216. Perilla oil prevents the excessive growth of visceral adipose tissue in rats by down-regulating adipocyte differentiation

Abstract

We examined the effect of dietary oils with different fatty acid compositions on the growth of visceral adipose tissue in rats. Rats were fed for 4 mo starting at weaning a basal diet containing (12 g/100 g diet) perilla oil rich in (n-3) polyunsaturated fatty acids (PUFA), safflower oil rich in (n-6) PUFA, olive oil rich in monounsaturated fatty acid, or beef tallow rich in saturated fatty acids. The amount of food consumed and body weight gain did not differ among the four dietary groups. The weight of the epididymal fat pad and the serum triglyceride concentration in perilla oil-fed rats were significantly lower (P < 0.05) than those of olive oil- and beef tallow-fed groups. The product of [(volume of individual adipocytes) x (number of adipocytes in epididymal fat pad)], which presumably represents total adipocyte volume in the fat pad, was significantly lower (P < 0.05) in perilla oil-fed rats than in beef tallow- and olive oil-fed groups. Expression of the late genes of adipocyte differentiation, peroxisome proliferator-activated receptor [Alpha], adipocyte P2 and adipsin, was significantly (P < 0.05) down-regulated in epididymal fat tissue of rats that had been fed perilla oil rather than beef tallow or olive oil, whereas expression of the early gene, lipoprotein lipase, was not significantly affected. Greater levels (P < 0.05) of (n-3) PUFA in the membrane phospholipid fraction of the fat tissue were observed in perilla oil-fed rats than in the other dietary groups. These results suggest that perilla oil or (n-3) PUFA prevents excessive growth of adipose tissue in rats at least in part by suppressing the late phase of adipocyte differentiation. Abstract

Polyprenoic acid appears to prevent a second primary tumor in patients who have received treatment for liver cancer. Polyprenoic acid is a synthetic retinoid that is chemically related to vitamin A. Of 89 patients with liver cancer that had been treated surgically or with injections of ethanol, 44 took polyprenoic acid and 45 took a placebo. They were followed until they developed a second tumor, for a median of 38 months. During the study, 34 patients (38%) developed a second tumor. However, only 27% of those taking polyprenoic acid developed a second tumor compared to 49% in the placebo group. Polyprenoic acid did not significantly reduce the rate of tumor recurrence, but it substantially reduced the rate of secondary primary tumors. Only 16% of those taking polyprenoic acid developed a second primary tumor, compared to 44% of those taking a placebo.

225. Oral supplementation with branched-chain amino acids improves transthyretin turnover in rats with carbon tetrachloride-induced liver cirrhosis

Abstract

The hypothesis that dietary branched-chain amino acid (BCAA) supplementation improves the impaired protein turnover in male Donryu rats with carbon tetrachloride-induced liver cirrhosis was tested. We supplemented cirrhotic rats orally for 2 wk with BCAA solution [26.67 mg BCAA/(100 g body weight[center dot]d)], a conventional amino acid mixture [4.25 mg BCAA/(100 g body weight[center dot]d)] or saline and fed these three groups the AIN76 basal diet to have similar intakes of total energy and total nitrogen. Normal rats without liver cirrhosis were fed the basal diet similar to the above (noncirrhotic controls). After supplementation, rats were fed intravenous transthyretin (thyroxine-binding prealbumin) doubly labeled with 125I-tyramine-cellobiose and 131I. Kinetic indices including production rate of transthyretin were analyzed from plasma transthyretin disappearance curves. Tissue sites of transthyretin degradation were assayed using a trapped ligand technique by measuring 125I-tyramine-cellobiose levels. The production rate of transthyretin was significantly lower in cirrhotic rats supplemented with saline (mean 25.46 x [10.sup.-3] [center dot] [h.sup.-1]) compared with noncirrhotic controls (45.08 x [10.sup.-3] [center dot] [h.sup.-1]) (P < 0.05). This was corrected by supplementing cirrhotic rats with BCAA (37.05 x [10.sup.-3] [center dot] [h.sup.-1], P < 0.05) but not with conventional amino acid mixture (22.49 x [10.sup.-3] [center dot] [h.sup.-1]). The accelerated degradation of transthyretin in muscles of cirrhotic rats was improved by BCAA (P < 0.05). In conclusion, dietary supplementation with BCAA improves the impaired transthyretin turnover in rats with liver cirrhosis. J. Nutr. 126: 1412-1420, 1996.
