202. Exploratory assessment to evaluate seed sprouting under elevated CO₂ revealed improved biomass, physiology, and nutritional value of Trachyspermum ammi

Abstract

Elevated atmospheric CO2 (eCO(2)) can significantly enhance plant metabolism and improve their nutritional quality. Although several studies addressed the effect of eCO(2) on plants, to our knowledge, there are no studies that have applied eCO(2) to improve growth, chemical composition, and biological properties of ajwain (Trachyspermum ammi) during sprouting. Herein, eCO(2) (620 mu mol mol(-1)) was employed to enhance photosynthetic reactions. Improved photosynthesis induced primary and bioactive secondary metabolism, which led to improved biological activities of T. ammi sprouts in comparison with control sprouts and seeds. eCO(2)-treated sprouts showed significantly (p < 0.05) higher levels of most measured nutritional primary and secondary metabolites like soluble sugars, essential amino acids, organic acids, essential oils, phenolics, and flavonoids than control sprouts, which had significantly higher values than control seeds. eCO(2) significantly improved the antimicrobial properties of T. ammi sprouts against 11 out of 13 microbial species than control sprouts, which had more potent antimicrobial activities than seeds. The significant increase in the antioxidant potential of treated sprouts was indicated by improved levels of ferric reducing antioxidant power (FRAP), DPPH, and oxygen radical absorbance capacity (ORAC). In addition, the anticancer activity against four different human tumor cell lines was significantly (p < 0.05) boosted by both sprouting and eCO(2) exposure. Thus, the present study confirms the synergistic effect of sprouting with eCO(2) exposure as promising approaches to produce ajwain sprouts with rich nutraceuticals, effective phytochemicals, and enhanced biological activities.

Abstract

Plastic pollution is a worldwide concern causing the death of animals (mainly aquatic fauna) and environmental deterioration. Plastic recycling is, in most cases, difficult or even impossible. For this reason, new research lines are emerging to identify highly biodegradable bioplastics or plastic formulations that are more environmentally friendly than current ones. In this context, microbes, capable of synthesizing bioplastics, were revealed to be good models to design strategies in which microorganisms can be used as cell factories. Recently, special interest has been paid to haloarchaea due to the capability of some species to produce significant concentrations of polyhydroxyalkanoate (PHA), polyhydroxybutyrate (PHB), and polyhydroxyvalerate (PHV) when growing under a specific nutritional status. The growth of those microorganisms at the pilot or industrial scale offers several advantages compared to that of other microbes that are bioplastic producers. This review summarizes the state of the art of bioplastic production and the most recent findings regarding the production of bioplastics by halophilic microorganisms with special emphasis on haloarchaea. Some protocols to produce/analyze bioplastics are highlighted here to shed light on the potential use of haloarchaea at the industrial scale to produce valuable products, thus minimizing environmental pollution by plastics made from petroleum.

Abstract

Soil salinity is the major abiotic stress that disrupts nutrient uptake, hinders plant growth, and threatens agricultural production. Plant growth-promoting rhizobacteria (PGPR) are the most promising eco-friendly beneficial microorganisms that can be used to improve plant responses against biotic and abiotic stresses. In this study, a previously identified B. thuringiensis PM25 showed tolerance to salinity stress up to 3M NaCl. The Halo-tolerant Bacillus thuringiensis PM25 demonstrated distinct salinity tolerance and enhance plant growth-promoting activities under salinity stress. Antibiotic-resistant Iturin C (ItuC) and bio-surfactant-producing (sfp and srfAA) genes that confer biotic and abiotic stresses were also amplified in B. thuringiensis PM25. Under salinity stress, the physiological and molecular processes were followed by the over-expression of stress-related genes (APX and SOD) in B. thuringiensis PM25. The results detected that B. thuringiensis PM25 inoculation substantially improved phenotypic traits, chlorophyll content, radical scavenging capability, and relative water content under salinity stress. Under salinity stress, the inoculation of B. thuringiensis PM25 significantly increased antioxidant enzyme levels in inoculated maize as compared to uninoculated plants. In addition, B. thuringiensis PM25-inoculation dramatically increased soluble sugars, proteins, total phenols, and flavonoids in maize as compared to uninoculated plants. The inoculation of B. thuringiensis PM25 significantly reduced oxidative burst in inoculated maize under salinity stress, compared to uninoculated plants. Furthermore, B. thuringiensis PM25-inoculated plants had higher levels of compatible solutes than uninoculated controls. The current results demonstrated that B. thuringiensis PM25 plays an important role in reducing salinity stress by influencing antioxidant defense systems and abiotic stress-related genes. These findings also suggest that multi-stress tolerant B. thuringiensis PM25 could enhance plant growth by mitigating salt stress, which might be used as an innovative tool for enhancing plant yield and productivity. Abstract

