Summary Secreted proteins and peptides hold large potential both as therapeutics and as enzyme catalysts in biotechnology. The high stability of many secreted proteins helps maintain functional integrity in changing chemical environments and is a contributing factor to their commercial potential. Disulphide bonds constitute an important post‐translational modification that stabilizes many of these proteins and thus preserves the active state under chemically stressful conditions. Despite their importance, the discovery and applications within this group of proteins and peptides are limited by the availability of synthetic biology tools and heterologous production systems that allow for efficient formation of disulphide bonds. Here, we refine the design of two DisCoTune (Disulphide bond formation in E. coli with tunable expression) plasmids that enable the formation of disulphides in the highly popular Escherichia coli T7 protein production system. We show that this new system promotes significantly higher yield and activity of an industrial protease and a conotoxin, which belongs to a group of disulphide‐rich venom peptides from cone snails with strong potential as research tools and pharmacological agents.

Abstract Background Soil carbon-rich organic amendments (biochar, humic substances) may improve the quality and fertility of arable soil. Their co-application can additively enhance the beneficial effect on soil. Hypothetically, the pre-treatment of biochar, by aging via soaking in a solution of commercially available humic substances, could result in synergism, which may exceed the benefit from simple co-application of both amendments to the soil. Therefore, the aim of this study was to investigate the impact of biochar, humic substances, the combination of both, and the impact of biochar aged by humic substances solution on soil microbial activities and plant growth in a short-term pot experiment with lettuce. Results The aging of biochar decreased the C:N ratio as compared to non-activated biochar. The co-application of biochar and humic substances into the soil resulted in the highest microbial biomass carbon and respiration activity. The majority of enzyme activities (β-glucosidase, arylsulfatase, N-acetyl-β-d-glucosaminidase, phosphatase) were the highest in humic substances-amended soil. The application of humic substances and biochar with humic substances seemed to stimulate microbial growth and activity followed by the competition of microflora for nutrients with plants, whereas the aged biochar behaved differently. The plants treated by aged biochar achieved the highest values of dry aboveground and root biomass of all variants. However, the assumed rapid uptake of nutrients by plants resulted in lower nutrient availability for microflora, and a decline in microbial viability. Conclusions Based on this study, the positive effect of co-applied humic substances and biochar on soil fertility, quality, and health can be concluded. The usability of biochar aging by humic solution requires further study. Graphic abstract

The purpose of the paper is to create a multidimensional talent management model with embedded aspects of artificial intelligence in the human resource processes to increase employees' engagement and performance of the enterprise. The research was implemented on a sample of 317 managers/owners in Slovenian enterprises. Multidimensional constructs of the model include several aspects of artificial intelligence implementation in the organization's activities related to human resource management in the field of talent management, especially in the process of acquiring and retaining talented employees, appropriate training and development of employees, organizational culture, leadership, and reducing the workload of employees, employee engagement and performance of the enterprise. The results show that AI supported acquiring and retaining a talented employees, AI supported appropriate training and development of employees, appropriate teams, AI supported organizational culture, AI supported leadership, reducing the workload of employees with AI have a positive effect on performance of the enterprise and employee engagement. The results will help managers or owners create a successful work environment by implementing artificial intelligence in the enterprise, leading to increased employee engagement and performance of the enterprise. Namely, our results contribute to the efficient implementation of artificial intelligence into an enterprise and give owners or top managers a broad insight into the various aspects that must be taken into account in business management in order to increase employee engagement and enterprise’s competitive advantage.

Większość fotografii Smolski wykonał w rodzinnej Łuce. Artykuł przybliża obraz chłopskiego uniwersum nadnarwiańskich, prawosławnych wsi. Fotograf z Łuki wykonywał zdjęcia, które pokazywały prywatny świat chłopów. To, co zostało utrwalone na fotografiach, świadczy o sile obrzędów przejścia, które miały bardzo duże znaczenie w społeczności chłopskiej, ponieważ utrwalały więzi rodzinne i sąsiedzkie. Wyrażały etapy życia człowieka w określonym porządku w zamkniętym cyklu narodzin, ślubów i pogrzebów. Zebrane zdjęcia są także ważnym świadectwem mikrohistorii Białorusinów Podlasia. Fotografie mają wielką wartość ze względów historycznych, kulturowych i społecznych. Mogą mieć duże znaczenie w popularyzacji kultury białoruskiej na Podlasiu jako przykładu jej siły i wartości, a także w zachęcaniu do powrotu do korzeni.

The functional beverages (FBs) are an important segment of functional food products due to health benefits they provide and their appealing sensory characteristics, suitability and affordability. FBs market offers many opportunities for new product development (nutraceuticals, colorants, plant-based medicines and products) with desirable and effective composition of nutrients and bioactive molecules (BAMs) aimed to deliver health benefits and improve human well-being. Recently, the use of medicinal and aromatic plants (MAPs) in the production of FBs has become increasingly popular due to specific content of nutrients (amino and fatty acids) and BAMs (volatile and non-volatile) attributed to the biological effects and health benefits. BAMs are stored in leaves, flowers, fruits, seeds, barks and roots, and they mostly include phenolics (phenolic acids, flavonoids, tanins, anthocyanins, lignans and stilbenes), essential oils (EOs), terpenoids, alkaloids, phytosterols and saponins. The aromatic features of MAPs are mainly related to volatile compounds of EOs, but the presence of non-volatile compounds, such as phenolics, also contributes to the specific sensory properties. Phytochemical profiling of plant species containing specific and complex mixtures of BAMs, provides numerous opportunities for the development of new categories of FBs, but also opens new challenges in their isolation using conventional and advanced extraction techniques, as well as determination of potential biological effects. This review summarizes the categories of the most common FBs, BAMs from selected MAPs and their biological effects, extraction techniques suitable for production of plant extracts and EOs, product quality and prediction trends, and several directions towards future research on FBs development strategies.

