The lymphatic system is a common route for the spread of cancer and the identification of lymph node metastases is a key task during cancer surgery. This paper demonstrates the use of optical coherence tomography to construct parametric images of lymph nodes. It describes a method to automatically estimate the optical attenuation coefficient of tissue. By mapping the optical attenuation coefficient at each location in the scan, it is possible to construct a parametric image indicating variations in tissue type. The algorithm is applied to ex vivo samples of human axillary lymph nodes and validated against a histological gold standard. Results are shown illustrating the variation in optical properties between cancerous and healthy tissue.

Leptin, a hormone produced by adipose tissue, regulates energy balance in the hypothalamus and is involved in fertility, immune response and carcinogenesis. The existence of disorders related to leptin deficit and leptin overabundance calls for the development of drugs activating or inhibiting the leptin receptor (ObR). We synthesized four proposed receptor-binding leptin fragments (sites I, IIa and IIb, III), their reportedly antagonist analogs, and a peptide chimera composed of the two discontinuous site II arms. To assess the pharmacological utility of leptin fragments, we studied the peptides' ability to stimulate the growth of ObR-positive and ObR-negative cells. The combined site II construct and site III derivatives selectively reversed leptin-induced growth of ObR-positive cells at mid-nanomolar concentrations. However, these peptides appeared to be partial agonists/antagonists as they activated cell growth in the absence of exogenous leptin. A designer site III analog, featuring non-natural amino acids at terminal positions to decrease proteolysis and a blood-brain barrier (BBB) penetration-enhancing carbohydrate moiety, proved to be full agonist to ObR, i.e., stimulated proliferation of different ObR-positive but not ObR-negative cells in the presence or absence of leptin. This glycopeptide bound to isolated ObR on solid-phase assays and activated ERK-1/2 signaling in ObR-positive MCF-7 cells at 100-500 nM concentrations. The glycopeptide was stable in mouse serum, readily crossed endothelial/astrocyte cell layers in a cellular BBB model, and was distributed into the brain of Balb/c mice after intraperitoneal administration. These characteristics suggest a potential pharmaceutical utility of the designer site III glycopeptide in leptin-deficient diseases.

The photodynamic activity of a carrier-sensitizer system consisting of heterotopic colloidal nanoparticles (diameter 100-1000 nm) of a cationic amphiphilic cyclodextrin, heptakis(2-omega-amino-O-oligo(ethylene oxide)-6-hexylthio)-beta-CD (SC6CDNH2) encapsulating the anionic 5,10,15,20-tetrakis(4-sulfonatophenyl)-21H,23H-porphyrin (TPPS) is investigated by an interdisciplinary approach involving the combination of time-resolved absorption and emission techniques with in vitro studies on cultured tumor cells. In a range of TPPS:SC6CDNH2 molar ratios between 1:10 and 1:50 these nanoparticles preserve the photodynamic properties of the entrapped photoactive agent. In fact, the triplet state of TPPS is efficiently populated, very long-lived and, as a consequence, able to produce singlet oxygen (the essential species for the photodynamic action) with quantum yield comparable to the free TPPS. Photodynamic efficacy of the carrier/sensitizer system is proven by in vitro studies on tumor Hela cells treated with TPPS:SC6CDNH2 at different molar ratio, showing significant cells death upon illumination with visible light.

A structural change from fractal to nanorod J-aggregates of tetrakis(4-sulfonatophenyl)porphyrin has been obtained by acting on the intermolecular interaction potential. The size and shape of these self-assembled porphyrin clusters have been monitored under different experimental conditions, by means of polarized and depolarized dynamic light scattering and small and wide angle elastic light scattering. At sufficiently low porphyrin concentration and high ionic strength, the shielded repulsive potential seems to be responsible for the fractal structure of the aggregates. On the contrary, at low ionic strength (nonshielded potential) and high porphyrin concentration, these species self-assemble in a rodlike arrangement. The length of the so-formed rod-shaped aggregates decreases on increasing porphyrin concentration. Moreover, both fractals and rods display a structure-dependent optical activity induced by a chiral template.

Well-defined arrays of porphyrins attached to a rigid polyisocyanide backbone have been synthesized and their physical and optical properties studied. The helical polymers are rigidified by an inter-side chain hydrogen-bonded network and have an average mass of 1.1 x 10(6) Daltons and a polydispersity index of 1.3. Each of the polymer strands contains four columns of around 200 stacked porphyrins and has an overall length of 87 nm. The chromophores are arranged in a left-handed helical fashion along the polymer backbone. Photophysical studies show that at least 25 porphyrins within one column are excitationally coupled.

