To evaluate the effects of quartz precipitation on the abundance and preservation of organic matter pores in marine shale reservoirs, the type of authigenic quartz and the source of silica, as well as the corresponding relation of the Lower Cambrian Shuijingtuo Formation shale in South China were investigated. Quartz in the Shuijingtuo shale occurs as four different types: detrital quartz, replacement of biosiliceous debris, euhedral quartz filled in interparticle pores, and microquartz dispersed in a clay matrix. Euhedral quartz (1–5 μm) and matrix-dispersed microquartz (100–400 nm) are found to be the dominant forms of authigenic quartz. The euhedral quartz accumulates along the interparticle pores, and the porous organic matter fills the interior of the space. Microquartz is mainly wrapped in porous organic matter. Two silica sources were revealed: biogenic silica and clay-derived silica. Biogenic Si is most likely the major source for authigenic quartz in the organic-rich (total organic carbon (TOC) > 2.55 wt.%) samples, which accounts for 23–57 wt.% (average 35 wt.%) of the total Si. Based on petrographic observations, we posit that the precipitation of large-sized euhedral quartz in the interparticle pores most likely originated from biogenic silica in the early stage of diagenesis and that the silica for the clay matrix-dispersed microquartz is provided by biogenic silica and clay-derived silica. The observation of SEM images indicates that the precipitation of early diagenetic euhedral quartz in the interparticle pores enhances rock stiffness, and the buttressing effect can protect the organic matter pores from compaction during the late-stage burial diagenesis. In contrast, the precipitation of late diagenetic microquartz in the clay matrix can lead to a reduction in the capacity of the accommodation space to host retained petroleum, consequently leading to a reduction in the development of organic matter pores and the generation of shale gas.

In this study, we analyzed the spatial and temporal characteristics of extreme precipitation in summer in southern Xinjiang (SXJ) using CN05.1 daily precipitation data, the National Centers for Environmental Prediction (NCEP) monthly average data, and ERA5 four-times-daily reanalysis data from 1961 to 2020, determined the interdecadal variation in extreme precipitation, and further explored the circulation mechanisms and anomalous water vapor transport characteristics associated with changes in extreme precipitation. The results showed that extreme precipitation, extreme precipitation days, and extreme precipitation contribution in SXJ all increased significantly, with the turnaround occurring in 1986. Enhanced vertical upward motion, greater convergence of water vapor at lower levels, and a more unstable atmospheric convection led to an increase in extreme precipitation along the southern border region. The eastward Pacific Ocean and southward Indian Ocean provided anomalous water vapor transport, and the boundary water vapor flux trend and its correlated distribution with extreme precipitation showed that southern boundary water vapor transport played an important role in the increase in extreme precipitation. At vertical heights, the dominant longitudinal water vapor transport fluxes were concentrated in the middle and upper layers, whereas the latitudinal water vapor transport fluxes were concentrated in the middle and lower layers.

The spatial and temporal resolution of remote sensing products in land surface temperature (LST) studies can be improved using the downscaling method. This is a crucial area of research as it provides basic data for the study of climate change. However, there have been few studies evaluating the applicability of downscaling methods using underlying surfaces of varying complexities. In this study, we focused on the semi–homogeneous underlying surface of Gurbantunggut Desert and evaluated the applicability of five classical, passive microwave, downscaling methods based on the machine learning of Catboost, using 365 days of AMSR–2 and MODIS data in 2019, which can be scanned once during the day and night. Our results showed four main points: (1) The correlation coefficients between feature vectors and the LST of the semi–homogeneous underlying surface were clearly different from those of the surrounding oases. The correlation coefficient of the semi–homogeneous underlying surface was high, and that of the surrounding oases was low. (2) At the same frequency, the correlation coefficient between vertically polarized BT and LST was greater than that between horizontally polarized BT and LST. Considering the semi–heterogeneous underlying surface, 23.8 GHz and 36.5 GHz may be more suitable for passive microwave LST retrieval than 89 GHz according to physical mechanisms. (3) The fine–scale LST downscaling accuracy achieved with all BT channels of AMSR–2 was higher than that achieved with the other four classical models. The day and night RMSE values verified with MYD11A1 data were 2.82 K and 1.38 K, respectively. (4) The correlation coefficients between downscaled LST and the soil temperature of the top layer of the site were the highest, with daytime–nighttime R2 values of 0.978 and 0.970, and RMSE values of 3.42 and 4.99 K, respectively. The all–channel–based LST downscaling method is very effective and can provide a theoretical foundation for the acquisition of all–weather, multi–layer soil temperature.

Rabies is a highly fatal disease, and it is vital to find effective ways to manage and control infection. There is a need for new effective antiviral drugs that are particularly effective treatments for rabies. Deoxynivalenol (DON) is known mainly for its toxicity, but at the molecular level, it can inhibit RNA and DNA replication, and there is increasing evidence that different doses of DON have a positive effect on inhibiting virus replication. Based on this, we evaluated the effect of DON on inhibiting the rabies virus in vitro. The inhibitory effect of DON on rabies virus activity was dose- and time-dependent, and 0.25 μg/mL of DON could inhibit 99% of rabies virus activity within 24 h. Furthermore, DON could inhibit the adsorption, entry, replication, and release of rabies virus but could not inactivate the virus. The inhibitory effect of DON on rabies virus may be achieved by promoting apoptosis. Our study provides a new perspective for the study of anti-rabies virus and expands the direction of action of mycotoxins.

Objective To assess and compare the performance of fluorine‐18‐labeled fluorodeoxyglucose positron emission tomography (18F‐FDG‐PET/ CT) and gallium‐68‐labeled tetraazacyclododecanetetraacetic acid‐DPhe1‐Tyr3‐octreotate (68Ga‐ DOTATATE) PET/CT in the targeted imaging of culprit tumors causing osteomalacia. Methods This was a clinical retrospective analysis. We analyzed 13 patients (five men, eight women; mean age, 49 years; range, 19–55 years) with suspicion of tumor‐induced osteomalacia (TIO) between March 2017 and October 2019. All patients underwent two functional imaging methods to locate the culprittumors. Studies were performed on a PET/CT scanner. The injection doses of 18F‐ FDG and 68Ga‐DOTATATE were 0.5mCi/kg and approximately 5.0mCi, respectively. In the two scans, the whole body was captured from head to toe 45 to 60 min after intravenous tracer injection. 68Ga‐DOTATATE PET/CT and 18F‐FDG PET/CT imaging results locate culprit tumors according to the following criteria: (i) abnormal foci uptake concentration was observed locally, and the uptake level was higher than the background level of the right lobe of the liver; (ii) combined CT showed or did not have obvious abnormal density changes; and (iii) non‐specific ingestion lesions due to fracture, arthritis, necrosis of femoral head are excluded. Compared with the results of pathological examination and clinical follow‐up, the sensitivity, specificity and accuracy of 68Ga‐DOTATATE PET/CT imaging and 18F‐FDG PET/CT imaging for TIO were analyzed. Results All patients had symptoms of osteomalacia and hypophosphatemia. The lag time (symptoms to PET diagnosis) ranged from 2 to 12 years. There were eight cases of TIO patients and five cases of non‐TIO patients confirmed by surgery, pathology and follow‐up. Among the eight TIO patients, there were six cases (75.0%) of PMTs, one case (12.5%) of giant cell tumor, one case (12.5%) of hemangiopericutoma. Most (n = 6, 75.0%) of the confirmed tumors in our patient population were in the lower extremities, followed by craniofacial regions (n = 1, 12.5%), and torso (n = 1, 12.5%), respectively. Among the five non‐TIO patients, there were two cases of Fanconi syndrome, one case of rickets, and two cases of sporadic osteomalacia hypophosphorus. The culprit tumors could be located either in the bone (n = 5, 62.5%) or the soft tissue (n = 3, 37.5%). 18F‐FDG PET/CT was able to localize the tumor in six (6/13, 46.1%) patients. 68Ga‐DOTATATE PET/CT detected tumor in 8 (83.3%) of 13 patients. The sensitivity of 68Ga‐DOTATATE PET/CT imaging and 18F‐FDG PET/CT imaging in the evaluation of TIO in our patient population were 100% (8/8) vs 75% (6/8). The specificity of the two different methods was 80% (4/5). The overall accuracy was 92.3% (12/13) vs 76.9% (10/13). Conclusions 68Ga‐DOTATATE PET/CT is very effective in assessing hypophosphatemia patients with TIO typical symptoms compared with 18F‐FDG. Therefore, in clinically suspected cases of hypophosphatemic osteomalacia, 68Ga‐DOTATATE PET/CT should be preferred as an imaging modality investigation to avoid delay in the treatment of this disease.