213. Trends in nationwide incidence of pediatric type 1 diabetes in Montenegro during the last 30 years

Significant and unexplained variations in type 1 diabetes (T1D) incidence through the years were observed all around the world. The update on this disorder’s incidence is crucial for adequate healthcare resource planning and monitoring of the disease. The aim of this study was to give an update on the current incidence of pediatric T1D in Montenegro and to analyze incidence changes over time and how the exposure to different factors might have affected it. This retrospective cohort study included a total of 582 patients younger than 15 years who were newly diagnosed with T1D during the past 30 years. The average age at diagnosis was 8.4 ± 3.91 years. The mean annual incidence of T1D in the Montenegro population during the whole study period of 30 years was 15.2/100,000 person-years. Slightly higher incidence rates were observed in male compared to female individuals, and the incidence increased with age, with the highest incidence in the 10–14 age group. If the model is observed as one without jointpoints, the annual percentage change (APC) for the total population is 3.1 (1.8–4.4); for male individuals, 3.8 (2.1–5.5); and for female individuals, 2.1 (0.6–3.5). In 2020, the first year of the coronavirus disease of 2019 (COVID-19) pandemic, in comparison to 2019, the incidence rate increased from 19.7/100,000 to 21.5/100,000, with the highest increase in the age group of 5–9 years. This is the first nationwide report on a 30-year period of T1D incidence trend in Montenegro. It suggests that T1D incidence among Montenegrin children is rising again and that there is a short-term influence of COVID-19 on new-onset T1D.

214. Gut community alterations associated with Clostridioides difficile colonization in hospitalized gastroenterological patients with or without inflammatory bowel disease

Clostridioides difficile colonization and development of infection commonly occur in inflammatory bowel disease (IBD) patients and can trigger flare-ups. Both conditions are inherently linked to disrupted gut microbiota. This study included 149 hospitalized gastrointestinal patients, which were divided into IBD (n = 48) and non-IBD patients (n = 101). Patients were tested for C. difficile colonization (qPCR and selective plating), and gut bacterial communities were analyzed with 16S amplicon sequencing. Blood test results were retrospectively collected from the medical records. IBD and non-IBD patients had comparable C. difficile colonization rates (31.7 and 33.3%, respectively). Compared to non-IBD C. difficile-non-colonized patients, IBD and C. difficile-colonized patients shared multiple common bacterial community characteristics including decreased diversity and reduced abundance of strict anaerobic bacteria. Furthermore, certain microbiota alterations were enhanced when IBD was accompanied by C. difficile colonization, indicating a synergistic effect between both medical complications. Conversely, certain microbial patterns were specific to C. difficile colonization, e.g., co-occurrence with Enterococcus, which was most common in IBD patients (81.3%).

Protein binding can impair the potency of human immunodeficiency virus (HIV) protease inhibitors. Therefore, the activity of a novel compound, CGP 61755, was studied in the absence or presence of alpha1-acid glycoprotein (alpha1AGP). In MT-2 cells, the activity loss was 4-fold (EC90 without alpha1AGP, 29 nM vs. 122 nM with alpha1AGP). In primary lymphocytes, the loss was 8-fold (EC90, 45 nM vs. 364 nM). In identical experiments, the activity loss in MT-2 cells and lymphocytes was 2- and 3-fold, respectively, for indinavir, 11- and 10-fold for saquinavir, and 11- and 48-fold for ritonavir. For SC-52151, a 17-fold loss was seen in MT-2 cells, whereas no EC90 with alpha1AGP was reached in lymphocytes. This study demonstrates that the impact of alpha1AGP on in vitro activity varies greatly among different HIV protease inhibitors. The magnitude of such differences is greater in human lymphocytes than in a standard cell line.

Familija Gesneriaceae obuhvata biljne vrste pretežno rasprostranjene u tropskim u subtropskim oblastima Starog i Novog sveta. Samo mali broj vrsta naseljava umerene oblasti, a među njima je i jedinih 5 vrsta ove familije u Evropi: Ramonda myconi, endemit Pirinejskog poluostrva, kao i R. nathaliae, R. serbica, Haberlea rhodopensis i Jankaea heldreichii, četiri endemične vrste Balkanskog poluostrva. Svih pet vrsta su relikti Tercijera, vremena kada je klima u Evropi bila toplija i vlažnija. Za vreme Ledenog doba, one su našle skrovište u klisurama i kanjonima gde ih najčešće nalazimo i danas. Ovi „živi fosili“ danas nepovoljne uticaje spoljašnje sredine u velikoj meri preživljavaju zahvaljujući poikilohidričnosti, pa se još nazivaju i „biljke koje vaskrsavaju“. Danas se dve balkanske vrste roda Ramonda odlikuju disjunktnim arealima. Ramonda nathaliae zastupljena je u Makedoniji, S Grčkoj, na obroncima planine Šare na Kosovu i u JI Srbiji. Najveći deo areala vrste R. serbica nalazi se u Albaniji, a prisutna je i na SZ Grčke, u Z Makedoniji, JZ i SI Crnoj Gori, JZ, JI i SI Srbiji, kao i na SZ Bugarske. Jedina zona u kojoj se areali dve vrste susreću nalazi se u JI Srbiji, gde na dva lokaliteta, Oblik i Radovanski Kamen, obrazuju simpatrijske populacije. Obe vrste prevashodno naseljavaju krečnjačku podlogu, s tim što se R. nathaliae može naći i na serpentinitu, škriljcima i granitu. Ova vrsta, takođe, bolje podnosi otvorenija staništa, a njene populacije se mogu naći i na većem rasponu nadmorskih visina, zbog čega se smatra otpornijom i tolerantijom. Ciljevi ovog rada su višestruki: da se utvrde morfološke osobine polena i semena sve tri vrste roda Ramonda, jer detalji njihove građe do sada nisu istraživani; da se procene vijabilnost njihovog polena i klijavost semena; da se utvrdi broj hromozoma, kao i veličina genoma kod tri vrste; da se proveri da li postoje hibridne jedinke u simpatrijskim populacijama; da se pomoću molekularnih markera utvrdi stepen sličnosti i filogeografske veze između vrsta R. nathaliae i R. serbica. U tu svrhu, polen i seme analizirani su pomoću svetlosnog i scanning elektronskog mikroskopa, broj hromozoma određen je standardnim kariološkim tehnikama, veličina genoma procenjena je pomoću protočne citometrije, a genetički diverzitet primenom molekularne AFLP tehnike... Family Gesneriaceae comprises plant species mostly distributed in the tropics and subtropics of both the Old and New Worlds. Only a small number of species inhabits temperate regions. Among them, there are the only 5 species of this family in Europe: Ramonda myconi, endemic of the Iberian Peninusla, as well as R. nathaliae, R. serbica, Haberlea rhodopensis i Jankaea heldreichii, four endemic species of the Balkan Peninsula. All five species are Tertiary relicts, remnants from the times when the climate was much warmer and more humid. During the Ice Age, they have found shelter in canyons and gorges where can be often found today as well. These „living fossils“ are poikilohydric, „resurrection plants“ what helps them survive the inhospitable environmental conditions. Balkan species of the genus Ramonda are today characterized by disjunct distribution. Ramonda nathaliae is restricted to Macedonia, N Greece, slopes of Mt. Šara in Kosovo and few localities in SE Serbia. The largest part of the R. serbica range is situated in Albania, but is also present in NW Greece, W Macedonia, SW and NE Montenegro, SW, SE and NE Serbia and NE Bulgaria. The ranges of two species cooccur only in two localities in SE Serbia, Oblik and Radovanski Kamen, establishing sympatric populations. Both species prefer limestone rocks, but R. nathaliae can also be found on serpentine, schist and granite. This species is often found in more open habitats and in higher altitudes why it is considered as more resistant and tolerant than R. serbica. The aims of this study are: to investigate morphology of both pollen and seeds of three Ramonda species; to estimate pollen viability and germination capacity of their seeds; to determine chromosome numbers and genome size of three species; to explore if there are hybrids in sympatric populations; to look into genetic diversity and phylogeography of R. nathaliae and R. serbica. The purpose of this paper is to analyze the negative impact of “mundialisation” on the banking sector. It is evident that the global banking system has become more fragile. As a response to progressing instability in financial architecture, policy makers have become interested in better understanding of high sensitivity in the financial sector, particularly in banks. One of the basic techniques for calculating banking fragility is stress - testing. In order to estimate the potentially harmful and unpredictable effects on their transactions, banks have created stress - testing models, which turned out to be an indispensable tool in the bank supervision process. Banking regulators have started using improved stress - testing methods for estimating aggregate risk and for defining the adequate capital level in the banking sector.

