201. Controlled Release of Simvastatin from Biomimetic ?-TCP Drug Delivery System

Abstract

Simvastatin have been shown to induce bone formation and there is currently a urgent need to develop an appropriate delivery system to sustain the release of the drug to increase therapeutic efficacy whilst reducing side effects. In this study, a novel drug delivery system for simvastatin by means of hydrothermally converting marine exoskeletons to biocompatible beta-tricalcium phosphate was investigated. Furthermore, the release of simvastatin was controlled by the addition of an outer apatite coating layer. The samples were characterized by x-ray diffraction analysis, fourier transform infrared spectroscopy, scanning electron microscopy and mass spectroscopy confirming the conversion process. The in-vitro dissolution of key chemical compositional elements and the release of simvastatin were measured in simulated body fluid solution showing controlled release with reduction of approximately 25% compared with un-coated samples. This study shows the potential applications of marine structures as a drug delivery system for simvastatin.

207. Bone regeneration of calvarial defect using marine calcareous-derived beta-tricalcium phosphate macrospheres

Abstract

The aim of this study was to examine the bone regeneration properties of beta-tricalcium phosphate hydrothermally converted from foraminifera carbonate exoskeleton in the repair of rat calvarial defect. These natural materials possess unique interconnected porous network with uniform pore size distribution, which can be potentially advantageous. In total, 20 adult male Wistar rats received full-thickness calvarial defect with a diameter of 5 mm. The rate of newly formed bone was measured radiologically by X-ray and micro-computed tomography and by histologic examination. After 2 weeks, the beta-tricalcium phosphate group exhibited full closure of the defect site, while control group remained unrestored at the end of the 6-week experimentation. It was observed that the newly regenerated bone thickened over the course of the experiment in the beta-tricalcium phosphate group. No soft tissue reaction was observed around the beta-tricalcium phosphate implant and the rats remained healthy. These results showed that repair of the calvarial defect can be achieved by biomimetic beta-tricalcium phosphate macrospheres, which hold potential for application as bone grafts for bone augmentation surgeries.

Abstract

Relata-se, pela primeira vez no Brasil, um caso de dermatite decorrente de infecção por micobactéria atípica do complexo Mycobacterium fortuitum-peregrinum, em espécie felina, sem raça definida, fêmea, com cinco anos de idade. Há oito meses, evoluía com lesões maculares equimóticas, nodulares, erosadas, ulceradas, acompanhadas de fístulas exsudativas, com intenso prurido e algia. Evidenciou-se a presença de micobactéria do complexo Mycobacterium fortuitum-peregrinum (grupo IV de Runyon ) identificada após evidenciação histopatológica, cultivo bacteriano e por testes bioquímicos. Após dois meses de terapia sistêmica com enrofloxacina (5mg/kg/Bid/VO) e tópica (triclosan e rifamicina) houve involução das lesões, com efeitos colaterais discretos. A case of dermatitis caused by atypical mycobateria of Mycobacterium fortuitum peregrinum complex was observed for the first time in Brazil. A five-year-old female mixed breed cat had a dermatitis process which had started eight months before. Lesions were characterized by macules, nodules, erosions, ulcers and exsudative fistulas, with intensive pruritus and pain. It had failed to respond to clinical treatment and surgical excision. The diagnosis was based on history, physical and dermatologic examination and complementary tests (cytologic, bacterioscopic, bacterial culture, histopathological, hemathological, ultrasonographic, radiographic and electrocardiographic). The presence of mycobacteria of Mycobacterium fortuitum-peregrinum complex (group IV of Runyon) was observed and identified after histopathological evidence, bacterial culture and biochemical tests. After two months of systemic therapy with enrofloxacin (5mg/kg, orally/twice a day) and topic treatment with triclosan and rifamicin, rapid improvement of the clinical appearance of the lesions were observed with low incidence of side-effects.

Abstract

In recent years, active research on explosives in exploring its effective use in the field of engineering is widely carried out. One way of instant initiation of explosive is by means of a metal wire explosion. The wire explosion technique generates plane shock waves produced by exploding parallel metal wire rows using a high voltage impulsive current. This method has been performed as mass shock compaction processing technology, such as shock synthesis of industrial diamond. The present study was undertaken to investigate the behavior of copper pipe by the detonation of PETN powder explosive initiated by metal wire explosion. This was carried out with the motivation to know the effect of PETN on the structure, which was detonated by metal wire explosion, and the detonation state of PETN. A copper wire exploded using metal wire explosion technique, was used to initiate the PETN filled in the copper pipe. The expansion process of the copper pipe by the detonation of PETN was evaluated by the optical observation using high-speed camera and the numerical analysis using LS-DYNA3D. The results on the expansion velocity of copper pipes showed a variation depending on the initiation and the amount of PETN used for the expansion.

