Under the condition of high speed, the deformation of the pulley will cause the metal strip to shift along the cone of the pulley, which will aggravate the wear between the various parts of the CVT, which will seriously affect the service life and transmission efficiency of the CVT. Therefore, the mechanical model of the pulley under the condition of high speed was established, the influence of speed ratio, torque, speed and other factors on the axial deformation of the CVT pulley was analyzed by using the finite element method, and the test was verified. The results show that the axial deformation of the pulley decreases with the increase of the speed ratio, and decreases first and then increases with the increase of torque and speed, and the test results are consistent with the analysis results.

Abstract Background Outbreaks of the coral predator Crown-of-Thorns Starfish (CoTS) pose a severe threat to coral reefs in the Indo-Pacific Ocean. In 2018, the South China Sea (SCS) experienced significant CoTS outbreaks, leading to extensive coral mortality across the Xisha, Zhongsha, Dongsha, and Nansha Islands, severely impacting the coral reef ecosystem. Results To explore the origins of these outbreaks, we conducted a comprehensive genomic analysis using data from genomic single nucleotide polymorphism sites (SNPs) and mitochondrial haplotypes. Our analysis reveals that CoTS populations in the SCS, which exhibit moderate genetic diversity and may have undergone positive selection or population expansion. There was limited genetic differentiation among CoTS populations from XS, ZS, and NS groups. Especially between the XS and ZS groups, there was almost no genetic differentiation. The populations from XS, ZS, and NS groups have strong genetic connections with populations in Vietnam and the Philippines. There was high gene flow from Vietnam to the Xisha Islands and from the Philippines to the Nansha Islands, suggesting that the CoTS populations in these regions primarily originate from these neighboring countries. Conclusion The comprehensive analyses of SNP and mitochondrial genomes have provided valuable insights into the population genetics of CoTS. This research has generated significant genomic resources and facilitated important studies on the genetics of the CoTS species. By identifying potential source populations and understanding the genetic basis of their spread, managers can develop more effective conservation strategies to protect vulnerable coral reef ecosystems in the SCS.

Abstract Local adaptation has been a central theme of eco-evolutionary research for decades. It is generally assumed that plant pathogens are locally adapted due to their standing interactions with biotic and abiotic factors in the ecosystem. Effectors, secreted small proteins encoded by pathogens, play critical roles in host–pathogen interactions, by activating host genotype-specific resistance, suppressing plant immunity, and playing other functions. In this study, we investigated the potential involvement of RXLR effector genes in ecological adaptation by examining the simple sequence repeat (SSR), virulence, and effector profiles in Phytophthora infestans isolates collected from two geographic regions differing in ecological environments. Genotypic analyses with SSR markers and virulence assays showed that the pathogen from the two regions shared genetic background but differed in virulence spectrums. High-throughput sequencing and expression analysis of 24 selected P. infestans isolates further showed variations in the RXLR effector repertoire, ranging from 536 to 548 for each isolate and the expression of effector genes was highly associated with the accumulation of homologous sRNA. Regional specific alleles were detected at 94 RXLR effector genes, and a specific accumulation of homologous 25–26 nt sRNAs was found at 67 RXLR effector genes. Two of the regional specific RXLR effector genes were confirmed to be virulence factors. Taken together, these results suggest that genomic and epigenetic variations in RXLR effector genes contribute significantly to the ecological adaptation of P. infestans populations and that regional specific effector genes will help to understand the adaptive landscape of pathogens and efficient use of host resistance genes.

Abstract In order to explore the application potential of artificial intelligence (AI) and text mining technology in educational policy analysis and evaluate their impact on the psychological perception of policy audiences, this study firstly introduces the application of AI and text mining technology in education. Secondly, it explores the application of psychological theories in educational policy analysis. Finally, this study constructs an educational policy text analysis model and verifies the feasibility of the optimized model through performance comparison experiments and case analysis. The experimental results show that the optimized model exhibits higher accuracy, recall rate, and F1 score compared to traditional models when handling educational policy text analysis tasks with different data volumes. This finding highlights the importance of optimizing models for specific tasks and the potential of improving the understanding and analysis capabilities of models for specific text types through careful adjustments. In addition, the application of psychological theories to the analysis of educational policy texts provides a new perspective and method for understanding the impact of policies on audience psychological states, which helps in formulating more effective and humanized policies. Therefore, the study has certain reference significance for the use of AI and text mining technology to support educational policy analysis and formulation, providing valuable insights and guidance for future related research and practice.

