Abstract

Background Variability in standard-of-care classifications precludes accurate predictions of early tumor recurrence for individual patients with meningioma, limiting the appropriate selection of patients who would benefit from adjuvant radiotherapy to delay recurrence. We aimed to develop an individualized prediction model of early recurrence risk combining clinical and molecular factors in meningioma. Methods DNA methylation profiles of clinically annotated tumor samples across multiple institutions were used to develop a methylome model of 5-year recurrence-free survival (RFS). Subsequently, a 5-year meningioma recurrence score was generated using a nomogram that integrated the methylome model with established prognostic clinical factors. Performance of both models was evaluated and compared with standard-of-care models using multiple independent cohorts. Results The methylome-based predictor of 5-year RFS performed favorably compared with a grade-based predictor when tested using the 3 validation cohorts (ΔAUC = 0.10, 95% CI: 0.03–0.018) and was independently associated with RFS after adjusting for histopathologic grade, extent of resection, and burden of copy number alterations (hazard ratio 3.6, 95% CI: 1.8–7.2, P < 0.001). A nomogram combining the methylome predictor with clinical factors demonstrated greater discrimination than a nomogram using clinical factors alone in 2 independent validation cohorts (ΔAUC = 0.25, 95% CI: 0.22–0.27) and resulted in 2 groups with distinct recurrence patterns (hazard ratio 7.7, 95% CI: 5.3–11.1, P < 0.001) with clinical implications. Conclusions The models developed and validated in this study provide important prognostic information not captured by previously established clinical and molecular factors which could be used to individualize decisions regarding postoperative therapeutic interventions, in particular whether to treat patients with adjuvant radiotherapy versus observation alone.

Abstract

This paper reviews and reinterprets the archaeological evidence of the building materials of the Antonine Wall. It presents a new perspective on the structural and geotechnical performance of the Wall with a particular focus on turf and earthen building materials and the role of the stone base in the moisture management of this largest turf-built monument surviving in the Roman world. Results demonstrate the heterogeneous character of the Wall and highlight the complexities of its construction. New avenues of future work are proposed as part of the Earthen Empire Leverhulme Research Project.

Abstract

There is growing awareness across the neuroscience community that the replicability of findings about the relationship between brain activity and cognitive phenomena can be improved by conducting studies with high statistical power that adhere to well-defined and standardised analysis pipelines. Inspired by recent efforts from the psychological sciences, and with the desire to examine some of the foundational findings using electroencephalography (EEG), we have launched #EEGManyLabs, a large-scale international collaborative replication effort. Since its discovery in the early 20th century, EEG has had a profound influence on our understanding of human cognition, but there is limited evidence on the replicability of some of the most highly cited discoveries. After a systematic search and selection process, we have identified 27 of the most influential and continually cited studies in the field. We plan to directly test the replicability of key findings from 20 of these studies in teams of at least three independent laboratories. The design and protocol of each replication effort will be submitted as a Registered Report and peer-reviewed prior to data collection. Prediction markets, open to all EEG researchers, will be used as a forecasting tool to examine which findings the community expects to replicate. This project will update our confidence in some of the most influential EEG findings and generate a large open access database that can be used to inform future research practices. Finally, through this international effort, we hope to create a cultural shift towards inclusive, high-powered multi-laboratory collaborations. © 2021 The Authors.

Abstract

In water distribution systems, valves play a crucial role in system reliability and security by providing a shutoff function when it is necessary to isolate subsystems. In this paper failure of shutoff valves to close is considered. The failure impact is quantified in terms of the expanding subnetwork and the increased number of customers out of service. To evaluate a system-wide valve failure impact, three methods are suggested: segment--valve matrix, decision tree analysis, and simulation. The segment--valve matrix shows the propagation of failure through the network as valves fail to close. The decision tree enumerates all possible valve failure combinations and corresponding event probabilities. Because the complete enumeration can be unwieldy, simulation procedures are offered that approximate the enumeration results closely. Abstract

A previously published measurement of the ground state resonance of 10He, populated by a reaction of a 59 MeV/u 14Be beam on a deuterated polyethylene target, was further analyzed to search for 4n emission resulting from 2p removal. No evidence for 4n events was found. A lower limit of about 1 MeV was determined for a possible resonance in 12He.
