Abstract

Classic Kaposi sarcoma (CKS) is an inflammatory-mediated neoplasm primarily caused by Kaposi sarcoma–associated herpesvirus (KSHV). Kaposi sarcoma lesions are characterized, in part, by the presence of proinflammatory cytokines and growth factors thought to regulate KSHV replication and CKS pathogenesis. Using genomic DNA extracted from 133 CKS cases and 172 KSHV-latent nuclear antigen-positive, population-based controls in Italy without HIV infection, we examined the risk of CKS associated with 28 common genetic variants in 14 immune-modulating genes. Haplotypes were estimated for IL1A, IL1B, IL4, IL8, IL8RB, IL10, IL12A, IL13, and TNF. Compared with controls, CKS risk was decreased with 1235T/−1010G–containing diplotypes of IL8RB (odds ratio, 0.49; 95% confidence interval, 0.30-0.78; P = 0.003), whereas risk was increased with diplotypes of IL13 containing the promoter region variant 98A (rs20541, alias +130; odds ratio, 1.88; 95% confidence interval, 1.15-3.08; P = 0.01) when considered in multivariate analysis. Risk estimates did not substantially vary by age, sex, incident disease, or disease burden. Our data provide preliminary evidence for variants in immune-modulating genes that could influence the risk of CKS. Among KSHV-seropositive Italians, CKS risk was associated with diplotypes of IL8RB and IL13, supporting laboratory evidence of immune-mediated pathogenesis. Abstract

Dengue is a major mosquito-borne disease in many tropical and sub-tropical countries worldwide, with entomological surveillance and control activities as the key management approaches. This study aimed to explore the spatial dispersal of the vector Aedes albopictus, captured by the modified sticky ovitrap (MSO) in residential areas with low-rise buildings in Selangor, Malaysia. Distribution maps were created and shown as temporally distinguished classes based on hotspot analysis by Getis-Ord; spatial autocorrelation assessed by semivariograms using the exponential Kernel function; and universal Kriging showing areas with estimated high and low vector densities. Distribution, hotspot and interpolated maps were analysed based on the total number of mosquitoes by month and week. All maps in the present study were generated and visualised in ArcMap. Spatial autocorrelation of Ae. albopictus based on the monthly occurrence of Ae. albopictus was found in March, April, October, November and December 2018, and when based on the weekly numbers, in weeks 1, 2, 3, 5, 7, 12, 14, 25, 26, 27, 31, 33, 42, 49 and 52. Semivariograms, based on the monthly and weekly numbers of Ae. albopictus, indicated spatial autocorrelation of the species extending between 50 and 70 m. The mosquito density maps reported in this study may provide beneficial information to facilitate implementation of more efficient entomological control activities.

Abstract

A 10-day experiment was performed to examine different mono, binary and ternary dietary combinations on survival and growth of D-shaped and umbone black-lip pearl oyster, Pinctada margaritifera, larvae. The three tropical microalgae species were the flagellate Isochrysis galbana clone T. Iso (CS-177) and diatoms Chaetoceros calcitrans (CS-178) and Chaetoceros muelleri (CS-176) which were fed to D-shaped and umbone larvae at a density of 7000 and 14 000 cells mL−1, respectively. A second experiment was performed to investigate the feasibility of replacing T. Iso with a lipid emulsion for both D-shaped and umbone larvae for 10 and 12 days, respectively. The treatments included only T. Iso, unfed and lipid emulsion to substitute T. Iso at levels of 10% (LIP10), 30% (LIP30) and 100% (LIP100). In the first experiment, results showed that a monospecific diet of T. Iso led to significantly higher ( P < 0.05) survival and growth of D-shaped larvae than all the other treatments. Meanwhile, D-shaped larval survival was significantly lower when only fed C. calcitrans as well as growth for those fed C. calcitrans or in combination with C. muelleri. However, for umbone larvae, survival and growth were significantly higher when fed a binary combination of T. Iso and C. muelleri or the ternary combination of T. Iso, C. muelleri and C. calcitrans compared with all other treatments. For the second experiment, results showed that with increasing lipid emulsion replacement, survival of both D-shaped and umbone larvae significantly decreased ( P < 0.05); however, the LIP100 treatment was not significantly different ( P > 0.05) from the unfed treatment. For D-shaped larvae, no significant growth difference was detected ( P > 0.05) between the T. Iso and LIP10 fed treatments while for umbone larvae, the T. Iso, LIP10 and LIP30 were not significantly different ( P > 005). These results indicate that microalgae combinations appear more necessary for later staged P. margaritifera larvae. In addition, the use of a lipid emulsion appeared to provide some nutrition to the larvae, although more research should be conducted to improve the use of such replacements. [ABSTRACT FROM AUTHOR]