AimsSex hormones play an important role in the pathogenesis of cardiovascular disease (CVD). This cross-sectional study aimed to explore the associations of dehydroepiandrosterone (DHEA) and dehydroepiandrosterone sulfate (DHEAS) with coronary heart disease (CHD) and stroke in middle-aged and elderly patients with type 2 diabetes mellitus (T2DM).Materials and MethodsA total of 995 patients with T2DM were included in the study analysis. Serum levels of DHEA and DHEAS were quantified using liquid chromatography–tandem mass spectrometry. Binary logistic regression analyses were performed to assess the associations of DHEA and DHEAS with CHD and stroke. Receiver operating characteristic (ROC) curve analysis was performed to determine the optimal DHEA and DHEAS cutoff values for the detection of CHD in men with T2DM.ResultsIn men with T2DM, after adjustment for potential confounders in model 3, the risk of CHD decreased with an increasing serum DHEA level [odds ratio (OR) = 0.38, quartile 4 vs. quartile 1; 95% confidence interval (CI) = 0.16–0.90; p = 0.037 for trend). Consistently, when considered as a continuous variable, this association remained significant in the fully adjusted model (OR = 0.59, 95% CI = 0.40–0.87, p 0.05).ConclusionsSerum DHEA and DHEAS were significantly and negatively associated with CHD in middle-aged and elderly men with T2DM. This study suggests potential roles of DHEA and DHEAS in CHD pathogenesis.

Abstract Integrated network analysis pipeline (iNAP) is an online analysis pipeline for generating and analyzing comprehensive ecological networks in microbiome studies. It is implemented in two sections, that is, network construction and network analysis, and integrates many open‐access tools. Network construction contains multiple feasible alternatives, including correlation‐based approaches (Pearson's correlation and Spearman's rank correlation along with random matrix theory, and sparse correlations for compositional data) and conditional dependence‐based methods (extended local similarity analysis and sparse inverse covariance estimation for ecological association inference), while network analysis provides topological structures at different levels and the potential effects of environmental factors on network structures. Considering the full workflow, from microbiome data set to network result, iNAP contains the molecular ecological network analysis pipeline and interdomain ecological network analysis pipeline (IDENAP), which correspond to the intradomain and interdomain associations of microbial species at multiple taxonomic levels. Here, we describe the detailed workflow by taking IDENAP as an example and show the comprehensive steps to assist researchers to conduct the relevant analyses using their own data sets. Afterwards, some auxiliary tools facilitating the pipeline are introduced to effectively aid in the switch from local analysis to online operations. Therefore, iNAP, as an easy‐to‐use platform that provides multiple network‐associated tools and approaches, can enable researchers to better understand the organization of microbial communities. iNAP is available at http://mem.rcees.ac.cn:8081 with free registration.

The potent cytotoxic property of Vγ2Vδ2 T cells makes them attractive for adoptive T cell transfer therapy. The transfusing of the expanded Vγ2Vδ2 T cells into cancer patients shows well-tolerated, but the clinical response rates are required to be improved, implying that there is still an unmet efficacy with low toxicity for this novel anti-tumor therapy. In this study, we test the anti-tumor efficacy of a Y-body-based bispecific antibody (bsAb) Vγ2 x PD-L1 that preferentially redirects Vγ2Vδ2 T cells to combat PD-L1 positive tumor cells. With nanomolar affinity levels to Vγ2Vδ2 T cells and PD-L1+ tumor cells, Vγ2 x PD-L1 bridges a Vγ2Vδ2 T cell with a SKOV3 tumor cell to form a cell-to-cell conjugation. In a PD-L1-dependent manner, the bsAb elicits effective activation (CD25+CD69+), IFNγ releasing, degranulation (CD107a+), and cytokine production (IFNγ+ and TNFα+) of expanded Vγ2Vδ2 T cells. The activations of the Vγ2Vδ2 T cells eliminate PD-L1-expressing human cancer cell lines, including H1975, SKOV3, A375, H1299, and H2228 cells, but not PD-L1 negative cells including HEK-293 (293) cells and healthy PBMCs. Finally, we show that combining Vγ2 x PD-L1 with adoptively transferring Vγ2Vδ2 T cells inhibits the growth of existing tumor xenografts and increases the number of Vγ2Vδ2 T cells into the tumor bed. Vγ2 x PD-L1 represents a promising reagent for increasing the efficacy of adoptively transferred Vγ2Vδ2 T cells in the treatment of PD-L1 positive malignant tumors.

BackgroundThe associations of dehydroepiandrosterone (DHEA) and dehydroepiandrosterone sulfate (DHEAS) with diabetic kidney disease (DKD) remained unclear. Thus, this cross-sectional study aimed to explore the associations of DHEA and DHEAS with the risk of DKD in patients with T2DM.MethodsThe information of 1251 patients with T2DM were included in this study. Serum DHEA and DHEAS were quantified using liquid chromatography-tandem mass spectrometry assays. Multivariate logistic regression analyses were used to assess the associations of DHEA and DHEAS with DKD as well as high urine albumin to creatinine ratio (ACR).ResultsIn men with T2DM, the risk of DKD decreased with an increasing DHEA concentration after adjustment for traditional risk factors; the fully adjusted OR (95% CI) for tertile3 vs tertile1 was 0.37 (0.19-0.70; P = 0.010 for trend). Similarly, when taking high ACR as the outcome, low DHEA levels were still significantly associated with increased odds of high ACR (OR, 0.37; 95% CI, 0.19–0.72 for tertile3 vs tertile1; P = 0.012 for trend). The restricted cubic spline showed that the risk of DKD gradually decreased with the increment of serum DHEA levels (P-overall = 0.007; P-nonlinear = 0.161). DHEAS was not independently associated with the risk of DKD in men. In contrast, no significant relationships were found between DHEA and DHEAS and the risk of DKD in women (all P > 0.05).ConclusionsIn men with T2DM, low serum DHEA levels were independently related to the risk of DKD after adjustment for traditional risk factors. Our finding highlights the potential role of DHEA in the development of DKD in men with T2DM.

Dust storms and dust aerosols seriously affect environmental variation and climate change at regional and global scales. Accordingly, these hazards are the current focus in studies related to Earth science. The near-surface layer is an important link for the upward transmission of dust aerosols. However, the difficulty associated with obtaining real-time observation data from this layer has markedly hindered the progress of related research. In sand source areas, the topographic relief of natural dunes is easily ignored, despite serving as an essential factor affecting wind-driven dust emission, transport, and deposition. In this study, we explored the similarities and differences in horizontal dust flux (Q) between Xiaotang and Tazhong using observation data. In Xiaotang, the variation in the Q value with height was found to fit a power function; however, in Tazhong, the Q value did not show a significant gradient change. Such phenomena are caused by the secondary sand source generated by the undulation of natural dunes. The median particle diameter of the dust lifted from the ground during dust storms was essentially the same between Xiaotang and Tazhong, ranging from 74 to 82 μm in Tazhong and from 53 to 81 μm in Xiaotang. The maximum wind speed in Xiaotang was greater than that in Tazhong, resulting in a larger Q value for each particle size range in Xiaotang. The coarse sediment grain was identified as the main factor controlling the vertical variation trend of Q. Further, fine particles were found to have a minor impact.

BackgroundSmall molecule RNAs (miRNAs) could induce downregulation of α-synuclein (SNCA) expression by binding the 3’ untranslated region of SNCA, thus playing an important role in the pathogenesis of Parkinson’s disease (PD). Recent studies suggest that SNCA-related miRNAs in saliva are promising PD biomarkers. Research on those miRNAs in plasma is rare in patients with PD.ObjectiveTo detect the plasma expression levels of three SNCA related miRNAs (miR-7, miR-153, and miR-223) in PD, and to explore their diagnostic value and associations with clinical phenotype.MethodsMiR-7, miR-153, and miR-223 levels were detected in the plasma of 75 PD patients and 73 normal controls (NCs) via real-time quantitative polymerase chain reaction. The receiver operating characteristic (ROC) curves were delineated to evaluate their diagnostic value in PD. In addition, their associations with demographic, key motor, and non-motor symptoms were explored by serial scales.ResultsThe expression levels of plasma miR-153 and miR-223 were significantly decreased in patients with PD relative to NCs. The area under the ROC curve separating PD from NCs was 63.1% for miR-153 and 86.2% for miR-223, respectively. The plasma miR-153 level in de novo PD was lower than that in treated patients (p = 0.006), its level increased gradually with disease duration (r = 0.358, p = 0.002) and Unified Parkinson’s Disease Rating Scale Part III score (r = 0.264, p = 0.022). Plasma miR-223 level was decreased in patients with clinical possible rapid eye movement sleep behavior disorder (cpRBD) compared with those without cpRBD (p < 0.001), and its level was negatively associated with RBDSQ score (r = -0.334, p = 0.003). Multiple linear regression analysis revealed that disease duration (p = 0.049) was the independently associated factor of miR-153 level; whereas, RBDSQ (p = 0.009) was related to miR-223 level in PD.ConclusionPlasma miR-153 and miR-223 levels could be potential biomarkers of PD.

