Background: Circulating tumor cells (CTCs) and relevant autophagy Beclin-1 genes expression are critical biomarkers for tumorigenesis and tumor progress. Here we investigated the relationship of dynamic changes of CTCs and Beclin-1 expression of CTCs with renal cell carcinoma (RCC) prognosis. Materials and methods: A total of 69 patients with RCC were enrolled and divided into two groups based on the postoperative status of distant metastasis, including metastasis-free group (n = 58) and metastatic group (n = 11). Demographic characteristics of each patient were recorded in detail. All 69 enrolled patients had received multiple CTC tests and peripheral blood samples were obtained at three different time points (1 day before operation, 6 months and 12 months after operation). Peripheral blood samples were drawn before each time point and CTCs were separated by using Can Patrol CTC enrichment technique. CTCs were divided into epithelial, mesenchymal and mixed phenotype based on different surface biomarkers. RNA in situ hybridization assay was used to detect the expression of Beclin1 gene. Results: The percentages of epithelial, mesenchymal and mixed CTCs were 11.64%, 28.04% and 60.32%, respectively. There were no significant differences of initial CTCs counts between metastasis-free group (8.43 ± 5.15) and metastatic group (7.71 ± 3.82) (P > 0.05). As for metastatic group, the number of mixed CTCs at 12 months postoperatively was significantly higher than that of mixed CTCs preoperatively and 6 months postoperatively (P < 0.05). In the metastatic group, the number of Beclin1 positive CTCs was significantly higher than that of Beclin1 negative CTCs preoperatively (P < 0.05), moreover, there were several significantly changes of Beclin1 positive CTCs with different types and at different time points. Conclusion: The recurrence or metastasis of RCC was uncorrelated with initial CTCs counts, but probably related with the variation trend of CTCs, especially mesenchymal CTCs and Beclin1 positive CTCs. Stream processing applications continuously process large amounts of online streaming data in real time or near real time. They have strict latency constraints. However, the continuous processing makes them vulnerable to any failures, and the recoveries may slow down the entire processing pipeline and break latency constraints. The upstream backup scheme is one of the most widely applied fault-tolerant schemes for stream processing systems. It introduces complex backup dependencies to tasks, which increases the difficulty of controlling recovery latencies. Moreover, when dependent tasks are located on the same processor, they fail at the same time in processor-level failures, bringing extra recovery latencies that increase the impacts of failures. This paper studies the relationship between the task allocation and the recovery latency of a stream processing application. We present a correlated failure effect model to describe the recovery latency of a stream topology in processor-level failures under a task allocation plan. We introduce a recovery-latency aware task allocation problem (RTAP) that seeks task allocation plans for stream topologies that will achieve guaranteed recovery latencies. We discuss the difference between RTAP and classic task allocation problems and present a heuristic algorithm with a computational complexity of O(n log2n) to solve the problem. The enhancer of zeste 2 polycomb repressive complex 2 subunit (EZH2) gene has been recognized to be a proto‑oncogene and to be linked to human malignancies. However, the additional functions of EZH2 in renal cell carcinoma (RCC) are not completely understood. In the present study, a possible role of EZH2 in RCC was identified. EZH2 was demonstrated to promote the cell proliferation and invasion potential of 769‑P cells, and inhibition of EZH2 was demonstrated to prevent these two processes in 786‑O cells. Mechanically, EZH2 was demonstrated to increase the phosphorylation of signal transducer and activator of transcription 3 (STAT3) and upregulate 72 kDa type IV collagenase (MMP‑2) expression. When cells were treated with small interfering RNA targeting STAT3 or Stattic, a specific inhibitor of STAT3, the invasive ability of the cells was decreased and downregulation of MMP‑2 was observed. Based on these results, in the present study it was hypothesized that EZH2 may serve a critical role in the progression of RCC. A natural ursolic compound, 2α,3β-dihydroxy-urs-12-en-28-oic acid (corosolic acid, CRA) was isolated from the root of Actinidia valvata Dunn. ( A. valvata Radix). Since a large number of triterpenoid compound has marked anticancer effects toward various types of cancer cell lines in vitro , this study was carried out to investigate the anticancer effect of CRA in human gastric cancer cell line BGC823 cells and the underlying apoptotic mechanism of CRA was examined in BGC823 cell lines. The results showed that CRA significantly suppressed the viability of BGC823 cells in a concentration- and time-dependent manner. CRA also significantly increased the sub G1 population by cell cycle analysis in a concentration dependent manner. Exposure to CRA decreased p65, bcl-2, Fas, smac mRNA and protein expression, and increased IκBα, bax, survivin mRNA and protein expression. Results of immunofluorescence staining and EMSA further indicated CRA induced apoptosis by inhibiting nuclear translocation of nuclear factor NF-κB subunit p65. Consistently overall, our findings suggest that CRA