Abstract The practical capacity of lithium-oxygen batteries falls short of their ultra-high theoretical value. Unfortunately, the fundamental understanding and enhanced design remain lacking, as the issue is complicated by the coupling processes between Li2O2 nucleation, growth, and multi-species transport. Herein, we redefine the relationship between the microscale Li2O2 behaviors and the macroscopic electrochemical performance, emphasizing the importance of the inherent modulating ability of Li+ ions through a synergy of visualization techniques and cross-scale quantification. We find that Li2O2 particle distributed against the oxygen gradient signifies a compatibility match for the nucleation and transport kinetics, thus enabling the output of the electrode’s maximum capacity and providing a basis for evaluating operating protocols for future applications. In this case, a 150% capacity enhancement is further achieved through the development of a universalizing methodology. This work opens the door for the rules and control of energy conversion in metal-air batteries, greatly accelerating their path to commercialization.

Abstract Background Circular RNAs (circRNAs) are highly stable regulators, often accumulated in mammalian brains and thought to serve as “memory molecules” that govern the long process of aging. Mounting evidence demonstrated circRNA dysregulation in the brains of Alzheimer’s disease (AD) patients. However, whether and how circRNA dysregulation underlies AD progression remains unexplored. Methods We combined Poly(A)-tailing/RNase R digestion experimental approach with CARP, our published computational framework using pseudo-reference alignment for more sensitive and accurate circRNA detection to identify genome-wide circRNA dysregulation and their downstream pathways in the 5xFAD mouse cerebral cortex between 5 and 7 months of age, a critical window marks the transition from reversible to irreversible pathogenic progression. Dysregulated circRNAs and pathways associated with disease progression in 5xFAD cortex were systematically compared with circRNAs affected in postmortem subcortical areas of a large human AD cohort. A top-ranked circRNA conserved and commonly affected in AD patients and 5xFAD mice was depleted in cultured cells to examine AD-relevant molecular and cellular changes. Results We discovered genome-wide circRNA alterations specifically in 5xFAD cortex associated with AD progression, many of which are commonly dysregulated in the subcortical areas of AD patients. Among these circRNAs, circGigyf2 is highly conserved and showed the highest net reduction specifically in the 7-month 5xFAD cortex. CircGIGYF2 level in AD patients’ cortices negatively correlated with dementia severity. Mechanistically, we found multiple AD-affected splicing factors that are essential for circGigyf2 biogenesis. Functionally, we identified and experimentally validated the conserved roles of circGigyf2 in sponging AD-relevant miRNAs and AD-associated RNA binding proteins (RBPs), including the cleavage and polyadenylation factor 6 (CPSF6). Moreover, circGigyf2 downregulation in AD promoted silencing activities of its sponged miRNAs and enhanced polyadenylation site processing efficiency of CPSF6 targets. Furthermore, circGigyf2 depletion in a mouse neuronal cell line dysregulated circGigyf2-miRNA and circGigyf2-CPSF6 axes and potentiated apoptotic responses upon insults, which strongly support the causative roles of circGigyf2 deficiency in AD neurodegeneration. Conclusions Together, our results unveiled brain circRNAs associated with irreversible disease progression in an AD mouse model that is also affected in AD patients and identified novel molecular mechanisms underlying the dysregulation of conserved circRNA pathways contributing to AD pathogenesis.

