125I-Concanavalin A was found to bind to purified Golgi membranes in a manner which was time-dependent, proportional to the amount of Golgi protein, saturable, and inhibited by alpha-methyl-D-glucoside and unlabeled concanavalin A. Approximately 2 nmol of 125I-concanavalin A were bound per mg of membrane protein with a mean Ka of 0.2 x 10(7) M-1. After binding of concanavalin A to the membranes, all unbound lectin was removed by repeated washing prior to assay of galactosyltransferase activity to assure that bulk concanavalin A would not bind to the sugar acceptor during the enzyme assay. Therefore, the effects of the lectin were not due to altered acceptor concentrations in the assay. As a result of concanavalin A binding the activity of the galactosyltransferase of these membranes was enhanced by 70 to 100%. The enhancement was abolished by treatment of the concanavalin A-bound Golgi with alpha-methylglucoside. Kinetically this stimulation was reflected as an altered Km for N-acetylglucosamine and UDP-galactose. The data are suggestive that the stimulation of the enzyme results indirectly from perturbation of the membranes on binding of the lectin.

The same three low molecular weight copper-binding proteins which have been purified from the soluble fractions of Cu2+-loaded livers (200 mug Cu2+ per g wet weight) were also found to be present in livers of normal rats (4 mug Cu2+ per g wet weight) but in much smaller quantities. Therefore, Cu2+ loading enhances the amount of preexisting proteins rather than inducing the synthesis of new proteins. Amino acid analyses of each of the three showed them to be different from the metallothioneins which are present on loading with other trace metals including Cd2+ and Zn2+.

Recent electrophysiological and pharmacological studies have confirmed previous clinical evidence that the gene defect in cystic fibrosis is strongly expressed in the sweat gland. This has provided a major impetus to efforts to culture the cells of this tissue in order to provide a source of experimental material for molecular studies. Toward this end, eccrine sweat glands were isolated from collagenase treated skin specimens and the secretory coil and the reabsorptive duct separated. Segments of each portion of the gland were transferred to a plastic or collagen substrate and covered with serum-containing or serum-free defined growth media. Epithelial cell outgrowth took place in both media but fibroblast overgrowth occurred in the presence of serum at concentrations as low as 1%. In serum-free medium both secretory and reabsorptive cells formed tightly joined epithelial sheets, first as monolayers and later as multilayers consisting of at least six cell layers. Growth continued for approximately fifteen generations each of about two and a half days. Remarkably large domes or hemicysts with diameters as great as two cm were formed, apparently attesting to the retention of the capacity of the cells to actively transport ions and water. Ultrastructurally, cells which grew out from the secretory coil resembled the fluid secreting clear cells; neither dark cells nor myoepithelial cells were propagated.

Insulin and insulin receptors may play a role in the modulation of cytotoxic effector cell function. We have investigated the effects of insulin on ADCC. At low effector-to-target cell ratios, the addition of insulin resulted in a dose-dependent reduction of erythrocyte target cell lysis but not lysis of tumor target cells. In a patient with insulin resistance, total lipodystrophy, and a demonstrable reduction in specific insulin binding to circulating monocytes, ADCC for erythrocyte target cells was unaffected by the addition of insulin. In addition, incubation of normal effector cells in patient's serum similarly abrogated the effect of insulin. The failure of insulin to inhibit ADCC for erythrocyte target cells may serve as a reliable and simple assay for the demonstration of altered insulin receptors or for the detection of anti-insulin receptor antibodies.

Multidrug resistance is a complex pleiotropic phenotype of cross-resistance and collateral sensitivity to unrelated compounds observed in many mammalian cell mutants selected for resistance to single agents. In Chinese hamster ovary cells, colchicine-resistant mutants expressing this phenotype have been characterized extensively. Such mutants arise apparently from a single genetic event, and the basis of this phenotype appears to be localized at the membrane level, resulting in altered drug permeability. Expression of a 170,000-dalton surface glycoprotein (P-glycoprotein) has been identified to correlate with the multidrug-resistance phenotype. Selection of a second mutation in colchicine-resistant mutants, for resistance to phytohemagglutinin, results in an alteration of the carbohydrate moiety in P-glycoprotein and other surface components. This mutation does not noticeably affect the multi-drug-resistance phenotype. The altered permeability of mutant cells to drugs, however, can be modulated by nonionic detergents or metabolic inhibitors. These findings are consistent with a molecular mechanism of multidrug resistance whereby the pleiotropic response of the cell is mediated by an overexpression of a cell-surface protein, the P-glycoprotein.