218. From Waste to Green: Water-Based Extraction of Polyphenols from Onion Peel and Their Adsorption on Biochar from Grapevine Pruning Residues

Onion peels (OP) are rich in bioactive compounds with a plethora of benefits for human health, but this valuable material is often wasted and underutilized due to its inedibility. Likewise, grapevine pruning residues are commonly treated as agricultural waste, but biochar (BC) obtained from this material has favorable characteristics as an adsorbent. This study investigated the potential of BC in removal of targeted polyphenolic compounds from OP extracts. The OP extracts were obtained adhering to green chemistry principles using deionized water amplified by three methods: maceration (MAC), ultrasound-assisted extraction (UAE), and microwave-assisted extraction (MAE). The extraction efficiency on the polyphenolic profile and antioxidant capacity was investigated with different extraction temperatures and solid-to-liquid (s/l) ratios. For further analysis, UAE at 90 °C with an s/l ratio of 1:100 was used due to higher polyphenolic compound yield. The BC adsorption capacity of individual polyphenols was fitted with the Langmuir and Freundlich isotherm models. Quercetin-3,4′-diglucoside obtained the highest R2 coefficient in both models, and the highest qmax value. The optimum conditions in the dosage experiment suggested an amount of 0.5 g of BC using 3 g/L extracts. The studied BC showed a high affinity for targeted phytochemicals from OP extracts, indicating its potential to be applied for the green adsorption of valuable polyphenolic compounds.

In the present work, the functionalisation of austenitic stainless steel, AISI 316L surfaces via nanosecond Nd:YAG laser texturing in order to modify the surface morphology with crosshatch and dimple patterns is presented. A tribological analysis under lubrication with sunflower and jojoba oil with and without the addition of a solid lubricant, MoS2 nanotubes, was performed. In conjunction with friction/wear response laser-textured surface wettability, oil spreadability and oil retention capacity were also analysed. It was shown that the crosshatch pattern generally exhibited lower friction than the dimple pattern, with the addition of MoS2 nanotubes not having any significant effect on the coefficient of friction under the investigated contact conditions. This was found in addition to the better oil spreadability and oil retention capacity results of the crosshatch-textured surface. Furthermore, texturing reduced the wear of the stainless-steel surfaces but led to an approximately one order of magnitude larger wear rate of the steel counter-body, primarily due to the presence of hard bulges around the textured patterns. Overall, the crosshatch pattern showed better oil retention capacity and lower friction in combination with different vegetable oils, thus making it a promising choice for improving tribological performance in various environmentally friendly applications.

Vocabulary is the key component of listening narrative comprehension, but its contribution has been scarcely investigated in bilingual children. This study aimed to examine (a) listening narrative comprehension and receptive vocabulary in L1 (Italian) and L2 (English) in preschool and first grade children; (b) the specific contribution of receptive vocabulary to listening narrative comprehension in both languages. Participants were 30 preschoolers and 32 first graders, who are all Italian children attending an international school in English. In both languages, receptive vocabulary was assessed through PPVT-R and listening narrative comprehension through TOR 3-8. The results showed that listening narrative comprehension was age appropriate in both languages but higher in L1. Lower vocabulary in L1 than L2 was found, and this difference is higher for preschoolers than for first grades; finally, two regressions performed on listening narrative comprehension in each language showed that only vocabulary in the same language accounted for listening narrative comprehension. Children obtain higher performance in L1; however, after a few years of L2 exposure in the educational context, language skills fall within the normal range, with some weakness in vocabulary. Vocabulary contribution to listening narrative comprehension is similar in both languages and specific for each.

Fresh-cut potatoes (FCP), like other fresh-cut (minimally processed) vegetables, are a convenient but highly perishable product. Unlike most fresh-cut vegetables, which are “ready-to-eat”, FCP must be cooked before consumption. Therefore, in addition to the safety (chemical and microbiological), quality and sensory characteristics of raw FCP, the same requirements should be applied for cooked potatoes. It is known that many factors play a role in meeting all these requirements: (i) selection of cultivars less susceptible to browning; (ii) use of anti-browning and antimicrobial agents and/or certain physical methods against browning and microbial growth; (iii) packaging and cold storage conditions. In recent studies on FCP, scientists have attempted to deepen their knowledge of the mechanisms of browning prevention to better understand changes at the molecular level as well. The main objective of this review is to provide a comprehensive overview of recent research, which aimed at deepening knowledge of the various changes that occur in potatoes during processing, and to develop new approaches that could help improve quality and extend FCP shelf life. It also discusses the effects of subsequent cooking of FCP on sensory and other properties, as well as on chemical constituents.