Abstract

Descreve-se, pela primeira vez no Brasil, um caso de criptococose canina com acometimento oftalmo e dermatopático, com diagnóstico intra-vitam, em animal da raça Pastor Alemão, fêmea, com 24 meses de vida, criado em São Paulo, em contacto com outros cinco cães assintomáticos, que havia se infectado pelo contato com dejetos de pombos (Columba livia). Evoluia há 90 dias, com quadro tegumentar e ósseo, sintomas e lesões características, sendo, ainda, detectada forma assintomática de coriorretinite, de início unilateral. O diagnóstico foi estabelecido pelos dados da anamnese, dos exames físico, dermatológico e complementares (radiográfico, cultivo micológico, histopatologia de pele) tendo-se evidenciado e isolado cepa de Criptococcus neoformans var. neoformans. Após nove meses de terapia com itraconazol (9mg/kg/SID/VO) houve involução total do quadro, sem qualquer efeito adverso à droga. A case of cryptococcosis with ocular and cutaneous involvement is reported by the first time in Brazil in a dog. A two-year-old female German Shepherd living in a hold house with other five healthy dogs was infected through the contact with pigeon (Columba livia) feces. The illness started 90 days previously with cutaneous and bone involvement resulting in characteristic clinical signs and lesions, in addition to a initially unilateral asymptomatic chorioretinitis. The diagnosis was established based on anamnesis, physical and dermatologic examinations and complementary exams (radiographs, skin biopsy and histopathology) and the isolation of a Cryptococcus neoformans var. neoformans strain was accomplished. There was a complete resolution of the disease after nine months of therapy using itraconazole (9mg/kg, q24h, PO) and no side effect was observed.

Abstract

Foraminifera carbonate exoskeleton was hydrothermally converted to biocompatible and biodegradable zinc-tricalcium phosphate (Zn-TCP) as an alternative biomimetic material for bone fracture repair. Zn-TCP samples implanted in a rat tibial defect model for eight weeks were compared with unfilled defect and beta-tricalcium phosphate showing accelerated bone regeneration compared with the control groups, with statistically significant bone mineral density and bone mineral content growth. CT images of the defect showed restoration of cancellous bone in Zn-TCP and only minimal growth in control group. Histological slices reveal bone in-growth within the pores and porous chamber of the material detailing good bone-material integration with the presence of blood vessels. These results exhibit the future potential of biomimetic Zn-TCP as bone grafts for bone fracture repair. © 2013 by the authors; licensee MDPI.

Abstract

The HERschel Inventory of The Agents of Galaxy Evolution (HERITAGE) of the Magellanic Clouds will use dust emission to investigate the life cycle of matter in both the Large and Small Magellanic Clouds (LMC and SMC). Using the Herschel Space Observatory's PACS and SPIRE photometry cameras, we imaged a 2x8 square degree strip through the LMC, at a position angle of ~22.5 degrees as part of the science demonstration phase of the Herschel mission. We present the data in all 5 Herschel bands: PACS 100 and 160 {\mu}m and SPIRE 250, 350 and 500 {\mu}m. We present two dust models that both adequately fit the spectral energy distribution for the entire strip and both reveal that the SPIRE 500 {\mu}m emission is in excess of the models by 6 to 17%. The SPIRE emission follows the distribution of the dust mass, which is derived from the model. The PAH-to-dust mass (f_PAH) image of the strip reveals a possible enhancement in the LMC bar in agreement with previous work. We compare the gas mass distribution derived from the HI 21 cm and CO J=1-0 line emission maps to the dust mass map from the models and derive gas-to-dust mass ratios (GDRs). The dust model, which uses the standard graphite and silicate optical properties for Galactic dust, has a very low GDR = 65(+15,-18) making it an unrealistic dust model for the LMC. Our second dust model, which uses amorphous carbon instead of graphite, has a flatter emissivity index in the submillimeter and results in a GDR = 287(+25,-42) that is more consistent with a GDR inferred from extinction., Comment: To be published in Astronomy & Astrophysics, Herschel First Results Issue

