Abstract

International audience; Burkina Faso is a highly endemic area for Hepatitis B virus (HBV) which remains a major challenge for blood safety with >13% of candidate blood donors being chronically infected. However, little is known about the molecular epidemiology of the viral strains currently circulating. In this study, 99 HBV strains from HBsAg positive candidate blood donors in Ougadougou were genetically characterized by sequencing the pre-S/S region of the viral genome. Phylogenetic analyses revealed a 25% prevalence of HBV quasi-subgenotype A3 (A3QS ) co-circulating with the confirmed dominant HBV genotype E (72%). HBV/A3QS sequences formed a sub-cluster closely related to West-African sequences previously characterized, and showed a low intra-group genetic diversity (0.75%) suggesting a relatively recent spreading of HBV/A3QS strains in Burkina Faso. Low genetic diversity of genotype E strains compared to A3QS was confirmed. Mixed infections with the two genotypes were identified in 3% of the donors tested and contributed to artifacts during PCR amplification of the viral genome leading to erroneous apparent intergenotype recombinant sequences. While the co-circulation of two HBV genotypes in a restricted area may favor the emergence of intergenotype recombinant variants, strictly controlled molecular experimental procedures should be used to accurately characterize HBV circulating recombinant forms. Abstract

The present study carried out in the Saint Camille Medical Centre of Ouagadougou and related to the prevalence of Rotaviruses, Adenoviruses and enteric parasites infections among HIV-seropositive and HIV-seronegative children allowed to include 66 children aged from 2 to 60 months presenting acute diarrhea. The results revealed that 10.60% were infected by HIV. Stool samples analysis by direct microscopy and immunochromatographic tests (Rota-strip and Adeno-strip) showed 42.42% of global infections, among these, 18.18% parasitic infections and 24.24% viral infections, predominantly rotaviruses (22.73 and 1.52% for Rotaviruses and Adenoviruses, respectively). Among parasitic infections, Protozoan were most common than Helminthes (12.12% versus 6.06%). According to present results, Rotaviruses appeared as the major etiological agents in children aged from 2 to 11 months. However high rate of protozoan and helminthes was found in children aged from 12 to 60 months. Thus, virus, particularly Rotavirus must be painstaking as the principal etiologic agent of infant diarrhea in our country.

Abstract

INTRODUCTION: les plantes constituent une grande source de principes actifs qui peuvent être utilisés pour traiter de nombreuses maladies, dont le diabète. L'objectif de cette étude était de recenser les plantes utilisées en médecine traditionnelle pour traiter le diabète dans la région Maritime du Togo. METHODES: de janvier 2013 à juin 2014, une enquête ethnobotanique a été réalisée auprès de 164 guérisseurs traditionnels dans la région Maritime par des interviews directes à l'aide d'un questionnaire semi structuré. RESULTATS: les données recueillies ont permis d'identifier 112 espèces végétales appartenant à 51 familles. Les familles les plus représentées ont été les Caesalpiniaceae / Fabaceae avec 9 espèces, suivie des Euphorbiaceae et des Compositae avec 8 espèces chacune. Les espèces les plus citées ont été Allium sativum, Alium cepa, Guilandina bonduc, Moringa oleifera et de Picralima nitida qui ont eu une valeur usuelle de 0,05. En termes de recettes, 132 recettes sont préparées à partir des 112 espèces de plantes. Les recettes à plantes uniques ont été au nombre de 78, tandis que 54 recettes sont obtenues par des associations de plantes. Les parties de plantes les plus utilisées ont été les feuilles suivies par les racines. La principale méthode de préparation reste la décoction. CONCLUSION: la région maritime du Togo dispose d'une biodiversité floristique importante en matière de plantes antidiabétiques. Ces résultats constituent une bonne base de données pour le criblage biologique dans la recherche de molécules antidiabétiques à base des plantes.