Modifications of natural DNA by the dinuclear platinum(II) organometallic complex [[Pt(Me)Cl(Me(2)SO)](2)(mu-N-N)] [where N-N=H(2)N(CH(2))(6)NH(2)] (ORGANObisPt) were studied by methods of molecular biophysics. These methods include DNA binding studies using atomic absorption spectrophotometry, HPLC analysis of enzymatically digested DNA, interstrand cross-linking employing gel electrophoresis under denaturing conditions, DNA unwinding studied by gel electrophoresis, mapping of DNA adducts by transcription assay, DNA melting curves measured by absorption spectrophotometry and conformational analysis of platinated DNA by differential pulse polarography. The results indicate that the complex ORGANObisPt binds irreversibly to DNA. Its DNA binding mode is, however, different from that of the formally equivalent [[cis-PtCl(NH(3))(2)](2)(mu-N-N)] (1,1/c,c), which exhibits antitumor activity including that in the tumor cells resistant to cisplatin. Interestingly, ORGANObisPt binds to DNA considerably faster than 1,1/c,c and cisplatin. In addition, when ORGANObisPt binds to DNA it exhibits a strong base sequence specificity to guanine residues. ORGANObisPt forms mainly monofunctional adducts on double-helical DNA. It forms also a small amount of DNA interstrand cross-links (approximately 2%), i.e. a radically smaller amount in comparison with the complex 1,1/c,c. Importantly, these interstrand cross-links of ORGANObisPt are capable of terminating RNA synthesis in vitro, while its major monofunctional adducts are not. In addition, the adducts of ORGANObisPt affect the conformation of DNA, but in a different way than its dinuclear analogue 1,1/c,c or cisplatin. Some structural features of ORGANObisPt, such as the charge or nature of the trans and cis activating groups relative to the labile chloride, might be responsible for the altered DNA binding mode and biological activity in comparison with the 1,1/c,c compound.

Natural carrageenans of diverse structural types isolated from the red seaweed Gigartina skottsbergii were recently identified as potent and selective inhibitors of herpes simplex virus types 1 and 2 (HSV-1 and -2). The mu/nu-carrageenan 1C3 was tested in vitro for its ability to select resistant variants. After serial passages of HSV-1 strain F in Vero cells in the presence of increasing concentrations of 1C3, viruses emerged that were approximately 2- to 10-fold more resistant to 1C3 inhibition than parental virus; these viruses formed large plaques with an altered syncytial phenotype (1C3-syn). Plaque-purified syncytial variants isolated from passages 13 and 14 have shown variable levels of resistance to 1C3, as well as to the other antiviral carrageenans isolated from G. skottsbergii and to other sulfated polysaccharides with known antiviral activity, such as heparin and dextran sulfate 8000, but all the clones were susceptible to acyclovir. The syn phenotype was not related to polysaccharide resistance. All the 1C3-syn variants formed large syncytia in Vero and CV-1 cells but did not induce fusion in other cell types. The growth efficiency in Vero cells, as well as the virulence for mice by intracerebral or intraperitoneal inoculation of 1C3-syn variants, showed no significant alterations in comparison with the parental virus. The syncytial properties were not affected by cyclosporine or melittin, suggesting that an alteration on glycoprotein gB could be responsible for the syn phenotype induced by 1C3., (Copyright 2002 Wiley-Liss, Inc.)

The complex trans-bis(dimethylsulfoxide)chloromethylplatinum(II) (1) is fairly soluble in water, where it undergoes multiple equilibria involving the formation of geometrically distinct [Pt(H(2)O)(DMSO)Cl(CH(3))] aqua-species. On reacting an aqueous solution of 1 with monodentate nitrogen donor ligands L, such as pyridines or amines, two well distinct patterns of behavior can be recognized: (i) a single stage fast substitution of one DMSO by the entering ligand, yielding a complex of the type trans(C,N)-[Pt(DMSO)(L)Cl(CH(3))] which contains four different groups coordinated to the metal and which undergoes a slow conversion into its cis-isomer, (ii) a double substitution affording cationic complex ions of the type cis-[Pt(L)(2)(DMSO)(CH(3))](+). When this latter reaction is carried out using sterically hindered ligands, slow rotation of the bulk ligand around the Pt[bond]N bond allows for the identification of head-to-head and head-to-tail rotamers in solution, through (1)H NMR spectrometry. The addition of chloride anion to 1 leads to the anionic species cis-[Pt(DMSO)Cl(2)(CH(3))](-), where a molecule of DMSO still remains coordinated to the metal center, despite its quite fast rate of ligand exchange (k(exch) with free DMSO=12+/-1 s(-1)). The reaction of complex 1 with bidentate ligands, such as ethylenediamine (en) or simple amino acids, leads to the cationic species [Pt(en)(DMSO)(CH(3))](+) or to the neutral [Pt(DMSO)(N[bond]O)(CH(3))], (where N[bond]-O[double bond]GlyO(-), AlaO(-)).