Aiming at the problems such as low structural bearing efficiency, structural weight gain, heavy load and cabin noise caused by high rear backbraced engine in BWB layout, based on novel materials and pultruded rod stitched efficient unitized structure(PRSEUS), a multi-disciplinary optimization design method for unconventional circular body structures with aerodynamic, noise, vibration and technological constraints was studied to improve the bearing efficiency and lightweight of structures. Based on the adaptive agent model construction technology, an agent model for the performance analysis of the aerodynamic and noise subsystem of the airframe structure is established to predict the aerodynamic load and noise performance of the airframe structure, and the basic requirements of the aerodynamic and noise performance are transformed into constraints such as geometric size and mechanical response, so that the multi-disciplinary problems are concentrated in a set of finite element analysis model with efficient mathematical programming method. The time-consuming problem of multi-disciplinary high-precision model analysis is solved, and the multi-disciplinary optimization design of the wing-body integrated body structure is realized. Through the finite element analysis of the body structure, the bearing efficiency of the structure has been greatly improved, and the weight loss rate of the central body structure has reached 8.7% under the premise of meeting the constraints of various disciplines.

Abstract As an emerging multi‐energy consumption subject, data centres (DCs) are bound to become crucial energy users for integrated energy systems (IES). Therefore, how to fully tap the potential of the collaborative operation between DCs and IES to improve total energy efficiency and economic performance is becoming a pressing need. In this article, the authors research an optimization coordinated by the energy scheduling and information service provision within the scenario of an integrated energy system with a data centre (IES‐DC). The mathematical model of IES‐DC is first established to reveal the energy conversion process of the electricity‐heat‐gas IES and the DC's energy consumption affected by the scale of active IT equipment. For dynamical providing multi‐energy and computing service by coordinating scheduling energy and information equipment, the formulations of IES‐DC scheduling, which is described as a Markov decision process (MDP), are presented, and it is solved by introducing the twin‐delayed deep deterministic policy gradient (TD3), which is a model‐free deep reinforcement learning (DRL) algorithm. Finally, the numerical studies show that compared with benchmarks, the proposed method based on the TD3 algorithm can effectively control the operation of energy conversion equipment and the number of active servers in IES‐DC.

PTBs equipped with advanced technologies such as oscillating-rotating and high-frequency sonic mechanisms have been examined for their potential to enhance oral hygiene. Special attention is given to vulnerable groups, including the elderly and individuals with intellectual disabilities, to assess how PTBs meet their specific oral health needs. A comprehensive literature search was conducted in databases including PubMed, Cochrane Library, Embase, and Google Scholar using keywords such as “power toothbrush”, “electric toothbrush”, “manual toothbrush”, “plaque removal”, “gingivitis”, “calculus”, “dental stains”, “oral hygiene”, “elderly”, and “intellectual disabilities”. Studies published between 2000 and 2024 were selected based on their relevance to the PTB and MTB comparison, with an emphasis on outcomes related to oral hygiene efficacy. As this review is narrative rather than systematic, it focuses on synthesizing existing knowledge without applying strict inclusion or exclusion criteria. The results indicate that PTBs generally outperform MTBs in reducing plaque, gingivitis, and stains, though the benefits for special populations are less pronounced but still significant. However, practical issues such as user experience and mechanical reliability of PTBs warrant further investigation. In conclusion, this review enhances the understanding of PTB effectiveness, guides consumer choices, and informs future technological advancements in dental care practices.Keywords: power toothbrushes, manual toothbrushes, plaque removal, gingival health, dental calculus, oral hygiene

