Abstract

Q fever is a highly infectious disease with a worldwide distribution. Its causative agent, the intracellular bacterium Coxiella burnetii, infects a variety of vertebrate species, including humans. Its evolutionary origin remains almost entirely unknown and uncertainty persists regarding the identity and lifestyle of its ancestors. A few tick species were recently found to harbor maternally-inherited Coxiella-like organisms engaged in symbiotic interactions, but their relationships to the Q fever pathogen remain unclear. Here, we extensively sampled ticks, identifying new and atypical Coxiella strains from 40 of 58 examined species, and used this data to infer the evolutionary processes leading to the emergence of C. burnetii. Phylogenetic analyses of multi-locus typing and whole-genome sequencing data revealed that Coxiella-like organisms represent an ancient and monophyletic group allied to ticks. Remarkably, all known C. burnetii strains originate within this group and are the descendants of a Coxiella-like progenitor hosted by ticks. Using both colony-reared and field-collected gravid females, we further establish the presence of highly efficient maternal transmission of these Coxiella-like organisms in four examined tick species, a pattern coherent with an endosymbiotic lifestyle. Our laboratory culture assays also showed that these Coxiella-like organisms were not amenable to culture in the vertebrate cell environment, suggesting different metabolic requirements compared to C. burnetii. Altogether, this corpus of data demonstrates that C. burnetii recently evolved from an inherited symbiont of ticks which succeeded in infecting vertebrate cells, likely by the acquisition of novel virulence factors.

Here, we address this evolutionary issue in the case of Q fever. Its causative agent, the intracellular bacterium Coxiella burnetii, is extremely infectious to humans and a variety of animals. However, uncertainty persists regarding its evolutionary origin, including the identity and lifestyle of its ancestors. In this article, we show that C. burnetii arose from a rare evolutionary transformation of a maternally-inherited endosymbiont of ticks into a specialized and virulent pathogen of vertebrates. While arthropod symbionts are typically transmitted maternally and thought not to be infectious to vertebrates, we establish here that one Coxiella symbiont has evolved the necessary adaptations to exploit the vertebrate cell, leading to the emergence of Q fever.

Abstract

International audience; M-TiO2 (M = Ag, Cu) nanocomposite layers were grown by pulsed direct liquid injection chemical vapor deposition (DLICVD) on various substrates to produce bactericidal surfaces with long term activity. Monodisperse Ag nanoparticles (NPs) with an average size of 5-10 nm are embedded in an anatase matrix. A bactericidal behavior determined by the JIS Z 2801 standard test was found for Ag-TiO2 films for Ag ≤ 1 at. % and above. Higher Ag content is not necessary since efficiency is already at its maximum (relative activity 100%). By contrast, using Cu as antibacterial agent, a larger size distribution of metal particles was found (20 to 400 nm). Cu-TiO2 films exhibit a bactericidal behavior if their thickness is higher than 100 nm and Cu content ≥ 3.5 at. %. These coatings are still antibacterial after 5 months of aging and their efficiency has decreased by only 35%.

Abstract

National audience; Emerging infectious diseases are actually of major concern worldwide. Most of them are zoonotic and their epidemiology must consequently be studied conjointly in humans and animals. We present here a long-term research program led in the Camargue. This area situated in the Rhone delta, crossroads of many birds‘ migration routes is a biodiversity hotspot. It appears vulnerable to disease emergence due to its key location and to the common use of wetlands by wild and domestic animals as well as humans. Since 2004, a health ecology program is led in the Camargue mainly focusing on Influenza A viruses. First, pathogen dynamics are studied in their wild reservoir. For example, a seasonal pattern of avian influenza A virus circulation in waterfowl was highlighted, an infection peak being generally observed in early fall, probably due to the massive arrival of young and possibly immunologically naive birds. Seasonality of avian Influenza viruses is currently studied in gulls. Second, pathogen transmission between wild and domestic birds is investigated. Interestingly, no highly pathogenic avian influenza virus was detected in wild birds of the region from 2006 to 2009 suggesting that migrating birds do not play an important role in the transmission of these viruses. On the other hand, the potential impact of domestic duck releases for shooting on avian influenza A viruses dynamics in wild waterfowl is currently studied. Finally, researches are led on pathogen exchanges between wildlife and humans. Antibio-resistant bacteria, similar to those circulating in humans in France, have been detected in gulls showing that they could be contaminated by human pathogens. This multidisciplinary program already proved that animal health, human health and ecology issues can be fruitfully linked to gain insight in the epidemiology of emerging pathogens. Abstract