Resonance light scattering (RLS) is a phenomenon due to an enhancement of the scattered light in close proximity to an absorption band. The effect is easily detectable in the case of strongly absorbing chromophores, which are able to interact, thus leading to large aggregates (Pasternack, R. F.; Collings, P. J. Science 1995, 269, 935). The measurement of absorption spectra from solutions containing such resonant systems can lead to misleading results. In this paper, a simple method is described to obtain absorption spectra of aggregated species with a fairly good correction of the scattering component. The RLS spectrum, obtained using a common spectrofluorimeter, is correlated to the extinction spectrum of the same sample, allowing for an estimation of the scattering contribution to the total extinction spectrum. The method has been successfully applied both on real samples containing aggregated chromophores, such as porphyrins, chlorophyll a and gold colloids, and by simulating extinction spectra.

The ionic methylplatinum(II) complexes [Pt(Me)(L)(dmphen)]X (dmphen = 2,9-dimethyl-1,10-phenanthroline, L = Me(2)SO, X = PF(6)(-) 1a, BF(4)(-) 1b, CF(3)SO(3)(-) 1c, ClO(4)(-) 1d, B(C(6)H(5))(4)(-) 1e, [B(3,5-(CF(3))(2)C(6)H(3))(4)](-) 1f; L = n-Bu(2)SO, X = CF(3)SO(3)(-) 1g; L = PPh(3), X = PF(6)(-) 2a, BF(4)(-) 2b, CF(3)SO(3)(-) 2c, ClO(4)(-) 2d, B(C(6)H(5))(4)(-) 2e, [B(3,5-(CF(3))(2)C(6)H(3))(4)](-) 2f; X = CF(3)SO(3)(-), L = CyNH(2) 3a, i-PrNH(2) 3b, 2,6-Me(2)py 3c, EtNH(2) 3d, AsPh(3) 3e, dimethylthiourea (Me(2)th) 3f and the uncharged [Pt(Me)(X)(dmphen)] (X = SCN(-) 4a, SeCN(-) 4b) complexes have been synthesized and fully characterized. In chloroform, as well as in acetone or methanol, complexes 1a-1g, 2a-2h (X = Cl(-) g, NO(2)(-) h, formed "in situ"), and 3e show dynamic behavior due to the oscillation of the symmetric chelating ligand dmphen between nonequivalent bidentate modes. All the other compounds feature a static structure in solution. The crystal structure of 2a shows a tetrahedral distortion of the square planar coordination geometry, a loss of planarity of the dmphen ligand, and, most notably, a rotation of the dmphen moiety, around the N1-N2 vector, to form a dihedral angle of 42.64(8) degrees with the mean coordination plane. The hexafluorophosphate ion lies on the side of the phenanthroline ligand. The interionic structures of 2a, 2b, and 2f were investigated in CDCl(3) at low temperature by (1)H-NOESY and (19)F[(1)H]-HOESY NMR spectroscopies. Whereas PF(6)(-) (2a) and BF(4)(-) (2b) show strong contacts with the cation [Pt(Me)(PPh(3))(dmphen)](+), being located preferentially on the side of the phenanthroline ligand, the [B(3,5-(CF(3))(2)C(6)H(3))(4)](-) (2f) ion does not form a tight ion pair. The dynamic process was studied by variable-temperature NMR spectroscopy for 1a-1f and 2a-2h in CDCl(3). The activation energies DeltaG(298) for the sulfoxide complexes 1a-1f are lower than those of the corresponding phosphine complexes 2a-2f by approximately 10 kJ mol(-)(1). The nature of the counteranion exerts a tangible influence on the fluxionality of dmphen in both series of complexes 1 and 2. The sequence of energies observed for 2a-2h encompasses an overall difference of about 16 kJ mol(-)(1), increasing in the order Cl(-) approximately NO(2)(-) << CF(3)SO(3)(-) < ClO(4)(-) < B(C(6)H(5))(4)(-) < BF(4)(-) approximately PF(6)(-) < B(3,5-(CF(3))(2)C(6)H(3))(4)(-). Acetone and methanol have an accelerating effect on the flipping. Concentration-dependent measurements, carried out in CDCl(3) for 2a with n-Bu(4)NPF(6) and the ligands dmphen, n-Bu(2)SO, sec-Bu(2)SO, and sec-Bu(2)S showed that the rate of the fluxional motion is unaffected by added n-Bu(4)NPF(6), whereas in the other cases this increases linearly with increasing ligand concentration, according to a pattern of behavior typical of substitution reactions. Dissociative and associative mechanisms can be envisaged for the observed process of flipping. Dissociation can be prevalent within the ion pair formed by a "noncoordinating" anion with the metallic cationic complex in chloroform. Among the possible associative mechanisms, promoted by polar solvents or by relatively strong nucleophiles, a consecutive displacement mechanism is preferred to intramolecular rearrangements of five-coordinate intermediates.