Abstract Background The cardiometabolic index (CMI) is a novel metric for assessing cardiometabolic health and type 2 diabetes mellitus (DM), yet its relationship with insulin resistance (IR) and prediabetes (preDM) is not well-studied. There is also a gap in understanding the nonlinear associations between CMI and these conditions. Our study aimed to elucidate these associations. Methods We included 13,142 adults from the National Health and Nutrition Examination Survey (NHANES) 2007–2020. CMI was calculated by multiplying the triglyceride-to-high density lipoprotein cholesterol (TG/HDL-C) by waist-to-height ratio (WHtR). Using weighted multivariable linear and logistic regression explored the relationships of CMI with glucose metabolism markers, IR, preDM, and DM. Nonlinear associations were assessed using generalized additive models (GAM), smooth curve fittings, and two-piecewise logistic regression. Results Multivariate regression revealed positive correlations between CMI and glucose metabolic biomarkers, including FBG (β = 0.08, 95% CI: 0.06–0.10), HbA1c (β = 0.26, 95% CI: 0.22–0.31), FSI (β = 4.88, 95% CI: 4.23–5.54), and HOMA-IR (β = 1.85, 95% CI: 1.56–2.14). There were also significant correlations between CMI and increased risk of IR (OR = 3.51, 95% CI: 2.94–4.20), preDM (OR = 1.49, 95% CI: 1.29–1.71), and DM (OR = 2.22, 95% CI: 2.00-2.47). Inverse nonlinear L-shaped associations were found between CMI and IR, preDM, and DM, with saturation inflection points at 1.1, 1.45, and 1.6, respectively. Below these thresholds, increments in CMI significantly correlated with heightened risks of IR, preDM, and DM. Conclusions CMI exhibited inverse L-shaped nonlinear relationships with IR, preDM, and DM, suggesting that reducing CMI to a certain level might significantly prevent these conditions.

Abstract At present, the speed of the reel of soybean combine harvester is mainly adjusted manually, and basically does not have the ability to automatically adjust the speed of the reel according to changes in crops or working speed. In the maize-soybean strip intercropping mode, soybean plants are densely planted. During harvesting, the feeding rate increases, making it even more necessary to adjust the reel speed according to the growth status of the crops. This article uses PLC as the main control platform to design a reel speed control system for the existing combine harvester. Tests have shown that the control system can meet the requirements for reel speed control. The relative error of the reel control system’s speed test is 2.6%, the relative error of the operating speed is 6.77%, and the maximum relative error of the automatic control of the reel speed is 5.38%. Compared with traditional reel, it reduces header loss by 2.12%. The soybean combine harvester reel speed control system designed in this article can realize automatic adjustment of the reel speed, reduce feeding losses caused by changes in crop height, changes in feed volume, and untimely adjustment of the reel, and can reduce header loss, increased crop yield.

Abstract Background The WD40 domain, one of the most abundant in eukaryotic genomes, is widely involved in plant growth and development, secondary metabolic biosynthesis, and mediating responses to biotic and abiotic stresses. WD40 repeat (WD40) protein has been systematically studied in several model plants but has not been reported in the Capsicum annuum (pepper) genome. Results Herein, 269, 237, and 257 CaWD40 genes were identified in the Zunla, CM334, and Zhangshugang genomes, respectively. CaWD40 sequences from the Zunla genome were selected for subsequent analysis, including chromosomal localization, phylogenetic relationships, sequence characteristics, motif compositions, and expression profiling. CaWD40 proteins were unevenly distributed on 12 chromosomes, encompassing 19 tandem duplicate gene pairs. The 269 CaWD40s were divided into six main branches (A to F) with 17 different types of domain distribution. The CaWD40 gene family exhibited diverse expression patterns, and several genes were specifically expressed in flowers and seeds. Yeast two-hybrid (Y2H) and dual-luciferase assay indicated that CaWD40-91 could interact with CaAN1 and CaDYT1, suggesting its involvement in anthocyanin biosynthesis and male sterility in pepper. Conclusions In summary, we systematically characterized the phylogeny, classification, structure, and expression of the CaWD40 gene family in pepper. Our findings provide a valuable foundation for further functional investigations on WD40 genes in pepper.

Abstract Traditional Chinese medicine (TCM) is considered to be one of the most comprehensive and influential form of traditional medicine. It plays an important role in clinical treatment and adjuvant therapy for cancer. However, the complex composition of TCM presents challenges to the comprehensive and systematic understanding of its antitumor mechanisms, which hinders further development of TCM with antitumor effects. Omics technologies can immensely help in elucidating the mechanism of action of drugs. They utilize high-throughput sequencing and detection techniques to provide deeper insights into biological systems, revealing the intricate mechanisms through which TCM combats tumors. Multi-omics approaches can be used to elucidate the interrelationships among different omics layers by integrating data from various omics disciplines. By analyzing a large amount of data, these approaches further unravel the complex network of mechanisms underlying the antitumor effects of TCM and explain the mutual regulations across different molecular levels. In this study, we presented a comprehensive overview of the recent progress in single-omics and multi-omics research focused on elucidating the mechanisms underlying the antitumor effects of TCM. We discussed the significance of omics technologies in advancing research on the antitumor properties of TCM and also provided novel research perspectives and methodologies for further advancing this research field.

