Abstract

Flame retardants mitigate the threat of fire from inherently flammable wood and wood products responsible for sustaining a high standard of living. Wood is one of the aesthetically pleasing, highly sustainable as well as eco-friendly materials. It is not only an integral part of structures, but also the main source of home appliances, furniture, industrial applications, and offices all over the world. The coating provides protection to wood materials against outdoor weathering, photochemical degradation, and fire. Therefore, to minimize the degradation of wood for the increasing long life and survival ability, various coatings were synthesized and applied to enhance their performance for the wood application. This review provides a historical overview that leads to the structural properties of wood and the most promising surface treatments that will help pave the way for developing more effective and non-intrusive flame retardants in the future. Abstract

A novel melamine-phosphate tri-functional acrylate (MPTO) was successfully synthesized via cyclization reaction of hexamethylolmelamine (HMM) with phosphorous oxychloride (POCl3) followed by addition reaction of hydroxyethylmethacrylate (HEMA). The molecular structure of MPTO was identified by FTIR and 1H-NMR, 13C-NMR, and GC-MS spectra. The synthesized MPTO oligomer was impregnated with polyurethane acrylate to make various formulations and UV cured on wood and galvanized steel panels. The results of UV cured films from differential scanning calorimeter (DSC) and thermo-gravimetric analysis (TGA) exhibited increase in thermal stability and highest char yield of 18.4% at 800°C. The diffused diffraction peak appearing around 2ϴ = 16–22° corresponds to an amorphous structure which leads to irregular arrangement with formation of strong cross-linking polymer network. Furthermore, all coating films show prominent flame retardancy with UL-94 V-0 rating and maximum limiting index (LOI) values of 34.8%. The polyurethane acrylate (PU) coatings cured with MPTO that exhibited excellent mechanical properties were estimated using various tests such as adhesion, pencil hardness, solvent resistance, flexibility, and corrosion test. The coating performance revealed that MPTO can improve the mechanical, thermal, and flame retardant properties because of its unique structure identity of melamine-phosphate moiety and long aliphatic segment of an acrylate ester. Abstract

Glioblastoma (GBM) is the most aggressive form of adult brain tumors with a poor median survival of less than 15 months. The alkylating agent temozolomide is used for chemotherapy for GBM. Regardless of the rigorous therapeutic regime, almost all cases develop resistance and recur. The mechanisms of the inherent and acquired resistance are less understood and demand in-depth research. We performed a pooled and bar-coded shRNA screen (5045 shRNAs) in a glioma cell line to identify mediators of temozolomide resistance in GBM. The data was analyzed by the HitSelect algorithm, and the top ranking genes imparting resistance (n=257) and sensitivity (n=256) were further subjected to gene set enrichment analysis (GSEA), Gene Ontology (GO) annotation and survival analysis in The Cancer Genome Atlas (TCGA) GBM cohort. An enrichment of translation and protein synthesis-related pathways was found in sensitivity genes, while enrichment of proliferation, metabolic processes, cytokine-cytokine receptor interaction and response to stress was found in resistance genes. Suppressor of Lin-12-Like Protein 1 (SEL1L) was identified as one of the top ranking genes in our screen that caused resistance to temozolomide. SEL1L is a component gene of the endoplasmic-reticulum-associated protein degradation (ERAD). It is known to play an important role in clearing misfolded proteins from cells, thus maintaining proteostasis. SEL1L was found to be a poor prognostic marker in the TCGA GBM cohort treated with temozolomide, as identified by univariate and Kaplan- Meier analysis. In vitro SEL1L knockdown resulted in a significant reduction of proliferation and in an increased sensitivity towards temozolomide. Upon temozolomide treatment of cells in the SEL1L knockdown condition, an increase in apoptosis was observed as measured by Annexin V staining. In vivo tumor showed increased SEL1L protein levels as compared to control brain and mice tumor that responded to temozolomide. We subjected to GSEA, (i) the genes that exhibited survival significance in the TCGA GBM cohort with temozolomide, as well as (ii) the genes that showed differential expression in the SEL1L low and high expression groups. Interestingly, both these analyses identified significant enrichment of overlapping pathways like TNFA mediated NF-kappaB signaling, EMT, IL6 STAT3 signaling, G2M checkpoint and E2F target genes, further highlighting the role of SEL1L in temozolomide resistance. This underscores that SEL1L is an important mediator for temozolomide resistance and is a poor prognosticator in GBM. SEL1L high expression correlates with various resistance-related signaling pathways. Our study emphasizes on the interplay between these pathways and the ERAD machinery in maintaining proteostasis in response to chemotherapeutic stress.