The rates of chloride for triphenylphosphine substitution have been measured in dichloromethane for a series of cyclometalated [Pt(N-N-C)Cl] complexes containing a number of terdentate N-N-C anionic ligands, derived from deprotonated alkyl-, phenyl-, and benzyl-6-substituted 2,2'-bipyridines. These rates have been compared with those of the corresponding [Pt(N-N)(C)Cl] (N-N = 2,2'-bipyridine; C = CH3 or C6H5) complexes having the same set of donor atoms but less constrained arrangements of the ligands. The reactions of the cyclometalated compounds occur as a single-stage conversion from the substrate to the ionic pair [Pt(N-N-C)(PPh3)]Cl products. There is no evidence by 1H and 31P(1H) NMR spectroscopy for the formation of other Pt(II) species or of concurrent ring-opening processes. In contrast, in the monoalkyl- or monoaryl-2,2'-bipyridine complexes, chloride substitution is followed by subsequent slower processes which involve the detachment of one arm of the chelated 2,2'-bipyridine, fast cis to trans isomerization of the cis-[Pt(PPh3)2(eta 1-bipy)(R)]+ transient intermediate, and, eventually, the release of free bipy, yielding trans-[Pt(PPh3)2(R)Cl] (R = Me or Ph). All reactions are first-order with respect to complex and phosphine concentration, obeying the simple rate law kobsd = k2[PPh3]. The values of the second-order rate constant k2 do not seem particularly sensitive to the nature of the bonded organic moiety (alkyl or aryl), to its structure (cyclometalated or not), to the size of the ring, or to the number of alkyl substituents on it. The effects are those foreseen on the basis of an associative mode of activation. The only exception to this pattern of behavior is constituted by the complex [Pt(bipy phi-H)Cl] (bipy phi = 6-phenyl-2,2'-bipyridine), which features a significant rate enhancement with respect to the analogue [Pt(bipy)(Ph)Cl] complex. The results of this work, together with those of a previous paper, suggest that there is not a specific role of cyclometalation in controlling the reactivity, unless an in-plane aryl ring becomes part of the pi-acceptor system of the chelated 2,2-bipyridine, behaving as a cyclometalated analogue of the nitrogen terdentate 2,2':6',2"-terpyridine.