Abstract

Introduction: cette etude a eu pour objectifs de diagnostiquer la tuberculose pulmonaire par l'examen microscopique et par la PCR des crachats et de determiner les bases moleculaires de la resistance a la rifampicine et a l'isoniazide. Methodes: le diagnostic du Complexe Mycobacterium Tuberculosis (CMTB) a ete effectue par microscopie apres coloration au Ziehl Nielsen et par PCR en temps reel en utilisant le kit d'identification du complexe MTB (Sacace Biotechnologie, Italie). Les resistances a la Rifampicine et a l'Isoniazide ont ete etudiees par la technique de la PCR en utilisant le kit MTB resistance 8 (Sacace, Biotechnologie). Resultats: sur les 59 patients diagnostiques pour la tuberculose pulmonaire, 59,3% etaient positifs en microscopie optique et 44,1% etaient positifs par PCR en Temps reel. Les resistances a la rifampicine ( rpo B) et a l'isoniazide (k at G et inh A) ont ete observees chez 9 patients. La resistance a la rifampicine etait due aux mutations (Asp516Val, Ser531Trp, Leu533Pro) et celle a l'isoniazide par les substitutions Ser315Thr du gene kat G et C209T du gene inh A. Les multi resistances a la rifampicine et a l'isoniazide ont ete observees dans 55,5% des echantillons et concernaient les associations : rop BAsp513Val + inh AC209T et rpo BLeu533Pro + kat GSer315Thr. Conclusion: la PCR en temps reel qui permet l'identification des alleles mutants rpoB, katG et inhA de M. tuberculosis est un outil de diagnostic epidemiologique de grande importance car elle permet de determiner le niveau de resistance a la rifampicine et a l'isoniazide. Abstract

Background: Burkina-Faso's HIV/AIDS program is one of the most successful in Africa, with a declining HIV prevalence and treatment outcomes that rival those of developed countries. Prevention of mother-to-child transmission (PMTCT) guidelines in Burkina-Faso, initiated in the year 2000, were revised in 2004, 2006 and 2010. The guideline document has since undergone several stages of improvement, largely based on recommendations from WHO, with adaptations by local experts in the field. Option B+ adopted since August 2014 in Burkina-Faso has enabled maintenance of mothers on longer treatment and increasing their survival and that of their children. Through this review, we describe the achievements and challenges of HIV PMTCT programs in Burkina-Faso. Aims of study: This study had the following objectives: 1) describing the historical perspective of PMTCT implementation in Burkina-Faso; 2) presenting the effectiveness of interventions at improving PMTCT service delivery and promoting retention of mothers and babies in care; and 3) determining the impact of male partner involvement on PMTCT in Burkina-Faso. Abstract

Sapoviruses (Says) are a common cause of gastroenteritis in children. In sub-Saharan Africa, there is a scarcity of information regarding SaV as an etiological agent of diarrhea. Here, we investigated the prevalence, molecular characterization and clinico-epidemiological features of SaV infections in children less than 5 years of age with diarrhea in Burkina Faso. We further investigated the role of type 1 histo blood group antigens as susceptibility factors. In total, 309 fecal and 208 saliva samples from diarrheal children in Ouagadougou, Burkina Faso, were collected between May 2009 and March 2010. Say was detected using real-time PCR, and genogrouped/genotyped by PCR or sequencing. Saliva samples were ABO, Lewis and secretor phenotyped using in house ELISA assays. We found a high prevalence (18%) and large genetic diversity with all 4 human genogroups, and 9 genotypes/genoclusters circulating during the study period. The SaV infections were generally associated with milder symptoms, and neither ABH, Lewis or secretor phenotypes affected susceptibility to Say infections. Abstract