222. Fungicidal Protection as Part of the Integrated Cultivation of Sugar Beet: An Assessment of the Influence on Root Yield in a Long-Term Study

Despite the major role of non-chemical treatments in integrated plant protection, fungicides often need to be applied as a crop protection treatment in sugar beet farming. They should be used based on a good understanding of the requirements and effectiveness of the active ingredients. In 11-year field experiments, the effect that one and three foliar applications of fungicides containing various active ingredients (triazoles, benzimidazoles, strobilurines) had on sugar beet root yields was assessed, depending on various thermal and rainfall conditions. It was found that in eight of the 11 years, foliar application of fungicides increased yields compared to unprotected plants, and three foliar treatments during the growing season were more effective than a single application. The negative correlation of the root yield of fungicidally protected plants with total June rainfall was weaker than the same relationship for unprotected plants. At the same time, the positive correlation between the yield of fungicidally protected sugar beets and average June air temperature was stronger than the same relationship for unprotected plants. The research results indicate the need to conduct long-term field experiments and to continuously improve integrated production principles for sugar beet, especially regarding the rational use of pesticides.

223. Inhibitory Control, Social Cognition, and Peer Social Competence among Children with and without a Migration Background in Italy

Executive functions and social cognition (i.e., the mental operations that underlie social interaction) are essential for children’s successful interactions, yet their role in peer social competence among children with and without a migration background is still underexplored. This study investigated the influence of inhibitory control and two domains of social cognition (theory of mind and facial emotion recognition) on children’s peer social competence exploring moderation by children’s migration background. First graders in Italy (n = 321, M age = 6.4 years, 45% girls, and 39% with a migration background) completed standardized tests, while teachers rated students’ peer social competence. The results indicated that immigrant-origin children were perceived as being less socially competent than their non-immigrant counterparts. Among children with a migration background, better inhibitory control and facial emotion recognition skills were linked to increased peer social competence. Following the presentation of these results, we will discuss the implications for both developmental theory and practice.

224. Optimization of the Spray-Drying Encapsulation of Sea Buckthorn Berry Oil

The aim of this study was to evaluate the effect of spray-drying parameters on the physicochemical properties of encapsulated sea buckthorn berry oil. Different carriers (gum arabic, β-cyclodextrin, and their mixture (1:1, w/w)), inlet air temperatures (120, 150, and 180 °C), and carrier-to-oil ratios (2, 3, and 4, w/w) were evaluated. The obtained powders were characterized in terms of the product yield (36.79–64.60%), encapsulation efficiency (73.08–93.18%), moisture content (0.23–3.70%), hygroscopicity (1.5–7.06 g/100 g), solubility (19.55–74.70%), bulk density (0.25–0.44 g/L), total carotenoid content (mg/100 g dm), and antioxidant capacity (871.83–1454.39 μmol TE/100 g dm). All physicochemical properties were significantly affected by the carrier-to-oil ratio and inlet air temperature. Higher carrier-to-oil ratios increased the product yield, encapsulation efficiency, solubility, and bulk density and decreased the powder hygroscopicity. Elevating the drying temperatures during spray drying also increased the product yield, encapsulation efficiency, and solubility, while it decreased the powder moisture content, total carotenoid content, and antioxidant capacity. Based on the physicochemical properties, the use of β-cyclodextrin as a carrier, a drying temperature of 120 °C, and a carrier-to-oil ratio of 4 were selected as optimal conditions for the production of sea buckthorn berry oil powder. The obtained powder is a valuable material for a wide range of applications in the food and nutraceutical industries.

Agricultural waste, which is produced in large quantities annually, can be a threat to the environment. Biochar (BC) production represents a potential solution for reducing the amount of grapevine pruning residues and, accordingly, the impact on the environment and climate change. Biochar produced by the process of pyrolysis from grapevine pruning residues was investigated and characterized to be applied as an adsorbent of polyphenolic compounds with the aim of using the waste from viticultural production to obtain a quality product with adsorption and recovery potential. Standards of caffeic acid (CA), gallic acid (GA), and oleuropein (OLP) were used as polyphenolic representatives. The obtained data were fitted with the Langmuir and Freundlich isotherms models to describe the adsorption process. The best KL (0.39) and R2 (0.9934) were found for OLP using the Langmuir model. Furthermore, the adsorption dynamics and recovery potential of BC were investigated using an adapted BC column and performed on an HPLC instrument. The adsorption dynamics of biochar resulted in the adsorption of 5.73 mg CA g−1 of BC, 3.90 mg GA g−1 of BC, and 3.17 mg OLP g−1 of BC in a 24 h contact. The online solid phase extraction of the compounds performed on an HPLC instrument yielded a recovery of 41.5 ± 1.71% for CA, 61.8 ± 1.16% for GA, and 91.4 ± 2.10% for OLP. The investigated biochar has shown a higher affinity for low-polar compound adsorption and, consequently, a higher polar compound recovery suggesting its potential as an efficient polyphenolic compound adsorbent.

226. Artificial Intelligence and Business Studies: Study Cycle Differences Regarding the Perceptions of the Key Future Competences

The purpose of this article is to identify the differences in various aspects of the perception of artificial intelligence by students of economics and business studies at different levels of study and, on this basis, to formulate recommendations both to the higher education institutions themselves, which educate in the field of economic and business sciences, as well as to curriculum designers. First, we utilized descriptive statistics to analyze the responses for each construct among undergraduate and postgraduate students. In the second part, we employed the Kolmogorov-Smirnov and Shapiro-Wilk tests to assess the normality of data distribution. Finally, in the third part, we employed the non-parametric Mann-Whitney U test to identify the differences between undergraduate and postgraduate students. The results show that statistically significant differences can be identified especially in how students of both study levels see and understand the importance of AI. Although we did not identify significant differences between students of both levels in how they see their role in the future labor market, which will be (or already is) characterized by artificial intelligence, we must emphasize that students of both levels evaluate their roles modestly in this respect. Therefore, on this basis, we have made recommendations for more active development and integration of AI in the study process; the article presents important suggestions for improving education to prepare students for the business world of artificial intelligence.

