Abstract

This work is concerned with the internal stabilization of the FitzHugh-Nagumo system defined on a bounded domain. We give a sufficient condition under which such system can be exponentially stabilized by only one feedback controller acting on a nonempty open subdomain in the first equation.

Abstract

Characterization of land displacement induced by long-term overexploitation of groundwater is necessary to ensure sustainable water supply in Beijing, China. The northern part of the Beijing Plain is an important water source area and is also designed for groundwater recharge from South-to-North Water Diversion Project. We aim to depict the process of characterizing land displacement under complex hydrogeological and geological context in the region using remote sensing and geographic information system. Interferometric synthetic aperture radar time-series analysis was used to detect land displacement from 2003 to 2010. Statistic linear regression equations between groundwater level and land displacement were built based on linear consolidation principle. The spatial difference of Pearson correlation coefficient ( R) and slope ( k) were discriminated to quantify the response of land displacement to groundwater level change. The results show that there are two major displacement cones with annual rates up to −40 and −24 mm year. R and k had a negative and positive correlation with increasing land displacement, respectively. A larger R reflects that the groundwater level has a closer relation with the occurrence of land displacement. The weak correlation is due to the delay in the propagation of the pressure drawdown in the fine-sediment layers or lens from the pumped aquifers where the pressure is measured. Thick compressible layer has more potential for land displacement. Results of this study are necessary to clarify the land displacement characteristics, to make full use of abundant spatial-temporal dataset, and ultimately to support hazard prevention and mitigation decisions. 238. Labeling Assembly of Hydrophilic Methionine into Nanoparticle for Mild‐Heat Mediated Immunometabolic Therapy

Abstract

Methionine metabolism has a significant impact on T cells’ survival and activation even in comparison to arginine, a well‐documented amino acid in metabolic therapy. However, hydrophilic methionine is hardly delivered into TME due to difficult loading and rapid diffusion. Herein, the labeling assembly of methionine into nanoparticle is developed to overcome high hydrophilicity for mild‐heat mediated immunometabolic therapy. The strategy is to first label methionine with protocatechualdehyde (as the tag) via reversible Schiff‐base bond, and then drive nanoassembly of methionine (MPC@Fe) mediated by iron ions. In this fashion, a loading efficiency of 40% and assembly induced photothermal characteristics can be achieved. MPC@Fe can accumulate persistently in tumor up to 36 h due to tumor‐selective aggregation in acidic TME. A mild heat of 43 °C on tumor by light irradiation stimulated the immunogenic cell death and effectively generated CD8+T cells. Notably, MPC@Fe assisted by mild heat promoted 4.2‐fold of tumor‐infiltrating INF‐γ+CD8+T cells, leading to an inhibition ratio of 27.3‐fold versus the free methionine. Such labeling assembly provides a promising methionine delivery platform to realize mild heat mediated immunometabolic therapy, and is potentially extensible to other amino acids. Hydrophilic methionine is barely delivered into the tumor microenvironment due to difficult loading and rapid diffusion. To solve it, a labeling assembly strategy, first labeling methionine with protocatechualdehyde (as the tag), and then driving nanoassembly of Met (MPC@Fe) mediated by iron ions, is presented. A series of biological efficacy is achieved via labeling assembly.

246. Reversible PEGylation and Schiff-base linked imidazole modification of polylysine for high-performance gene delivery.

Abstract

Gene carriers made from polylysine are of interest in relation to gene therapy but suffer from the lack of transfection efficiency due to limited stability and endosomal escape ability. To address this problem, we designed and developed Schiff-base linked imidazole modified polylysine with a reversible-PEGylation catiomer (SL-ImPEG-SS-PLL) for high efficiency gene delivery. The reversible PEGylation was introduced for in vivo circulation, as well as selective PEG detachment to augment the cellular internalization, while introducing Schiff-base linked imidazole residues into polylysine was expected to accelerate the endosomal escape of the DNA payload, as well as facilitate intracellular DNA unpacking and release, thus significantly enhancing gene delivery efficiency. The size alteration of the SL-I15mPEG-SS-PLL/pDNA polyplexes in the presence of 10 mM GSH suggests stimulus-induced PEG detachment under tumor relevant reduction conditions. Acid–base titration assays indicate that imidazole residues confer on polylysine remarkable buffering ability. The agarose gel retardation assay suggests that the Schiff-base linkages provide an increased DNA binding ability to protect DNA against nucleases and timely intracellular DNA unpacking to permit DNA dissociation from polylysine and access to transcriptional machinery. Biological efficacy assessment of this multifunctional carrier, using pEGFP and pGL-3 as reporter genes, indicates comparable to or even higher transfection efficiencies than gold standard PEI and their transfection efficiencies are slightly affected by serum. More importantly, in vivo transfection of pEGFP reveals that GFP expression was found not only in some of the important organs, such as the liver, spleen, kidneys and lungs, but also in the transplanted carcinoma. These experimental results suggest that the reversible PEGylation and Schiff-base linked imidazole modification make SL-ImPEG-SS-PLL a great potential candidate for an effective and biocompatible gene delivery system. Abstract

Stroke accelerates inflammatory monocyte recruitment to the endothelium and consequent atheroprogression via high-mobility group box 1-receptor for advanced glycation end products signaling. Notably, Hmgb1 interacts with multiple toll-like receptors (TLRs) and promotes TLR4-mediated proinflammatory myeloid cell activation. Therefore, TLR-associated mechanism(s) within monocytes may play a role in Hmgb1-driven poststroke atheroprogression.