Algae are frequently exposed to toxic metals, and zinc (Zn) is one of the major toxicants present. We exposed two green microalgae, Chlorella sorokiniana and Scenedesmus acuminatus, to sub-lethal concentrations (1.0 and 0.6 mM) of Zn for seven days. Algal responses were analysed at the level of growth, oxidative stress, and antioxidants. Growth parameters such as cell culture yield and pigment content were less affected by Zn in C. sorokiniana, despite the fact that this alga accumulated more zinc than S. acuminatus. Also, C. sorokiniana, but not S. acuminatus, was able to acclimatize during long-term exposure to toxic concentrations of the test metals (specific growth rate (µ) was 0.041/day and total chlorophyll was 14.6 mg/mL). Although, Zn induced oxidative stress in both species, C. sorokiniana experienced less stress than S. acuminatus. This could be explained by a higher accumulation of antioxidants in C. sorokiniana, where flavonoids, polyphenols, tocopherols, glutathione (GSH) and ascorbate (ASC) content increased. Moreover, antioxidant enzymes glutathione S transferase (GST), glutathione reductase (GR), superoxide dismutase (SOD), peroxidase (POX) and ascorbate peroxidase (APX), showed increased activities in C. sorokiniana. In addition to, and probably also underlying, the higher Zn tolerance in C. sorokiniana, this alga also showed higher Zn biosorption capacity. Use of C. sorokiniana as a bio-remediator, could be considered.

243. Green fabrication of nanocomposite doped with selenium nanoparticle–based starch and glycogen with its therapeutic activity: antimicrobial, antioxidant, and anti-inflammatory in vitro

Abstract

Synthesis of nanoparticles (NPs) was market strictly attention in the materials and their applications where offered multifunction materials. The synthesis process of NPs takes a wide range of methodology. Nowadays, the green synthesis methods are the pioneer as well as acceptable towered the environment. In this present work, the green nanocomposites were prepared using an ecofriendly method. Additionally, the prepared nanocomposite was loaded with selenium nanoparticles (SeNPs) using the green process as well. The produced nanocomposites were characterized using UV–visible, attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy, X-ray diffraction (XRD), scan electron microscope (SEM), energy-dispersive X-ray analyses (EDX), transmission electron microscope (TEM), and dynamic light scattering (DLS). The prepared nanocomposite recorded the nanostructure around 90 nm, and the selenium loaded nanocomposite 78 nm. The loaded nanocomposite (LNC) with SeNPs displayed good antioxidant activity using 2,2-diphenyl-1- picrylhydrazyl (DPPH) radical scavenging test with IC5068 µg/mL compared with free nanocomposite (FNC) IC50(239.1 µg/mL). Anti-inflammatory potential represented by hemolysis inhibition % was promoted by LNC with SeNPs (31.7% and 69.1% at 100 and 1000 µg/mL, respectively) compared to hemolysis inhibition 20.6 and 64.2% at 100 and 1000 µg/mL of FNC, respectively. B. subtilis, S. aureus, E. faecalis, P. vulgaris, P. aeruginosa, E. coli, S. typhimurium, and C. albicanswere more affected by LNC than FNC with increasing fold 16.55, 26.75, 30.60, 41.81, 53.10, 40.77, 10.98, and 41.10%, respectively. Unfortunately, neither FNC nor LNC showed any activity against black mould M. circinelloideand mycotoxogenic fungi A. nigerand A. flavus. From the current findings, LNC with SeNPs exhibited highest in vitro therapeutic potential than FNC.