Treatment of liver plasma membranes with phospholipase A2 or high doses of concanavalin A enhances the activity of Mg2+ATPase assayed at temperatures greater than 30 degrees C. The effects of the two treatments are not additive. Both the removal of phospholipids and binding of the lectin increase the degree of polarization of fluorescence of the lipid-soluble fluorophores, diphenylhexatriene and beta-parinaric acid, suggesting that decreased lipid fluidity may activate Mg2+-ATPase. In fact modification of lipid fluidity by reconstitution of phospholipase-treated membranes with phosphatidylcholines of defined fatty acid composition or by addition of cis-vaccenic acid showed a strong inverse correlation between Mg2+ATPase activity and lipid fluidity as monitored by fluorescence polarization. However, despite the ability of concanavalin A to nonspecifically order membrane lipid, its effect on Mg2+ATPase is apparently not mediated in this manner because other enzyme-activating lectins such as Ricinus communis agglutinin and wheat germ agglutinin are without effect on lipid fluidity. The facts that lectins of lower valency than tetravalent native concanavalin A such as divalent succinyl concanavalin A are far less effective in activating the enzyme and that paraformaldehyde treatment also activates suggests that cross-linking of membrane proteins is responsible. Hence, the diminution in activity of this membrane enzyme due to the disordering effect of heat in the physiological temperature range can be counteracted by isothermally increasing the order of either membrane lipid or protein.

We have prepared a lambda gt10 cDNA library with the mRNA isolated from fetal calf brains which were actively myelinating. Using two oligonucleotides made according to the known amino acid sequence of myelin proteolipid protein (PLP or lipophilin), we have isolated several cDNA clones for this major intrinsic membrane protein of myelin. One of these clones, designated as pLP1, is found to contain 444 bp of coding sequence for the C-terminal half of PLP and 486 bp of 3' untranslated sequence. Using pLP1 as a hybridization probe, we have studied the regulation of PLP at the mRNA level during rat brain development. Our results show that the relative amounts of mRNA for PLP and that for the major extrinsic protein of the myelin membrane, myelin basic protein, increase coordinately during the course of myelination in the brain.

Previous studies have shown that calcium channel blockers can overcome, at least partially, multidrug resistance (MDR). This study was undertaken to attempt to determine the mechanisms whereby these agents bring about this effect. Their influence on the uptake and retention of several cancer drugs and on the toxic actions of these compounds was assessed employing MDR cell lines from several species. The wild-type drug sensitive parent cells proved to be more susceptible than the multidrug resistant variants to the effects of calcium channel blockers on cancer drug accumulation. This was shown for verapamil, nifedipine and the calmodulin inhibitor trifluoperazine acting on human, mouse and Chinese hamster ovary (CHO) cell lines. The enhancement of drug accumulation by calcium antagonists was similar to that caused by non-ionic detergents. Furthermore, verapamil was unable to alter 45Ca2+ accumulation in sensitive or resistant cells, suggesting that these agents act in a calcium-independent manner. Verapamil accumulation in multidrug resistant cells was reduced compared to sensitive cells. In spite of this reduced accumulation, however, verapamil alone was much more toxic to multidrug resistant cells than to the sensitive cells. This suggests that calcium channel blockers are specifically toxic to MDR cells by virtue of an interaction with the MDR cell surface distinct from that involved in promoting cellular accumulation.