Study region: Ishim-Tobol River Basin, Northern Kazakhstan Study focus: In this study, an ensemble reconstruction of the June–July streamflow from the Ishim-Tobor River was carried out using random forest (RF), K-nearest neighbor (KNN) and multiple linear regression (MLR) models. The reliability of the ensemble reconstruction was verified by a comparison with other regional reconstructions and historical records. A correlation analysis and vapor fluxes were applied to visualize the significant influence of atmospheric circulation on the study area. The cumulative distribution functions (CDFs) examined the distribution of the high (low) flows highlighted by the reconstruction. New hydrological insights for the region: Our study analyzes the application of machine learning algorithms and a traditional MLR model to hydrological reconstruction. The single model reconstruction contained information and results on streamflow variability were not sufficient. Consequently, we integrated the three models into the ensemble reconstruction. The extended streamflow record reveals the basin's hydrological changes over the past 229 years. From 1788–2016, the reconstructed streamflow was perennially below the mean value, which indicates more prominent drought and water deficit conditions within the basin. This phenomenon was significantly influenced by water vapor transport from the North Atlantic and Arctic Oceans. If future climate scenarios lead to drought in the basin, then surface water demand will not be satisfied for 7 out of 10 years.

Dust aerosol vertical profiles are very essential to accurately evaluate their climate forcing and trans-subcontinental transportation to downstream areas. We initiated a joint comprehensive field experiment to investigate the vertical profiles and optical characteristics of dust aerosol in the hinterland of Taklimakan Desert (TD) during summer 2019. After smoothing the raw signals, the CHM15k ceilometer could distinctly detect a moderate intensity of dust layer, cloud layer, and subsequent rainfall process. The results showed that dust events frequently occurred in TD during the entire period; the overall mean PM2.5, PM10, PM2.5/PM10, and Ångström exponent are 110.4 ± 121.0 μg/m3, 317.2 ± 340.0 μg/m3, 0.35 ± 0.07, and 0.28 ± 0.12, respectively, suggesting that dust particles are predominant aerosol types in TD. There was an obvious summertime dust stagnation layer persistently hanged over the desert at 1.5–3.0-km height. A deep and intense daytime convective structure was also detected by the ceilometer, with maximum aerosol mixing layer height of ∼3 km that appeared at 12:00 UTC, which was in favor of lifting the ground-generated dust particles into the upper atmosphere. The normalized range-corrected signal log10(RCS), aerosol extinction σ(z), and backscattering coefficient β(z) of the ceilometer were higher than 6.2, 0.5 km−1, and 0.01 km−1 sr−1 for heavy dust storms, respectively, and the corresponding vertical optical range was smaller than 1.0 km. The aerosol lidar ratio was equal to 50 sr, which was greatly different from those of clear-sky cases. The retrieved σ(z) values were about 2.0, 0.5, 0.14, and 0.10 km−1 at 200-m height, respectively, under strong dust storm, blowing dust, floating dust, and clear-sky conditions. This indicated that the aerosol extinction coefficients under dust events were about 3–10 times greater than those of clear-sky cases. The statistics of the aerosol optical parameters under different dust intensities in TD were very helpful to explore and validate dust aerosols in the application of climate models or satellite remote sensing.

Purpose: To investigate the characteristics of macular structures and microcirculation of posterior staphyloma (PS) and explored factors related to PS in eyes with high myopia.Methods: There were 114 eyes of 82 patients in this study. Using 1:1 matching of the axial length of myopic eyes, patients were divided into no PS (NPS) and PS groups. Comprehensive ophthalmic examinations were performed on all patients. Structural parameters were acquired using swept source optical coherence tomography (SS-OCT). OCT angiography (OCTA) was used to measure the microcirculation parameters. Generalized estimated equation and linear correlation analysis were used for the statistical analysis.Results: Patients with PS had a thinner retinal thickness (RT) and choroid thickness (CT) (all p < 0.05) in the measurement areas and a significantly lower subfoveal scleral thickness (SFST) (p < 0.001) than those without PS. Retinal deep vascular complex density (DVD) (all p < 0.05) and choriocapillaris perfusion area (CCPA) (all p < 0.001) were significantly lower in the measurement areas of the PS group than in those of the NPS group. There was no significant difference in the retinal superficial vascular density between the two groups. Generalized estimating equation indicated that SFST (B = 0.079, p = 0.001), parafoveal RT (B = −0.162, p = 0.041), foveal CT (B = 0.292, p = 0.013), parafoveal CT (B = −0.157, p = 0.023), foveal CCPA (B = 0.691, p = 0.003) and parafoveal CCPA (B = −0.026, p = 0.004) were significantly correlated with PS. Age (r = −0.323, p = 0.001), spherical equivalent refraction (SER) (r = 0.289, p = 0.004), subfoveal CT (r = 0.398, p < 0.001), foveal DVD (r = 0.346, p < 0.001), foveal CT (r = 0.429, p < 0.001), and foveal CCPA (r = 0.387, p < 0.001) were strongly correlated with SFST.Conclusions: The macular structures and microcirculation in the PS group were different from those in the NPS group. SFST, CT, and CCPA were significantly correlated with PS. Lower SFST in PS was correlated with abnormalities of CT and microcirculation.

Abstract Background The NF-κB signalling pathway has been reported to be related to liver fibrosis, and we investigated whether the NF-κB signalling pathway is involved in liver fibrosis caused by secreted phospholipase A2 of Clonorchis sinensis (CssPLA2). Furthermore, expression of the receptor of CssPLA2 on the cell surface of hepatic stellate cells (HSCs) may greatly contribute to liver fibrosis. Methods CssPLA2 was administered to BALB/c mice by abdominal injection. The levels of markers of NF-κB signalling pathway activation in mouse liver tissue were measured by quantitative RT-PCR, ELISA and western blot. Additionally, HSCs were incubated with CssPLA2, and an NF-κB signalling inhibitor (BAY 11-7082) was applied to test whether the NF-κB signalling pathway plays a role in the effect of CssPLA2. Then, the interaction between CssPLA2 and its receptor transmembrane 7 superfamily member 3 (TM7SF3) was confirmed by co-immunoprecipitation (co-IP) and GST pull-down. To determine how TM7SF3 influences the ability of CssPLA2 to cause liver fibrosis, a TM7SF3 antibody was used to block TM7SF3. Results The levels of the NF-ΚB signalling pathway activation markers TNF-α, IL-1β and phospho-p65 were increased by CssPLA2 in the context of liver fibrosis. In addition, the interaction between TM7SF3 and CssPLA2 was confirmed by co-IP and GST pull-down. When TM7SF3 was blocked by an antibody targeting 1–295 amino acids of TM7SF3, activation of HSCs caused by CssPLA2 was alleviated. Conclusions The NF-ΚB signalling pathway is involved in the activation of HSCs by CssPLA2. TM7SF3, the receptor of CssPLA2, plays important roles in liver fibrosis caused by CssPLA2.