Abstract

Objective: A new precise, accurate, sensitive and robust reverse phase high performance liquid chromatography (RP-HPLC) method was developed for Cefoperazone and Tazobactam in marketed preparation. Method: The Chromatographic separation was achieved on Thermo BDS Hypersil C18 column (250 × 4.6 mm i.d.5 μ) at ambient temperature. A Binary mobile phase consisting of 10 mM potassium dihydrogen phosphate buffer, pH 2.8 and Acetonitrile (80:20, v/v) was delivered through a column at a flow rate of 1 ml/min. Estimation was performed at a coveted wavelength of 254 nm. Result: The method was linear over the concentration range of 40-200 μg/ml and correlation coefficient (r2) was found to be 0.9997 for Cefoperazone (CEFO) and 5-25 μg/ml for Tazobactam (TAZO), correlation coefficient was found to be 0.9996. The retention times of the CEFO and TAZO were found to be 3.667 min and 4.737 min. respectively. The percentage content for Cefoperazone 98.94±1.0 and of Tazobactam was 99.69±0.96 in the marketed formulation. The low value of % Relative Standard Deviation (%RSD) was 0.224 and 0.018 for Cefoperazone and Tazobactam, respectively, indicates the reproducibility of this method. The low value of Limit of detection (LOD) and Limit of quantitation (LOQ) suggests the sensitiveness of method. Conclusion: It can be concluded from the outcomes that the proposed RP-HPLC method was found to be simple, basic, accurate, robust and precise for the normal analysis of Cefoperazone and Tazobactam in bulk and sterile dried injection dosage forms. This method was validated as per ICH guidelines.

Abstract

Background: Pyridinium and imidazolium-based compounds show diverse applications with basic skeleton designs in order to achieve improved optical and thermal behavior. Aim: The aim of the study was to design and develop a greener, efficient protocol towards newer pyridinium and imidazolium compounds, and investigate optical, solvatochromic, thermal, and theoretical properties. Objectives: The purpose is to study the optical properties of pyridinium and imidazolium compounds, for which we illustrate the solvent polarity effect on the absorption and emission behavior as a function of orientation polarizability and ET(30) solvent parameters. The study focuses on thermal stability and computes the molecular orbital orientation and HOMO- LUMO energies using theoretical simulation by the DFT approach. Methods: The structures were confirmed by FT-IR, Mass, 1H NMR and 13C NMR, and optical properties were investigated using a UV-Visible spectrophotometer and fluorometer. The thermal behavior was investigated using thermal gravimetric analysis, and molecular orbital orientation and energies were determined using GAUSSIAN 16 software. Results: The newer compounds with good thermal stability and optical behavior have been synthesized and characterized. The study interprets the intermolecular electron transfer amongst the molecules and the effect of solvents on their excitation and emission properties. The experimental and theoretical study illustrates the optical, thermal, and electronic properties of both compounds. Conclusion: The present work describes the solvatochromic optical behavior of pyridinium bromide and imidazolium bromide synthesized by a microwave-assisted, greener and efficient strategy. The solvatochromic study interprets the presence of non-specific solutesolvent interactions. The photophysical, thermal, and DFT study revealed that both pyridinium and imidazolium compounds are used for optoelectronic applications. Moreover, the work could be helpful to researchers for developing new skeletons for optoelectronic applications.

