209. Electroluminescence studies on longwavelength indium arsenide quantum dot microcavities grown on gallium arsenide

Abstract

A comprehensive study of the electroluminescence of four GaAs/AlGaAs microcavity devices with InAs/GaInAs quantum dot active regions emitting near 1.3 micrometer was conducted. The four molecular beam epitaxial grown samples with AlAs oxide aperture confinement layers were fabricated, characterized, and optically modeled. Optical power transmission of the samples was modeled using Matlab and compared with measured transmission data. Resonant cavity light emitting diodes (RCLEDs) and three vertical cavity surface emitting laser (VCSEL) samples were fabricated and electro-optically characterized over a range of injection currents and temperatures. Devices achieved continuous wave room temperature lasing at 1.28 micrometer with an output power of more than 3 mW, a threshold current of 2.3 mA, and a slope efficiency of 10.3 W/A. The characteristic temperature was 49.4 K and the wall plug efficiency at was a maximum of over 36%. The minimum threshold current, 1.25 mA, was at a temperature of -10 deg C. The cavity resonance wavelength was tuned too short for the peak wavelength of the active region gain curve which limited the temperature at which the VCSELs produced lasing to about room temperature.

Abstract

This practice paper explores the co-production of knowledge in a collaborative PhD studentship funded by the Economic and Social Research Council (ESRC) and Age UK, which examined the formal volunteering undertaken by older adults in England. Some third sector organisations, especially larger ones such as Age UK, are both producers and users of social science knowledge. In this paper we critically reflect on the co-production of knowledge, and the ways in which both student and supervisors experience the co-creation of knowledge.

222. Variation in commercial smoking mixtures containing third-generation synthetic cannabinoids.

Abstract

Variation in ingredients (qualitative variation) and in quantity of active compounds (quantitative variation) in herbal smoking mixtures containing synthetic cannabinoids has been shown for older products. This can be dangerous to the user, as accurate and reproducible dosing is impossible. In this study, 69 packages containing third-generation cannabinoids of seven brands on the UK market in 2014 were analyzed both qualitatively and quantitatively for variation. When comparing the labels to actual active ingredients identified in the sample, only one brand was shown to be correctly labelled. The other six brands contained less, more, or ingredients other than those listed on the label. Only two brands were inconsistent, containing different active ingredients in different samples. Quantitative variation was assessed both within one package and between several packages. Within-package variation was within a 10% range for five of the seven brands, but two brands showed larger variation, up to 25% (Relative Standard Deviation). Variation between packages was significantly higher, with variation up to 38% and maximum concentration up to 2.7 times higher than the minimum concentration. Both qualitative and quantitative variation are common in smoking mixtures and endanger the user, as it is impossible to estimate the dose or to know the compound consumed when smoking commercial mixtures. 231. CandidatusLiberibacter asiaticus Minimally Alters Expression of Immunity and Metabolism Proteins in Hemolymph of Diaphorina citri, the Insect Vector of Huanglongbing

Abstract

Huanglongbing (HLB), also known as citrus greening disease, is the most serious disease of citrus plants. It is associated with the Gram-negative bacterium ‘CandidatusLiberibacter asiaticus’ (CLas), which is transmitted between host plants by the hemipteran insect vector Diaphorina citriin a circulative, propagative manner involving specific interactions with various insect tissues including the hemolymph, fluid that occupies the body cavity akin to insect blood. High resolution quantitative mass spectrometry was performed to investigate the effect of CLas exposure on D. citrihemolymph at the proteome level. In contrast to the broad proteome effects on hundreds of proteins and a diverse array of metabolic pathways previously reported in gut and whole insect proteome analyses, the effect of CLas on the hemolymph was observed to be highly specific, restricted to key immunity and metabolism pathways, and lower in magnitude than that previously observed in the whole insect body and gut. Vitellogenins were abundantly expressed and CLas-responsive. Gene-specific RNA expression analysis suggests that these proteins are expressed in both male and female insects and may have roles outside of reproductive vitellogenesis. Proteins for fatty acid synthesis were found to be up-regulated, along with metabolic proteins associated with energy production, supported at the organismal level by the previously published observation that D. citriindividuals experience a higher level of hunger when reared on CLas-infected plants. Prediction of post-translational modifications identified hemolymph proteins with phosphorylation and acetylation upon CLas exposure. Proteins derived from the three most prominent bacterial endosymbionts of the psyllid were also detected in the hemolymph, and several of these have predicted secretion signals. A DNAK protein, the bacterial HSP70, detected in the hemolymph expressed from Wolbachia pipientiswas predicted to encode a eukaryotic nuclear localization signal. Taken together, these data show specific changes to immunity and metabolism in D. citrihemolymph involving host and endosymbiont proteins. These data provide a novel context for proteomic changes seen in other D. citritissues in response to CLas and align with organismal data on the effects of CLas on D. citrimetabolism and reproduction.

