The metabolism of [2-(14)C]- and [ring-U-(14)C]2,4-dichlorophenoxyacetic acid (2,4-D) has been studied in cell suspension cultures of soybean (Glycine max L.) and wheat (Triticum aestivum L.). 2,4-D was extensively metabolized by both cultures, the rates of metabolism and the metabolite patterns remaining constant over at least 60 (soybean) and 25 (wheat) growth cycles. Amino acid conjugates were the predominant metabolite fraction in soybean cells whereas β-D-glucoside conjugates predominated in wheat cells. The two cell types studied also differed in the amino acid compositions of the amino acid conjugate fractions and in the aglycone patterns of the β-D-glucoside fractions. Novel metabolite fractions of both wheat and soybean cells included a polar water-soluble metabolite fraction and a covalently modified protein fraction of low molecular weight.

Cell suspension cultures of soybean (Glycine max L.) and wheat (Triticum aestivum L.) incorporated 2,4-dichlorophenoxyacetic acid (2,4-D) into a metabolite fraction which was insoluble in ethanol, water, and hot sodium dodecylsulphate. Further treatment with hot dimethylformamide solubilized a material which by the following criteria appeared to consist of 2,4-D derivatives covalently bound to lignin: i) co-chromatography of radioactivity and of UV-absorbing material upon gel permeation chromatography; ii) spectral similarity with authentic lignins (IR- and UV-spectra, phloroglucinol reaction), 2,4-D appeared to be incorporated as the intact molecule, as shown by comparison of ring- and sidechain-labeled 2,4-D and by detection of monohydroxylated and intact 2,4-D as the major radioactive products of acid hydrolysis. The same compounds were released from the metabolite material which could not be solubilized in dimethylformamide. The incorporation of xenobiotics or their metabolites into lignin, followed by deposition in the cell wall, is suggested as a general pathway for 'local excretion' and detoxification by plant cells.

The differential response of cultured parsley cells to u.v. irradiation and elicitor treatment is a paradigm for analysis of specific plant defense responses. We demonstrate that freshly isolated parsley protoplasts, in the absence of detectable cell wall, maintain fully the ability to be activated by these important environmental factors. Stimulated protoplasts synthesize typical qualitative patterns and amounts of potentially protective flavonoid glycosides and coumarin phytoalexins following either u.v. irradiation or treatment with fungal elicitor, respectively. Induced accumulation of mRNAs and enzymes of the phenylpropanoid biosynthetic pathways is nearly identical in protoplasts and cells. Stimulation of protoplasts with elicitor requires only a short period of contact, which is not sufficient for cell wall regeneration. Importantly, there is no activation of these pathways during protoplast preparation. These results establish that parsley protoplasts respond appropriately to two physically distinct stimuli and might serve as an especially suitable system for the analysis of signal transduction and gene activation.

Different components of a crude cell-wall preparation from the phytopathogenic fungus, Phytophthora megasperma f. sp. glycinea, act as elicitors of phytoalexin accumulation in parsley (Petroselinum crispum) and soybean (Glycine max). Treatments of cultured parsley cells and protoplasts or soybean cells and cotyledons with proteinase-digested or deglycosylated elicitor preparations identify proteinaceous constituents as active eliciting compounds in parsley, which are inactive in soybean. The proteinase-treated elicitor as well as a defined heptaglucan are active in soybean but do not stimulate phytoalexin synthesis in parsley. Soybean and parsley cells therefore not only perceive different signals from cell walls of Phytophthora megasperma f. sp. glycinea, but are unable to respond to the fungal compounds primarily recognized by the other plant.

We have determined the primary structures of two 4-coumarate: CoA ligase (4CL) isoenzymes in parsley (Petroselinum crispum) by sequencing near full-length cDNAs corresponding to the two 4CL genes, Pc4CL-1 and Pc4CL-2, present in this plant. Comparison of the cDNA and genomic nucleotide sequences showed that each 4CL gene is organized in five exons separated by introns of varying lengths. The positions of introns are the same in both genes and 97-99% of the corresponding nucleotide sequences are identical. The two isoenzymes, which are nearly identical in their primary structures, were separated by ion-exchange chromatography, and were found to be indistinguishable with regard to substrate specificity. Assignment to Pc4CL-1 and Pc4CL-2 was achieved by comparison with catalytically active 4CL proteins, isolated from Escherichia coli cells which had been transformed with plasmids harboring the corresponding cDNAs.