Independent lines of Chinese hamster ovary cells resistant to the antineoplastic drug, daunorubicin, were obtained by clonal isolation in increasing drug concentrations. A single daunorubicin-resistant phenotype typified by reduced cellular drug accumulation was observed. These mutants displayed a complex phenotype of resistance to a variety of unrelated drugs. Such properties are similar to those of membrane-altered colchicine-resistant lines (V. Ling and L.H. Thompson, J. Cell. Physiol., 83: 103-116, 1974). Analysis of the plasma membrane components of the daunorubicin-resistant clones by gel electrophoresis revealed a prominent cell surface glycoprotein with a molecular weight of about 170,000. This component was immunologically cross-reactive with the cell surface P-glycoprotein of about the same molecular weight, previously identified in colchicine-resistant cells. Thus, it appears that the mechanism of resistance characterized by P-glycoprotein expression could be the basis of many drug-resistant phenotypes.

The membrane glycoprotein enzyme, alkaline phosphatase was induced in cultured human fibroblasts by dibutyryl cyclic AMP, sodium butyrate, the serum glycoprotein fetuin, the Tamm-Horsfall urinary glycoprotein, and by a number of inhibitors of DNA synthesis. The uninduced basal enzyme activity increased at later stages of growth when the cells became confluent. Induction by dibutyryl cyclic AMP or fetuin was most effective when the agents were added after the cells had reached stationary phase and was maximal after at least two days of exposure. The levels of induction resulting from the addition of pairs of the agents, dibutyryl cyclic AMP, n-butyrate and fetuin were additive indicating that these have different modes of action. The inhibitors of DNA synthesis, cytosine arabinoside, hydroxyurea, and methothrexate were less effective inducers. Bromodeoxyuridine which also has non-DNA mediated effects induced to the same extent as dibutyryl cyclic AMP. Similar experiments with sex- and age-matched cell strains derived from patients with cystic fibrosis failed to detect differences in the levels of induction from those observed in normal cells. In addition, the combined inductive effects of Tamm-Horsfall glycoprotein, isoproterenol and theophylline, were similar with normal and cystic fibrosis cells.

Several inherited disorders in humans and in rodents result in myelin dysgenesis and a deficiency of the molecular constituents of myelin. A complementary DNA to one of the two major myelin proteins, myelin proteolipid protein (also known as lipophilin), has been used with Southern blot analysis of somatic cell hybrid DNA to map the human proteolipid protein gene to the middle of the long arm of the human X chromosome (bands Xq13-Xq22) and to assign the murine proteolipid protein gene to the mouse X chromosome. Comparison of the gene maps of the human and mouse X chromosomes suggests that myelin proteolipid protein may be involved in X-linked mutations at the mouse jimpy locus and has implications for Pelizaeus-Merzbacher disease, a human inherited X-linked myelin disorder.

Metallothionein was purified under anaerobic conditions from livers of human fetuses ranging from 19 weeks gestational age to term. Homogeneous metallothionein obtained in the absence of reducing agents from the soluble fraction of the tissue which contained 24% and 85% of the total liver copper and zinc, respectively, had less than 1 g atom of copper and about 3.1 g atoms of zinc/mol. Extraction with 1% 2-mercaptoethanol of the insoluble fraction of the tissue (containing 76% and 15% of the liver copper and zinc, respectively) yielded a metallothionein with approximately 2.5 g atoms of copper and 1.3 g atoms of zinc. When the whole tissue was extracted similarly by the procedure of Rydén and Deutsch ((1978) J. Biol. Chem. 253, 519), nearly equal proportions of the two metals were obtained in the soluble fraction from which metallothionein, with about 3.0 g atoms of zinc and 1.5 g atoms of copper, was purified. Thus, zinc- and copper-rich forms of metallothionein are differently distributed between the soluble and insoluble fractions of fetal liver. Nevertheless, the predominant metal in metallothionein of human fetal liver is zinc as is the case in the adult; significant amounts of copper are also present, consistent with the elevated quantity of this metal in the fetal tissue relative to the adult. Cadmium was not detected. Zinc is much more easily removed from the protein than is copper.

Nonresponse to chemotherapy may result from the acquisition of multidrug resistance by malignant cells. Overexpression of the 170,000 dalton cell surface P-glycoprotein is associated with this phenotype and this appears to result from amplification of a multigene family coding for this protein. A cDNA encoding a conserved portion of P-glycoprotein has been cloned from hamster cells, and this was used in the present study to localize human P-glycoprotein gene sequences to chromosome 7q36.