The purpose of the paper is to present a model of factors affecting the successful project implementation by introducing agility and artificial intelligence to increase the company’s competitiveness. In the model, the multidimensional constructs describing the implementation of an agile work environment and artificial intelligence technologies and tools were developed. These multidimensional constructs are agile work environment, agile leadership, agile team skills and capabilities, improving the work of the leader in the project, adopting AI technologies in the project, and using AI solutions in a project. Their impact on successful project implementation and on the company competitiveness was tested. The fundamental reason for conducting this research and developing the model is to enhance the understanding of factors that contribute to the successful implementation of projects and to increase a company’s competitiveness. Our developed model encompasses multidimensional constructs that describe the agile work environment and the utilization of AI technologies. By examining the impact of these constructs on both successful project implementation and company competitiveness, we aimed to establish a comprehensive framework that captures the relationship between agility, AI, and successful project implementation. This model serves as a valuable tool for companies seeking to improve their project implementation processes and gain a competitive edge in the market. The research was based on a sample of 473 managers/owners in medium-sized and large companies. Structural equation modeling was used to test the hypotheses. In today’s turbulent environment, the results will help develop guidelines for a successful combination of agile business practices and artificial intelligence to achieve successful project implementation, increasing a company’s competitiveness.

Abstract Background It is encouraging to see a substantial increase in individuals surviving cancer. Even more so since most of them will have a positive effect on society by returning to work. However, many cancer survivors have unmet needs, especially when it comes to improving their quality of life (QoL). Only few survivors are able to meet all of the recommendations regarding well-being and there is a body of evidence that cancer survivors’ needs often remain neglected from health policy and national cancer control plans. This increases the impact of inequalities in cancer care and adds a dangerous component to it. The inequalities affect the individual survivor, their career, along with their relatives and society as a whole. The current study will evaluate the impact of the use of big data analytics and artificial intelligence on the self-efficacy of participants following intervention supported by digital tools. The secondary endpoints include evaluation of the impact of patient trajectories (from retrospective data) and patient gathered health data on prediction and improved intervention against possible secondary disease or negative outcomes (e.g. late toxicities, fatal events). Methods/design The study is designed as a single-case experimental prospective study where each individual serves as its own control group with basal measurements obtained at the recruitment and subsequent measurements performed every 6 months during follow ups. The measurement will involve CASE-cancer, Patient Activation Measure and System Usability Scale. The study will involve 160 survivors (80 survivors of Breast Cancer and 80 survivors of Colorectal Cancer) from four countries, Belgium, Latvia, Slovenia, and Spain. The intervention will be implemented via a digital tool (mHealthApplication), collecting objective biomarkers (vital signs) and subjective biomarkers (PROs) with the support of a (embodied) conversational agent. Additionally, the Clinical Decision Support system (CDSS), including visualization of cohorts and trajectories will enable oncologists to personalize treatment for an efficient care plan and follow-up management. Discussion We expect that cancer survivors will significantly increase their self-efficacy following the personalized intervention supported by the m-HealthApplication compared to control measurements at recruitment. Surface characteristics of printing substrates are of the utmost importance to all types of paper that interact with ink. During all types of printing processes, the behaviour of the liquid phase (ink or dye) on the paper is directly defined by the paper cellulose-based surface. The printed ink spreads and penetrates more into paper fibres when the paper surface is rougher and more permeable. Contact angle measurements by sessile drop method are considered the most appropriate for determining the paper sheet surface energy. Paper as hydrophilic material has a high absorption rate resulting in a low contact angle. The objective of this study was to evaluate the surface free energy of laboratory-made papers containing straw pulp obtained from residues after the harvest of the most cultivated cereals in Croatia (wheat, barley and triticale). The obtained surface free energy results are promising for straw pulp usage in the manufacture of printing paper.

This study investigated the function of the cruciate ligaments during a forward lunge movement. The mechanical roles of the anterior and posterior cruciate ligament (ACL, PCL) during sagittal plane movements, such as forward lunging, are unclear. A forward lunge movement contains a knee joint flexion and extension that is controlled by the quadriceps muscle. The contraction of the quadriceps can cause anterior tibial translation, which may strain the ACL at knee joint positions close to full extension. However, recent findings suggest that it is the PCL rather than the ACL which is strained during forward lunging. Thus, the purpose of the present study was to establish a musculoskeletal model of the forward lunge to computationally investigate the complete mechanical force equilibrium of the tibia during the movement to examine the loading pattern of the cruciate ligaments. A healthy female was selected out of a group of healthy subjects, who all performed a forward lunge on a force platform, targeting a knee flexion angle of 90˚. Skin-markers were placed on anatomical landmarks on the subject and the movement was recorded by five video cameras. The three-dimensional kinematic data describing the forward lunge movement were extracted and used to develop a biomechanical model of the lunge movement. The model comprised two legs including femur, crus, rigid foot segments and the pelvis. Each leg had 35 independent muscle units, which were recruited according to a minimum fatigue criterion. This approach allowed a full understanding of the mechanical equilibrium of the knee joint, which revealed that the PCL had an important stabilizing role in the forward lunge movement. In contrast, the ACL did not have any significant mechanical function during the lunge movement. Furthermore, the results showed that m. gluteus maximus may play a role as a knee stabilizer in addition to the hamstring muscles.

Research Question (RQ): Are there statistically significant differences in work motivation, satisfaction and engagement among older employees before the COVID-19 pandemic and during the COVID-19 pandemic? Purpose: The aim of the paper is to examine if there are statistically significant differences in work motivation, satisfaction and engagement among older employees before the COVID-19 pandemic and during the COVID-19 pandemic. The aim of the paper is also to identify which factors contribute to increasing work motivation, satisfaction and engagement during the COVID-19 pandemic. Method: The differences in work motivation, satisfaction and engagement among older employees before the COVID-19 pandemic and during the COVID-19 pandemic were tested with the nonparametric test for two related samples. To analyze the differences between individual statements, we used the non-parametric Wilcoxon signed-rank test. Results: The results show that there are statistically significant differences in work motivation, satisfaction and engagement among older employees before the COVID-19 pandemic as compared to the during COVID-19 pandemic. Organisation: The results have a significant impact on a more detailed review of the creation of new working conditions for employees during the Covid-19 pandemic and especially in the period after the Covid-19 pandemic. Society: During the COVID-19 pandemic, companies and society can focus on how to constructively manage the new challenges that the COVID-19 pandemic brings to companies and what they can learn from it. For companies and leaders or HRM in particular, the pandemic can be used as a starting point for positive and future-oriented developments. Originality: With our findings we contribute to the discussion on a constructive and future oriented approach to the COVID-19 pandemic, both for practices around the management of employees as well as for future research. Also, our research highlights which factors increase work motivation, work satisfaction and work engagement during COVID-19 and how to create appropriate working conditions during the COVID-19 and after the COVID-19 pandemic to keep employees motivated, satisfied and engaged. Limitations/further research: Our sample was limited to Slovenian companies and older employees. Also, in our research we limited on work motivation, work satisfaction and work engagement. Therefore, for further research we propose the examination of differences in others constructs before the COVID-19 pandemic and employees during the COVID-19 pandemic in Slovenian companies and also, the examination of differences in constructs between Slovenia and other countries.

