203. Human Bone Marrow Mesenchymal Stem Cell-Derived Exosomes Attenuate Blood-Spinal Cord Barrier Disruption via the TIMP2/MMP Pathway After Acute Spinal Cord Injury

Abstract

After spinal cord injury (SCI), destruction of the blood-spinal cord barrier (BSCB) results in infiltration of blood cells, such as neutrophils and macrophages, leading to permanent neurological dysfunction. Previous studies have shown that human bone marrow mesenchymal stem cell (BMSC)-derived exosomes have a beneficial neuroprotective effect in SCI models. However, whether BMSC-Exos contribute to the integrity of the BSCB has not been clarified. The purpose of this study was to investigate the mechanism of BMSC-Exo-induced changes in the permeability of the BSCB after SCI. Here, we first used BMSC-Exos to treat an SCI rat model, showing that BMSC-Exos can inhibit BSCB permeability damage and improve spontaneous repair. Next, we found that tissue inhibitors of matrix metalloproteinase 2 (TIMP2) have been shown to play an important role in the function of BMSC-Exos by inhibiting the matrix metalloproteinase (MMP) pathway, thereby reducing the reduction of cell junction proteins. Therefore, we constructed siTIMP2 to knock out TIMP2 in BMSC-Exos, which caused the activity of BMSC-Exos to be significantly weakened. Finally, we constructed an in vitro model of BSCB with HBMECs and verified that TIMP2 in BMSC-Exos in vitro can also alleviate BSCB damage. This proof-of-principle study demonstrates that BMSC-Exos can preserve the integrity of the BSCB and improve functional recovery after SCI through the TIMP2/MMP signaling pathway.

Abstract

Objectives Many older adults receive informal care from multiple caregivers, including support from a primary caregiver and a secondary caregiver network (SCN). This study examined the association between SCN support and primary caregiver burden, and whether the association varies across women and men, Black and White. Methods Data came from the 2015 National Health and Aging Trend Study and the National Study of Caregiving, including non-Hispanic White and Black men and women who were identified as primary caregivers (n = 967) and their secondary caregivers (n = 2,253). SCN support was indicated by (a) care domain overlap and (b) proportion of caregiving by SCN. Multiple regression models were estimated for the analyses. Results Both SCN support variables were found to reduce primary caregiver burden, and the effect of proportion of caregiving by SCN was found to vary by gender–race groups. With the increase of the proportion of caregiving by SCN, both Black and White women caregivers tend to experience faster decrease in caregiver burden than Black men. Discussion Our findings support the role of SCN in reducing primary caregiver burden and demonstrate that the benefit of SCN support varies across the 4 gender–race groups. Abstract

Localized tissue hypoxia is a consequence of vascular compromise or rapid cellular proliferation and is a potent inducer of compensatory angiogenesis. The oxygen-responsive transcriptional regulator hypoxia-inducible factor 2 α (HIF-2 α) is highly expressed in vascular ECs and, along with HIF-1 α, activates expression of target genes whose products modulate vascular functions and angiogenesis. However, the mechanisms by which HIF-2 α regulates EC function and tissue perfusion under physiological and pathological conditions are poorly understood. Using mice in which Hif2a was specifically deleted in ECs, we demonstrate here that HIF-2 α expression is required for angiogenic responses during hindlimb ischemia and for the growth of autochthonous skin tumors. EC-specific Hif2a deletion resulted in increased vessel formation in both models; however, these vessels failed to undergo proper arteriogenesis, resulting in poor perfusion. Analysis of cultured HIF-2 α - deficient ECs revealed cell-autonomous increases in migration, invasion, and morphogenetic activity, which correlated with HIF-2 α -dependent expression of specific angiogenic factors, including delta-like ligand 4 (Dll4), a Notch ligand, and angiopoietin 2. By stimulating Dll4 signaling in cultured ECs or restoring Dll4 expression in ischemic muscle tissue, we rescued most of the HIF-2 α -dependent EC phenotypes in vitro and in vivo, emphasizing the critical role of Dll4/Notch signaling as a downstream target of HIF-2 α in ECs. Abstract