The dependence of the specific activities of human alpha-L-fucosidase and four other lysosomal glycosidases on cell growth was determined in long-term lymphoid cell lines in which the polymorphic forms of alpha-L-fucosidase were identified by isoelectric focusing. In cultured lymphoblasts of Fu-1 phenotype the alpha-L-fucosidase synthesis during cell growth was found to be proportional to the general amount of protein. However, in cells derived from individuals possessing at least one FUCA*2 allele, the amount of enzyme increased progressively during the log-phase of growth, reaching a maximum at the transition to the stationary phase. The maximum increase of enzyme concentration was observed in the mid-log phase of exponential growth. The results may be explained by the existence of a regulatory locus FUCT which maps in close proximity to the structural gene. The FUCT locus which controls the amount of enzyme in the cell appears to be specific for alpha-L-fucosidase.

Genetic linkage analysis with polymorphic DNA markers (restriction fragment length polymorphisms: RFLPS) has allowed the assignment of the cystic fibrosis (CF) locus to the long arm of chromosome 7, within the region of band q31. Two of these markers, MET and D7S8, are tightly linked to the disease locus. Although recent data suggest that they are located on opposite sides of CF, the two can be separated by as much as 5 centimorgans. To obtain a better description of the CF locus and, eventually, to identify the affected gene, additional DNA markers are required to connect MET and D7S8, physically. We have screened the flow-sorted chromosome-7-specific library and thus far isolated 28 new probes from the 7q31 region by DNA hybridization analysis that uses a series of somatic cell hybrids containing various portions of human chromosome 7. Together with the previously identified markers, MET, D7S8, D7S13 and D7S16, these new markers should provide a fine genetic and physical map for the chromosomal region surrounding CF. DNA segments can then be sequentially cloned by chromosome walking from points closest to the CF locus and examined for genes that are preferentially expressed in tissues known to be affected in the disease.

The membranes of mammalian cells are composed of an ordered array of lipids and proteins, the latter containing carbohydrate residues directed towards the exterior and important in the interaction of cells with each other and with external proteins. This external (plasma) membrane and other more simple membranes within the cell are damaged in all diseases which compromise the integrity of the cell. However, in many cases, chemical or functional changes in these membranes are central to the pathogenesis of the disease. These processes are illustrated, and a classification of membrane-related diseases is proposed. This includes: receptor-related diseases such as type II familial hypercholesterolaemia, Grave's disease, some lysosomal storage diseases and some forms of diabetes and obesity; structural instability as manifested by red cell abnormalities and multiple sclerosis; changes in lipid state as in muscular dystrophy and multiple sclerosis; altered permeability or transport as in cystic fibrosis, diseases associated with specific transport defects, and the action of many bacterial toxins, and abnormality of the cytoskeleton-membrane interface as in Chediak-Higashi disease and some diseases associated with red cell abnormalities. Different mechanisms can contribute to the membrane disorder in a single disease state and many of these are described to illustrate this diversity.

The binding to isolated hepatocyte plasma membranes of radioactively labelled inhibitors of microfilamentous and microtubular protein function ([3H]cytochalasin B and [3H]colchicine, respectively) was studied as one means of assessing the degree of association of these proteins with cell surface membranes. [3H]Cytochalasin B which behaved identically to the unlabelled compound with respect to binding to these membranes was prepared by reduction of cytochalasin A with NaB3H4. The binding was rapid, readily reversible, proportional to the amount of membrane and relatively insensitive to changes of pH or ionic strength. At 10(-6) M [3H]cytochalasin B, glucose of p-chloromercuribenzoate, an inhibitor of glucose transport inhibited binding by about 20%; treatment of membranes with 0.6 M KI which depolymerizes F actin to G actin caused about 60% inhibition of binding. These two types of inhibition were additive indicating two separate classes of binding sites, one associated with sugar transport and one with microfilaments. Filamentous structures with the diameter of microfilaments (50 A) were seen in electron micrographs of thin sections of the membranes. At concentrations greater than 10(-5) M [3H]cytochalasin B, binding was proportional to drug concentration, characteristic of non-specific adsorption or partitioning. Intracellular membranes of the hepatocyte also bound [3H]cytochalasin B, those of the smooth endoplasmic reticulum to a greater extent than plasma membranes. [3H]Colchicine bound to plasma membranes in proportion to the amount of membrane and at a rate compatible with binding to tubulin. However, other properties of the binding including effects of temperature, drug concentration and antisera against tubulin were different from those of binding to tubulin. Hence, no evidence was obtained for association of microtubular elements with these membranes. Despite this there appeared to be an interdependence between microtubule and microfilament inhibitors: vinblastine sulfate stimulated [3H]cytochalasin B binding and cytochalasin B stimulated 3H colchicine binding. [3H]Colchicine also bound to intracellular membranes, especially smooth microsomes.

