Although effective rabies virus vaccines have been existing for decades, each year, rabies virus infections still cause around 50.000 fatalities worldwide. Most of these cases occur in developing countries, where these vaccines are not available. The reasons for this are the prohibitive high costs of cell culture or egg grown rabies virus vaccines and the lack of a functional cold chain in many regions in which rabies virus is endemic. Here, we describe the excellent temperature resistance of a non-replicating mRNA based rabies virus vaccine encoding the rabies virus glycoprotein (RABV-G). Prolonged storage of the vaccine from -80°C to up to +70°C for several months did not impact the protective capacity of the mRNA vaccine. Efficacy after storage was demonstrated by the induction of rabies specific virus neutralizing antibodies and protection in mice against lethal rabies infection. Moreover, storing the vaccine at oscillating temperatures between +4° and +56°C for 20 cycles in order to simulate interruptions of the cold chain during vaccine transport, did not affect the vaccine's immunogenicity and protective characteristics, indicating that maintenance of a cold chain is not essential for this vaccine.

The Ilha Grande National Park, Paraná, Brazil, is located in the Upper Paraná River and has characteristics typical of a floodplains. This protected area includes lagoons connected and disconnected to the Paraná River, although the latter also connect during periods of high water level, thus composing a heterogeneous group of lacustrine environments. The enormous potential the flora and fauna diversities are still little known to the region, as can be seen through benthic invertebrates, inclunding bivalves mollusks. The granulometric composition of these floodplain lagoons was formed mainly by mud and very fine sand. Furthermore, organic matter composition was predominantly of fine particulate. The other abiotic factors differed from lagoons located within the island of the park to those located in the left margin of Paraná River. The results demonstrated the importance of abiotic factors such as the physical composition of granulometric texture, organic matter and macrophyte banks, to the establishment of bivalves in these floodplain lagoons. We recorded bivalves of Pisidium (native), Diplodon (native), and Corbicula (invasive). The highest values of Diplodon sp. density were observed at São João/C lake, for Pisidium sterkianum (Pilsbry, 1897) at São João/M lake, and to Jatobá/C lagoon with high density of invasive species Corbicula fluminea (Müller, 1774). This study to obtain conduct the first records of freshwater bivalves in floodplains lagoon in the Ilha Grande National Park, and provides contributions to better understanding the ecology of these mollusks. The recording of native species in the region of Upper Paraná River floodplain after a lomg period without new records, demonstrated the importance of protecting the lagoons of the Ilha Grande National Park as they can be a possible refuge to some species of native freshwater bivalves.

There is a global need for effective and affordable rabies vaccines, which is unmet by current vaccines due to limitations in their production capacities, required administration schedules, storage requirements, and cost. Many different experimental approaches previously used for bacterial and viral vaccines have been applied to rabies, but with variable success. One of the most promising new concepts is the use of messenger RNA (mRNA) in encoding the main rabies virus antigen, the envelope glycoprotein (RABV-G). CureVac has applied their proprietary technology platform for the production of mRNA to this problem, resulting in the rabies vaccine candidate CV7201. Following preclinical studies in mice and pigs showing that CV7201 could induce neutralizing immune responses that protected against rabies virus, different dosages and routes of administration of CV7201 were tested in a phase 1 human study. This clinical study proved that mRNA vaccination was safe and had an acceptable reactogenicity profile, but immune responses depended on the mode of administration, and they did not unequivocally support CV7201 for further development as a prophylactic vaccine with this particular formulation. Further, preclinical studies using RABV-G mRNA encapsulated in lipid nanoparticles (LNPs) showed an improved response in both mice and nonhuman primates, and these encouraging results are currently being followed up in clinical studies in humans. This review summarizes the recent advances in mRNA vaccines against rabies.

One of the central themes in ecology is the relative importance of local and regional processes for determining the communities’ structure, since both processes may behave as filters in the composition of local communities. Thus, this study analyzed the influence of environmental factors on the benthic invertebrate community distribution in different channels of the Upper Paraná River floodplain, through quarterly samplings conducted from March to December 2010. Through the biotic and abiotic data, we performed a Canonical Correspondence Analysis, where it was possible to visualize the centers of Ivinhema and Paraná rivers and Ipoitã channel separate from other points by high values of depth and velocity and taxa typical of lotic environments, such as Harpacticoida, Haplotaxidae, and Narapidae, and the center of the Curutuba channel, with L. fortunei. One may conclude that flow velocity, granulometric texture, and sediment organic matter were structuring factors of the benthic community, determining the distribution of invertebrates both among the various channels and between the marginal and central regions in these environments, providing greater or lesser availability of resources and environmental heterogeneity.