Abstract

Background: The presence of Human Herpesvirus-8 (HHV8) DNA is predictive of Kaposi sarcoma (KS) among patients with HIV-associated or iatrogenic immunosuppression. However, correlates of HHV8-DNA detection in the general population remain undefined. Methods: We assessed correlates of HHV8-DNA detection among Italian adults without KS who had antibodies against HHV8-latent nuclear antigen by immunofluorescence assay. HHV8-K6 DNA sequences were detected in peripheral blood mononuclear cells using TaqMan CR. Results: Of the 158 subjects 26 (16.5%) had detectable HHV8-DNA [median copies/million cells, 53; (13-2128)]. Adjusted for age, sex, and laboratory, HHV8-DNA was detected more frequently in participants with >7 total residents in the childhood home [OR = 3.7 (1.5-9.1)], >2 younger siblings [OR = 2.6 (1.1-6.5)], and current cardiovascular [OR = 3.6 (1.3-9.7)] or renal [OR = 3.1 (1.2-8.0)] disease. Finally, SIRMM can be viewed as an user friendly problem solving environment.

Abstract

Background HIV-1 genotypic susceptibility scores (GSSs) were proven to be significant prognostic factors of fixed time-point virologic outcomes after combination antiretroviral therapy (cART) switch/initiation. However, their relative-hazard for the time to virologic failure has not been thoroughly investigated, and an expert system that is able to predict how long a new cART regimen will remain effective has never been designed. Methods We analyzed patients of the Italian ARCA cohort starting a new cART from 1999 onwards either after virologic failure or as treatment-naïve. The time to virologic failure was the endpoint, from the 90th day after treatment start, defined as the first HIV-1 RNA > 400 copies/ml, censoring at last available HIV-1 RNA before treatment discontinuation. We assessed the relative hazard/importance of GSSs according to distinct interpretation systems (Rega, ANRS and HIVdb) and other covariates by means of Cox regression and random survival forests (RSF). Prediction models were validated via the bootstrap and c-index measure. Results The dataset included 2337 regimens from 2182 patients, of which 733 were previously treatment-naïve. We observed 1067 virologic failures over 2820 persons-years. Multivariable analysis revealed that low GSSs of cART were independently associated with the hazard of a virologic failure, along with several other covariates. Evaluation of predictive performance yielded a modest ability of the Cox regression to predict the virologic endpoint (c-index≈0.70), while RSF showed a better performance (c-index≈0.73, p < 0.0001 vs. Cox regression). Variable importance according to RSF was concordant with the Cox hazards. Conclusions GSSs of cART and several other covariates were investigated using linear and non-linear survival analysis. RSF models are a promising approach for the development of a reliable system that predicts time to virologic failure better than Cox regression. Such models might represent a significant improvement over the current methods for monitoring and optimization of cART.