Abstract Background Diabetic osteoporosis (DOP) is a systemic metabolic bone disease caused by diabetes mellitus (DM). Adipose-derived stem cells (ASCs) play an important role in bone regeneration. Our previous study confirmed that ASCs from DOP mice (DOP-ASCs) have a lower osteogenesis potential compared with control ASCs (CON-ASCs). However, the cause of this poor osteogenesis has not been elucidated. Therefore, this study investigated the underlying mechanism of the decline in the osteogenic potential of DOP-ASCs from the perspective of epigenetics and explored methods to enhance their osteogenic capacity. Methods The expression level of JNK1-associated membrane protein (JKAMP) and degree of DNA methylation in CON-ASCs and DOP-ASCs were measured by mRNA expression profiling and MeDIP sequencing, respectively. JKAMP small interfering RNA (siRNA) and a Jkamp overexpression plasmid were used to assess the role of JKAMP in osteogenic differentiation of CON-ASCs and DOP-ASCs. Immunofluorescence, qPCR, and western blotting were used to measure changes in expression of Wnt signaling pathway-related genes and osteogenesis-related molecules after osteogenesis induction. Alizarin red and ALP staining was used to confirm the osteogenic potential of stem cells. Bisulfite-specific PCR (BSP) was used to detect JKAMP methylation degree. Results Expression of JKAMP and osteogenesis-related molecules (RUNX2 and OPN) in DOP-ASCs was decreased significantly in comparison with CON-ASCs. JKAMP silencing inhibited the Wnt signaling pathway and reduced the osteogenic ability of CON-ASCs. Overexpression of JKAMP in DOP-ASCs rescued the impaired osteogenic capacity caused by DOP. Moreover, JKAMP in DOP-ASCs contained intragenic DNA hypermethylated regions related to the downregulation of JKAMP expression. Conclusions Intragenic DNA methylation inhibits the osteogenic ability of DOP-ASCs by suppressing expression of JKAMP and the Wnt signaling pathway. This study shows an epigenetic explanation for the reduced osteogenic ability of DOP-ASCs and provides a potential therapeutic target to prevent and treat osteoporosis.

Aiming at the time-consuming problem of the full-wave (FW) simulation of the scattering characteristics of the traditional graphene reconfigurable reflectarray antenna, a fast prediction method of electromagnetic (EM) response based on deep learning is proposed. The convolutional neural network (CNN) method in deep learning is effectively used in the research of this paper. This method first discretizes the input vector (patch geometry, chemical potential, frequency, incident angle, etc.) of the graphene reflectarray antenna, and then preprocesses the data into a two-dimensional image suitable for CNN training, and finally uses CNN to train the model instead of extensive FW simulation calculations, the EM response of the reflectarray antenna is calculated. The training results of three algorithms of support vector regression (SVR), radial basis function network (RBFN) and CNN are comprehensively compared. The experimental results show that CNN method has good performance and accuracy in the EM response prediction of the graphene reconfigurable reflectarray antenna, with an accuracy of over 99%, and can also save at least 99% of time.

Terrestrial vegetation response to surface water availability is important for land–atmosphere interactions. However, the current understanding of how the vegetation responds to surface water remains limited since the physical processes happening within the biosphere and hydrosphere are highly coupled. It is even more difficult to measure such interactions for the processes related to surface soil moisture (SSM)—the central variable that interacts the most intimately with vegetation—since the observations of SSM are often scarce and uneven. Here, we use the satellite observations of vegetation optical depth (VOD) and SSM to map the response time scales of vegetation to surface water anomalies. We use the stability theory to derive vegetation memory time (${{{\bf \tau }}_{{{\bf ReS}}}}$) to reveal the global pattern of vegetation memory to surface water anomalies. That is, the time vegetation takes to return back to its equilibrium when an anomaly dissipates to a certain level (e.g., the e-folding level). We also estimate the plant reactive time (${{{\bf \tau }}_{{{\bf ReA}}}}$)—the time when impacts of surface anomaly reach its peak to evaluate the overall resilience of terrestrial vegetation to surface water anomalies. The results show that ${{{\bf \tau }}_{{{\bf ReS}}}}$ tends to be longer in herbaceous biomes, whereas ${{{\bf \tau }}_{{{\bf ReA}}}}$ is longer in biomes with tree cover. Such anticorrelation of ${{{\bf \tau }}_{{{\bf ReS}}}}$ and ${{{\bf \tau }}_{{{\bf ReA}}}}$ indicates that the herbaceous biomes may be more vulnerable to surface water perturbations during climate extremes. Our study provides a global quantification on vegetation—soil moisture feedbacks—enabling comparison with earth system models.

Right ventricular–pulmonary artery coupling (RV-PA coupling) refers to the relationship between RV contractility and RV afterload. Normal RV-PA coupling is maintained only when RV function and pulmonary vascular resistance are appropriately matched. RV-PA uncoupling occurs when RV contractility cannot increase to match RV afterload, resulting in RV dysfunction and right heart failure. RV-PA coupling plays an important role in the pathophysiology and progression of cardiovascular diseases. Therefore, early and accurate evaluation of RV-PA coupling is of great significance for a patient’s condition assessment, clinical decision making, risk stratification, and prognosis judgment. RV-PA coupling can be assessed by using invasive or noninvasive approaches. The aim of this review was to summarize the pathological mechanism and evaluation methods of RV-PA coupling, the advantages and disadvantages of each method, and the application value of RV-PA coupling in various cardiovascular diseases.

Dust in sand dunes is an effective and important source of dust emission. The Taklimakan Desert (TD) is one of the main sources of global dust: the sand dunes account for approximately 85% of the total area of the TD. However, the dust concentration, emission characteristics, and physical factors of different parts of the sand dunes in the TD during the day and night, as well as dust and non-dust days, remain unclear. In this study, dust observations were collected over a 3 month period in the TD to investigate the physical processes by which dust moves across a surface and generates PM10 and PM2.5 from the top and bottom of sand dunes. The results showed that the daily average maximum concentrations of particulate matter with diameters below 2.5 and 10 µm (PM2.5 and PM10) in the dune during the observation period reach ~90 and ~190 µg·m−3, respectively. Dust emission generated in the saltation process (maximum emission of PM10 was 3–5 mg·m−2·s−1) in the TD dunes was larger than that in other areas and had a good correlation with u* (friction velocity), where u* = 0.4 m·s−1 was the threshold of sand dune dust emission. The wind speed at the top of dunes was larger than that at the bottom, which was conducive to the accumulation of PM10 at the top of the dune. Furthermore, the MLH (mixed layer height) decreased after sunset and the turbulence weakens, which was conducive to the retention of dust in the air. Moreover, the dust made the solar radiation at the top of the dune −15 W·m−2 (average) lower than at the bottom. These results provided a new understanding of dune emissions in the TD and could be used for dust modeling in regional and global models.

We investigated the expression and biological function of retinoic acid inducible gene I (RIG-I) in esophageal squamous cell carcinoma (ESCC). Materials and methods: An immunohistochemical analysis was performed on 86 pairs of tumor tissue and adjacent normal tissue samples of patients with ESCC. We generated RIG-I-overexpressing ESCC cell lines KYSE70 and KYSE450, and RIG-I- knockdown cell lines KYSE150 and KYSE510. Cell viability, migration and invasion, radioresistance, DNA damage, and cell cycle were evaluated using CCK-8, wound-healing and transwell assay, colony formation, immunofluorescence, and flow cytometry and Western blotting, respectively. RNA sequencing was performed to determine the differential gene expression between controls and RIG-I knockdown. Tumor growth and radioresistance were assessed in nude mice using xenograft models. RIG-I expression was higher in ESCC tissues compared with that in matched non-tumor tissues. RIG-I overexpressing cells had a higher proliferation rate than RIG-I knockdown cells. Moreover, the knockdown of RIG-I slowed migration and invasion rates, whereas the overexpression of RIG-I accelerated migration and invasion rates. RIG-I overexpression induced radioresistance and G2/M phase arrest and reduced DNA damage after exposure to ionizing radiations compared with controls; however, it silenced the RIG-I enhanced radiosensitivity and DNA damage, and reduced the G2/M phase arrest. RNA sequencing revealed that the downstream genes DUSP6 and RIG-I had the same biological function; silencing DUSP6 can reduce the radioresistance caused by the overexpression of RIG-I. RIG-I knockdown depleted tumor growth in vivo, and radiation exposure effectively delayed the growth of xenograft tumors compared with the control group. RIG-I enhances the progression and radioresistance of ESCC; therefore, it may be a new potential target for ESCC-targeted therapy.

With the cyclical development of emerging technologies, in reality, the evolution dynamics of their innovation networks will inevitably show obvious time attributes. Numerous network analyses of real complex systems usually focus on static networks; however, it is difficult to describe that most real networks undergo topological evolutions over time. Temporal networks, which incorporate time attributes into traditional static network models, can more accurately depict the temporal features of network evolution. Here, we introduced the time attribute of the life cycle of emerging technology into the evolution dynamics of its innovation network, constructed an emerging technology temporal innovation network from a temporal network perspective, and established its evolution model in combination with the life cycle and key attributes of emerging technology. Based on this model, we took 5G technology as an example to conduct network evolution simulation, verified the rationality of the above model building, and analyzed the cyclical evolution dynamics of this network in various topological structures. The results show that the life cycle of emerging technology, as well as multiple knowledge attributes based on the key attributes of emerging technology, are important factors that affect network evolution by acting on node behaviors. Within this study, we provide a more realistic framework to describe the internal mechanism of the cyclical evolution of emerging technology innovation network, which can extend the research on innovation network evolution from the single topological dynamics to the topological–temporal dynamics containing time attributes and enrich the research dimensions of innovation network evolution from the perspective of temporal evolution.

