Abstract

The proportional subdistribution hazards model (i.e. Fine-Gray model) has been widely used for analyzing univariate competing risks data. Recently, this model has been extended to clustered competing risks data via frailty. To the best of our knowledge, however, there has been no literature on variable selection method for such competing risks frailty models. In this paper, we propose a simple but unified procedure via a penalized h-likelihood (HL) for variable selection of fixed effects in a general class of subdistribution hazard frailty models, in which random effects may be shared or correlated. We consider three penalty functions, least absolute shrinkage and selection operator (LASSO), smoothly clipped absolute deviation (SCAD) and HL, in our variable selection procedure. We show that the proposed method can be easily implemented using a slight modification to existing h-likelihood estimation approaches. Numerical studies demonstrate that the proposed procedure using the HL penalty performs well, providing a higher probability of choosing the true model than LASSO and SCAD methods without losing prediction accuracy. The usefulness of the new method is illustrated using two actual datasets from multi-center clinical trials.

Abstract

We previously reported a potent small molecule Mer tyrosine kinase inhibitor UNC1062. However, its poor PK properties prevented further assessment in vivo. We report here the sequential modification of UNC1062 to address DMPK properties and yield a new potent and highly orally bioavailable Mer inhibitor, 11, capable of inhibiting Mer phosphorylation in vivo, following oral dosing as demonstrated by pharmaco-dynamic (PD) studies examining phospho-Mer in leukemic blasts from mouse bone marrow. Kinome profiling versus more than 300 kinases in vitro and cellular selectivity assessments demonstrate that 11 has similar subnanomolar activity against Flt3, an additional important target in acute myelogenous leukemia (AML), with pharmacologically useful selectivity versus other kinases examined.

Abstract

Summary: Regression methodology has been well developed for competing risks data with continuous event times, both for the cause‐specific hazard and cumulative incidence functions. However, in many applications, including those from the Surveillance, Epidemiology, and End Results (SEER) program of the National Cancer Institute, the event times may be observed discretely. Naive application of continuous time regression methods to such data is not appropriate. We propose maximum likelihood inferences for estimation of model parameters for the discrete time cause‐specific hazard functions, develop predictions for the associated cumulative incidence functions, and derive consistent variance estimators for the predicted cumulative incidence functions. The methods are readily implemented using standard software for generalized estimating equations, where models for different causes may be fitted separately. For the SEER data, it may be desirable to model different event types on different time scales and the methods are generalized to accommodate such scenarios, extending earlier work on continuous time data. Simulation studies demonstrate that the methods perform well in realistic set‐ups. The methodology is illustrated with stage III colon cancer data from SEER. [ABSTRACT FROM AUTHOR]

Abstract

Tumor cells with p53 inactivation frequently exhibit chemotherapy resistance, which poses a long-standing challenge to cancer treatment. Here we unveiled a previously unrecognized role of TET2 in mediating p53-loss induced chemotherapy resistance in colon cancer. Deletion of TET2 in p53-null colon cancer cells enhanced DNA damage and restored chemotherapy sensitivity. By taking a two-pronged approach that combined pharmacological inhibition with genetic depletion, we discovered that p53 destabilized TET2 at the protein level by promoting its autophagic degradation. At the molecular level, we further revealed a physical association between TET2 and p53 that facilitated the nucleoplasmic shuttling of TET2, as well as its recruitment to the autophagosome for degradation. Our study has unveiled a functional interplay between TET2 and p53 during anti-cancer therapy. Our findings establish the rationale for targeting TET2 to overcome chemotherapy resistance associated with mutant p53 tumors.

241. Pseudo-Cyclization through Intramolecular Hydrogen Bond Enables Discovery of Pyridine Substituted Pyrimidines as New Mer Kinase Inhibitors

Abstract

Abnormal activation or overexpression of Mer receptor tyrosine kinase has been implicated in survival signaling and chemoresistance in many human cancers. Consequently, Mer is a promising novel cancer therapeutic target. A structure-based drug design approach using a pseudo-ring replacement strategy was developed and validated to discover a new family of pyridinepyrimidine analogs as potent Mer inhibitors. Through SAR studies, 10 (UNC2250) was identified as the lead compound for further investigation based on high selectivity against other kinases and good pharmacokinetic properties. When applied to live cells, 10 inhibited steady-state phosphorylation of endogenous Mer with an IC50 of 9.8 nM and blocked ligand-stimulated activation of a chimeric EGFR-Mer protein. Treatment with 10 also resulted in decreased colony-forming potential in rhabdoid and NSCLC tumor cells, thereby demonstrating functional anti-tumor activity. The results provide a rationale for further investigation of this compound for therapeutic application in patients with cancer.

243. Market performance and the loss aversion behind green management

Abstract

As societies become more concerned with environmental protection, some firms go beyond their legal obligations and proactively adopt their own green management practices even if those practices cannot produce short-term benefits. As a result, researchers have begun to question the underlying motives of these firms. Leveraging corporate loss aversion, we argue that firms that outperform their peers become more risk-seeking to avoid any future loss of competitive advantage. As a result, they become more likely to engage in stakeholder management in the form of green management practices. Specifically, we explore the Green Power Partnership (GPP) which solicits firms to purchase green power. Using the data for the green power consumed by firms that are participating in the GPP program in the United States, we find that firms with performance above social aspiration are likely to increase their use of green power. We also find that related diversification has a negative influence on such motivation.