In the presence of 8 mM external Ca++, the K+ permeability of human red cell ghosts increases provided K+ is also present in the medium. This increase does not represent K+/K+ exchange but a stimulation of net K+ efflux. The stimulation is half-maximal at 0.7 +/- 0.15 mM (n=5). At concentrations above 4.0 mM, external K+ inhibits net K+ efflux. Similar stimulatory and inhibitory effects of external K were also observed in intact cells after exposure to Pb++ or to Ca++ in the presence of fluoride, iodoacetate plus adenosine, or propranolol, suggesting that a common K+ -activated K+ -specific transfer system may be involved under all of these various circumstances. Internal K+ also stimulates net K+ efflux from ghosts, but it is uncertain whether internal K+ is an absolute requirement for the K+ permeability increase. In contrast to external Na+ which slightly stimulates K+ efflux, internal Na+ inhibits. The inhibition by internal Na+ is abolished by sufficiently high concentrations of external K+, showing that K+ binding to the outer membrane surface and Na+ binding to the internal surface are mutually interdependent. In red cell ghosts the Ca++ -K+ -stimulated net K+ efflux increases with increasing pH until a plateau is reached between pH 7.2 and 8.0. In fluoride-poisoned intact cells, the Ca++-K+ stimulated flux passes through a maximum around pH 6.8. Neither internal nor external Mg++ interferes with the combined effects of Ca++ and K+. Similarly, external EDTA has no influence at concentrations which are far lower than the Ca++ concentration required to produce a maximal response. In contrast, low concentrations of internal EDTA prevent the permeability change.

Cultured lymphoblasts derived from infants with Menkes' disease exhibit the same increased avidity for copper as do fibroblasts and most extrahepatic tissues from these patients. The Menkes' cells preferentially take up not only copper but also, on exposure to elevated metal concentrations, the other metallothionein-binding metals, zinc and cadmium. Menkes' lymphoblasts contain larger amounts of metallothionein than normal cells following exposure to each of these metals; the amount bound to this protein quantitatively accounted for the total cellular increment in metal in Menkes' cells. Induction of metallothionein synthesis caused both normal and Menkes' cells to subsequently take up increased amounts of 67Cu. These observations suggest that an enhanced capacity of Menkes' cells to accumulate metallothionein may be responsible for their increased uptake and retention of copper.

The plasma membranes of hamster, mouse, and human tumor cell lines that display multiple resistance to drugs were examined by gel electrophoresis and immunoblotting. In every case, increased expression of a 170,000-dalton surface antigen was found to be correlated with multidrug resistance. This membrane component is of identical molecular size and shares some immunogenic homology with the previously characterized P-glycoprotein of colchicine-resistant Chinese hamster ovary cells. This finding may have application to cancer therapy.

Colchicine-resistant Chinese hamster ovary (CHO) cell mutants whose resistance results from reduced drug permeability have been isolated previously in our laboratories. This reduced permeability affects a wide range of unrelated drugs, resulting in the mutants displaying a multiple drug resistance phenotype. A 170,000-dalton cell surface glycoprotein (P-glycoprotein) was identified, and its expression appears to correlate with the degree of resistance. In this study we were able to confer the multiple drug resistance phenotype on sensitive mouse L cells by DNA-mediated gene transfer of DNA obtained from the colchicine-resistant mutants. P-glycoprotein was detected in plasma membranes of these DNA transformants by staining with an antiserum raised against membranes of mutant CHO cells. These results are consistent with a causal relationship between P-glycoprotein expression and the multiple drug resistance phenotype.