The ligand exchange rate constants for the reactions [Pt(bph)(SR2)2] + 2*SR2 --> [Pt(bph)(*SR2)2] + 2SR2 (bph = 2,2'-biphenyl dianion; R = Me and Et) and cis-[PtPh2(SMe2)2] + 2*SMe2 --> cis-[PtPh2(*SMe2)2] + 2SMe2 have been determined in CDCl3 as a function of ligand concentration and temperature, by 1H NMR isotopic labeling and magnetization transfer experiments. The rates of exchange show no dependence on ligand concentration and the kinetics are characterized by largely positive entropies of activation. The kinetics of displacement of the thioethers from [Pt(bph)(SR2)2] with the dinitrogen ligands 2,2'-bipyridine and 1,10-phenanthroline (N-N) to yield [Pt(bph)(N-N)], carried out in the presence of sufficient excess of thioether and N-N to ensure pseudo-first-order conditions, follow a nonlinear rate law k(obsd) = a[N-N]/(b[SR2] + [N-N]). The general pattern of behavior indicates that the rate-determining step for substitution is the dissociation of a thioether ligand and the formation of a three-coordinated [Pt(bph)(SR2)] intermediate. The value of the parameter a, which measures the rate of ligand dissociation, is constant and independent of the nature of N-N, and it is in reasonable agreement with the value of the rate of ligand exchange at the same temperature. Theoretical ab initio calculations were performed for both [Pt(bph)(SMe2)2] and cis-[PtPh2(SMe2)2], and for their three-coordinated derivatives upon the loss of one SMe2 ligand. The latter optimize in a T-shaped structure. Calculations were performed in the HF approximation (LANL2DZ basis set) and refined by introducing the correlation terms (Becke3LYP model). The activation enthalpies from the optimized vacuum-phase geometries are 52.3 and 72.2 kJ moll compared to the experimental values in CDCl3 solution, 80 +/- 1 and 93 +/- 1 kJ mol(-1) for [Pt(bph)(SMe2)2] and cis-[PtPh2(SMe2)2], respectively. The electrostatic potential maps of both parent compounds show a remarkable concentration of negative charge over the platinum atom which exerts a repulsion force on an axially incoming nucleophile. On the other hand, the strength of the organic carbanions trans to the leaving group and the stabilization of the T-shaped intermediate in the singlet ground state may also rationalize the preference for the dissociative mechanism. All of the kinetic and theoretical data support the latter hypothesis and indicate, in particular, that dissociation from the complex containing the planar 2,2'-biphenyl dianion is easier than from its analogue with single aryl ligands. Electron back-donation from filled d orbitals of the metal to empty pi* of the in-plane cyclometalated rings is weak or absent and is not operative in promoting an associative mode of activation.

Tacaribe virus (TACV) is an arenavirus that is genetically and antigenically closely related to Junin virus (JUNV), the aetiological agent of Argentine haemorrhagic fever (AHF). It is well established that TACV protects experimental animals fully against an otherwise lethal challenge with JUNV. To gain information on the nature of the antigens involved in cross-protection, recombinant vaccinia viruses were constructed that express the glycoprotein precursor (VV-GTac) or the nucleocapsid protein (VV-N) of TACV. TACV proteins expressed by vaccinia virus were indistinguishable from authentic virus proteins by gel electrophoresis. Guinea pigs inoculated with VV-GTac or VV-N elicited antibodies that immunoprecipitated authentic TACV proteins. Antibodies generated by VV-GTac neutralized TACV infectivity. Levels of antibodies after priming and boosting with recombinant vaccinia virus were comparable to those elicited in TACV infection. To evaluate the ability of recombinant vaccinia virus to protect against experimental AHF, guinea pigs were challenged with lethal doses of JUNV. Fifty per cent of the animals immunized with VV-GTac survived, whereas all animals inoculated with VV-N or vaccinia virus died. Having established that the heterologous glycoprotein protects against JUNV challenge, a recombinant vaccinia virus was constructed that expresses JUNV glycoprotein precursor (VV-GJun). The size and reactivity to monoclonal antibodies of the vaccinia virus-expressed and authentic JUNV glycoproteins were indistinguishable. Seventy-two per cent of the animals inoculated with two doses of VV-GJun survived lethal JUNV challenge. Protection with either VV-GJun or VV-GTac occurred in the presence of low or undetectable levels of neutralizing antibodies to JUNV.

The effects of the carrageenans 1T1 (lambda-type), 1C1 (kappa/iota-type) and 1C3 (upsilon/nu-type), isolated from the red seaweed Gigartina skottsbergii, on herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) infection of murine astrocytes were investigated. The three compounds were effective inhibitors of virus replication, as determined by a virus yield inhibition assay, in astrocytes as well as in Vero cells. The 50% inhibitory concentration was in the range of 0.9-3.6 and 0.4-3.2 microg/ml for astrocytes and Vero cells, respectively, whereas the 90% inhibitory concentration ranged from 6.5 to 17.0 microg/ml in astrocytes and from 3.5 to 22.2 microg/ml in Vero cells. No cytotoxicity was detected at concentrations of up to 1,000 microg/ml, indicating high selectivity indices for these compounds. Inhibition of viral cytopathology and antigen expression was also detected in the presence of the carrageenans. The increase in the expression of glial fibrillary acidic protein (GFAP), an activated astrocyte marker, produced during the course of HSV-1 infection in astrocytes, was reversed in the presence of 1C1 and 1C3. By contrast, the lambda-carrageenan 1T1 increased the expression of GFAP, independently of HSV-1 infection.