222. Efficacy of triple dose albendazole treatment for soil-transmitted helminth infections.

Abstract

In Malaysia, soil-transmitted helminth (STH) infections still persist among indigenous communities. In the past, local studies have focused mostly on epidemiologic aspects of STH infections with a scarcity of information on the efficacy of deworming treatment. The present study consisted of 2 phases: a cross-sectional phase on current epidemiological status and risk factors of STH infections and a longitudinal study over 6 weeks on triple dose albendazole efficacy against STH infections. A total of 253 participants were recruited at baseline and a pre-tested questionnaire was administered to obtain information on socio-demographics, environmental and behavioural risk factors. Stool samples were evaluated using a modified Kato-Katz technique. Cure rate (CR) and egg reduction rate (ERR) were assessed at 3 weeks following a 3-day course of 400mg albendazole treatment and infection status were observed again at 6 weeks. Baseline positivity of trichuriasis, ascariasis and hookworm infections were 56.1%, 11.9% and 20.2%, respectively. Multivariate analysis showed age below 18 years old (P = 0.004), without latrine in house (P = 0.042) and indiscriminate defecation (P = 0.032) were associated with STH infections. In the longitudinal study (N = 89), CR for trichuriasis was 64.6%, while CR of 100% was observed for both ascariasis and hookworm. ERR was above 90% for all three STH species. A rapid increased of Trichuris trichiura egg output was observed at 6 weeks. In conclusion, STH infections are highly prevalent among indigenous communities. Children and teenagers, poor sanitation and hygiene behaviour were determinants for STH infections. Triple dose albendazole is found to be efficacious against Ascaris lumbricoides and hookworm infections but has moderate curative effect with high ERR against T. trichiura. Although triple dose albendazole regimen has logistic challenges and may not be a routine option, consideration of this treatment regime may still be necessary in selective communities to reduce high intensity of T. trichiura infection.

Abstract

The study of water pathways from the soil to the atmosphere through plants—the so-called soil–plant–atmosphere continuum (SPAC)—has always been central to agronomy, hydrology, plant physiology, and other disciplines, using a wide range of approaches and tools. In recent years, we have been witnessing a rapid expansion of interweaving monitoring activities and model development related to SPAC in climatic, ecological, and applications other than the traditional agrohydrological, and it is therefore timely to review the current status of this topic and outline future directions of research. The initiative for the special section of Vadose Zone Journal on SPAC emanated from several sessions we recently organized in international conferences and meetings. With a view to the specific research questions covered in this special section, this article introduces and reviews SPAC underlying issues and then provides a brief overview of the invited contributions. We have grouped together the 15 contributions under three main sections related to the local, field, and landscape spatial scales of interests. Within these sections, the papers present their innovative results using different measuring techniques (from classic tensiometers and TDR sensors to more advanced and sophisticated equipment based on tomography and geophysics) and different modeling tools (from mechanistic models based on the Richards equation to more parametrically parsimonious hydrologic balance models). They provide a snapshot of the current state of the art while emphasizing the significant progress attained in this field of research. New technological developments and applications are also highlighted.

