203. The Role of NG2 Glial Cells in ALS Pathogenesis

Abstract

A growing body of evidence suggests that glial cells are critical for the support of normal neuronal function, and in particular astrocyte and oligodendroglial dysfunction may contribute to a number of neurodegenerative diseases, including ALS. One important function of glial cells is to transport nutrients from capillaries to neurons. Much of the nutritional support is in the form of glucose; however our lab and others have provided strong evidence that lactate support from oligodendrocytes via monocarboxylate transporters (MCTs) is a major contributor to neuronal metabolism and survival in vivo. Preliminary data from our laboratory indicates that genetic knockdown or pharmacological inhibition of glial specific monocarboxylate transporter 1 (MCT1) leads to loss of spinal cord motor neurons in vitro and in vivo. In addition, our lab and others recently showed in SOD1 mutant mice NG2+ cells, the progenitor cells for oligodendrocytes, upregulate NG2 and show enhanced proliferation in the ventral horn gray matter where motor neurons die. In addition, NG2+ cells formed large numbers of newly generated immature, non-myelinating oligodendroglia, but no astrocytes or neurons. Finally, when we specifically protected oligodendrocyte injury in the SOD1 animal model by deleting mutant SOD1 in NG2+ cells, we were able to delay disease onset by 126 days- many months longer than any other genetic ablation approach to date. We hypothesize that NG2 glial cells play a significant role in ALS pathogenesis. In particular, we are testing how human NG2 glial cells derived from ALS patient iPS cells will impact human motor neurons, also derived from human iPS cells., The original document contains color images.

Abstract

The Na+-dependent glutamate transporter GLT-1 (EAAT2) shows selective expression in astrocytes, and neurons induce the expression of GLT-1 in astrocytes. In an unpublished analysis of GLT-1 promoter reporter mice, we identified an evolutionarily conserved domain of 467 nucleotides ~ 8 kb upstream of the GLT-1 translation start site that is required for astrocytic expression. Using in silico approaches, we identified Pax6 as a transcription factor that could contribute to the control of GLT-1 expression by binding within this region. We demonstrated the expression of Pax6 protein in astrocytes in vivo. Lentiviral transduction of astrocytes with exogenous Pax6 increased the expression of enhanced green fluorescent protein ( eGFP) in astrocytes prepared from transgenic mice that use a bacterial artificial chromosome containing a large genomic region surrounding the GLT-1 gene to control expression of eGFP. It also increased GLT-1 protein and GLT-1-mediated uptake, whereas there was no effect on the levels of the other astroglial glutamate transporter, glutamate aspartate transporter (GLAST). Transduction of astrocytes with an sh RNA directed against Pax6 reduced neuron-dependent induction of GLT-1 or eGFP. Finally, we confirmed Pax6 interaction with the predicted DNA-binding site in electrophoretic mobility assays and chromatin immunoprecipitation (Ch IP). Together, these studies show that Pax6 contributes to the regulation of GLT-1 through an interaction with these distal elements and identify a novel role of Pax6 in astrocyte biology. Abstract

TGFbeta-like trophic factors have been shown to be protective in acute neuronal injury paradigms. In the current studies, we analyzed and compared members of this growing family, including GDNF, neurturin, nodal, persephin, TGFbeta1 as well as neuroimmunophilin ligand GPl-1O46, for protection against chronic glutamate toxicity and the mechanisms for protection. In parallel, we developed a novel organotypic spinal cord culture system to study the ability of these factors to promote motor axon outgrowth. Using these systems, we were able to differentiate the neuroprotective effect of the TGFbeta-like factors from their motor axon outgrowth-promoting activity. GDNF, neurturin, persephin, and nodal all protect against excitotoxic motor neuron degeneration. Low amounts of GDNF (1 ng/ml) and high concentrations of neurturin induced vigorous motor axon outgrowth. In contrast, nodal, persephin and TGFbetal did not induce motor axon outgrowth. Finally, we have ascertained that the neuroprotective properties of some of these compounds (GDNF and GPl-1 046) may lie in their ability to induce rapidly synthesis of the glutamate transporter EAAT2.