In order to diagnose rotavirus infection, stool samples from 134 children assisted at the Hospital B. Houssay, Vte. López, Pcia. de Buenos Aires, Argentina, have been analysed employing the Rotazyme II Diagnostic Kit (Abbot Lab.). The percentage of positive samples reached 38.8% detecting 30 and 22 positive samples in 1994 (n(o) = 84) and 1995 (n(o) = 50), respectively. The frequency of the agent peaked during the cold season of the year. The age group between 6 and 23 months was the most affected, whereas the lowest percentage of viral identification was detected in children under 6 months. No differences were detected due to the gender of children. Viral RNA analysis pointed out 7 different electropherotypes, all of them showing long electropherotype patterns.

The effect of gamma radiation on poliovirus infectivity seeded in sludge samples was investigated in order to determine the radiation dose required to inactivate 90% of viral infectivity (D10). Sludges were obtained from anaerobic pretreated sewages produced by San Felipe, a wastewater treatment facility located at the Tucuman province, Argentina. A D10 of 3.34 kGy was determined for poliovirus type III, Sabin strain, suspended in sludge samples. This value dropped to 1.92 kGy when the virus was suspended in water. A virucidal effect associated to sludges was also demonstrated. These results will be of interest when considering the dose of gamma radiation to be applied to wastewater sludges in order to preserve the environment from viral contamination.

A series of new organometallic platinum(II) complexes containing terdentate polypyridine ligands has been prepared and characterized. Their absorption spectra in 4:1 (v/v) MeOH/EtOH fluid solution at room temperature and luminescence in the same matrix at 77 K have been investigated. The new species are [Pt(terpy)Ph]Cl (3, terpy = 2,2':6',2"-terpyridine, Ph = phenyl), [Pt(Ph-terpy)Cl]Cl (4, Ph-terpy = 4'-phenyl-2,2':6',2"-terpyridine), [Pt(Ph-terpy)Me]Cl (5), and [Pt(Ph-terpy)Ph]Cl (6). The results have been compared with those for [Pt(terpy)Cl]Cl (1) and [Pt(terpy)Me]Cl (2). NMR data evidence that all the complexes but 3 and 6 oligomerize in solution leading to stacked species. The absorption spectra are dominated by moderately intense metal-to-ligand charge-transfer (MLCT) bands in the visible region and by intense ligand-centered (LC) bands in the UV region. All the compounds are luminescent in a 4:1 (v/v) MeOH/EtOH rigid matrix at 77 K, exhibiting a structured emission within the range 460-600 nm. This feature is assigned to formally (3)LC excited states which receive substantial contribution from closely lying (3)MLCT levels. Complexes 1, 2, 4, and 5 also exhibit a relatively narrow and unstructured luminescence band within the range 680-800 nm, which dominates the luminescence spectrum on increasing concentration and exciting at longer wavelengths. The band is assigned to a dsigma(metal) --> pi(polypyridine) ((3)MMLCT) state, originating from metal-metal interactions occurring in head-to-tail dimers (or polymers). A third broad band is shown by 1 and 4 under all concentration conditions and by 2 and 5 only in concentrated solutions and is attributed to excimeric species originating from pi-pi interactions due to stacking between polypyridine ligands.

A sulphated galactan (SG) with low molecular weight (app. 2800) was isolated from extracts of Cryptopleura ramosa, a red seaweed from the South American coasts. The compound was a selective inhibitor of HSV-1 and HSV-2 replication in Vero cells with 50% inhibitory concentrations (IC50) in the range 1.6-4.2 micrograms/ml and a 50% cytotoxic concentration (CC50) of 476 micrograms/ml. SG was also effective against HSV-1 in cells of neural origin such as murine astrocytes. The mode of action of SG could be ascribed to an inhibitory action on virus adsorption. Furthermore, SG did not inhibit the blood coagulation process at concentrations highly exceeding the IC50.