Abstract

A panel of monoclonal antibodies (mAbs) was used for the characterisation of leucocyte subpopulations during the ontogeny of common carp ( Cyprinus carpio , L.) and sea bass ( Dicentrarchus labrax , L.). In carp the leucocytes were monitored in different lymphoid organs by immunofluorescence and flow cytometry using specific mAbs for early T cells (WCL9), B cells (WCI12), monocytes/macrophages (WCL15) and thrombocytes (WCL6).Early T cells were very numerous (77 %) in thymus during the first weeks post fertilisation (p.f.), but also present in other organs, especially head kidney. Subsequently, these cells disappeared from all organs, except the thymus (40%). B cells appeared in the head kidney from the second week p.f., and later on in the spleen and blood, but their number remained low in the thymus and gut. Thrombocytes were detected in cell suspensions of spleen from the first week p.f. and their percentage increased until the 4 thweek (30%) and then decreased in spleen (10%), but increased in blood (30%). Monocyte/macrophage-like cells were present in all organs from the first week p.f. and their percentage gradually increased until the 8 thweek p.f..By using mAb WCL15 on fixed tissue the in situ distribution of monocytes/macrophages was studied in thymus, head kidney, spleen and gut from 2 days until 60 weeks p.f.. Macrophages were found from day 2 p.f. in the head kidney and in the dorsal part of the yolk sac epithelium. From 1 week onwards, macrophages were also found scattered in the thymus and gut and during the second week also in spleen. The number of macrophages increased in all lymphoid tissues until the 6 th-8 thweek p.f., then decreased except in the thymus, where they became localised mainly at the cortical-medullary boundary.By using mAb WCL38 increasing numbers of mucosal T cells were observed in cell suspensions of gills and intestine from the first week p.f. onwards. With immuno-histochemistry the early appearance of mucosal T cells could be confirmed in the gills and intestine but was also detected in skin. With mAb WCL9 the ontogeny of the carp thymus was studied and special attention was paid to the development of cortex and medulla. The differentiation between cortex and medulla started in the 4 thweek p.f.. Ultrastructural study of the developing thymus confirmed these data and permitted the analysis of the distribution and morphological differences of the epithelial cells. In addition, numerous apoptotic cells appeared in the cortex from the 4 thweek p.f. onwards, while lower numbers were observed in medulla.In sea bass, mAbs DLT15 and DLIg3 specific for T and B cells, respectively, were employed to describe the morphology and distribution of these cells. Enriched leucocyte fractions from different tissues (thymus, spleen, head kidney, intestine and blood) were used. High numbers of T cells were detected in thymus and intestine, whereas B cells were more numerous in blood, head kidney and spleen. Furthermore, an ontogenetic study with DLT15 revealed that the thymus was the first organ with T cells, followed by head kidney and spleen. Although some differences were observed between the two commercially important fish species studied, the data presented in this thesis are of phylogenetic interest and can be used for the development of vaccination strategies in young fish.

237. Enhancing the Expressiveness of the Bunge-Wand-Weber Ontology

Abstract

In practical terms, conceptual modeling is at the core of systems analysis and design. The plurality of modeling methods available has however been regarded as detrimental, and as a strong indication that a common view or theoretical grounding of modeling is wanting. This theoretical foundation must universally address all potential matters to be represented in a model, which consequently suggested ontology as the point of departure for theory development. The Bunge–Wand–Weber (BWW) ontology has become a widely accepted modeling theory. Its application has simultaneously led to the recognition that, although suitable as a meta-model, the BWW ontology needs to be enhanced regarding its expressiveness in empirical domains. In this paper, a first step in this direction has been made by revisiting BUNGE’s ontology, and by proposing the integration of a “hierarchy of systems” in the BWW ontology for accommodating domain specific conceptualizations.

Organizations generally are not responding effectively to rising IT security threats because people issues receive inadequate attention. The stark example of IT security is just the latest strategic IT priority demonstrating deficient IT leadership attention to the social dimension of IT. Universities in particular, with their devolved people organization, diverse adoption of IT, and split central/local federated approach to governance and leadership of IT, demand higher levels of interpersonal sophistication and strategic engagement from their IT leaders. An idealized model for IT leaders for the 21st century university is proposed to be developed as a framework for further investigation. The testing of this model in an action research study is proposed.

Organizations invest substantial resources in Enterprise Systems (ES) expecting positive outcomes for the organization. Implementing an ES is a lengthy-costly undertaking, with general upheaval for many of the organizations. Many organizations therefore are seriously considering rapid implementations of ES to reduce related resources. A rapid ES implementation requires effective management of knowledge (KM) as the extent of the engagement of external and internal parties (consultants and vendors with the client) is limited. This research paper introduces a theoretical model to assess the impact of KM in a rapid implementation of SAP R/3 that had completed in a record time of three weeks. Using the Adaptive Structuration Theory (AST) this paper proposes a theoretical model 1) to identify the KM enablers and KM strategies of an rapid ES implementation that facilitate knowledge creation, retention and transfer and 2) to determine the importance of knowledge transfer modes in a rapid ES implementation.