Erythropoietin receptor (EPOR) is widely expressed in healthy and malignant tissues. In certain malignancies, EPOR stimulates tumor growth. In healthy tissues, EPOR controls processes other than erythropoiesis, including mitochondrial metabolism. We hypothesized that EPOR also controls the mitochondrial metabolism in cancer cells. To test this hypothesis, we generated EPOR-knockdown cancer cells to grow tumor xenografts in mice and analyzed tumor cellular respiration via high-resolution respirometry. Furthermore, we analyzed cellular respiratory control, mitochondrial content, and regulators of mitochondrial biogenesis in vivo and in vitro in different cancer cell lines. Our results show that EPOR controls tumor growth and mitochondrial biogenesis in tumors by controlling the levels of both, pAKT and inducible NO synthase (iNOS). Furthermore, we observed that the expression of EPOR is associated with the expression of the mitochondrial marker VDAC1 in tissue arrays of lung cancer patients, suggesting that EPOR indeed helps to regulate mitochondrial biogenesis in tumors of cancer patients. Thus, our data imply that EPOR not only stimulates tumor growth but also regulates tumor metabolism and is a target for direct intervention against progression.

233. Social determinants of health and emergency department utilization among adults receiving buprenorphine for opioid use disorder

Background: Individuals with opioid use disorder (OUD) use the emergency department (ED) at high rates. Medication treatment for OUD (MOUD) is associated with reduced ED utilization. However, individuals receiving MOUD still utilize ED services at higher rates than the general population. The objective of this study is to compare the psychosocial and clinical characteristics of those who do and do not utilize ED services based on the Healthy People 2030 framework regarding social determinants of health (SDoH) among a sample of individuals receiving MOUD. Methods: Participants receiving buprenorphine for OUD at an outpatient addiction clinic completed a cross-sectional survey between July and September 2019. A 6-month prospective medical record review was conducted. The primary outcome was ED visit (yes/no) during the 6-month study period. Demographic, psychosocial, and clinical characteristics were gathered from survey measures and chart abstraction. Chi square and T-tests tested differences by ED utilization. Results: Participants (n=142) were 54.9% female and 68.8% Black, with an average age of 43.2 years (SD=12.5). Of the participants, 38.7% visited the ED in the study period, primarily for infectious or musculoskeletal causes. Participants with an ED visit were more likely to be Black (p=.011), have less social support (p=.030), more medical comorbidities (p=.008) including chronic pain (p=.045), and more visits with an addiction provider in the study period (p=.009). Conclusions: Factors associated with ED utilization among individuals receiving buprenorphine for OUD include low social support and medical comorbidities, including chronic pain. More research is needed on modifiable SDoH that influence ED utilization.

Izhodišče: Katetrska vstavitev aortne zaklopke (TAVI) je v zadnjih letih postala prevladujoča metoda za zdravljenje degenerativne aortne stenoze pri starejših bolnikih. Prispevek prikazuje prve rezultate zdravljenja s TAVI v mednarodnem kardiovaskularnem centru MC Medicor in jih primerja z najpomembnejšimi mednarodnimi raziskavami zadnjih let. Metode: V retrospektivno raziskavo smo vključili zaporedno serijo bolnikov, pri katerih smo opravili TAVI. Podatke smo pridobili iz lastnega registra perkutanih kardioloških posegov. Rezulati: Med 12. decembrom 2016 in 6. junijem 2020 smo TAVI opravili pri 109 bolnikih s povprečno starostjo 81 let in EuroScore II 3.95. Srednji ultrazvočni gradient na aortni zaklopki je znašal 46 mm Hg, izračunana površina ustja pa 0,8 cm2. Pri vseh bolnikih, razen pri enem, smo uporabili perkutani pristop preko femoralne arterije. Balonsko predilatacijo smo izvedli v 36,7 %, novo zaklopko vsadili v 98,2 % in jo balonsko postdilatirali v 15,6 %. Srednji ultrazvočni gradient po TAVI je znašal 9 mm Hg. Zmerna aortna regurgitacija je bila prisotna v 3,7 % (95 % IZ 1,0–9,1 %), huda pa pri nobenem od bolnikov. Nov srčni spodbujevalec je potrebovalo 22,9 % bolnikov (95 % IZ 15,4–32,0 %). En bolnik je imel tranzitorno ishemično kap. Urgentno perikardiocentezo smo opravili v 2,7 % (95 % IZ 0,6–7,8 %), srčno operacijo v 1,8 % (95 % IZ 0,2–6,5 %), žilno operacijo v 0,9 % (95 % IZ 0,0–5,0 %) in hemostazo s prekritimi žilnimi opornicami v 1,8 % (95 % IZ 0,2–6,5 %). Bolnišnična in 30-dnevna umrljivost sta bili 1,8 % (95 % IZ 0,2–6,5 %), enoletna 3,1 % (95 % IZ 0,0–6,5 %), dvoletna 5,6 % (95 % IZ 0,0–11,4 %) in triletna 14,2 % (95 % IZ 0,0–29,6 %). Zaključek: Začetni rezultati TAVI v Medicorju so ugodni in primerljivi z najpomembnejšimi mednarodnimimi raziskavami zadnjih let.

This paper proposes a method for changing pixel shape by converting a CMYK raster image (pixel) to an HSB vector image, replacing the square cells of the CMYK pixels with different vector shapes. The replacement of a pixel by the selected vector shape is done depending on the detected color values for each pixel. The CMYK values are first converted to the corresponding RGB values and then to the HSB system, and the vector shape is selected based on the obtained hue values. The vector shape is drawn in the defined space, according to the row and column matrix of the pixels of the original CMYK image. Twenty-one vector shapes are introduced to replace the pixels depending on the hue. The pixels of each hue are replaced by a different shape. The application of this conversion has its greatest value in the creation of security graphics for printed documents and the individualization of digital artwork by creating structured patterns based on the hue.