Background. The live oral rotavirus (RV) vaccines have shown a reduced efficacy in Africa. Recent in vitro studies have shown binding of the RV surface protein (VP4) to histo-blood group antigens (HBGAs) in an RV genotype-dependent manner, suggesting them to be putative receptors for RV. The diversity of HBGA phenotypes in different ethnic populations, combined with prevalence/absence of specific RV genotypes, led us to hypothesize whether the genetic variations in HBGAs in a population limit susceptibility to certain RV genotypes, plausibly leading to reduced vaccine efficacy. Methods. Association between HBGAs status and susceptibility to RV P genotypes was investigated in children in Burkina Faso and Nicaragua. In total, 242 children with diarrhea in Burkina Faso and Nicaragua were investigated, 93 of whom were RV positive. Results. In Burkina Faso, the P[8] RV strains (n = 27) infected only Lewis-and secretor-positive children (27/27; P less than .0001), but no Lewis-negative children. In contrast, the P[6] strains (n = 27) infected predominantly Lewis-negative children (n = 18; P less than.0001) but also Lewis-positive children, irrespective of their secretor status. The results from Nicaragua confirmed that all P[8]-infected children (n = 22) were secretor Lewis positive. Conclusions. As VP4 of genotype P[8] is a component of current RV vaccines, our finding that Lewis-negative children are resistant to P[8] strains provides a plausible explanation for the reduced vaccine efficacy in populations with a high percentage of Lewis-negative individuals, such as in Africa. Abstract

Background: A significant fraction of cases of diarrhea, a leading cause of childhood mortality worldwide, remain unexplained. Objectives: To identify viruses in unexplained cases of diarrhea using an unbiased metagenomics approach. Study design: Viral nucleic acids were enriched from the feces from 48 cases of unexplained diarrhea from Burkina Faso, sequenced, and compared against all known viral genomes. Results: The full genome of a highly divergent astrovirus was sequenced in a sample co-infected with parechovirus 1. RT-PCR identified a single astrovirus infection in these 48 patients indicating a low prevalence. Human astrovirus-BF34 was most closely related to mamastrovirus species 8 and 9 also found in human with which it shared 62%, 74%, and 57% amino acid identities over its protease, RNA dependent RNA polymerase and capsid proteins, respectively. Conclusions: Burkina Faso astrovirus is proposed as prototype for a novel species in the genus Mamastrovirus, here tentatively called Mamastrovirus 20, representing the fifth human astrovirus species.

Abstract

The aim of this study is to compare/contrast the colostrum lipid composition of 53 Burkinabe women, collected in 2005 at the Maternity of Centre Medical Saint Camille in Ouagadougou (Burkina Faso), with similar data obtained in breast milk, five years ago and then to show the evolution of this important aliment. The fatty acid composition of colostrum samples was determined by Gas-liquid Chromatography-Mass Spectrometry. Saturated lipids (C8:0-C:14-0) showed a progressive increasing trend in the Burkinabe woman colostrum with respect those already measured five years ago. The C15:0-C24:0 fractions were found costantly higher, but their trends were in progressive decrement. The 18:2n-6 fraction (linoleic acid) reached the highest value in the third day post partum. The 18:3n-3 was constantly higher in the second and third days. The 20:4n-6 (arachidonic acid) and LC n-6 PUFA were lower ever since the first day, but with a trend to increase. Also 22: 6n-3 and LC n-3 PUFA were costantly lower. The 18:2n-6/18:3n-3 and LC n-6/LC n-3 ratios were lower and higher, respectively, if compared with those already measured five years ago. These results suggest the need to improve alimentary habits of mothers in order to restore the balanced n-6/n3 PUFA ratio in their colostrums.

250. Antioxidant and antibacterial activities of polyphenols from ethnomedicinal plants of Burkina Faso

Abstract

Polyphenols from four medicinal plants of Burkina Faso, Combretum micranthum, Khaya senegalensis, Pterocarpus erinaceus and Sida acuta, were screened for their antioxidant and antimicrobial activities against pathogenic bacteria. The medicinal plants displayed different polyphenols contents and antioxidant activities. The bark of P. erinaceus had the highest antioxidant activity. Some microorganisms were susceptible to polyphenol extracts with minimal bactericidal concentration values between 20 and 2000 mg/ml while other microorganisms appeared to be resistant to the extracts. Microbicide and microbiostatic activities of the extracts were dependent on the type of strains. Results suggest that these plants are not only interesting sources for antimicrobial activities but also potential sources of phenolic antioxidants.Key words: Antioxidants, polyphenols, antimicrobials, medicinal plants.African Journal of Biotechnology Vol. 4 (8), pp. 823-828