240. From ad-hoc to engineered collaboration in virtual workspaces

Abstract

Distributed collaboration over the Internet has become increasingly common in recent years, supported by various technologies such as virtual workspace systems. Often such collaboration is ad-hoc, and virtual workspaces are set up anew for each new instance of collaboration. We propose that much of the ad-hoc collaboration can be captured and transformed into patterns for reuse in future collaboration. This paper presents the results of the past five years of our work in this area. We introduce the notion of patterns of virtual collaboration; present a framework for extracting patterns of work in virtual workspace systems; and introduce an information model of virtual collaboration. We then present an overview of our data and process mining methods and reverse engineering techniques for discerning work processes carried out through virtual workspace systems. Finally we present our visual mining techniques that we use to discern aspects of work processes in virtual workspaces.

Abstract

Lmphomyeloid organs of three common Antarctic fish species, Trematomus bernacchii, Trematomus nicolai and Chionodraco hamatus, were analysed. Contrary to species living in temperate sea water, the thymus of polar fishes were flattened, incompletely lobated and scarcely distinguishable by normal histology into cortical and medullary regions. Functional regionalisation, however, was suggested by differences in the sizes of thymocytes from the outer to the inner thymus zone. Another particularity was observed in the thymus of Trematomus species: next to lymphocytes, numerous erythroid cells circulated and differentiated in the parenchyma. Only two main types of epithelial cells could be found by cytological analysis: (i) limiting cells that surround the haematopoietic tissue and (ii) reticular cells that constitute the frame where the lymphoid and erythroid cells can proliferate and differentiate. The reticular cells could not be distinguished in cortical and medullary subtypes as observed in temperate-water fish. Numerous Hassall's corpuscles, probably with a scavenging role, were also observed in the thymus. The head kidney housed haematopoietic tissue, lacked any excretory tubules, and had a huge blood supply, characteristic of polar fish species. It appeared mainly lymphopoietic in C. hamatus but contemporary erythropoietic and lymphopoietic in Trematomus species. The ultrastructural analysis revealed the presence of both reticular and limiting epithelial cells. Reticular epithelial cells (REC) characteristically showed numerous vesicles with a granular content and cell debris. Numerous lymphoblasts, lymphocytes and plasma cells were observed among the REC. Erythropoiesis occurred in all polar species analysed, but in C. hamatus the erythroblasts did not differentiate because they had a fast senescence. The spleen appeared mainly erythropoietic, with scarcely developed areas of white pulp, in Trematomus species; the erythropoiesis was scarcely evident in C. hamatus

Abstract

Biogeochemical cycling of elements largely occurs in dissolved state, but many elements may also be bound to natural nanoparticles (NNP, 1–100 nm) and fine colloids (100–450 nm). We examined the hypothesis that the size and composition of stream water NNP and colloids vary systematically across Europe. To test this hypothesis, 96 stream water samples were simultaneously collected in 26 forested headwater catchments along two transects across Europe. Three size fractions (~1–20 nm, >20–60 nm, and >60 nm) of NNP and fine colloids were identified with Field Flow Fractionation coupled to inductively coupled plasma mass spectrometry and an organic carbon detector. The results showed that NNP and fine colloids constituted between 2 ± 5% (Si) and 53 ± 21% (Fe; mean ± SD) of total element concentrations, indicating a substantial contribution of particles to element transport in these European streams, especially for P and Fe. The particulate contents of Fe, Al, and organic C were correlated to their total element concentrations, but those of particulate Si, Mn, P, and Ca were not. The fine colloidal fractions >60 nm were dominated by clay minerals across all sites. The resulting element patterns of NNP <60 nm changed from North to South Europe from Fe‐ to Ca‐dominated particles, along with associated changes in acidity, forest type, and dominant lithology.
