Abstract

Changes in labile carbon (LC) pools and microbial communities are the primary factors controlling soil heterotrophic respiration (Rh ) in warming experiments. Warming is expected to initially increase Rh but studies show this increase may not be continuous or sustained. Specifically, LC and soil microbiome have been shown to contribute to the effect of extended warming on Rh . However, their relative contribution is unclear and this gap in knowledge causes considerable uncertainty in the prediction of carbon cycle feedbacks to climate change. In this study, we used a two-step incubation approach to reveal the relative contribution of LC limitation and soil microbial community responses in attenuating the effect that extended warming has on Rh . Soil samples from three Tibetan ecosystems-an alpine meadow (AM), alpine steppe (AS), and desert steppe (DS)-were exposed to a temperature gradient of 5-25°C. After an initial incubation period, soils were processed in one of two methods: (a) soils were sterilized then inoculated with parent soil microbes to assess the LC limitation effects, while controlling for microbial community responses; or (b) soil microbes from the incubations were used to inoculate sterilized parent soils to assess the microbial community effects, while controlling for LC limitation. We found both LC limitation and microbial community responses led to significant declines in Rh by 37% and 30%, respectively, but their relative contributions were ecosystem specific. LC limitation alone caused a greater Rh decrease for DS soils than AMs or ASs. Our study demonstrates that soil carbon loss due to Rh in Tibetan alpine soils-especially in copiotrophic soils-will be weakened by microbial community responses under short-term warming.

Abstract

The main working parts of the cleaning device of a rice combine harvester can be controlled by an established control strategy in real time based on the monitored grain sieve loss. This is an efficient way to improve their cleaning adaptability, since as a consequence, the main working parameters of combine harvesters can automatically adapt to crop and environment changes, and the corresponding cleaning performance can be improved. To achieve the target of cleaning control based on the monitored grain sieve loss, a fuzzy control system was developed, which selected S7-1200 PLC as the main control unit to build the lower computer hardware system, utilized ladder language to complete the system compilation, and used LabVIEW 14.0 software to design the host&ndash, computer interface. The effects of fan speed, guide plate angle, and sieve opening on the grain sieve loss and grain impurity ratio have been investigated through a large number of bench tests. The relevance level of the operating parameters on the performance parameters has been determined also, and finally, a fuzzy control model was developed for the cleaning system. The experiment results indicated that the designed fuzzy control model can control the cleaning section settings, such as fan speed and guide plate angle automatically, and reduce the grain sieve loss to some extent.

240. Spatiotemporal Distributions of Fluoride and Arsenic in Rivers with the Role of Mining Industry and Related Human Health Risk Assessments in Kyrgyzstan

Abstract

To determine the spatiotemporal distributions of fluoride and arsenic in the rivers within the mining districts of Kyrgyzstan and health risks associated with the use of these rivers, a total of 169 water samples were collected from the main rivers and tributaries of Kyrgyzstan from 2016 to 2018. Through the cold and hot spot analysis, multivariate statistical analysis and health risk assessment model, the results indicated that the fluoride and arsenic concentrations in river waters increased year by year from 2016 to 2018. According to the World Health Organization (WHO) drinking water guidelines, 0%, 1.09% and 1.06% of the analyzed samples exceeded the limit for fluoride (1.5 mg/L), and 0%, 1.09% and 2.13% of the samples exceeded the limit for arsenic (10 µg/L) in 2016, 2017 and 2018, respectively. The gathering areas for high fluoride concentrations were mainly distributed in the Issyk-Kul Basin, Chu River Valley and Fergana Basin, while high arsenic concentrations were mainly observed in the Chu River Valley and southern Fergana Basin. Although fluoride and arsenic were not found to exceed the limits simultaneously, the two pollutants accumulated high values in the southern Fergana Basin in 2018, which indicated the risk of joint poisoning. From 2016 to 2018, arsenic concentrations in the river water of Kyrgyzstan created a high risk of carcinogenesis by the ingestion intake exposure route, which resulted in the total risk of health hazards to children and adults caused by fluoride and arsenic to exceed the maximum acceptable ranges. Therefore, the national environmental protection department in Kyrgyzstan should strengthen the dynamic monitoring of arsenic concentration in the rivers, and related scientific research institutions should conduct more in-depth research on the migration and transformation process of pollutants, the toxic mechanism, and even the substantive countermeasures in the future.

