy, negatively affects geological education development in classical universities of our country. Decline in funding for regional geological works forces students to attend professional retraining after graduation. This tendency can be dangerous for the economy as it can result in serious economic losses and lack of explored resources portfolio for mining.