Abstract

• Design of solar air cavity-receiver with RPC absorbers is presented. • 5 kW prototype was experimentally tested under concentrated thermal radiation. • Parameters: RPC material, pore size, concentration ratio and air mass flow rate. • SiSiC 10 PPI achieved peak efficiency of 0.69 at 1133 °C air outlet temperature. Concentrated solar energy can be used as the source of high-temperature heat for industrial processes, but the challenge is to design a solar receiver that can effect such a thermal conversion efficiently. This study reports on the engineering design and experimental testing of a 5 kW solar cavity-receiver containing a reticulated porous ceramic (RPC) structure that can absorb high-flux radiation volumetrically and heat up, by convection, an air flow serving as the heat transfer fluid. The thermal performance, characterized by the thermal efficiency and the air outlet temperature, was determined experimentally for four parameters, namely: RPC material (silicon-infused silicon carbide or SiSiC, alumina, and ceria), mean pore size (range 0.8–2.5 mm, corresponding to 10–30 pores per inch or PPI, at 0.90 porosity), solar concentration ratio (range 1965–3900 suns over a 4 cm-diameter cavity aperture, supplied by a high-flux solar simulator), and air mass flow rate (range 2–10 kg/h). Thermal efficiencies between 0.22 and 0.69 were obtained at steady-state air outlet temperatures ranging from 1160 to 450 °C. Larger pores enhance heat transfer while variable porosity across the RPC can reduce temperature gradients and potentially contribute to the design optimization. The highest efficiency of 0.69 was achieved by the SiSiC 10 PPI cavity at an air outlet temperature of 1133 °C and air mass flow rate of 9.9 kg/h. The solar receiver design proved to deliver a high-temperature air flow (>1000 °C) with a reasonably high thermal efficiency (>0.65). [ABSTRACT FROM AUTHOR]

Abstract

Schwannomatosis (SWNTS) is a genetic cancer predisposition syndrome that manifests as multiple and often painful neuronal tumors called schwannomas (SWNs). While germline mutations in SMARCB1 or LZTR1, plus somatic mutations in NF2 and loss of heterozygosity in chromosome 22q have been identified in a subset of patients, little is known about the epigenomic and genomic alterations that drive SWNTS-related SWNs (SWNTS-SWNs) in a majority of the cases. We performed multiplatform genomic analysis and established the molecular signature of SWNTS-SWNs. We show that SWNTS-SWNs harbor distinct genomic features relative to the histologically identical non-syndromic sporadic SWNs (NS-SWNS). We demonstrate the existence of four distinct DNA methylation subgroups of SWNTS-SWNs that are associated with specific transcriptional programs and tumor location. We show several novel recurrent non-22q deletions and structural rearrangements. We detected the SH3PXD2A-HTRA1 gene fusion in SWNTS-SWNs, with predominance in LZTR1-mutant tumors. In addition, we identified specific genetic, epigenetic, and actionable transcriptional programs associated with painful SWNTS-SWNs including PIGF, VEGF, MEK, and MTOR pathways, which may be harnessed for management of this syndrome. Abstract

Glioblastoma (GBM) is the most aggressive type of glioma. Temozolomide (TMZ) is currently the drug of choice used for post-operative chemotherapy of GBM. However, the presence of intrinsic and acquired resistance hinders the success of chemotherapy. To understand the TMZ resistant mechanisms in glioma, we investigated the alterations in cellular signaling pathways by performing transcriptome analysis of TMZ treated glioma cells. Gene Set Enrichment Analysis (GSEA) indicated a significant enrichment of Wnt/β-catenin signaling besides many other pathways in TMZ treated cells. Further, we demonstrate that TMZ treatment increased the activity from TOPflash reporter, (a Wnt responsive reporter), enhanced the levels of pGSK-3β (S9) and reduced the levels of p-β-catenin (S33/37/T41) with a concomitant increase in transcript and protein levels of Wnt targets in a concentration and time-dependent manner. While TMZ treated cells did not show alteration in any of the Wnt ligands, PI3K inhibitor (LY294002) treatment repressed Akt activation and abolished the TMZ–mediated induction of Wnt/β-catenin pathway. In addition, we show that Wnt/β-catenin signaling activation by TMZ is independent of ATM/Chk2 pathway. Further, we also demonstrate the activation of mTOR pathway after TMZ treatment. Thus, our results demonstrate that activation of Wnt/β-catenin pathway involves an ATM/Chk2- independent PI3K/Akt/GSK-3 cascade in TMZ treated cells and further provides mechanistic basis for the chemoresistance of glioma to TMZ.