Le Languedoc-Roussillon présente des caractéristiques particulières ou un niveau particulier de risques: le vieillissement de la population, particulièrement évident dans la région, ainsi que l'augmentation de la population en situation précaire posent des problèmes de nutrition et de prise en charge. L'alimentation de type méditerranéen, dont les bienfaits pour la santé sont reconnus, est progressivement remplacée par une alimentation mondialisée, standardisée et peu variée, ce qui entraîne, avec le développement de la sédentarité, une épidémie d'obésité/diabète avec, entre autres, des conséquences cardio-vasculaires. Parmi les maladies émergentes, il faut noter certaines formes de cancer, des maladies neuro-dégénératives (Alzheimer), et nombre de maladies infectieuses et parasitaires qui, du fait des échanges constants avec la région, risquent de se développer, les vecteurs ou réservoirs étant présents sur notre territoire. Une réflexion des responsables locaux et de la population est nécessaire.

Abstract

Few studies have analyzed the gut microbiota of child in unindustrialized countries, but none during the first month of life. Stool samples were collected from healthy newborns in hospitals of Gabon (n = 6) and Republic of the Congo (n = 9) at different time points during the first month of life: meconium, day 2 (D02), day 7 (D07) and day 28 (D28). In addition, one fecal sample was collected from each mother after delivery. Metagenomic sequencing was performed to determine the bacterial communities, and multiplex real-time PCR was used to detect the presence of seven enteric viruses (rotavirus a, adenovirus, norovirus I and II, sapovirus, astrovirus, enterovirus) in these samples. Bacterial diversity was high in the first days of life, and was dominated by the genus Prevotella. Then, it rapidly decreased and remained low up to D28 when the gut flora was composed almost exclusively of strictly anaerobic bacteria. Each infant’s fecal bacterial microbiota composition was significantly closer to that of their mother than to that of any other woman in the mothers’ group, suggesting an intrauterine, placental or amniotic fluid origin of such bacteria. Moreover, bacterial communities differed according to the delivery mode. Overall, the bacterial microbiota communities displayed a similar diversification and expansion in newborns within and between countries during the first four weeks of life. Moreover, six of the fifteen infants of this study harbored enteric viruses (rotavirus, enterovirus and adenovirus) in fecal samples, but never in the meconium. A maternal source for the viruses detected at D02 and D07 can be excluded because none of them was found also in the child’s mother. Abstract

Acquired carbapenemases currently pose one of the most worrying public health threats related to antimicrobial resistance. A NDM-1-producing Salmonella Corvallis was reported in 2013 in a wild raptor. Further research was needed to understand the role of wild birds in the transmission of bacteria resistant to carbapenems. Our aim was to investigate the presence of carbapenem-resistant Escherichia coli in gulls from southern France. In 2012, we collected 158 cloacal swabs samples from two gull species: yellow-legged gulls ( Larus michahellis) that live in close contact with humans and slender-billed gulls ( Chroicocephalus genei) that feed at sea. We molecularly compared the carbapenem-resistant bacteria we isolated through culture on selective media with the carbapenem-susceptible strains sampled from both gull species and from stool samples of humans hospitalized in the study area. The genes coding for carbapenemases were tested by multiplex PCR. We isolated 22 carbapenem-resistant E. coli strains from yellow-legged gulls while none were isolated from slender-billed gulls. All carbapenem-resistant isolates were positive for bla VIM-1 gene. VIM-1-producing E. coli were closely related to carbapenem-susceptible strains isolated from the two gull species but also to human strains. Our results are alarming enough to make it urgently necessary to determine the contamination source of the bacteria we identified. More generally, our work highlights the need to develop more bridges between studies focusing on wildlife and humans in order to improve our knowledge of resistant bacteria transmission routes.