Abstract

Aphids are important agricultural pests and also biological models for studies of insect-plant interactions, symbiosis, virus vectoring, and the developmental causes of extreme phenotypic plasticity. Here we present the 464 Mb draft genome assembly of the pea aphid Acyrthosiphon pisum. This first published whole genome sequence of a basal hemimetabolous insect provides an outgroup to the multiple published genomes of holometabolous insects. Pea aphids are host-plant specialists, they can reproduce both sexually and asexually, and they have coevolved with an obligate bacterial symbiont. Here we highlight findings from whole genome analysis that may be related to these unusual biological features. These findings include discovery of extensive gene duplication in more than 2000 gene families as well as loss of evolutionarily conserved genes. Gene family expansions relative to other published genomes include genes involved in chromatin modification, miRNA synthesis, and sugar transport. Gene losses include genes central to the IMD immune pathway, selenoprotein utilization, purine salvage, and the entire urea cycle. The pea aphid genome reveals that only a limited number of genes have been acquired from bacteria; thus the reduced gene count of Buchnera does not reflect gene transfer to the host genome. The inventory of metabolic genes in the pea aphid genome suggests that there is extensive metabolite exchange between the aphid and Buchnera, including sharing of amino acid biosynthesis between the aphid and Buchnera. The pea aphid genome provides a foundation for post-genomic studies of fundamental biological questions and applied agricultural problems.

Abstract

In May, 2006, on a Bank Holiday weekend, an 18-year-old woman presented to an inner-city London emergency department. She had been at a nightclub with friends and purchased tablets, which she understood to be Ecstasy or amfetamines, from a dealer. After ingesting fi ve tablets, she collapsed in the nightclub and appeared to have a seizure lasting 10 min. On arrival in the emergency department, she was agitated and had dilated pupils (8 mm), sinus tachycardia (156 bpm), and a blood pressure of 150/51 mm Hg. Her score on the Glasgow coma scale was 15 and she was apyrexial (35·9°C). She had no signifi cant past medical history and was on no regular medication.

Abstract

In this research, pasteurization of beer by nonthermal high pressure processing (HPP) and thermosonication (TS) were compared with thermal pasteurization. The inactivation of Saccharomyces cerevisiae ascospores in beer was studied and modelled for HPP at 200, 300 and 400 MPa, and for TS at 50, 55 and 60 °C with an acoustic energy density of 16.15 W/mL. The energy requirements for equivalent ascospore inactivation by HPP, TS, and thermal processes were compared. For the same processing time, ascospore inactivation was greatest with nonthermal HPP, followed by 60 °C TS, then 60 °C thermal processing. Nonlinear survival curves which could be described by the Weibull model were observed for both HPP and TS. To achieve a 2.5 log reduction in ascospores, HPP required 77 kJ/L compared with 189 kJ/L for thermal processing and 2612 kJ/L for TS. HPP and thermosonication may be alternatives to thermal beer pasteurization, achieving greater log reductions in S. cerevisiae ascospores with shorter processing times (TS and HPP) or less energy (HPP). It is argues that Confucian role ethics seems ill suited for this task. The author argues for a conception of Confucian autonomy that both satisfies role ethicists and supports feminist goals and practices.

Abstract

A suspicious white powder labeled 'idanyl-biphenyl-amninone,' which was seized by customs officials at the "channel island" of Jersey, UK, was brought to our laboratory for identification and characterization of its structure. The elucidation process required the use of several complementary analytical techniques, including gas chromatography-mass spectrometry, liquid chromatography coupled with high-resolution mass spectrometry, nuclear magnetic resonance spectroscopy, and X-ray crystallography. The unknown compound was ultimately identified as 1-(2,3-dihydro-1 H-inden-5-yl)-2-phenyl-2-(pyrrolidin-1-yl)-ethanone, a novel cathinone derivative. To the best of our knowledge, this compound has not been registered in the CAS or IUPAC databases. However, it has recently been marketed on the Internet as 'indapyrophenidone,' and we therefore propose this as the common name of the compound. The results of this study may serve forensic and clinical laboratories in the identification of its related compounds with similar backbone structure using the information reported in this article obtained by the application of advanced analytical techniques.