Abstract The average five-year survival rate for esophageal cancer, a common malignant tumor of the digestive system, is barely 20%. The majority of esophageal squamous cell carcinoma (ESCC) patients had already progressed to a locally advanced or even advanced stage at initial diagnosis, making routine surgery ineffective. Chemotherapy and immunotherapy are important neoadjuvant treatments for ESCC, however, it remains unknown how treatment will affect the immunological microenvironment, especially at the spatial level. Here, we presented the TME characters of ESCC from the temporal and spatial dimensions using scRNA-seq and ST, investigated the changes of immune cell clusters in the TME under neoadjuvant chemotherapy and preoperative immunotherapy, and explored the potential mechanisms. It was found that compared with chemotherapy, immunotherapy combined with chemotherapy increased the level of T cell proliferation, partially restored the function of exhausted T cells, induced the expansion of specific exhausted CD8 T cells, increased the production of dendritic cells (DCs), and supported the immune hot microenvironment of the tumor. We also found that CD52 and ID3 have potential as biomarkers of ESCC. Particularly, CD52 may be served as a predictor of the efficacy to screen the advantaged population of different regimens. Through multiple pathways, CAF2 and CAF5’s antigen-presenting role affected the other fibroblast clusters, resulting in malignant transformation. We analyzed the immune microenvironment differences between the two regimens to provide a more thorough description of the ESCC microenvironment profile and serve as a foundation for customized neoadjuvant treatment of ESCC.

The dissolved inorganic nitrogen (DIN) is a critical indicator of the organism growth and ecological balance within aquatic systems. Various water quality determinants influence the temporal variations in DIN concentration, making the identification of key drivers essential for a deeper understanding of nutrient cycling and the formulation of targeted management strategies. However, the impact of hydrological alterations on the key drivers of DIN and its seasonal variability remains unclear. This study investigates seasonal variations in DIN in Hanfeng Lake, a subtropical urban lake located in the Three Gorges Reservoir (TGR) area, over the period from 2021 to 2023. The results indicated that DIN concentration was seasonally significantly different, showing higher values in winter and spring than that in summer and autumn. Random Forest modelling indicated that the temporal variations in DIN concentration were closely related to the notable seasonal fluctuations in key water quality indicators such as the temperature (T), Secchi depth (SD), and concentrations of dissolved phosphorus (DP), dissolved silica (DSi), chlorophyll-a (Chl-a), which were predominantly attributable to hydrological alterations associated with reservoir management and external pollutant inputs from agricultural fertilization. Partial Least Squares Path Modeling confirmed that physical indicators, like T and SD, were the primary drivers of variations in DIN concentration, while DP, DSi, and Chl-a also had significant effects. Based on these insights, the study recommends to minimize the agricultural fertilizer application in spring and summer for decreasing external inputs, and optimize reservoir operation strategies in autumn and winter for controlling hydrological alterations.