Abstract

Crops with branching characteristics, such as rapeseed, are difficult to harvest owing to their specific plant geometry. To solve this problem, an harvesting reel with improved tine trajectory, considering the reel working process and a function suitable for oilseed rape, is proposed in this study. The improved tine, of which angle can be continuously adjusted to adapt to the plant structure and meet the operating requirements of the reel in the stages of pushing the reel into the crops, feeding plants into the auger, and releasing the reel. The kinematics of the tine trajectory between the new and standard reels were calculated and a simulation comparative analysis was performed, which proved that the new reel satisfies the theoretical requirements. Single-factor field experiments were conducted for the two types of reels. The test results demonstrated that the proposed reel has a lower header loss rate and is significantly affected by the reel speed than the reel height or machine forward speed. According to the multivariate orthogonal results, the combination of optimal parameters with minimum loss rate yielded a reel rotational speed of 1.88 rad s−1, machine speed of 0.6 m s−1, and reel vertical height of 1100 mm. The actual loss rate was found to authentically decrease to 1.04% in the supplementary field test under this condition, which verified that the parameters are reasonable. This study combines experiments and trajectory analysis to explain the advantages of the improved reel and provide a new study direction of reducing rapeseed harvesting losses. • Designed an reel with improved tine trajectory suitable for harvesting rapeseed. • Simulation analysis proved the improved reel is more favorable for rape harvest. • Field comparative experiments proved the improved reel has a lower header loss. Abstract

In agriculture, the body tilt of vehicles or platforms can be controlled by levelling technology to prevent rollover and improve efficiency when operated in rough terrain. Many researchers have carried out important study in this area based on mechanism development and optimization, but most of them are not effective to crawler vehicles. The objective of this research was to specifically develop a levelling mechanism for crawler combine harvester, since as a consequence, enhancing the operation performance and trafficability. This levelling method depends on a hydraulic four-point adjustable lifting chassis, which could adjust vehicle chassis by controlling hydraulic cylinders. Therefore, the mathematical models between chassis attitude (including ground clearance, the horizontal and longitudinal inclination angle) and each length of hydraulic cylinder were deduced. The principle of lifting mechanism of crawler vehicle chassis was analyzed by using virtual prototyping software ADAMS, as well as dynamic performance under three different attitude adjustment conditions. The maximum adjustment range of the ground clearance is 140 mm and the lateral inclination adjustment range is ±5.17°. Experiment showed that the designed levelling mechanism was feasible and with positive significance for crawler vehicles levelling.

Abstract

Abstract Background Japanese encephalitis (JE) is a serious zoonosis caused by the Japanese encephalitis virus (JEV) which is a mosquito-borne pathogen of the family Flavivirus. However, the application of several developed laboratory methods for the detection of JEV antigens or antibodies are limited by their requirements of laboratory operations, skilled technicians and special facilities. Results To develop a method for detecting JEV antigen in swine, human, mosquito and other clinical specimens specifically, conveniently and effectively, an antigen capture enzyme-linked immunosorbent assay (ELISA) was established in this study. Sensitivity, specificity, repeatability and stability of the developed method were evaluated, and 60 clinical samples were tested in this study. The results demonstrated that the antigen capture ELISA was capable in detecting JEV antigen with high sensitivity and specificity compared with conventional methods. 14 samples showed the positive result with coincidence rate of 70%, and 46 displayed negative result with coincidence rate of 100% as compared to that of reverse transcription-polymerase chain reaction (RT-PCR). Conclusions The developed ELISA assay provides a convenient and specific method for the large-scale determination of JEV antigen in infected swine, human and mosquito samples with high sensitivity and specificity.