The objective of this work was to verify the applicability of a method of infiltration used for deployment of septic tanks in scientific studies. Measures consisted in checking the time needed for the water infiltrate 1 cm in a ruler in 10-minute intervals. The values of infiltration were 100 liters/hour; 83.5 liters/hour; 50 liters/hour; 412.5 liters/hour; 125 liters/hour respectively for temporary crop, pasture, mechanized sugar cane, forest, non-mechanized sugar cane. The amount of infiltrated water varied according to type of occupation in the area, duration of such occupation, as well as soil management and conservation techniques adopted in these environments.

Um dos temas centrais em ecologia é a importância relativa dos processos locais e regionais na determinação da estrutura de comunidades, pois ambos os processos podem agir como filtros na composição das comunidades locais. Nesse sentido, este estudo analisou a influência dos fatores ambientais sobre a distribuição da comunidade de invertebrados bentônicos em diferentes canais da planície de inundação do alto rio Paraná, por meio de coletas trimestrais realizadas de março a dezembro de 2010. A partir de dados bióticos e abióticos foi realizada Análise de Correspondência Canônica, onde foi possível agrupar os centros dos rios Ivinhema e Paraná e o canal Ipoitã em relação aos demais pontos, principalmente pelos maiores valores de profundidade e velocidade e pelos táxons típicos de ambientes lóticos, como Harpacticoida, Haplotaxidae e Narapidae, e o centro do canal Curutuba, com L. fortunei. Pode-se concluir que a velocidade de fluxo, a textura granulométrica e matéria orgânica do sedimento foram fatores estruturadores da comunidade bentônica, determinando a distribuição dos invertebrados tanto entre os diferentes canais como entre as regiões marginais e centrais desses ambientes, proporcionando maior ou menor disponibilidade de recursos e heterogeneidade ambiental.

Marine isotope stages (MIS) 11 has been proposed as an analog for the present interglacial; however, terrestrial records of this time period are rare. Sediments from Lake El'gygytgyn (67°30' N, 172°5' E) in Far East Russia contain a 3.56 Ma record of climate variability from the Arctic. Here, we present the first terrestrial Arctic reconstruction of environmental and climatic changes from MIS 8 through 12 (289 to 464 ka) using organic geochemical proxies. Terrestrial vegetation changes, as revealed by plant leaf wax (n-alkane) indices and concentrations of arborinol (a biomarker for trees), show increased tree cover around the lake during interglacial periods, with higher concentrations observed during MIS 11 as compared to MIS 9. A similar pattern is also observed in records of aquatic productivity revealed by molecular indicators from dinoflagellates (dinosterol), eustigmatophyte algae (long-chain (C28–C32) 1, 15 n-alkyl diols) in addition to short-chain n-alkanes, where aquatic productivity is highest during MIS 11. Changes recorded in these molecular proxies show a similar structure to relative temperature variability as recorded by the MBT/CBT (Methylation of Branched Tetraether/Cyclization of Branched Tetraether) paleothermometer, based on branched glycerol dialkyl glycerol tetraethers (GDGTs). Additionally, relative MBT/CBT temperature changes generally track pollen and diatom δ18O temperature estimates, compiled by other studies, which suggest glacial-interglacial temperature changes of ~ 9 to 12 °C. These records of environmental and climatic change indicate Arctic sensitivity to external forcings such as orbital variability and atmospheric greenhouse gas concentrations. Overall, this study indicates that organic geochemical analyses of the Lake El'gygytgyn sediment archive can provide critical insight into the response of lake ecosystems and their sensitivity in high latitude regions.

River-floodplain ecosystems (RFEs) provide multiple ecosystem services. However, their importance may be underestimated because they are not summarized yet. In this paper, we review and update the benefits that RFEs provide to society, including supporting, regulating, provisioning, and cultural ecosystem services. Although considered a unique ecosystem service category, we advocate that supporting services, like soil formation, nutrient cycling, primary production, and habitat provisioning can be comprehended as ecosystem processes that generate other services. RFEs provide valuable regulating services, including water regulation, storm protection, erosion control, water purification, waste treatment, and disease control. The society also benefits from provisioning services from RFEs, such as water for drinking and irrigation, food (e.g., fishes and crops), fiber, ornamental and biochemical resources, and energy production. RFEs also provide cultural services including recreation, ecotourism, religiosity, and spirituality. Most ecosystem services from pristine and human-altered RFEs are primarily regulated by the flood pulse because it maintains temporal and spatial habitat variability, high biodiversity, and biotic and abiotic interactions. Despite providing many benefits to society, RFEs are seriously threatened, mainly due to river regulation, land-use changes, pollution and invasive species. Consequently, the multiple demands and uses of RFEs worldwide raise challenges of conservation and restoration. [ABSTRACT FROM AUTHOR]