BackgroundTeaching severe pelvic trauma poses a significant challenge in orthopedic surgery education due to the necessity of both clinical reasoning and procedural operational skills for mastery. Traditional methods of instruction, including theoretical teaching and mannequin practice, face limitations due to the complexity, the unpredictability of treatment scenarios, the scarcity of typical cases, and the abstract nature of traditional teaching, all of which impede students’ knowledge acquisition. ObjectiveThis study aims to introduce a novel experimental teaching methodology for severe pelvic trauma, integrating virtual reality (VR) technology as a potent adjunct to existing teaching practices. It evaluates the acceptability, perceived ease of use, and perceived usefulness among users and investigates its impact on knowledge, skills, and confidence in managing severe pelvic trauma before and after engaging with the software. MethodsA self-designed questionnaire was distributed to 40 students, and qualitative interviews were conducted with 10 teachers to assess the applicability and acceptability. A 1-group pretest-posttest design was used to evaluate learning outcomes across various domains, including diagnosis and treatment, preliminary diagnosis, disease treatment sequencing, emergency management of hemorrhagic shock, and external fixation of pelvic fractures. ResultsA total of 40 students underwent training, with 95% (n=38) affirming that the software effectively simulated real-patient scenarios. All participants (n=40, 100%) reported that completing the simulation necessitated making the same decisions as doctors in real life and found the VR simulation interesting and useful. Teacher interviews revealed that 90% (9/10) recognized the VR simulation’s ability to replicate complex clinical cases, resulting in enhanced training effectiveness. Abstract Intratumour heterogeneity significantly hinders the efficacy of anticancer therapies. Compared with drug perturbation experiments, which yield pharmacological data at the bulk cell level, single‐cell RNA sequencing (scRNA‐seq) technology provides a means to capture molecular heterogeneity at single‐cell resolution. Here, scPharm is introduced, a computational framework that integrates pharmacological profiles with scRNA‐seq data to identify pharmacological subpopulations of cells within a tumour and prioritize tailored drugs. scPharm uses the normalized enrichment scores (NESs) determined from gene set enrichment analysis to assess the distribution of cell identity genes in drug response‐determined gene lists. Based on the strong correlation between the NES and drug response at single‐cell resolution, scPharm successfully identified the sensitive subpopulations in ER‐positive and HER2‐positive human breast cancer tissues, revealed dynamic changes in the resistant subpopulation of human PC9 cells treated with erlotinib, and expanded its ability to a mouse model. Its superior performance and computational efficiency are confirmed through comparative evaluations with other single‐cell prediction tools. Additionally, scPharm predicted combination drug strategies by gauging compensation or booster effects between drugs and evaluated drug toxicity in healthy cells in the tumour microenvironment. Overall, scPharm provides a novel approach for precision medicine in cancers by revealing therapeutic heterogeneity at single‐cell resolution.

The rapid growth of renewable energy and electric vehicles (EVs) presents new development opportunities for power systems and energy storage devices. This paper presents a novel integrated Green Building Energy System (GBES) by integrating photovoltaic-energy storage electric vehicle charging station (PV-ES EVCS) and adjacent buildings into a unified system. In this system, the building load is treated as an uncontrollable load and primarily utilized to facilitate the consumption of surplus photovoltaic (PV) power generated by EVCS. First, a strategy for determining the maximum value of the energy storage system (ESS) capacity is presented. Subsequently, to coordinate the charging and discharging plans of ESS, and EVs, a bi-objective optimization model was established focusing on GBES power purchase costs and the load peak-valley difference. The proposed GBES efficiently utilizes the integrated energy system comprising charging stations and adjacent buildings, maximizing the use of photovoltaic energy and external power grids during low-cost periods. In experiments, we compare the proposed optimized charging strategy with the unordered charging case, the simulation results demonstrate that the proposed method for coordinating ESS and EVs charging can respectively reduce the cost of purchased power by 33.2% and the peak-to-valley difference in load by 47.6%.

Abstract Amorphous FePO4 (AFP) is a promising cathode material for lithium‐ion and sodium‐ion batteries (LIBs & SIBs) due to its stability, high theoretical capacity, and cost‐effective processing. However, challenges such as low electronic conductivity and volumetric changes seriously hinder its practical application. To overcome these hurdles, core‐shell structure synthesis emerges as a useful solution. In this work, we for the first time made this comprehensive review on the progresses of core‐shell amorphous FePO4 (CS‐AFP). This review summarizes 1) various synthesis methods such as template method, microemulsion method, and other methods, 2) characterization techniques, and 3) their involvement in improving electrochemical performance in LIBs and SIBs. In terms of further understanding the underlying mechanisms of advancing electrochemical performance of CS‐AFP, the future perspective on two main aspects were insighted: (i) in situ characterization and (ii) novel designs of materials and structure for CS‐AFP.