Abstract

The time to return to sport from acute hamstring strain injuries is associated with several functional and structural impairments. However, not all previous studies assessed the preinjury level before acute hamstring strain injuries directly. The purpose of this study was to examine the associations of the time to return to performance following acute hamstring strain injuries with deficits in running biomechanics, hamstring function and structure in collegiate sprinters by a prospective study. Using a prospective cohort design, 72 participants were recruited from a collegiate track and field team. At the preinjury assessment, a 60-m running-specific test, passive straight leg raise test and isometric knee flexion strength test were assessed at the beginning of the competitive season for three consecutive years (2017–2019). Afterwards, postinjury examinations were performed only in sprinters with acute hamstring strain injuries. Twelve sprinters strained their hamstring muscle (incidence rate of hamstring strain injuries: 16.7%); the majority (n = 10) were classified as grades 0–2. The running speed deficit of the running-specific test was associated with the time to return to performance as well as the passive straight leg raise test deficit. In the running-specific test, lower-limb kinetic deficits were more strongly associated with the time to return to performance compared to lower-limb kinematic deficits. A running-specific test may be considered one of the most convenient and valid tests for assessing rehabilitation progress after acute hamstring strain injuries. • Incidence rate of hamstring strain injuries was 16.7% during competition season. • Majority (83.3%) of injuries was grades 0–2 and the other (16.7%) was grade 3. • Running speed and kinetic deficits relate to time to return to performance. • Altered hamstring strength and flexibility relate to rehabilitation process. • Magnetic resonance image measures may not be related to rehabilitation process. Abstract

We present development of the collimated bipolar jets from the symbiotic prototype Z And that appeared and disappeared during its 2006 outburst. In 2006 July Z And reached its historical maximum at U ~ 8.0. During this period, rapid photometric variations with Dm ~ 0.06 mag on the timescale of hours developed. Simultaneously, high-velocity satellite components appeared on both sides of the H-alpha and H-beta emission line profiles. They were launched asymmetrically with the red/blue velocity ratio of 1.2 - 1.3. From about mid-August they became symmetric. Their spectral properties indicated ejection of bipolar jets collimated within an average opening angle of 6.1 degrees. We estimated average outflow rate via jets to dM(jet)/dt ~ 2xE10-6(R(jet)/1AU)**(1/2) M(Sun)/year, during their August - September maximum, which corresponds to the emitting mass in jets, M(jet, emitting) ~ 6xE-10(Rjet)/1AU)^{3/2} M(Sun). During their lifetime, the jets released the total mass of M(jet, total) approx 7.4x1E-7 M(Sun). Abstract

Background: Signalling of angiotensin II via angiotensin II type 1 receptor (AT1) promotes cardiac and renal fibrosis, but its role in lung fibrosis is little understood. Using a rat bleomycin (BLM) induced model of pulmonary fibrosis, we examined the expression of AT1 in the lung and the effect of an AT1 antagonist on pulmonary fibrosis.Methods: Adult male Sprague-Dawley rats were given 0.3 mg/kg BLM intratracheally. Two days earlier they had received 10 mg/kg/day of the AT1 antagonist candesartan cilexetil mixed in the drinking water. AT1 expression in the lungs was examined by immunohistochemistry and immunoblot methods. The effect of the AT1 antagonist on pulmonary fibrosis was studied by analysis of bronchoalveolar lavage (BAL) fluid, histopathology, and hydroxyproline assay.Results: Immunohistochemical studies showed overexpression of AT1 in inflammatory immune cells, alveolar type II cells, and fibroblasts. A quantitative assay for AT1 showed that AT1 expression was significantly upregulated in cells from BAL fluid after day 3 and in the lung homogenates after day 21. Candesartan cilexetil significantly inhibited the increase in total protein and albumin, as well as the increase in total cells and neutrophils in BAL fluid. On day 21 candesartan cilexetil also ameliorated morphological changes and an increased amount of hydroxyproline in lung homogenates. In addition, BLM increased the expression of transforming growth factor (TGF)-beta1 in BAL fluid on day 7; this increase was significantly reduced by candesartan cilexetil.Conclusion: AT1 expression is upregulated in fibrotic lungs. Angiotensin II promotes lung fibrosis via AT1 and, presumably, in part via TGF-beta1. [ABSTRACT FROM AUTHOR]