The biological properties mediated by the main envelope glycoprotein (GP1) of a mouse-attenuated mutant of Junin virus, named Cl67, were investigated in comparison with its parental strain XJCl3. In contrast to the parental strain, this mutant was unable to multiply in primary cultures of mouse embryo fibroblasts (MEFs). Impairment of Cl67 multiplication was associated with a lack of virus binding to MEF, probably due to an altered interaction between GP1 and cellular receptors. Antigenic and immunogenic characterization of GP1, performed by neutralization assays, demonstrated that, under certain conditions, polyclonal and monoclonal antibodies exhibited differential affinity and specificity for each virus. Cl67-infected Vero cells showed a marked pH-dependent fusion capability, suggesting more efficient low pH triggering of fusion by mutant virus GP1 in comparison with the parental strain.

A series of monoalkyl square-planar complexes of the type [Pt(N-N)(CH(3))(Me(2)SO)]PF(6) (1-14), where N-N represents chelating diamines or diimines of widely different steric and electronic characteristics, was synthesized, and the complexes were fully characterized as solids and in solution. The substrates were tailored to offer only one site of exchange to a neutral molecule, i.e. Me(2)SO, in a noncoordinating solvent. No evidence for fluxionality of the N-N ligands was found, except for the case of complex 11 formed by 2,9-dimethyl-1,10-phenanthroline. In solution this complex is fluxional with the phenanthroline oscillating between nonequivalent bidentate modes by a mechanism which involves rupture of the metal-nitrogen bond and rapid interconversion of two coordinatively unsaturated T-shaped 14-electron three-coordinate molecular fragments. Rates of this fluxion were measured by NMR spectroscopy from the exchange effects on the (1)H signals of the methyl and aromatic hydrogens. The DeltaG() value for the fluxion is 49.6 +/- 4 kJ mol(-)(1). Dimethyl sulfoxide exchange with all the complexes has been studied as a function of ligand concentration by (1)H NMR line-broadening, isotopic labeling, and magnetization transfer experiments with deuterated acetone as the solvent. Second-order rate constants were obtained from linear plots of k(obs) vs [Me(2)SO] and activation parameters were obtained from exchange experiments carried out at different temperatures. Second-order kinetics and negative entropies of activation indicate an associative mechanism. The lability of dimethyl sulfoxide in the complexes depends in a rather unexpected and spectacular way upon the nature of the coordinate N-N ligands, the difference in reactivity between the first (N-N = N,N,N',N'-tetramethyl-1,2-diaminoethane, k(2)(298) = (1.15 +/- 0.1) x 10(-)(6) mol(-)(1) s(-)(1)) and the last (N-N = 2,9-dimethyl-1,10-phenanthroline, k(2)(298) = (3.81 +/- 0.005) x10(4) mol(-)(1) s(-)(1)) members of the series being greater than 10 orders of magnitude, as a result of a well-known phenomenon of steric retardation (for the first complex) and an unprecedented case of steric acceleration (for the last complex). Other factors of primary importance in controlling the reactivity are (i) the presence of an extensive pi system on the ligand N-N, (ii) the ease with which this pi system interacts with nonbonding d electrons of the metal, and (iii) the flexibility and ease of elongation of the chelate bite distance. The basicity plays a somewhat minor role, except in the restricted range of the same class of compounds such as substituted phenanthrolines.

C167, a mouse-attenuated strain of Junin virus derived from the XJC13 strain, also displayed reduced virulence for the South American cricetid Calomys musculinus, a natural reservoir of this virus in nature. Intracerebral inoculation of C. musculinus with 500 PFU of C167 produced only 25% mortality, whereas the parental XJC13 killed 85% of the animals. The attenuation of C167 for this cricetid was lower than for albino mice. The multiplication of C167 in C. musculinus-derived embryo or kidney fibroblasts was diminished with respect to XJC13, allowing us to define C. musculinus cells as a semipermissive system for C167, whereas murine and Vero cells were restrictive and permissive cultures, respectively. As a consequence, C167 as well as XJC13 were able to establish a persistent infection in C. musculinus embryo fibroblasts and Vero cells, but the mutant could not induce a carrier state in murine cells. Thus, the degree of susceptibility of C. musculinus to C167 was linked to the semipermissiveness of cricetid cells to virus multiplication.