The aim of this study was to investigate the magnetic properties of mixed nanocrystalline Zn/manganese oxide compounds synthesized by a hydrothermal method. These compounds are designed as (ZnO)1−n(MnO)n, where index n ranges from 0.05 to 0.60. The results of magnetic measurements, including AC magnetic susceptibility as a function of temperature (up to 160 K) and frequency (from 7 Hz up to 9970 Hz), as well as DC magnetization in magnetic fields up to 9 T and temperature up to 50 K, are reported. We observed various types of magnetic behavior depending on the nominal weight content of MnO. Samples with a low nominal content (up to n = 0.10) of MnO exhibited Curie–Weiss behavior at higher temperatures. For samples with high nominal weight contribution (from n = 0.30 to 0.60), spin-glass-like or/and weak ferromagnetic behavior is observed.

239. A Three-Dose mRNA COVID-19 Vaccine Regime Produces Both Suitable Immunogenicity and Satisfactory Efficacy in Patients with Solid Cancers

Background: The recommended booster third dose of vaccination against COVID-19 in cancer patients seems reasonable to protect them against a severe disease course. A prospective study was designed to assess the immunogenicity, efficacy, and safety of COVID-19 vaccination in this cohort. Methods: Patients with solid malignancies on active treatment were followed up after the primary course and booster third dose of vaccination to assess their anti-SARS-CoV-2 S1 IgG levels, efficacy in the case of SARS-CoV-2 infection, and safety. Results: Out of 125 patients receiving the primary course of vaccination, 66 patients received a booster third dose of mRNA vaccine, with a 20-fold increase in median anti-SARS-CoV-2 S1 IgG levels compared to Ab levels six months post-primary course of vaccination (p < 0.0001). After the booster third dose, anti-SARS-CoV-2 S1 IgG levels were comparable to healthy controls (p = 0.113). There was a decline in Ab levels 3 (p = 0.0003) and 6 months (p < 0.0001) post-third booster dose. No patients had either a severe disease course or a lethal outcome in the case of SARS-CoV-2 infection after the third booster dose. Conclusion: The third booster vaccination dose against COVID-19 in solid cancer patients triggers substantial immunogenicity and is safe and effective for preventing a severe COVID-19 disease course.

The cortical silent period (cSP) is a period of electrical silence following a motor-evoked potential (MEP) in the electromyographic signal recorded from a muscle. The MEP can be elicited by transcranial magnetic stimulation (TMS) over the primary motor cortex site corresponding with the muscle. The cSP reflects the intracortical inhibitory process mediated by GABAA and GABAB receptors. The study aimed to investigate the cSP in the cricothyroid (CT) muscle after applying e-field-navigated TMS over the laryngeal motor cortex (LMC) in healthy subjects. Then, a cSP as a neurophysiologic feature for laryngeal dystonia was observed. We applied a single-pulse e-field-navigated TMS to the LMC over both hemispheres with hook-wire electrodes positioned in the CT muscle in nineteen healthy participants, which triggered the elicitation of contralateral and ipsilateral corticobulbar MEPs. The subjects were engaged in a vocalization task, and then we assessed the following metrics: LMC intensity, peak-to-peak MEP amplitude in the CT muscle, and cSP duration. The results showed that the cSP duration from the contralateral CT muscle was distributed from 40 ms to 60.83 ms, and from the ipsilateral CT muscle, from 40 ms to 65.58 ms. Also, no significant difference was found between the contralateral and ipsilateral cSP duration (t(30) = 0.85, p = 0.40), MEP amplitude in the CT muscle (t(30) = 0.91, p = 0.36), and LMC intensity (t(30) = 1.20, p = 0.23). To conclude, the applied research protocol showed the feasibility of recording LMC corticobulbar MEPs and observing the cSP during vocalization in healthy participants. Furthermore, an understanding of neurophysiologic cSP features can be used to study the pathophysiology of neurological disorders that affect laryngeal muscles, such as laryngeal dystonia.

Biocompatible mucoadhesive formulations that enable a sustained drug delivery at the site of action, while exhibiting inherent antimicrobial activity, are of great importance for improved local therapy of vaginal infections. The aim of this research was to prepare and evaluate the potential of the several types of azithromycin (AZM)-liposomes (180–250 nm) incorporated into chitosan hydrogel (AZM-liposomal hydrogels) for the treatment of aerobic vaginitis. AZM-liposomal hydrogels were characterized for in vitro release, and rheological, texture, and mucoadhesive properties under conditions simulating the vaginal site of application. The role of chitosan as a hydrogel-forming polymer with intrinsic antimicrobial properties was explored against several bacterial strains typical for aerobic vaginitis as well as its potential effect on the anti-staphylococcal activity of AZM-liposomes. Chitosan hydrogel prolonged the release of the liposomal drug and exhibited inherent antimicrobial activity. Additionally, it boosted the antibacterial effect of all tested AZM-liposomes. All AZM-liposomal hydrogels were biocompatible with the HeLa cells and demonstrated mechanical properties suitable for vaginal application, thus confirming their potential for enhanced local therapy of aerobic vaginitis.

Since certain constituents are not naturally present in pure fruit juices, incorporating herbal extracts can provide specific sensory properties to the beverages and improve their biopotential. In our previous research, it was found that sage (Salvia officinalis L.), wild thyme (Thymus serpyllum L.), and combinations of their extracts had the highest total phenolic content and a unique composition of volatile compounds, which can contribute to the aromatic and antioxidant qualities of functional products. Therefore, this research aimed to investigate the potential of sage and wild thyme extracts, as well as their mixture (wild thyme:sage at 3:1, v/v), to enrich fruit juices (apple, pineapple, and orange). Obtained beverages were evaluated for sensory properties as well as phenolic and headspace composition (UPLC-MS/MS and HS-SPME/GC-MS analysis) and antioxidant capacity (ORAC assay). The incorporation of wild thyme extract in pineapple juice provided the most harmonious flavor and the highest content of volatile compounds (on PDMS/DVB fiber). The orange juice formulations were the most enriched with phenolic and volatile compounds (on DVB/CAR/PDMS fibers). The highest antioxidant capacity was observed in the formulation with orange juice and sage extract (22,925.39 ± 358.43 µM TE). This study demonstrated that enriching fruit juices with sage and wild thyme extracts could create functional beverages with improved sensory and health-promoting properties, providing valuable insights for the food and beverage industry to meet the growing demand of health-conscious consumers for natural and functional products.