Myocardial fibrosis (MF) is a non-reversible process that occurs following acute or chronic myocardial damage. MF worsens myocardial deformation, remodels the heart and raises myocardial stiffness, and is a crucial pathological manifestation in patients with end-stage cardiovascular diseases and closely related to cardiac adverse events. Therefore, early quantitative analysis of MF plays an important role in risk stratification, clinical decision, and improvement in prognosis. With the advent and development of strain imaging modalities in recent years, MF may be detected early in cardiovascular diseases. This review summarizes the clinical usefulness of strain imaging techniques in the non-invasive assessment of MF.

ObjectiveTo assess the association between vitamin D status and all-cause mortality among type 2 diabetes patients.Research Design and MethodsWe prospectively followed 1,291 participants with type 2 diabetes aged 20–80 years during 2013–2018. Cox proportional hazard regression models were used to estimate the association between different vitamin D status and all-cause mortality risk among hospitalized patients with type 2 diabetes.ResultsDuring a median follow-up of 4.15 years (5,365 person-years in total), 61 cases of death were identified. PurposeSubtraction coronary CT angiography (CCTA) may reduce blooming and beam-hardening artifacts. This study aimed to assess its value in improving the diagnostic accuracy of readers with different experience levels.MethodWe prospectively enrolled patients with target segment who underwent CCTA and invasive coronary angiography (ICA). Target segment images were independently evaluated by three groups of radiologists with different experience levels with CCTA using ICA as the standard reference. Diagnostic accuracy was measured by the area under the curve (AUC), using ≥50% stenosis as the cut-off value.ResultsIn total, 134 target segments with severe calcification from 47 patients were analyzed. The mean specificity of conventional CCTA for each group ranged from 22.4 to 42.2%, which significantly improved with subtraction CCTA, ranging from 81.3 to 85.7% (all p < 0.001). The mean sensitivity of conventional CCTA for each group ranged from 83.3 to 88.0%. Following calcification subtraction, the mean sensitivity decreased for the novice (p < 0.001) and junior (p = 0.017) radiologists but was unchanged for the senior radiologists (p = 0.690). With subtraction CCTA, the mean AUCs of CCTA significantly increased: values ranged from 0.53, 0.54, and 0.61 to 0.70, 0.74, and 0.85 for the novice, junior, and senior groups (all p < 0.001).ConclusionSubtraction CCTA could improve the diagnostic accuracy of radiologists at all experience levels of CCTA interpretation.

Superphanes, compounds in which the two benzene rings clamped parallel on top of each other by six bridges, have been attracting considerable interest because of their structural beauty and symmetry. However, progress in the research of superphane chemistry and beyond has been seriously hampered by their poor availability. Herein, we report the facile and scalable synthesis of a collection of superphanes with structural diversity and their unique photophysical properties, as well as their unusual host–guest behavior. A set of secondary amine–linked superphanes are obtained via dynamic self–assembly of a hexakis–amine and various readily derived aromatic dialdehyde in one pot, followed by in–situ reduction with NaBH4. Further post–functionalization was exemplified by secondary amine to tertiary amine conversion. All superphanes under study were found to exhibit interesting fluorescence either in solution or in the solid state. Finally, the unusual host–guest binding properties was exemplified by the stabilization of a labile 2Cl–·H2O cluster with fully protonated 6a both in solution and in the solid state. With the easy and versatile synthesis, modification, as well as unique photophysical and host–guest properties reported herein, we believe that this study offers new possibilities for superphane chemistry, supramolecular host design, advanced functional materials and beyond.

The unfolded protein response (UPR) is one of the most important protein quality control mechanisms in cells. At least, three factors are predicted to activate the UPR in yeast cells during fermentation. Using UPRE-lacZ as a reporter, we constructed two indicator strains, KZ and WZ, based on Angel-derived K-a and W303-1A strains, respectively, and investigated their UPR response to tunicamycin, ethanol, and acetic acid. Then, four strains carrying plasmids BG-cwp2 and BG were obtained to realize the displaying and secretion of β-glucosidase, respectively. The results of cellobiose utilization assays indicated interactions between the UPR and the metabolic burden between the strain source, anchoring moiety, oxygen supply, and cellobiose concentration. Meanwhile, as expected, growth (OD600), β-glucosidase, and β-galactosidase activities were shown to have a positive inter-relationship, in which the values of the KZ-derived strains were far lower than those of the WZ-derived strains. Additionally, extra metabolic burden by displaying over secreting was also much more serious in strain KZ than in strain WZ. The maximum ethanol titer of the four strains (KZ (BG-cwp2), KZ (BG), WZ (BG-cwp2), and WZ (BG)) in oxygen-limited 10% cellobiose fermentation was 3.173, 5.307, 5.495, and 5.486% (v/v), respectively, and the acetic acid titer ranged from 0.038 to 0.060% (v/v). The corresponding maximum values of the ratio of β-galactosidase activity to that of the control were 3.30, 5.29, 6.45, and 8.72, respectively. Under aerobic conditions with 2% cellobiose, those values were 3.79, 4.97, 6.99, and 7.67, respectively. A comparison of the results implied that β-glucosidase expression durably induced the UPR, and the effect of ethanol and acetic acid depended on the titer produced. Further study is necessary to identify ethanol- or acid-specific target gene expression. Taken together, our results indicated that the host strain W303-1A is a better secretory protein producer, and the first step to modify strain K-a for cellulosic ethanol fermentation would be to relieve the bottleneck of UPR capacity. The results of the present study will help to identify candidate host strains and optimize expression and fermentation by quantifying UPR induction.

BackgroundCognitive decline is the most dominant and patient-oriented symptom during the development of Alzheimer’s disease (AD) and mild cognitive impairment (MCI). This study was designed to test the feasibility of hybrid convolutional neural networks and long-short-term memory (CNN-LSTM) modeling driven early decision-tailoring with the predicted long-term cognitive conversion in AD and MCI.MethodsCharacteristics of patients with AD or MCI covering demographic features, clinical features, and time-dependent neuropsychological-related features were fused into the hybrid CNN-LSTM modeling to predict cognitive conversion based on a 4-point change in the AD assessment scale-cognition score. Treatment reassignment rates were estimated based on the actual and predicted cognitive conversion at 3 and 6 months according to the prespecified principle; that is if the ADAS-cog score of the patient declines less than 4 points or increases at either follow-up time point, the medical treatment recommended upon their diagnosis would be considered insufficient. Therefore, it is recommended to upgrade the medical treatment upon diagnosis. Actual and predicted treatment reassignment rates were compared in the general population and subpopulations categorized by age, gender, symptom severity, and the intervention subtypes.ResultsA total of 224 patients were included in the analysis. The hybrid CNN-LSTM model achieved the mean AUC of 0.735 (95% CI: 0.701–0.769) at 3 months and 0.853 (95% CI: 0.814–0.892) at 6 months in predicting cognitive conversion status. The AUC at 6 months was significantly impacted when data collected at 3 months were withdrawn. The predicted cognitive conversion suggested a revision of medical treatment in 46.43% (104/224) of patients at 3 months and 54.02% (121/224) at 6 months as compared with 62.05% (139/224) at 3 months (p = 0.001) and 62.50% (140/224) at 6 months (p = 0.069) according to their actual cognitive conversion. No significant differences were detected between treatment reassignment rates estimated based on actual and predicted cognitive conversion in all directions at 6 months.ConclusionUsing the synergistic advances of deep learning modeling and featured longitudinal information, our hypothesis was preliminarily verified with the comparable predictive performance in cognitive conversion. Results provided the possibility of reassigned recommended treatment for those who may suffer from cognitive decline in the future. Considering the limited diversity of treatment strategies applied in this study, the real-world medical situation should be further simulated.