Experimental infection of three mouse strains with a non-pathogenic mutant of Junin virus named Cl67 was compared with respect to the parental XJCl3 strain. After intracerebral (ic) or intraperitoneal inoculation, XJCl3 was highly virulent for 2 day-old C3H/HeJ, OF1, and BALB/cJ mouse strains, whereas its derivative Cl67 was attenuated. Survival of the Cl67-infected mouse was associated with a restricted replication at the site of inoculation which would impair spread of virus. Thus, the reduced virulence of Cl67 for suckling mice is independent of the mouse strain and the route of viral entry. When Cl67 was preinoculated ic 10 days before the challenge inoculation with XJCl3 by the same route, mice were partially protected from lethal infection. Since neutralizing antibodies were first detected at 30 days post-infection, an interference mechanism is postulated as a mechanism of protection of the mice.

As a part of research on polieterocycles compounds with condensed nuclei, containing pyrimidine ring, as potential antalgic/anti-inflammatory drugs, pyrazolopyrimidobenzoxazole and pyrazolopyrimidobenzothiazole were prepared and tested. None of the substances shows significative activities in vivo.

The growth characteristics, cytopathogenicity and viral polypeptides of the virulent strain XJ of Junin virus (JV), its attenuated derivative XJC13 and another naturally attenuated JV strain, IV4454, were comparatively studied. IV4454 and XJC13 viruses showed the highest and lowest cytopathology for Vero cells, respectively, as measured by plaque morphology, cell viability and inhibition of host cell protein synthesis. The kinetics and electrophoretic patterns of viral polypeptides in infected cell extracts were very similar among the three strains, whereas differences were detected in the surface glycoprotein GP38 by peptide mapping after limited proteolysis.

Spectroscopic methods have been employed in order to understand the molecular basis of the decrease in enzymatic activity of the antiinflammatory enzyme copper-zinc superoxide dismutase (SOD) following the covalent binding of polyethyleneglycol (PEG) chains to the protein amino-groups. The PEG modification is a general method recently proposed to improve the therapeutic index of enzymes. 1H NMR spectra on the cobalt substituted PEG-modified SOD, Cu2Co2-PEG-SOD, have been recorded. The signals are quite broad with respect to the unmodified enzyme. This has been interpreted on the basis of the effect of molecular weight on the linewidth. The analysis has shown that the histidine hydrogens involved in metal binding at the enzyme active site are the same in both native and PEG-modified SOD. Similarly, circular dichroism and absorption spectra indicate that the overall conformation of the metal clusters is not perturbed upon modification. On the other hand, azide titration shows that the affinity constant of N-3 for SOD is largely reduced upon PEG modification (K = 154 M-1 and 75 M-1 for the native and modified SOD, respectively). These results indicate that the decrease in enzymatic activity upon surface modification with PEG is not caused by a perturbation of the active site geometry, but to a decrease in the channeling of the O2- ion towards the enzyme active site.

C167, a mutant derived from the XJC13 strain of Junin virus, is highly attenuated in its pathogenic properties for newborn mice. Whereas 10(2).PFU of XJC13 injected intracerebrally killed 100% of two-day-old mice, the mutant showed no detectable lethality. Survival of mice infected with C167 was associated with a reduced and delayed virus replication in brain and a defective spread of virus from the site of inoculation to the other tissues, including spleen, kidney, thymus, liver, peritoneal cells and serum. As an apparent consequence of the restricted replication of C167 in mice, no detectable interferon induction and low levels of neutralizing antibodies were observed. Analysis of multiplication kinetics of C167 and XJC13 in different cell cultures in vitro has confirmed that the attenuated phenotype of C167 was related to a specific inefficient replication in murine cells. This host-range restriction was due to a combination of adsorption and penetration blockage.

The virulence in neonatal mice of a temperature-sensitive mutant of Junin virus, named C167, was studied. The thermosensitive properties of this mutant were tested by titration on Vero cells at 37 and 40 degrees C. The ratio of infectivity 40/37 was approximately 100-fold lower for C167 with respect to XJC13 (Table 1). The attenuation of C167 was determined by measurement of mean survival time and 50% lethal dose after intracerebral injection of 2 and 11 day old mice. For C167 the lethality index (expressed ad the ratio TCID50/LD50) was greater than 580, while for XJC13 the index was 4.4 (Table 2). The lack of virulence of C167 was correlated with a restricted ability to replicate in suckling mouse brains. By contrast, the mutant and the parental virus grew to a similar titre in Vero cells (Figure 2). Both viruses were indistinguishable in cross-neutralization tests using hyperimmune antisera.