First, the content of PTEs was determined in plant biomass and in soil samples using: (a) Optical emission spectrometry with inductively coupled plasma to determine Sb, As, Cd, Cu, Ni, Pb, and Se, and (b) thermal decomposition, amalgamation, and atomic absorption spectrometry to determine Hg. The effectiveness of Melilotus albus Med. (M. albus) for phytoremediation was evaluated using the bioconcentration factor (BCF). The phytoextraction potential of M. albus was determined using bioaccumulation factor (BAC) and translocation factor (TF) values. The highest concentration of PTEs in roots was detected for zinc (10.56 mg/kg of dry weight, DW) and copper (5.128 mg/kg of DW). Similarly, the highest concentration in above-ground parts of the plant was detected for zinc and copper (12.638 and 4.0 mg/kg of DW, respectively). Although the values were relatively high, the effectiveness of the absorption of these PTEs by plant biomass from the soil was relatively very low. BAC and BCF were always lower than 1. On the other hand, the results suggested that M. albus effectively transports PTEs (only for Zn, Pb and Hg) from roots to shoots, because TF was always higher than 1. However, the accumulation of PTEs from soils with a natural abundance of PTEs was not excessive in comparison to conventional maize silage. Therefore, there is no potential risk of biomethane production in biogas plants when biomass from M. albus is used.

Acridine and its derivatives (9-chloroacridine and 9-aminoacridine) are investigated here, supported on FAU type zeolite Y, as a delivery system of anticancer agents. FTIR/Raman spectroscopy and electron microscopy revealed successful drug loading on the zeolite surface, while spectrofluorimetry was employed for drug quantification. The effects of the tested compounds on cell viability were evaluated using in vitro methylthiazol-tetrazolium (MTT) colorimetric technique against human colorectal carcinoma (cell line HCT-116) and MRC-5 fibroblasts. Zeolite structure remained unchanged during homogeneous drug impregnation with achieved drug loadings in the 18–21 mg/g range. The highest drug release, in the µM concentration range, with favourable kinetics was established for zeolite-supported 9-aminoacridine. The acridine delivery via zeolite carrier is viewed in terms of solvation energy and zeolite adsorption sites. The cytotoxic effect of supported acridines on HCT-116 cells reveals that the zeolite carrier improves toxicity, while the highest efficiency is displayed by zeolite-impregnated 9-aminoacridine. The 9-aminoacridine delivery via zeolite carrier favours healthy tissue preservation while accompanying increased toxicity toward cancer cells. Cytotoxicity results are well correlated with theoretical modelling and release study, providing promising results for applicative purposes.

Pollutants and infectious diseases can spread through air with airborne droplets and aerosols. A respiratory mask can decrease the amount of pollutants we inhale and it can protect us from airborne diseases. With the onset of the COVID-19 pandemic, masks became an everyday item used by a lot of people around the world. As most of them are for a single use, the amount of non-recyclable waste increased dramatically. The plastic from which the masks are made pollutes the environment with various chemicals and microplastic. Here, we investigated the time- and size-dependent filtration efficiency (FE) of aerosols in the range of 25.9 to 685.4 nm of five different natural materials whose FE was enhanced using electrospun poly(vinylidene fluoride-co-hexafluoropropylene) (PVDF) fibres. A scanning electron microscope (SEM) was used to determine the morphology and structure of the natural materials as well as the thickness of the PVDF fibres, while the phase of the electrospun fibres was determined by Raman spectroscopy. A thin layer of the electrospun PVDF fibres with the same grammage was sandwiched between two sheets of natural materials, and their FE increased up to 80%. By varying the grammature of the electrospun polymer, we tuned the FE of cotton from 82.6 to 99.9%. Thus, through the optimization of the grammage of the electrospun polymer, the amount of plastic used in the process can be minimized, while achieving sufficiently high FE.

248. Treatment of Myasthenia Gravis Patients with COVID-19: Review of the Literature

Coronavirus disease 2019 (COVID-19), caused by the late 2019 outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), causes a respiratory disease which could put myasthenia gravis (MG) patients at a greater risk of developing severe disease course, since infections and some drugs are a well-recognized trigger of symptom exacerbation in MG patients. Out of ten most commonly used past and present drugs used in COVID-19 treatment, two (quinolone derivatives and azithromycin) are known to worsen MG symptoms, whereas another two (tocilizumab and eculizumab) might have positive effect on MG symptoms. Colchicine, remdesivir, lopinavir, ritonavir and favipiravir seem to be safe to use, while data are insufficient for bamlanivimab, although it is also probably safe to use. Considering MG treatment options in patients infected with SARS-CoV-2, acetylcholine esterase inhibitors are generally safe to use with some preliminary studies even demonstrating therapeutic properties in regard to COVID-19. Corticosteroids are in general safe to use, even recommended in specific circumstances, whereas other immunosuppressive medications (mycophenolate mofetil, azathioprine, cyclosporine, methotrexate) are probably safe to use. The only exception is rituximab since the resulting B cell depletion can lead to more severe COVID-19 disease. Concerning plasmapheresis and intravenous immunoglobulins, both can be used in COVID- 19 while taking into consideration thromboembolic properties of the former and hemodynamic disturbances of the latter. As current data suggest, all known COVID-19 vaccines are safe to use in MG patients.

Abstract Background A recent study found serum neurofilament light chain (NfL) levels to be strongly associated with poor neurological outcome in patients after cardiac arrest. Our aim was to confirm these findings in an independent validation study and to investigate whether NfL improves the prognostic value of two cardiac arrest-specific risk scores. Methods This prospective, single-center study included 164 consecutive adult after out-of-hospital cardiac arrest (OHCA) patients upon intensive care unit admission. We calculated two clinical risk scores (OHCA, CAHP) and measured NfL on admission within the first 24 h using the single molecule array NF-light® assay. The primary endpoint was neurological outcome at hospital discharge assessed with the cerebral performance category (CPC) score. Results Poor neurological outcome (CPC > 3) was found in 60% (98/164) of patients, with 55% (91/164) dying within 30 days of hospitalization.