ObjectiveThis study aims to investigate the relationship between dry eye disease (DED) and anxiety, as well as DED and depression. Additionally, the influence of health anxiety (HA) on this relationship was determined.MethodsA total of 206 patients with DED were recruited from Tianjin Medical University Eye Hospital clinic and surveyed using demographic questionnaires, the Ocular Surface Disease Index (OSDI), Hospital Anxiety and Depression Scale (HADS), and Short Health Anxiety Inventory (SHAI). Additionally, they were examined using Keratograph 5M and assessed for DED by corneal fluorescein staining. Regression analysis and the bootstrap method were used to investigate the influence of HA on the relationship between DED and emotional disorders.ResultsAmong the 206 patients with DED, 52 (25.24%) and 56 (27.18%) patients showed depression and anxiety, respectively. The OSDI score and HA were positively correlated with depression and anxiety (P < 0.01). The direct effects of OSDI on depression and anxiety were significant (95% confidence interval [CI]: 0.017–0.069; 0.008–0.060). Additionally, the bootstrap test showed significant mediating effects of HA (95% CI: 0.001–0.016; 0.003–0.021). The results suggested that the severity of DED symptoms, as measured by the OSDI score, affected anxiety and depression by a direct and an indirect pathway mediated by HA.ConclusionsWe found a significant correlation between DED and anxiety and depression. Moreover, HA was a mediator of the relationship between DED symptoms and anxiety and depression.

Aims/Introduction. Sex steroid hormones play a significant role in the progression of cardiovascular and metabolic diseases, including non-alcoholic fatty liver disease (NAFLD). The aim of this cross-sectional study was to explore the associations of testosterone, androstenedione, dehydroepiandrosterone (DHEA), and dehydroepiandrosterone sulfate (DHEAS), with the risk of NAFLD in patients with type 2 diabetes mellitus (T2DM). Materials and Methods. There were 1155 patients with T2DM included in the analysis. Serum levels of total testosterone and the precursors of androgens, including androstenedione, DHEA, and DHEAS, were quantified using liquid chromatography-tandem mass spectrometry assays. Results. The risk of NAFLD decreased as total testosterone concentration increased in men with T2DM. After adjusting for age, current smoking, current drinking, body mass index, duration of T2DM, diastolic blood pressure, total cholesterol, triglycerides, low-density lipoprotein/high-density lipoprotein cholesterol ratio, uric acid, C-reactive protein, and sex hormones in model 4, the adjusted odds ratio (OR) and 95% confidence interval (CI) of NAFLD for tertile3 vs tertile1 was 0.37 (0.17–0.77; P=0.024 for trend). When taken as a continuous variable, this association was still robust in model 4 (OR, 0.58; 95% CI, 0.42–0.80; P0.05). Moreover, women showed no significant associations of total testosterone, androstenedione, DHEA, and DHEAS, with the odds of NAFLD (all P>0.05). Conclusions. Serum total testosterone was independently associated with the risk of NAFLD among men with T2DM. This study highlights the potential role of testosterone as a risk factor for NAFLD in patients with T2DM.

The variation of solar radiation has a profound effect on the surface energy balance and hydrological cycle. Although the relationship between solar radiation variation and its influencing factors has been extensively studied, they are seldom used in Xinjiang, the largest province in China. In this study, we investigated the spatial distribution and temporal variation in global radiation (Eg), water vapor content (WVC), aerosol optical depth (AOD), total cloud cover (TCC), and low-level cloud cover (LCC) in Xinjiang, northwestern China, between 1961 and 2015. The annual average Eg reported at all stations was 5126.3–6252.8 MJ·m−2 with a mean of 5672 MJ·m−2. The highest annual mean Eg of 6252.8 MJ·m−2 occurred in Hami, eastern Xinjiang, whereas the lowest annual mean Eg of 5126.3 MJ·m−2 occurred in Urumqi, northern Xinjiang. The annual Eg variation was mainly affected by WVC, AOD, TCC, and LCC. Decreases in annual, spring, summer, autumn, and winter Eg trends were recorded in Xinjiang at rates of −33.88 × 10−2, −1.92 × 10−2, −1.89 × 10−2, −3.47 × 10−2, and −3.56 × 10−2 MJ·m−2·decade−1, respectively, with decreasing ratios of 9.43%, 5.85%, 0.14%, 8%, and 20.55%, respectively. Increasing trends in annual WVC, AOD, TCC, and LCC were noted in Xinjiang at rates of 7.12 × 10−5 mm·decade−1, 2.74 × 10−6 decade−1, 8.77 × 10−5 % decade−1, and 5.73 × 10−5% decade−1, respectively. In addition, increasing trends in the annual Eg at Yining and Yanqi stations were observed. The Eg spatial distribution was complex in Xinjiang at the stations observed in this study, which were divided into six groups. Eg at group 1 showed an increasing trend associated with decreases in the WVC and TCC, whereas decreases in Eg were observed at groups 2–6, which could have been influenced by increases in AOD, TCC, and LCC.

Head and neck squamous cell carcinoma (HNSCC) is one of the most common cancer worldwide and seriously threats public health safety. Despite the improvement of diagnostic and treatment methods, the overall survival for advanced patients has not improved yet. This study aimed to sort out prognosis-related molecular biomarkers for HNSCC and establish a prognostic model to stratify the risk hazards and predicate the prognosis for these patients, providing a theoretical basis for the formulation of individual treatment plans. We firstly identified differentially expressed genes (DEGs) between HNSCC tissues and normal tissues via joint analysis based on GEO databases. Then a total of 11 hub genes were selected for single-gene prognostic analysis to identify the prognostic genes. Later, the clinical information and transcription information of HNSCC were downloaded from the TCGA database. With the application of least absolute shrinkage and selection operator (LASSO) algorithm analyses for the prognostic genes on the TCGA cohort, a prognostic model consisting of three genes (COL4A1, PLAU and ITGA5) was successfully established and the survival analyses showed that the prognostic model possessed a robust performance in the overall survival prediction. Afterward, the univariate and multivariate regression analysis indicated that the prognostic model could be an independent prognostic factor. Finally, the predicative efficiency of this model was well confirmed in an independent external HNSCC cohort.

Objective: This study aimed to investigate the relationship between dry eye disease (DED) with anxiety and depression. Additionally, the mediating effect of sleep quality on this relationship was explored.Methods: 321 patients with DED were recruited from Tianjin Medical University Eye Hospital clinic and surveyed using demographic questionnaires, the Ocular Surface Disease Index (OSDI), Hospital Anxiety and Depression Scale (HADS), and Pittsburgh Sleep Quality Index (PSQI). Regression analysis and the bootstrap method were used to investigate the influence of sleep on the relationship between DED, anxiety and depression.Results: Among the patients with DED, 86 (26.79%), 85 (26.48%), and 54 (16.82%) patients presented with anxiety, depression, and both anxiety and depression respectively. The OSDI and PSQI score were positively correlated with depression and anxiety (all p < 0.01). The direct effects of OSDI on depression and anxiety were significant (P < 0.01). Additionally, the bootstrap test showed significant mediating effects of subjective sleep quality [95% CI [0.003–0.016] (depression); [0.001–0.011] (anxiety)] and sleep latency [95% CI [0.001–0.010] (depression); [0.001–0.008] (anxiety)]. These results indicated that the severity of DED symptoms, as measured by the OSDI score, affected anxiety and depression through a direct and an indirect pathway mediated by subjective sleep quality and sleep latency.Conclusions: The results indicated that there was a significant correlation between DED and anxiety and depression. Moreover, subjective sleep quality and sleep latency were a mediator of the relationship between DED symptoms and anxiety and depression.

Objective: The pulmonary sequelae of coronavirus disease 2019 (COVID-19) have not been comprehensively evaluated. We performed a follow-up study analyzing chest computed tomography (CT) findings of COVID-19 patients at 3 and 6 months after hospital discharge.Methods: Between February 2020 and May 2020, a total of 273 patients with COVID-19 at the Shenzhen Third People's Hospital were recruited and followed for 6 months after discharge. Chest CT scanning was performed with the patient in the supine position at end-inspiration. A total of 957 chest CT scans was obtained at different timepoints. A semi-quantitative score was used to assess the degree of lung involvement.Results: Most chest CT scans showed bilateral lung involvement with peripheral location at 3 and 6 months follow-up. The most common CT findings were ground-glass opacity and parenchymal band, which were found in 136 (55.3%) and 94 (38.2%) of the 246 patients at 3 months follow-up, and 82 (48.2%) and 76 (44.7%) of 170 patients at 6 months follow-up, respectively. The number of lobes involved and the total CT severity score declined over time. The total CT score gradually increased with the increasement of disease severity at both 3 months follow-up (trend test P < 0.001) and 6 months follow-up (trend test P < 0.001). Patients with different disease severity represented diverse CT patterns over time.Conclusions: The most common CT findings were ground-glass opacity and parenchymal bands at the 3 and 6 months follow-up. Patients with different disease severity represent diverse CT manifestations, indicating the necessary for long-term follow-up monitoring of patients with severe and critical conditions.

