CCL13/MCP-4 belongs to the CC chemokine family, which induces chemotaxis in many immune cells. Despite extensive research into its function in numerous disorders, a thorough analysis of CCL13 is not yet accessible. The role of CCL13 in human disorders and existing CCL13-focused therapies are outlined in this study. The function of CCL13 in rheumatic diseases, skin conditions, and cancer is comparatively well-established, and some studies also suggest that it may be involved in ocular disorders, orthopedic conditions, nasal polyps, and obesity. We also give an overview of research that found very little evidence of CCL13 in HIV, nephritis, and multiple sclerosis. Even though CCL13-mediated inflammation is frequently linked to disease pathogenesis, it’s fascinating to note that in some conditions, like primary biliary cholangitis (PBC) and suicide, it might even act as a preventative measure.

Near-infrared (NIR) fluorescence imaging-guided surgery is increasingly concerned in gastrointestinal surgery because it can potentially improve clinical outcomes. This new technique can provide intraoperative image guidance for surgical margin evaluation and help surgeons examine residual lesions and small tumors during surgery. NIR fluorophores methylene blue (MB) is a promising fluorescent probe because of its safety and intraoperative imaging in the clinic. However, whether MB possesses the potential to perform intraoperative navigation of the stomach and gastric tumors needs to be further explored. Therefore, the current study mainly validated MB’s usefulness in animal models’ intraoperative imaging of stomach and gastric tumors. NIR fluorophores MB can exhibit specific uptake by the gastric epithelial cells and cancer cells. It is primarily found that MB can directly target the stomach in mice. Interestingly, MB was applied for the NIR imaging of gastric cancer cell xenografts, suggesting that MB cannot specifically target subcutaneous and orthotopic gastric tumors in xenograft models. Thus, it can be concluded that MB has no inherent specificity for gastric tumors but specificity for gastric tissues. Apparently, MB-positive and negative NIR imaging are meaningful in targeting gastric tissues and tumors. MB is expected to represent a helpful NIR agent to secure precise resection margins during the gastrectomy and resection of gastric tumors.

Objective To investigate the inhibition of house dust mite induced oxidative stress with human bronchial epithelial cell line BEAS-2B by aldose reductase inhibitor (ARIs). Methods BEAS-2B was divided into control group, model group, NF-κB inhibitor group (PDTC group) and ARIs intervention group (ARIs group). Oxidative stress reaction (MDA and ROS content, SOD activity) was detected by ELISA. The contents of IL-5, IL-13 and IFN-γ were measured by ELISA. MTT was used to detect cell viability. Cell apoptosis was detected by flow cytometry. The expressions of NF-κB, NF-κB P65 and P38 MAPK proteins were detected by Western blot. Results SOD activity, IFN-γ content and cell survival rate in model group were lower than those in control group (P＜0.05), MDA, IL-5, IL-13 content and cell apoptosis rate were higher than those in control group (P＜0.05). Compared with model group, SOD activity, IFN-γ content and cell survival rate in PDTC and ARIs groups were increased(P＜0.05), MDA, IL-5, IL-13 content and cell apoptosis rate were decreased (P＜0.05). Compared with control group, the expression of NF-κB, NF-κB P65 and P38 MAPK protein in model group was significantly increased(P＜0.05). The expression of NF-κB, NF-κB P65 and P38 MAPK protein in PDTC group and ARIs group were significantly decreased (P＜0.05). Conclusions Aldose reductase inhibitors improve the oxidative stress response of human bronchial epithelial cells induced by house dust mites, promote the proliferation of BEAS-2B cells and inhibit apoptosis.

Summary: Efficient gene delivery in an integrated drug delivery system is urgent for multimodal antitumor therapy. Herein, we describe a protocol for constructing a peptide-based siRNA delivery system to achieve tumor vascular normalization and gene silencing in 4T1 cells. We highlighted four major steps, including (1) synthesis of the chimeric peptide, (2) preparation and characterization of PA7R@siRNA micelleplexes, (3) in vitro tube formation assay and transwell cell migration assay, and (4) siRNA transfection in 4T1 cells. This delivery system is expected to be used to silence gene expression, normalize tumor vasculature, and perform other treatments based on the different peptide segments.For complete details on the use and execution of this protocol, please refer to Yi et al. Abstract Cardiac hypertrophy occurs initially in response to an increased cardiac load as a compensatory mechanism to maintain cardiac output. However, sustained pathological hypertrophy can develop into heart failure and cause sudden death. Fibroblast growth factor 20 (FGF20) is a member of the fibroblast growth factor family, which involved in apoptosis, aging, inflammation, and autophagy. The precise function of FGF20 in pathological cardiac hypertrophy is unclear. In this study, we demonstrated that FGF20 was significantly decreased in response to hypertrophic stimulation. In contrast, overexpression of FGF20 protected against pressure overload-induced cardiac hypertrophy. Mechanistically, we found that FGF20 upregulates SIRT1 expression, causing deacetylation of FOXO1; this effect promotes the transcription of downstream antioxidant genes, thus inhibits oxidative stress. In content, the anti-hypertrophic effect of FGF20 was largely counteracted in SIRT1-knockout mice, accompanied by an increase in oxidative stress. In summary, our findings reveal a previously unknown protective effect of FGF20 on pathological cardiac hypertrophy by reducing oxidative stress through activation of the SIRT1 signaling pathway. FGF20 is a potential novel molecular target for preventing and treating pressure overload-induced myocardial injury.

miR-30b, which is encoded by the gene located on chromosome 8q24.22, plays an important role in a variety of diseases. In most types of tumors, miR-30b significantly inhibits the proliferation, migration, and invasion of cancer cells through the regulation of target genes. Moreover, miR-30b can inhibit the PI3K/AKT signaling pathway through the regulation of EGFR, AKT, Derlin-1, GNA13, SIX1, and other target genes, thus inhibiting the EMT process of tumor cells and promoting apoptosis. In addition, miR-30 plays a significant role in alleviating drug resistance in tumor cells. Although the use of miR-30b as a clinical diagnostic indicator or anticancer drug is still facing great difficulties in the short term, with the deepening of research, the potential application of miR-30b is emerging.

Complex composition and abnormal growth state of cells and tissues in the solid tumor microenvironment often lead to high density and stiffness different from normal tissues, thus forming a solid physical barrier to prevent the penetration of nanomedicines. Charge-reversal nanomedicines are a kind of emerging smart nano-systems that can show a change of surface charge from negative to positive before and after reaching the tumor site, thus effectively enhancing tumor penetration by promoting transcytosis, while maintaining a high in vivo safety. In this review, we provide a detailed summary of the current classification of charge-reversal nanomaterials and their working mechanisms, as well as also comment on the prospects and challenges of the smart nano-systems in the direction of thoroughly solving the tumor penetration problem.