This thesis presents work on the development of model-based Bayesian approaches to object tracking and intent prediction. Successful navigation/positioning applications rely fundamentally on the choice of appropriate dynamic model and the design of effective tracking algorithms capable of maximising the use of the structure of the dynamic system and the information from sensors. While the tracking problem with frequent and accurate position data has been well studied, we push back the frontiers of current technology where an object can undergo fast manoeuvres and position fixes are limited. On the other hand, intent prediction techniques which extract higher level information such as the intended destination of a moving object can be designed, given the ability to perform successful tracking. Such techniques can play important roles in various application areas, including traffic monitoring, intelligent human computer interaction systems and autonomous route planning. In the first part of this thesis Bayesian tracking methods are designed based on a standard fix-rate setting in which the dynamic system is formulated into a Markovian state space form. We show that the combination of an intrinsic coordinate dynamic model and sensors in the object's body frame leads to novel state space models according to which efficient proposal kernels can be designed and implemented by the sequential Monte Carlo (SMC) methods. Also, sequential Markov chain Monte Carlo schemes are considered for the first time to tackle the sequential batch inference problems due to the presence of infrequent position data. Performance evaluation on both synthetic and real-world data shows that the proposed algorithms are superior to simpler particle filters, implying that they can be favourable alternatives to tracking problems with inertial sensors. The modelling assumption that leads to Markovian state space models can be restrictive for real-world systems as it stipulates that the state sequence has to be synchronised with the observations. In the second major part of this thesis we relax this assumption and work with a more natural class of models, termed variable rate models. We generalise the existing variable rate intrinsic model to incorporate acceleration, speed, distance and position data and introduce new variable rate particle filtering methods tailored to the derived model to accommodate multi-sensor multi-rate tracking scenarios. The proposed algorithms can achieve substantial improvements in terms of tracking accuracy and robustness over a bootstrap variable rate particle filter. Moreover, full Bayesian inference schemes for the learning of both the hidden state and system parameters are presented, with numerical results illustrating their effectiveness. The last part of the thesis is about designing efficient intent prediction algorithms within a Bayesian framework. A pseudo-observation based approach to the incorporation of destination knowledge is introduced, making the mathematics of the dynamical model and the observation process consistent with the Markov state process. Based on the new interpretation, two algorithms are proposed to sequentially estimate the probability of all possible endpoints. Whilst the synthetic maritime surveillance data demonstrate that the proposed methods can achieve comparable prediction performance with reduced computational cost in comparison to the existing bridging distribution based methods, the results on an extensive freehand pointing database, which contains 95 three-dimensional pointing trajectories, show that the new algorithms can outperform other state-of-the-art techniques. Some sensitivity tests are also performed, confirming the good robustness of the introduced methods against model mismatches.

Abstract

The specific biological function of transient receptor potential vanilloid 1 (TRPV1) in pathogenesis of lung adenocarcinoma (LUAD) remains unclear. In this study, TRPV1 expression in tumor tissues, primary cells and cell lines of LUAD, as well as the mechanism mediating its hyperexpression were systematically studied. Multiple models and techniques were adopted to elucidate the relationship between TRPV1 hyperexpression and tumor recurrence and metastasis. Results showed that TRPV1 expression was increased in tumor tissues and primary tumor cells of LUAD patients. The increased expression was associated with worse overall survival outcome and raised HIF1α levels. TRPV1 expression in A549 and NCI-H292 cells was increased after pretreatment with cigarette smoke extract or spermine NONOate. Moreover, A549 cells with TRPV1 overexpression has enhanced tumor growth rates in subcutaneous grafted tumor models, and increased intrapulmonary metastasis after tail vein infusion in nude BALB/c nude mice. Mechanistically, TRPV1 overexpression in A549 cells promoted HIF1α expression and nuclear translocation by promoting CREB phosphorylation and activation of NOS1-NO pathway, ultimately leading to accelerated cell proliferation and stronger invasiveness. In addition, based on photothermal effects, CuS-TRPV1 mAb effectively targeted and induced apoptosis of TRPV1-A549 cells both in vivo and in vitro, thereby mitigating tumor growth and metastasis induced by xenotransplantation of TRPV1-A549 cells. In conclusion, TRPV1 hyperexpression in LUAD is a risk factor for tumor progression and is involved in proliferation and migration of tumor cells through activation of HIF1α. Our study also attempted a new strategy inhibiting the recurrence and metastasis of LUAD: by CuS-TRPV1 mAb precisely kill TRPV1 hyperexpression cells through photothermal effects.