Abstract In this study, the effect of ash removal on Shenfu bituminous coal was investigated. The coal was pretreated by hydrofluoric acid (HF) pickling, and the raw/pretreated coal chars were prepared at 900 °C in a fixed bed reactor. The structure of coal and char were detected by Fourier transform infrared (FTIR) and Raman spectroscopy. The reactivity was tested in a thermogravimetric analyzer, including coal pyrolysis and char gasification. The reaction kinetics was analyzed through the Coats–Redfern method, master plots, the model-free and model-fitting method. The results show that the HF pickling can remove silicon from coal efficiently, and the macromolecular framework of coal is quite stable according to FTIR. The Raman parameters imply some carbonaceous structure on coal surface changed. For slow pyrolysis of coal, the effect of heating rate is considered. The changes of pyrolysis characteristics and kinetics are insignificant. For char gasification, the reactivity under isothermal and non-isothermal condition are discussed with an emphasis in different residence time of devolatilization process. In kinetic control region (low temperature), the activation energy (E a) is very close (about 240 kJ/mol) for all chars. With the temperature increases, the reactivity of raw coal char is more easily suffered by diffusion. The random pore model is more suitable for the ash-free coal char, and the char with long residence time has a larger value of structural parameter ψ and smaller value of pre-exponential factor A. The E a calculated by model-fitting and model-free method were in good agreement.

Abstract Background Kashin–Beck disease (KBD) is a disabling osteoarticular disease involving growth and joint cartilage. Early diagnosis can effectively prevent the progress of the disease. However, the early diagnosis of it is still very difficult. Our aim was to study the knee joint lesions of a rat KBD model using ultra-high field magnetic resonance imaging (MRI) and compare it with X-ray imaging to analyze the possible MRI manifestations of KBD, and to further explore ways to determine the pathological damage of KBD in the early stage. Methods A total of 96 Wistar rats were selected and randomly divided into 4 groups: normal diet (Group A), KBD-affected diet (Group B), normal diet+T-2 toxin (Group C), and KBD-affected diet+T-2 toxin (Group D). T-2 toxin was administered at a dose of 0.1 mg/kg/day. In the 4th week, 8th week, and 12th week, eight rats randomly selected in each group were sacrificed by cervical dislocation after undergoing X-ray and 7.0 T MRI imaging, and then knee joints were harvested, sliced, and subjected to hematoxylin-eosin (H&E) staining. Results Characteristic image changes including of continuity interruption and early closure and fusion of epiphyseal plates were observed on T1WI in rat model of KBD. The total necrosis rates in the H&E stain of group A to group D were 4.35, 52.38, 33.3, and 73.68%, respectively. The positive rate of image change under 7.0 T MRI was 0.833 VS. that under X-ray was 0.33 (P = 0.001). Conclusions MRI at 7.0 T is highly sensitive to the early pathological changes of the epiphysis, epiphyseal plate, and metaphyseal end, which can improve imaging positive rate of KBD and decrease the rate of missed diagnosis. This imaging modality can be used for research on early joint lesions and for early diagnosis of KBD.

Two-dimensional (2D) materials have dominated nanoscience for the last two decades. Among all 2D materials, graphene, MoS2, and h-BN are extremely popular and have been tentatively scaled up to fabricate nanocomposites, energy storage devices, flexible electronics, etc., ex situ and in situ mechanical characterization of 2D crystals can help us understand their mechanical behavior and measure their mechanical properties, which are of great significance in both fundamental science and practical engineering. To date, a great effort has been devoted to both theoretical and experimental mechanics with a focus on unveiling mechanical behaviors and quantifying mechanical properties. Beyond original research, several insightful review works have been published with a specific focus on the mechanics of 2D materials. To have a complementary contribution to the overview of the mechanics of 2D materials, we would like to review the developed experimental techniques being used to mechanically characterize 2D materials. The working mechanism and associated advantages and disadvantages of the techniques will be briefly discussed. Based on the existence of arguments in mechanical properties and behaviors of 2D crystals, and immature mechanical characterization of 2D materials, more intensive and comprehensive studies are expected toward a full understanding of these novel and promising materials.

Abstract Background The novel coronavirus disease 2019 (COVID-19) broke out globally. Early prediction of the clinical progression was essential but still unclear. We aimed to evaluate the timeline of COVID-19 development and analyze risk factors of disease progression. Methods In this retrospective study, we included 333 patients with laboratory-confirmed COVID-19 infection hospitalized in the Third People’s Hospital of Shenzhen from 10 January to 10 February 2020. Epidemiological feature, clinical records, laboratory and radiology manifestations were collected and analyzed. 323 patients with mild-moderate symptoms on admission were observed to determine whether they exacerbated to severe-critically ill conditions (progressive group) or not (stable group). We used logistic regression to identify the risk factors associated with clinical progression. Results Of all the 333 patients, 70 (21.0%) patients progressed into severe-critically ill conditions during hospitalization and assigned to the progressive group, 253 (76.0%) patients belonged to the stable group, another 10 patients were severe before admission. we found that the clinical features of aged over 40 (3.80 [1.72, 8.52]), males (2.21 [1.20, 4.07]), with comorbidities (1.78 [1.13, 2.81]) certain exposure history (0.38 [0.20, 0.71]), abnormal radiology manifestations (3.56 [1.13, 11.40]), low level of T lymphocytes (0.99 [0.997, 0.999]), high level of NLR (0.99 [0.97, 1.01]), IL-6 (1.05 [1.03, 1.07]) and CRP (1.67 [1.12, 2.47]) were the risk factors of disease progression by logistic regression. Conclusions The potential risk factors of males, older age, with comorbidities, low T lymphocyte level and high level of NLR, CRP, IL-6 can help to predict clinical progression of COVID-19 at an early stage.

Abstract Hereditary spherocytosis (HS) is often misdiagnosed due to lack of specific diagnostic methods. Our study summarized clinical characteristics and described the diagnostic workflow for mild and moderate HS in Chinese individuals, using data from 20 adults, 8 of whom presented a familial history for HS. We used scanning electron microscopy (SEM) to diagnose HS. Abstract Background Pheochromocytoma/paraganglioma (PPGL) is a rare neuroendocrine tumor. Succinate dehydrogenase (SDH) deficiency has been confirmed to be associated with PPGL in various studies. SDHB mutations play an important role in PPGL. However, genetic screening of PPGL patients has not been widely carried out in clinics in China, and only a few related studies have been reported. Case presentation We report a case of a 23-year-old woman with paraganglioma (PGL) caused by a novel missense SDHB mutation, c.563 T > C (p.Leu188Pro), who presented with paroxysmal hypertension. Computed tomography (CT) and magnetic resonance imaging (MRI) revealed a PGL in the right retroperitoneum and no metastasis. The patient was treated with surgical excision and did not have postsurgerical paroxysmal hypertension. In addition, we searched the literature related to variations in SDHB genes in Chinese patients with PPGL using multiple online databases, including PubMed, China Hospital Knowledge Database and Wanfang Data. Ultimately, 14 studies (published between 2006 and 2019) comprising 34 cases of SDHB-related PGL or pheochromocytoma (PCC) were found. In total, 35 patients were enrolled in this study, and 25 mutations were identified. The common genetic alterations of SDHB in China were c.136C > T (11.4%), c.18C > A (11.4%) and c.725G > A (8.5%). Some carriers of SDHB mutations (28.1%) developed metastatic PPGL, and a high frequency of head and neck PGLs (HNPGLs) (59.4%) was reported. Conclusions We describe a classic case with a novel SDHB c.563 T > C mutation. Based on our literature review, common SDHB gene mutations in Chinese PPGL patients are c.136C > T, c.18C > A and c.725G > A.

Abstract Background The anteroposterior view of the lumbar plain radiograph (AP-LPR) was chosen as the original and first radiographic tool to determine and classify lumbosacral transitional vertebra with morphological abnormality (MA-LSTV) according to the Castellvi classification. However, recent studies found that AP-LPR might not be sufficient to detect or classify MA-LSTV correctly. The present study aims to verify the reliability of AP-LPR on detecting and classifying MA-LSTV types, taking coronal reconstructed CT images (CT-CRIs) as the gold criteria. Methods Patients with suspected MA-LSTVs determined by AP-LPR were initially enrolled. Among them, those who received CT-CRIs were formally enrolled to verify the sensitivity of AP-LPR on detecting and classifying MA-LSTV types according to the Castellvi classification principle. Results A total of 298 cases were initially enrolled as suspected MA-LSTV, among which 91 cases who received CT-CRIs were enrolled into the final study group. All suspected MA-LSTVs were verified to be real MA-LSTVs by CT-CRIs. However, 35.2% of the suspected MA-LSTV types judged by AP-LPR were not consistent with the final types judged by CT-CRIs. Two suspected type IIIa and 20 suspected type IIIb MA-LSTVs were verified to be true, while 9 of 39 suspected type IIa, 9 and 3 of 17 suspected type IIb, and 11 of 13 suspected type IV MA-LSTVs were verified to truly be type IIIa, IIIb, IV and IIIb MA-LSTVs by CT-CRIs, respectively. Incomplete joint-like structure (JLS) or bony union structure (BUS) and remnants of sclerotic band (RSB) between the transverse process (TP) and sacrum were considered to be the main reasons for misclassification. Conclusion Although AP-LPR could correctly detect MA-LSTV, it could not give accurate type classification. CT-CRIs could provide detailed information between the TP and sacrum area and could be taken as the gold standard to detect and classify MA-LSTV.

