A system of two dynamic plant chambers and a downstream reaction chamber has been set up to investigate the emission of biogenic volatile organic compounds (BVOC) and possible effects from pollutants such as ozone. The system can be used to compare BVOC emissions from two sets of differently treated plants, or to study the photochemistry of real plant emissions under polluted conditions without exposing the plants to pollutants. The main analytical tool is a proton-transfer-reaction time-of-flight mass spectrometer (PTR-TOF-MS) which allows online monitoring of biogenic emissions and chemical degradation products. The identification of BVOCs and their oxidation products is aided by cryogenic trapping and subsequent in situ gas chromatographic analysis. • The exploitation of natural variation in Arabidopsis thaliana (Arabidopsis) provides a huge potential for the identification of the molecular mechanisms underlying this variation as a result of the availability of a vast array of genetic and genomic resources for this species. Eighty-six Arabidopsis accessions were screened for natural variation in flooding tolerance. This forms the first step towards the identification and characterization of the role of candidate genes contributing to flooding tolerance. • Arabidopsis accessions at the 10-leaf stage were subjected to complete submergence in the dark. Survival curves were plotted to estimate median lethal times as a measure of tolerance. Flooding-associated survival parameters, such as root and shoot oxygen content, initial carbohydrate content and petiole elongation under water, were also measured. • There was a significant variation in submergence tolerance among Arabidopsis accessions. However, the order of tolerance did not correlate with root and shoot oxygen content or initial amounts of shoot starch and total soluble sugars. A negative correlation was observed between submergence tolerance and underwater petiole elongation. • Arabidopsis accessions show considerable variation in the ability to tolerate complete submergence, making it a good species in which to identify and characterize genes and to study mechanisms that contribute to survival under water. In vitro cultivated pear, Pyrus communis L. cv. Beurré Hardy, pedicels cut above their primary abscission layer can form a secondary abscission layer, especially under the influence of auxins or cytokinins in the culture medium. The maximum percentage of abscission reached by auxin application was always higher than that by cytokinin. The presence of the flower was of no consequence to the abscission. Characteristic differences in abscission were observed between pear and apple pedicels. In contrast to apple (1) secondary abscission in pear could also be induced by cytokinins, and (2) the site of abscission in pear was dependent on the auxin concentration. At lower auxin concentrations abscission was induced in the basal parts of the pedicels inserted in the medium, whereas at higher auxin concentrations the abscission layer was formed in the terminal parts of the pedicels above the culture medium. In vitro cultivated apple pedicels without a primary abscission layer can form a secondary (adventitious) abscission layer, especially under the influence of auxins. In the apple cv. Cox's Orange Pippin abscission can only be induced by auxins, while the site of the abscission layer, a few millimetres from the basal ends of the pedicels, is fixed and independent of the auxin concentration. The auxin treatment has to last at least 5-6 days to induce abscission, which is not affected by the presence of a flower. A secondary layer does not occur when pedicels are placed inverted on the media. Although abscission occurs both in light and in darkness, it is strongly promoted by light. Abscission is also accelerated by raising the temperature from 9°C to 21-25°C. High concentrations of 2,3,5-triiodobenzoic acid reduce the percentage of auxin-induced abscission. The effect of various growth substances on regeneration and growth of bulblets on excised bulb scale segments of Hyacinthus orientalis L. cv. Pink Pearl was studied. Bulblet formation was promoted by the auxins IAA and IBA but was inhibited by NAA at high concentrations. IAA hardly affected bulblet weight, whereas IBA and NAA at high concentrations decreased it. Cytokinins bad hardly any effect on bulblet regeneration and bulblet weight. Gibberellins inhibited both bulblet regeneration and bulblet weight; GA4+7 acted much stronger than GA3. During the last 3 years a method has been developed to reproduce Anthurium andraeanum Lind, through callus cultures. The procedural sequence is as follows: callus induction on excised leaf fragments, callus subculture on solid and in liquid media, adventitious sprout formation in callus on solid media, and rooting of excised sprouts. Abstract Background Recently, excellent results are reported on laparoscopic lavage in patients with purulent perforated diverticulitis as an alternative for sigmoidectomy and ostomy. The objective of this study is to determine whether LaparOscopic LAvage and drainage is a safe and effective treatment for patients with purulent peritonitis (LOLA-arm) and to determine the optimal resectional strategy in patients with a purulent or faecal peritonitis (DIVA-arm: perforated DIVerticulitis: sigmoidresection with or without Anastomosis). Methods/Design In this multicentre randomised trial all patients with perforated diverticulitis are included. Upon laparoscopy, patients with purulent peritonitis are treated with laparoscopic lavage and drainage, Hartmann's procedure or sigmoidectomy with primary anastomosis in a ratio of 2:1:1 (LOLA-arm). Patients with faecal peritonitis will be randomised 1:1 between Hartmann's procedure and resection with primary anastomosis (DIVA-arm). The primary combined endpoint of the LOLA-arm is major morbidity and mortality. A sample size of 132:66:66 patients will be able to detect a difference in the primary endpoint from 25% in resectional groups compared to 10% in the laparoscopic lavage group (two sided alpha = 5%, power = 90%). Endpoint of the DIVA-arm is stoma free survival one year after initial surgery. In this arm 212 patients are needed to significantly demonstrate a difference of 30% (log rank test two sided alpha = 5% and power = 90%) in favour of the patients with resection with primary anastomosis. Secondary endpoints for both arms are the number of days alive and outside the hospital, health related quality of life, health care utilisation and associated costs. Discussion The Ladies trial is a nationwide multicentre randomised trial on perforated diverticulitis that will provide evidence on the merits of laparoscopic lavage and drainage for purulent generalised peritonitis and on the optimal resectional strategy for both purulent and faecal generalised peritonitis. Abstract Background With the availability of infliximab, nowadays recurrent Crohn's disease, defined as disease refractory to immunomodulatory agents that has been treated with steroids, is generally treated with infliximab. Infliximab is an effective but expensive treatment and once started it is unclear when therapy can be discontinued. Surgical resection has been the golden standard in recurrent Crohn's disease. Laparoscopic ileocolic resection proved to be safe and is characterized by a quick symptom reduction. The objective of this study is to compare infliximab treatment with laparoscopic ileocolic resection in patients with recurrent Crohn's disease of the distal ileum with respect to quality of life and costs. Methods/design The study is designed as a multicenter randomized clinical trial including patients with Crohn's disease located in the terminal ileum that require infliximab treatment following recent consensus statements on inflammatory bowel disease treatment: moderate to severe disease activity in patients that fail to respond to steroid therapy or immunomodulatory therapy. Patients will be randomized to receive either infliximab or undergo a laparoscopic ileocolic resection. Primary outcomes are quality of life and costs. Secondary outcomes are hospital stay, early and late morbidity, sick leave and surgical recurrence. In order to detect an effect size of 0.5 on the Inflammatory Bowel Disease Questionnaire at a 5% two sided significance level with a power of 80%, a sample size of 65 patients per treatment group can be calculated. An economic evaluation will be performed by assessing the marginal direct medical, non-medical and time costs and the costs per Quality Adjusted Life Year (QALY) will be calculated. For both treatment strategies a cost-utility ratio will be calculated. Patients will be included from December 2007. Discussion The LIR!C-trial is a randomized multicenter trial that will provide evidence whether infliximab treatment or surgery is the best treatment for recurrent distal ileitis in Crohn's disease.