induces apoptosis via inhibition of NF-κB (p65) expression level and activation of IκBα in BGC cells as a potent anticancer candidate for gastric cancer treatment. Varroa destructor , a key biotic threat to the Western honey bee, has played a major role in colony losses over the past few years worldwide. Overuse of traditional acaricides, such as tau-fluvalinate and flumethrin, on V. destructor has only increased its tolerance to them. Therefore, the application of essential oils in place of traditional pesticides is an attractive alternative, as demonstrated by its high efficiency, lack of residue and tolerance resistance. To study the acaricidal activity of essential oils, we used clove oil ( Syzygium aromaticum L.), a typical essential oil with a wide range of field applications, and examined its effects on the enzyme activities of Ca 2+ -Mg 2+ -ATPase, glutathione-S-transferase (GST) and superoxide dismutase (SOD) and its effects on the water-soluble protein content of V. destructor body extracts after exposure to 0.1 μl and 1.0 μl of clove oil for 30 min. Our results showed that the water-soluble protein content significantly decreased after the treatments, indicating that the metabolism of the mites was adversely affected. The bioactivity of GSTs increased significantly after a low dosage (0.1 μl) exposure but decreased at a higher dosage (1.0 μl), while the activities of SOD and Ca 2+ -Mg 2+ -ATPase were significantly elevated after treatments. These results suggest that the protective enzyme SOD and detoxifying enzymes Ca 2+ -Mg 2+ -ATPase and GST contributed to the stress reaction of V. destructor to the essential oils and that the detoxification ability of V. destructor via GST was inhibited at higher dosages. Our findings are conducive to understanding the physiological reactions of V. destructor to treatment with essential oils and the underlying mechanisms behind the acaricidal activities of these natural products. Williamson's mouse deer, Tuagulus williamsoni (Kloss), is one of the smallest ungulates among tragulid species found in northern Thailand, and Yunnan Province, China. Here we describe Sarcocystis menglaensis n. sp., infecting two of 14 (14.3%) Williamson's mouse deer from south-western China. By light microscopy, sarcocysts of S. menglaensis are microscopic, up to 2,170 μm in length, and have a striated sarcocyst wall with 1.5-3.6 μm long palisade-like protrusions. Transmission electron microscopy observations revealed that sarcocyst wall is of 'type 10f', and has numerous villar protrusions folded over the cyst wall. The villar protrusions contained microtubules dispersed throughout the protrusions. Phylogenetic analysis based on 18S rDNA and mitochondrial cox1 gene sequences indicated that S. menglaensis shared a close affinity with species of Sarcocystis Lankester, 1982 from ruminants, which utilise felids as definitive hosts. Perception is the interaction interface between an intelligent system and the real world. Without sophisticated and flexible perceptual capabilities, it is impossible to create advanced artificial intelligence (AI) systems. For the next-generation AI, called 'AI 2.0', one of the most significant features will be that AI is empowered with intelligent perceptual capabilities, which can simulate human brain's mechanisms and are likely to surpass human brain in terms of performance. In this paper, we briefly review the state-of-the-art advances across different areas of perception, including visual perception, auditory perception, speech perception, and perceptual information processing and learning engines. On this basis, we envision several R&D trends in intelligent perception for the forthcoming era of AI 2.0, including: (1) human-like and transhuman active vision; (2) auditory perception and computation in an actual auditory setting; (3) speech perception and computation in a natural interaction setting; (4) autonomous learning of perceptual information; (5) large-scale perceptual information processing and learning platforms; and (6) urban omnidirectional intelligent perception and reasoning engines. The GLABROUS1 enhancer-binding protein (GeBP) gene family encodes typical transcription factors containing an atypical leucine zipper motif, and plays a critical role in regulating plant growth and development, as well as multiple abiotic stresses response, but little is known about the role of GeBP transcription factors in apple. In this study, seven GeBP family members were identified in apple by genome-wide analysis, expression response analysis showed that MdGeBPs had different tissue expression patterns and were responsive to a variety of hormonal and abiotic stresses. Subsequently, MdGeBP3 was cloned and transgenic Arabidopsis and apple callus were obtained for further functional characterization. MdGeBP3 protein was localized in the nucleus, and overexpression of MdGeBP3 was more sensitive to cytokinins and less sensitive to ABA. In addition, ectopic expression of MdGeBP3 in Arabidopsis resulted in reduced drought tolerance, promoted adventitious root development and delayed flowering. Overall, these results suggest that MdGeBP3 may play an important role in regulating apple growth and development, as well as abiotic stress responses. • MdGeBP family was identified in apple for the first time. • MdGeBP3 was more sensitive to cytokinins and less sensitive to ABA. • MdGeBP3 resulted in reduced drought tolerance. • MdGeBP3 promoted adventitious root development and delayed flowering.