Background We conducted a matched case–control study in China during the 2013/14–2015/16 influenza seasons to estimate influenza vaccine effectiveness (VE) by dose among children aged 6 months to 8 years. Methods Cases were laboratory-confirmed influenza infections identified through the influenza-like illness sentinel surveillance network in Guangzhou. Age- and sex-matched community controls were randomly selected through the expanded immunization program database. We defined priming as receipt of ≥1 dose of influenza vaccine during the immediate prior season. Results In total, 4,185 case–control pairs were analyzed. Among children 6–35 months, VE for current season dose(s) across the three seasons during 2013/14–2015/16 were 59% (95% Confidence Interval: 44–71%), 12% (−11%,30%), 54% (32–69%); among unprimed children 6–35 months, VE for 1 vs 2 current season doses were 45% (8–67%) vs 65% (46–78%), −2% (−53%,32%) vs 19% (−11%,40%), and 37% (−24%,68%) vs 61% (32–78%). Among children aged 3–8 years, VE for current season dose(s) across study seasons were 62% (36–78%), 43% (22–58%), 32% (1–53%). VE for unprimed children receiving 1 dose only in current season was insignificant or lower than among all children. Conclusion Findings support utility of providing second dose (“booster dose”) of seasonal influenza vaccine to unprimed children aged 6–35 months, and the need to study further dose effect of a booster dose among unprimed children aged 3–8 years in China.

Abstract Background Polycystic ovary syndrome (PCOS) is a complex endocrine syndrome with poorly understood mechanisms. To provide patients with PCOS with individualized therapy, it is critical to precisely diagnose the phenotypes of the disease. However, the criteria for diagnosing the different phenotypes are mostly based on symptoms, physical examination and laboratory results. This study aims to compare the accuracy and efficacy of diagnosing PCOS by integrating metabolomic markers with common clinical characteristics. Methods This is a prospective, multicenter, analyst-blinded, randomized controlled trial. Participants will be grouped into (1) people without PCOS (healthy control group), (2) patients diagnosed with PCOS based on clinical indices (experimental group 1), and (3) patients diagnosed with PCOS based on metabolomic indices (experimental group 2). A total of 276 participants, including 60 healthy people and 216 patients with PCOS, will be recruited. The 216 patients with PCOS will be randomly assigned to the two experimental groups in a 1:1 ratio, and each group will receive a different 6-month treatment. The primary outcome for the experimental groups will be the effect of PCOS treatment. Discussion The results of this trial should help to determine whether using metabolomic indices is more accurate and effective than using clinical characteristics in diagnosing the phenotypes of PCOS. The results could provide a solid foundation for the accurate diagnosis of different PCOS subgroups and for future research on individualized PCOS therapy. Background The traditional open surgery in the treatment of spinal tuberculosis has the disadvantages of greater trauma, with more intraoperative bleeding and long postoperative hospitalization days. With the development of minimally invasive spine surgery (MISS) and spinal endoscopic systems, the minimally invasive spine surgery technique was increasingly being used by surgeons because it minimizes trauma, and patients experience faster recovery. The purpose of this study was to discuss the efficacy and clear indications of this treatment. Methods The clinical data of thirty‐one patients with thoracolumbar tuberculosis treated by posterior percutaneous pedicle screw fixation combined with full‐endoscopic debridement and intravertebral bone grafting were analyzed retrospectively. The preoperative and postoperative imaging manifestations and clinical results were compared, and the postoperative complications were observed with a follow‐up for more than 1 year. Results Compared with values before the operation, the visual analogue scale (VAS) score and Oswestry Disability Index (ODI) score were significantly decreased at 3 months after operation and at the last follow‐up. The erythrocyte sedimentation rate (ESR) and C‐reactive protein (CRP) level were decreased at 3 months after surgery and at the last follow‐up, and no significant reduction was observed in the Cobb angle at 3 months after operation or at the last follow‐up. In addition, imaging examinations showed that the necrotic material in vertebral bodies was adequately resected. Conclusion One‐stage percutaneous pedicle screw fixation combined with full‐endoscopic focus debridement and intravertebral bone grafting can achieve the purpose of removing the focus of thoracolumbar tuberculosis, restoring neurological function, and reconstructing spinal stability. It was a safe and feasible surgical method and its surgical indications were clarified.

Abstract Background Calcium-dependent protein kinase (CPK) is one of the main Ca2+ combined protein kinase that play significant roles in plant growth, development and response to multiple stresses. Despite an important member of the stress responsive gene family, little is known about the evolutionary history and expression patterns of CPK genes in pineapple. Results Herein, we identified and characterized 17 AcoCPK genes from pineapple genome, which were unevenly distributed across eight chromosomes. Based on the gene structure and phylogenetic tree analyses, AcoCPKs were divided into four groups with conserved domain. Synteny analysis identified 7 segmental duplication events of AcoCPKs and 5 syntenic blocks of CPK genes between pineapple and Arabidopsis, and 8 between pineapple and rice. Expression pattern of different tissues and development stages suggested that several genes are involved in the functional development of plants. Different expression levels under various abiotic stresses also indicated that the CPK family underwent functional divergence during long-term evolution. AcoCPK1, AcoCPK3 and AcoCPK6, which were repressed by the abiotic stresses, were shown to be function in regulating pathogen resistance. Conclusions 17 AcoCPK genes from pineapple genome were identified. Our analyses provide an important foundation for understanding the potential roles of AcoCPKs in regulating pineapple response to biotic and abiotic stresses

Smart meter is a typical edge device that measures and records the energy data. The usage of smart meter data can improve the cyber physical relationship between the smart grid and cyber physical system. Millions of smart meters have been installed all around the world and the malfunction detection of large volume meter is a big issue. On site checking is a costing work and cannot meet the requirement for large scale meters. Online malfunctional meter detection and verification based on meter data analytics is a solution to the meter detection problem. For the purpose of detecting malfunctional smart meter, the low-voltage energy system model is studied and a meter error estimation method is proposed in this paper. This method adopts a decision tree to filter the abnormal data and classify data with different energy loss levels. Then clustering the data to obtain the data set with different energy usage behavior. A meter data matrix is constructed and meter error can be calculated from the solution of the matrix equation. A Recursive algorithm is adopted to solve the equation and estimate the meter error. The meter error above the regulation threshold will be classified as a malfunctional meter. The proposed approach has achieved higher accuracy in the experiment.

A new technology called in vivo glia-to-neuron conversion has emerged in recent years as a promising next generation therapy for neural regeneration and repair. This is achieved through reprogramming endogenous glial cells into neurons in the central nervous system through ectopically expressing neural transcriptional factors in glial cells. Previous studies have been focusing on glial cells in the grey matter such as the cortex and striatum, but whether glial cells in the white matter can be reprogrammed or not is unknown. To address this fundamental question, we express NeuroD1 in the astrocytes of both grey matter (cortex and striatum) and white matter (corpus callosum) to investigate the conversion efficiency, neuronal subtypes, and electrophysiological features of the converted neurons. We discover that NeuroD1 can efficiently reprogram the astrocytes in the grey matter into functional neurons, but the astrocytes in the white matter are much resistant to neuronal reprogramming. The converted neurons from cortical and striatal astrocytes are composed of both glutamatergic and GABAergic neurons, capable of firing action potentials and having spontaneous synaptic activities. In contrast, the few astrocyte-converted neurons in the white matter are rather immature with rare synaptic events. These results provide novel insights into the differential reprogramming capability between the astrocytes in the grey matter versus the white matter, and highlight the impact of regional astrocytes as well as microenvironment on the outcome of glia-to-neuron conversion. Since human brain has large volume of white matter, this study will provide important guidance for future development of in vivo glia-to-neuron conversion technology into potential clinical therapies.
