211. Anaerobic Membrane Bioreactors for Municipal Wastewater Treatment, Sewage Sludge Digestion and Biogas Upgrading: A Review.

Abstract

Anaerobic membrane bioreactors (AnMBRs) are formed through the combination of anaerobic digestion and membrane technology. The upgraded technology separates SRT from HRT in the anaerobic digester, shortening the treatment period, reducing the digester's volume and improving effluent quality. Furthermore, AnMBRs have a strong tolerance for the existing forms of objects and can handle liquids, high-solid materials and gases. Up to now, AnMBRs have been utilized in the treatment of various types of industrial and municipal wastewater, organic solid waste and also biogas upgrading, and they have achieved excellent performance. However, there are few studies which have discussed their multiple utilization, especially following the flow of wastewater treatment. This study summarizes the application of AnMBRs in their diverse roles in the municipal wastewater treatment process. The discussion revolves around energy generation and the fouling issue of AnMBRs in the treatment of municipal wastewater, the digestion of sewage sludge generated in conventional municipal wastewater treatment and the upgrading of biogas after anaerobic digestion. In addition to controlling operating parameters, strategies used to improve the treatment effectiveness are also introduced. Lastly, online methods for preventing membrane fouling, which is the main operational obstacle to AnMBRs' wider spread, are also discussed. 214. Stocking density affects the growth performance and metabolism of Amur sturgeon by regulating expression of genes in the GH/IGF axis

Abstract

The effects of stocking density on the growth and metabolism of Amur sturgeon were assessed. Amur sturgeon were grown for 70 days at three different stocking densities (low stocking density, LSD: 5.5 kg/m3; medium stocking density, MSD: 8.0 kg/m3; and high stocking density, HSD: 11.0 kg/m3), and the biometric index, muscle composition, and serum biochemical parameters were evaluated. In addition, pituitary, liver, and muscle samples were collected for gene cloning and expression analyses. After 70 days of growth, the fish maintained at HSD had significantly lower final body weight and specific growth rate, and a higher feed conversion ratio than those of the fish in the MSD and LSD groups. The HSD group had the lowest lipid and protein concentrations in serum and muscle. The serum cortisol concentration increased significantly in the HSD group, indicating that the stress-response system was activated in these fish. There was no change in the concentration of serum insulin-like growth factor 2 (IGF-2), while the concentrations of serum growth hormone (GH) and insulin-like growth factor 1 (IGF-1) decreased in the HSD group. The full-length cDNAs of GHand IGF-2 genes (995-bp and 1 207-bp long, respectively), were cloned and analyzed. In the HSD group, the expressions of GHin the pituitary and growth hormone receptor (GHR) and IGF-1in the liver were down-regulated at the end of the 70-day experiment. In the HSD group, the transcript level of IGF-2significantly decreased in the liver, but did not change in muscle. Overall, our results indicated that a HSD negatively affects the growth performance and leads to changes in lipid and protein metabolism in Amur sturgeon. The down-regulated expression of genes related to the GH/IGF axis may be responsible for the poor growth performance of Amur sturgeon under crowding stress.

233. A Preclinical Systematic Review of the Effects of Chronic Exercise on Autophagy-Related Proteins in Aging Skeletal Muscle

Abstract

Background: Exercise is one of the most effective interventions for preventing and treating skeletal muscle aging. Exercise-induced autophagy is widely acknowledged to regulate skeletal muscle mass and delay skeletal muscle aging. However, the mechanisms underlying of the effect of different exercises on autophagy in aging skeletal muscle remain unclear.Methods: A systematic review was performed following an electronic search of SCOPUS, PubMed, Web of Science, ScienceDirect, and Google Scholar and two Chinese electronic databases, CNKI and Wan Fang. All articles published in English and Chinese between January 2010 and January 2022 that quantified autophagy-related proteins in aging skeletal muscle models.Results: The primary outcome was autophagy assessment, indicated by changes in the levels of any autophagy-associated proteins. A total of fifteen studies were included in the final review. Chronic exercise modes mainly comprise aerobic exercise and resistance exercise, and the intervention types include treadmill training, voluntary wheel running, and ladder training. LC3, Atg5-Atg7/9/12, mTOR, Beclin1, Bcl-2, p62, PGC-1α, and other protein levels were quantified, and the results showed that long-term aerobic exercise and resistance exercise could increase the expression of autophagy-related proteins in aging skeletal muscle (p < 0.05). However, there was no significant difference in short term or high-intensity chronic exercise, and different types and intensities of exercise yielded different levels of significance for autophagy-related protein expression.Conclusion: Existing evidence reveals that high-intensity exercise may induce excessive autophagy, while low-intensity exercise for a short period (Intervention duration

245. LINC00518 Interference Inhibits Non–Small Cell Lung Cancer by Upregulating miR216b-5p Expression

Abstract

Introduction Non–small cell lung cancer (NSCLC) accounts for the majority of lung cancer cases, and effective treatment for this disease is still lacking. This study aimed to explore the potential role of LINC00518 and miR216b-5p on cell proliferation and tumor growth in NSCLC. Methods The expression of LINC00518, miR216b-5p, MMP7, and MMP9 in NSCLC cell lines was determined by RT-qPCR analysis, which was also used to confirm the transfection effects. After transfection, proliferation, clone-formation ability, migration, and invasion of NSCLC cells were detected by CCK8, clone-formation, wound-healing, and transwell assays, respectively. Western blot analysis was used to detect the expression of MMP7, MMP9, Ki67, and PCNA. A xenograft model was constructed by subcutaneous injection of transfected NSCLC cells into nude mice. Results The results indicated that LINC00518 expression was increased and miR216b-5p expression decreased in NSCLC cell lines, and A549 cells were chosen for the next experiments. LINC00518 interference inhibited proliferation, invasion, and migration of A549 cells, together with the progression of NSCLC in vivo. In addition, LINC00518 directly targeted miR216b-5p. Downregulation of miR216b-5p weakened the inhibitory effect of LINC00518 interference on proliferation, invasion, and migration of A549 cells, as well as progression of NSCLC in vivo. Discussion In conclusion, LINC00518 interference inhibits NSCLC, which is partially reversed by downregulation of miR216b-5p expression.
