Abstract

The physiological role of C-type natriuretic peptide (CNP) was examined. CNP is a 22-amino acid peptide that is structurally related to atrial natriuretic peptide (ANP) and brain natriuretic peptid and has been isolated from various mammalian tissues. Comparisons of its effects on blood vessels with those of ANP suggest that CNP is a vascular relaxing factor and thus has a role in cardiovascular tone distinct from that of ANP.

Abstract

Endothelial nitric oxide synthase (eNOS) is regulated both by caveolin-1 and 17[Beta]-estradiol ([E.sub.2]). Temporal relationships between effects of estrogen on caveolin-1 and nitric oxide (NO) are not known. Therefore, this study was designed to determine whether estrogen regulates caveolin-1 and, if so, whether such regulation corresponds to changes in nitrite/nitrate ([NO.sub.x]) production. Bovine aortic endothelial cells (BAECs) were cultured in the absence and presence of 17[Beta]-estradiol or 17[Alpha]-estradiol ([10.sup.-8] and [10.sup.-10] M) for 12, 24, and 48 h. eNOS protein expression and [NO.sub.x] production increased significantly after 24 h but not after 12-h treatment with 17[Beta]- and not 17[Alpha]-estradiol. Both mRNA and protein for caveolin-1 were increased significantly only after 48-h treatment with [E.sub.2], but eNOS protein and [NO.sub.x] production were decreased compared with cells treated for 24 h. These increases in caveolin-1 were inhibited by the estrogen receptor antagonist ICI-182,780 ([10.sup.-6] M). Results of this study suggest that [E.sub.2] stimulates caveolin-1 transcription and translation through estrogen receptor-mediated mechanisms. Am J Physiol Heart Circ Physiol 279: H1913--H1921, 2000.--Nicotine patches are available as an over-the-counter medication for aid in smoking cessation. This study was designed to determine how nicotine patch therapy over time and dose ranges used in smoking cessation programs in humans affects endothelium-dependent relaxations. Dogs were treated with nicotine patches (11, 22, or 44 mg/day) for 2 and 5 wk. Circulating nicotine and oxidized products of nitric oxide (NOx) were measured. Coronary arteries were prepared for measurement of isometric force and aortic endothelial cells were prepared for measurement of mRNA or NO synthase (NOS) activity. Circulating nicotine increased with increasing concentrations of nicotine patches. After 5 wk of treatment with 22 mg/day patches, circulating NOx was reduced but NOS activity was increased. NOS mRNA was similar among groups. Only after 5 wk of treatment with 22 mg/day patches were endothelium-dependent relaxations reduced to [[Alpha].sub.2]-adrenergic agonists, ADP, and the calcium ionophore A-23187. These results suggest a time and biphasic dose effect of nicotine treatment on endothelium-dependent responses that may be related to bioavailability of NO. This complex relationship of duration and dose of nicotine treatment may explain, in part, discrepancies in effects of nicotine on endothelium-dependent responses. canine; coronary artery; nitric oxide; nitric oxide synthase

Physiol. 46): H1618-H1621, 1999.--Nitric oxide (NO) is an important endothelium-derived relaxing factor that functions via activation of soluble guanylyl cyclase and cGMP generation in vascular smooth muscle. Recently, studies have described the synthesis and secretion of C-type natriuretic peptide (CNP) from endothelial cells. This peptide also mediates relaxation via cGMP but through activation of particulate guanylyl cyclase. We tested the hypothesis that endothelium-dependent relaxations to acetylcholine or bradykinin in isolated canine coronary arteries involve both releases of NO and CNP. Rings of canine coronary arteries were incubated with either inhibitors of NO production ([N.sup.G]-monomethyl-L-arginine, L-NMMA) or the natriuretic peptide receptor antagonist HS-142-1. CNP caused concentration-dependent relaxations of rings with and without endothelium. These relaxations were attenuated by HS-142-1. Relaxations to acetylcholine and bradykinin were attenuated by L-NMMA alone but not attenuated by HS-142-1 alone. Coinhibition with L-NMMA and HS-142-1 significantly inhibited acetylcholine- and bradykinin-induced relaxation to a magnitude greater than either inhibitor alone. In summary, a novel interaction between the NO and the natriuretic peptide system is demonstrated by increased attenuation of endothelium-dependent relaxations to acetylcholine and bradykinin when both NO synthase and natriuretic peptide receptors are inhibited. These investigations support the concept of release of multiple endothelium-derived factors in response to acetylcholine- and bradykinin-receptor stimulation in endothelial cells, which may include CNP, as well as NO. Abstract

In June 2015, the National Institutes of Health (NIH) released a Guide notice (NOT-OD-15-102) that highlighted the expectation of the NIH that the possible role of sex as a biologic variable be factored into research design, analyses, and reporting of vertebrate animal and human studies. Anticipating these guidelines, the NIH Office of Research on Women's Health, in October 2014, convened key stakeholders to discuss methods and techniques for integrating sex as a biologic variable in preclinical research. The workshop focused on practical methods, experimental design, and approaches to statistical analyses in the use of both male and female animals, cells, and tissues in preclinical research. Workshop participants also considered gender as a modifier of biology. This article builds on the workshop and is meant as a guide to preclinical investigators as they consider methods and techniques for inclusion of both sexes in preclinical research and is not intended to prescribe exhaustive/specific approaches for compliance with the new NIH policy.-Miller, L.
